Claims (62)
1. Гибридный регуляторный элемент, содержащий совокупность сегментов гетерологичного регуляторного элемента, где совокупность сегментов гетерологичного регуляторного элемента позволяет получить уникальный профиль экспрессии в ткани.
2. Гибридный регуляторный элемент по п. 1, где гибридный регуляторный элемент содержит
(a) полинуклеотид, последовательность которого на по меньшей мере 85 процентов идентична любой из последовательности нуклеиновой кислоты под SEQ ID NO: 11-30 и 35-60;
(b) полинуклеотидную последовательность любой из SEQ ID NO: 11-30 и 35-60 или
(c) фрагмент любой из SEQ ID NO: 11-30, 35-47 и 50-59.
3. Гибридный регуляторный элемент по п. 1, где по меньшей мере один из сегментов получен из
(a) полинуклеотидных последовательностей, изложенных под любым из SEQ ID NO: 1-10, 31-34 и 61-74; или
(b) полинуклеотидной последовательности, содержащей последовательность, характеризующуюся по меньшей мере 85% идентичностью последовательности с полинуклеотидными последовательностями, изложенными под любым из SEQ ID NO: 1-10, 31-34 и 61-74.
4. Гибридный регуляторный элемент по п. 1, где гетерологичная совокупность сегментов регуляторного элемента предусматривает совокупность по меньшей мере трех сегментов родительского регуляторного элемента.
5. Гибридный регуляторный элемент по п. 1, где гетерологичная совокупность сегментов регуляторного элемента предусматривает совокупность по меньшей мере четырех сегментов родительского регуляторного элемента.
6. Гибридный регуляторный элемент по п. 1, где профиль экспрессии в ткани представляет собой профиль экспрессии, характерный для корня, или профиль экспрессии, характерный для надземной части.
7. Гибридный регуляторный элемент по п. 1, где профиль экспрессии в ткани представляет собой универсальный профиль экспрессии в надземной части.
8. Гибридный регуляторный элемент по п. 1, где гибридный регуляторный элемент позволяет получить универсальный профиль экспрессии в корне, а каждый родительский регуляторный элемент позволяет получить экспрессию в ткани корня или надземной части, характерную для участка или части участка.
9. Гибридный регуляторный элемент по п. 1, где гетерологичная совокупность сегментов регуляторного элемента предусматривает более одной копии одного и того же сегмента из одного родительского регуляторного элемента.
10. Гибридный промотор по п. 1, где гетерологичная совокупность сегментов регуляторного элемента предусматривает сегменты родительского регуляторного элемента из более чем одного вида.
11. Гибридный промотор по п. 1, где гетерологичная совокупность сегментов регуляторного элемента предусматривает сегменты родительского регуляторного элемента из по меньшей мере одного родительского регуляторного элемента, полученного из вируса, или родительского регуляторного элемента растительного происхождения.
12. ДНК-конструкция, содержащая гибридный регуляторный элемент по п. 1.
13. ДНК-конструкция по п. 12, где гибридный регуляторный элемент функционально связан с представляющим интерес полинуклеотидом.
14. Кассета экспрессии, содержащая ДНК-конструкцию по п. 13.
15. Клетка-хозяин, содержащая кассету экспрессии по п. 14.
16. Клетка-хозяин по п. 14, где клетка-хозяин представляет собой растительную клетку, бактериальную клетку или клетку животного.
17. Трансгенное растение, содержащее кассету экспрессии по п. 14.
18. Гибридный регуляторный элемент, содержащий по меньшей мере одну полинуклеотидную последовательность, выбранную из группы, включающей
(a) полинуклеотид, последовательность которого на по меньшей мере 85 процентов идентична любой из последовательностей нуклеиновой кислоты под SEQ ID NO: 11-30 и 35-60;
(b) полинуклеотид под любым из SEQ ID NO: 11-30 и 35-60 или
(c) фрагмент SEQ ID NO: 11-30 и 35-60;
где указанная полинуклеотидная последовательность инициирует транскрипцию в растительной клетке.
19. ДНК-конструкция, содержащая гибридный регуляторный элемент по п. 18.
20. ДНК-конструкция по п. 16, где гибридный регуляторный элемент функционально связан с представляющим интерес полинуклеотидом.
21. Кассета экспрессии, содержащая ДНК-конструкцию по п. 19.
22. Клетка-хозяин, содержащая кассету экспрессии по п. 21.
23. Клетка-хозяин по п. 22, где клетка-хозяин представляет собой растительную клетку, бактериальную клетку или клетку животного.
