Abstract

Det beskrives en fremgangsmåte for den stereoselektive fremstillingen av en hetero-. bicyklisk alkohol entantiomer,at en i det vesentlige ren enantiomer med den generelle formel. hvor X er O, S, NH eller N-(Ci-C)alkyl, Yj og Y2 hver er uavhengig hydrogen eller substiruenter valgt fra halogen, (Ci-C)alkyl, (Ci-C)alkoksy, (Ci-C)halogenalkyl, formyl, nitro og cyano og Catomet har enten R- eller S-konfigurasjonen, fremstilles fra en forbindelse med den generelle formelhvor Rj er hydrogen eller en egnet beskyttende gruppe og Rer hydrogen eller hvor Ri og Rsammen danner en eventuelt mono- eller di-(Ci-C)alkyl substituert metylenbro, ved hjelp av følgende suksessive reaksjonstrinn: (i) reaksjon med en forbindelse med den generelle formelhvor Z er en hydroksygruppe eller en egnet avgangsgruppe, Rer en hydroksy-beskyttende gruppe og Rer et halogenatom, eller hvor Rog Rsammen utgjør en valensbinding eller et biradikal med formelen -C(Ri ])2-0-, hvor R] 1 er en lineær eller ' forgrenet ( C\ -C4)alkylgruppe og hvor Catomet enten har R- eller S-konfigurasjonen, (ii) den forbindelse som er dannet underkastes en reaksjon for fjerning av beskyttelsen og en ringslutningsreaksjon og (iii) beskyttelsen fjernes eventuelt fra hydroksygruppen i det ringsluttede produktet.Det beskrives videre enantiomerisk rene mellomprodukter, fremstillingen av disse mellomproduktene og fremstillingen av en startforbindelse.
