Abstract
Det er beskrevet templatfikserte ß- hårnålpeptidohermere syklo(-Tyr-His-X-Cys-Ser-Ala- DPro-Dab-Arg-Tyr-Cys-Tyr-Gln-Lys-DPro-Pro), med disulfidbinding mellom Cys4 og Cys11, og farmasøytisk akseptable salter derav, hvor X er Ala eller Tyr, som har CXCR4 antagoniserende egenskaper og som kan anvendes til å forhindre HIV-infeksjoner i friske individer eller for å forsinke eller stoppe virusprogresjon i infiserte pasienter; eller hvor kreft er mediert eller resulterer fra CXCR4-reseptoraktivitet; eller hvor immunologiske sykdommer er mediert eller resulterer fra CXCR4-reseptoraktivitet; eller for og behandle immunundertrykkelse; eller særlig for stamcellemobilisering av perifere blodstamceller og/eller mesenchymale celler (MSC) og/eller andre stamceller hvor retensjonen avhenger av CXCR4- reseptoren. Disse ß-hårnålpeptidhermerne kan bli fremstilt ved en prosess som er basert på en blandet faststoff- og løsningsfase syntetisk strategi, ved å bruke fremgangsmåter som er velkjent for de med adekvat kunnskap i peptidkjemi.
Description
Den foreliggende oppfinnelse tilveiebringer templatfikserte β-hårnålpeptidhermere som har CXCR4-antagoniserende aktivitet og som blir omfattet av den generelle beskrivelsen, men som ikke spesifikt er beskrevet i WO 2004/096840 A1.
DeMarco et al., Bioorganic & Medicinal Chemistry 14, 8396-8404 (2006), viser utvalgte β-hårnål hermere CXCR4 inhibitorer som er nyttige I behandling av HIV infeksjoner, kreft inflammasjon og i stamcelletransplantasjonsterapi, så vel som syntese av disse sykliske peptidhermerne.
WO 02/070547 A1 tilveiebringer β-hårnålpeptidhermere som har inkorporert templatfikserte kjeder av 8 til 16 α-aminosyre residuer og som har bredspekter antimikrobiel og antikreft aktivitet. Det er også beskrevet synteseprosesser for tillaging av disse peptidhermerne i parallell bibliotek format.
WO 01/16161 A1 tilveiebringer en prosess for syntese av templatfikserte βhårnålpeptidhermere som omfatter en templatefiksert kjede av fra 4 til 20 αaminosyre residuer. Prosessen er basert på syntetisk strategi med en blandet fast- og løsningsfase.
β-hårnålpeptidhermere ifølge oppfinnelsen er syklo(-Tyr-His-X-Cys-Ser-Ala-<D>Pro-Dab-Arg-Tyr-Cys-Tyr-Gln-Lys-<D>Pro-Pro), disulfidbinding mellom Cys4 og Cys11, og farmasøytisk akseptable salter derav, hvor X er Ala eller Tyr.
Ifølge oppfinnelsen kan de overnevnte β-hårnålhermere og farmasøytisk akseptable salter derav fremstilles med en prosess som omfatter:
(a) kobling av en egnet funksjonalisert fast understøttelse med et egnet N-beskyttet derivat av Pro;
(b) fjerning av den N-beskyttende gruppen fra produktet oppnådd i trinn (a); (c) kobling av produktet således oppnådd med et egnet N-beskyttet derivat av<D>Pro;
(d) fjerning av den N-beskyttende gruppen fra produktet som således ble oppnådd;
(e) kobling av produktet således oppnådd med et egnet N-beskyttet derivat av aminosyren som i det ønskede endeprodukt er i posisjon 14, dvs. Lys, hvor aminogruppen som er til stede i dens sidekjede likeledes er egnet beskyttet;
(f) fjerning av den N-beskyttende gruppen fra produktet som således ble oppnådd;

(k) fjerning av enhver beskyttende gruppe som er tilstede på funksjonelle grupper til ethvert ledd i kjeden av aminosyreresiduer; og
(l) hvis ønsket, konvertering av produktet således oppnådd til et farmasøytisk akseptabelt salt eller konvertering av et farmasøytisk akseptabelt, eller uakseptabelt salt som således er oppnådd til den tilsvarende frie forbindelse eller til et annet, farmasøytisk akseptabelt salt.
Trinnene i overnevnte prosess kan utføres ved fremgangsmåter som er velkjente for enhver person med tilstrekkelig kunnskap i peptidkjemi.
β-hårnålpeptidhermere ifølge oppfinnelsen kan anvendes i et stort område av anvendelser til å forhindre HIV-infeksjoner hos friske individer eller for å forsinke eller stoppe virusprogresjon i infiserte pasienter; eller der hvor kreft blir mediert av eller resulterer fra CXCR4-reseptoraiktivitet; eller der hvor immunologiske sykdommer blir mediert av eller resulterer fra CXCR4-reseptoraktivitet; eller for å behandle immunundertrykkelse; eller for å behandle inflammasjon, eller særlig for stamcellemobilisering av perifere blodstamceller og/eller mesenchymale celler (MSC) og/eller andre stamceller viss bibeholdelse avhenger av CXCR4-reseptoren.
β-hårnålpeptidhermere ifølge oppfinnelsen kan administreres i seg selv eller kan anvendes som en egnet formulering sammen med bærere, fortynningsmidler eller eksipienser som er velkjent på området.
Særlig kan β-hårnålpeptidhermere ifølge oppfinnelsen anvendes som en behandling for å øke frigjøring av hematopoietiske stamceller (HSC) fra benmargen for å anvendes ved allogen eller autolog transplantasjon.