24. Трансгенное растение, содержащее кассету экспрессии по п. 21.
25. Способ создания гибридного регуляторного элемента, включающий
(a) идентификацию нативного регуляторного элемента с профилем экспрессии, характерным для участка;
(b) идентификацию мотивов нативного регуляторного элемента и
(c) сборку по меньшей мере двух сегментов с получением гибридного регуляторного элемента, который инициирует транскрипцию в растительной клетке.
26. Способ по п. 25, дополнительно предусматривающий экспрессию представляющего интерес гетерологичного полинуклеотида, функционально связанного с гибридным регуляторным элементом.
27. Способ по п. 26, где представляющая интерес гетерологичная полинуклеотидная последовательность предусматривает генный продукт, который придает устойчивость к гербицидам, соли, холоду, засухе, патогенам или насекомым.
28. Способ по п. 26, где представляющая интерес гетерологичная полинуклеотидная последовательность экспрессируется в профиле экспрессии, характерном для корня, или профиле экспрессии, характерном для надземной части.
29. Способ по п. 25, где гибридный регуляторный элемент содержит
(a) полинуклеотид, последовательность которого на по меньшей мере 85 процентов идентична любой из последовательностей нуклеиновой кислоты под SEQ ID NO: 11-30 и 35-60;
(b) полинуклеотид под любым из SEQ ID NO: 11-30 и 35-60 или
(c) фрагмент любой из SEQ ID NO: 11-30, 35-47 и 50-59.
30. Способ по п. 25, где нативный регуляторный элемент содержит
(а) полинуклеотидную последовательность, содержащую последовательность, характеризующуюся по меньшей мере 85% идентичностью последовательности с полинуклеотидными последовательностями, изложенными под любым из SEQ ID NO: 1-10, 31-34 и 61-74; или
(b) полинуклеотидные последовательности, изложенные под любым из SEQ ID NO: 1-10, 31-34 и 61-74.
31. Способ направленной экспрессии полинуклеотидной последовательности в растении или растительной клетке, при этом указанный способ включает введение в растение или растительную клетку кассеты экспрессии, содержащей гибридный регуляторный элемент, функционально связанный с представляющей интерес гетерологичной полинуклеотидной последовательностью, где указанный гибридный регуляторный элемент позволяет получить уникальный профиль экспрессии в ткани представляющей интерес гетерологичной полинуклеотидной последовательности.
32. Способ по п. 31, где представляющая интерес гетерологичная полинуклеотидная последовательность экспрессируется в профиле экспрессии, характерном для корня, или профиле экспрессии, характерном для надземной части.
33. Способ по п. 31, где гибридный регуляторный элемент содержит
(a) полинуклеотид, последовательность которого на по меньшей мере 85 процентов идентична последовательности нуклеиновой кислоты под любым из SEQ ID NO: 11-30 и 35-60;
(b) полинуклеотид под любым из SEQ ID NO: 11-30 и 35-60 или
(c) фрагмент SEQ ID NO: 11-30, 35-47 и 50-59.
34. Способ по п. 31, где гибридный регуляторный элемент содержит по меньшей мере один сегмент из родительского регуляторного элемента, причем родительский регуляторный элемент содержит
(a) полинуклеотидные последовательности, изложенные под SEQ ID NO: 1-10, 31-34 и 61-74; или
(b) полинуклеотидную последовательность, содержащую последовательность, характеризующуюся по меньшей мере 85% идентичностью последовательности с полинуклеотидными последовательностями, изложенными под SEQ ID NO: 1-10, 31-34 и 61-74.
35. Способ по п. 31, где гибридный регуляторный элемент содержит совокупность по меньшей мере трех сегментов родительского регуляторного элемента.
36. Способ по п. 31, где гибридный регуляторный элемент содержит совокупность по меньшей мере четырех сегментов родительского регуляторного элемента.
37. Способ по п. 31, где профиль экспрессии в ткани представляет собой профиль экспрессии, характерный для корня, или профиль экспрессии, характерный для надземной части.
38. Способ по п. 37, где гибридный регуляторный элемент позволяет получить универсальный профиль экспрессии в корне, и родительские регуляторные элементы позволяют получить экспрессию в ткани корня или надземной части, характерную для участка или части участка.
39. Способ по п. 31, где гибридный регуляторный элемент содержит более одной копии сегмента из одного родительского регуляторного элемента.
40. Способ по п. 31, где гибридный регуляторный элемент содержит сегменты родительского регуляторного элемента из более чем одного вида.