Den akutte behandling med infusert HSC er i stor grad brukt for å gjenopprette immunfunksjoner hos pasienter som har mottatt myeloablativ terapi under behandling av kreftsykdommer så som multippelt myelom og non-Hodgkins lymfom. Pasienter eller donorer blir behandlet med HCS mobiliseringsmiddel, så som en forbindelse ifølge oppfinnelsen, og cellene blir deretter innsamlet fra perifert blod ved afarese. HCS blir transplantert tilbake etter f.eks. kjemoterapibehandling i pasienten (autolog transplantasjon) eller fra donor til mottaker (allogen transplantasjon), for således å fremme gjenopprettelse av immunfunksjonen (Frühauf et al., Br. J. Haematol. 122, 360-375 (2003)).
Andre anvendelser av HSC-behandlingen inkluderer, men er ikke begrenset til terapeutisk angiogenese i tilfelle av f.eks. hjerteanfall (Shepherd RM et al., Blood 2006 108(12):3662-3667).
β-hårnålpeptidhermere ifølge oppfinnelsen kan også anvendes til å behandle eller forhindre HIV-infeksjoner eller kreft så som brystkreft, hjernekreft, prostatakreft, lungekreft, nyrekreft, nevroblastom, non-Hodgkins lymfom, ovariekreft, multippelt myelom, kronisk lymfocyttisk leukemi, pankreaskreft, melanom, angiogenese og hematopoietiske vev; eller inflammatoriske lidelser så som astma, allergisk rinitt, hypersensitivitetslungesykdommer, hypersensitivitetspneumonitt, eosinofile pneumonier, forsinket type hypersensitivitet, interstitiell lungesykdom (ILD), idiopatisk pulmonal fibrose, ILD assosiert med reumatoid artritt, systemisk lupus erytematosus, ankyloserende spondylitt, perifer vaskulær sykdom, systemisk sklerose, Sjøgrens syndrom, von Hippel Lindau sykdom, systemisk anafylakse eller hypersensitivitetsresponser, medikamentallergier, reumatoid artritt, psoriatisk artritt, Behcets syndrom, mukositt, Crohns sykdom, multippel sklerose, myastenia gravis, juvenil igangsettende diabetes, glomerulonefritt, autoimmun troiditt, graftavstøtning, inkludert allograftavstøtning eller graft-versus-vert sykdom, inflammatoriske tarmsykdommer, inflammatoriske dermatoser; eller til å behandle immunundertrykkelse, inkludert immunundertrykkelse indusert med grafttransplantasjonsavstøtning.
β-hårnålpeptidhermere ifølge oppfinnelsen kan administreres enkeltvis, som blandinger av flere enn én β-hårnålpeptidhermer, i kombinasjon med, som tilfellet kan være, andre HSC-mobiliseringsmidler, eller anti-HIV-midler, eller antimikrobielle midler, eller antikreftmidler, eller antiinflammatoriske midler, og/eller i kombinasjon med andre farmasøytisk aktive midler.
Farmasøytiske sammensetninger som omfatter β-hårnålpeptidhermere ifølge oppfinnelsen kan fremstilles ved hjelp av konvensjonell blanding, oppløsning, granulering, belagt tablettfremstilling, finmaling, emulgering, innkapsling, innfanging eller lyofiliseringsprosesser. Farmasøytiske sammensetninger kan formuleres på konvensjonell måte ved å bruke én eller flere fysiologisk akseptable bærere, fortynningsmidler, eksipienser eller hjelpestoffer som fremmer prosessering av de aktive β-hårnålpeptidhermere til preparater som kan brukes farmasøytisk. Riktig formulering avhenger av administrasjonsfremgangsmåten som velges.
For topisk administrering kan β-hårnålpeptidhermere ifølge oppfinnelsen formuleres som løsninger, geler, salver, kremer, suspensjoner, etc. som er velkjent på området.
Systemiske formuleringer inkluderer de konstruert for administrering ved injeksjon, f.eks. subkutan, intravenøs, intramuskulær, intratekal eller intraperitoneal injeksjon, så vel som de konstruert for transdermal, transmukosal, oral eller pulmonal administrering.
For injeksjoner kan β-hårnålpeptidhermere ifølge oppfinnelsen formuleres i adekvate løsninger, fortrinnsvis i fysiologisk kompatible buffere så som Hanks løsning, Ringers løsning, eller fysiologisk saltløsningsbuffer. Løsningene kan inneholde formeldannende midler så som suspensjons-, stabiliserende og/eller dispergerende midler.
Alternativt kan β-hårnålpeptidhermerne ifølge oppfinnelsen være i pulverform for kombinasjon med en egnet bærer, f.eks. sterilt pyrogenfritt vann, før anvendelse.
For transmukosal administrering blir penetrerende midler som er egnet for den barrieren som skal penetreres brukt i formuleringen som kjent på området.
For oral administrering kan forbindelsene lett formuleres ved å kombinere de aktive β-hårnålpeptidhermerne ifølge oppfinnelsen med farmasøytisk akseptable bærere velkjent på området. Slike bærere muliggjør at β-hårnålpeptidhermere ifølge oppfinnelsen kan formuleres som tabletter, piller, drasjeer, kapsler, væsker, geler, sirup, slurryer, suspensjoner, etc. for oralt inntak av en pasient som skal behandles. For orale formuleringer, slik som for eksempel pulvere, kapsler og tabletter, så inkluderer passende eksipienser fyllmidler slik som sukkertyper, for eksempel laktose, sukrose, mannitol og sorbitol, cellulosepreparater slik som maisstivelse, hvetestivelse, risstivelse, potetstivelse, gelatin, tragantgummi, metylcellulose, hydroksypropylmetylcellulose, natriumkarboksymetylcellulose, granulerende midler, og bindemidler. Hvis ønsket kan oppløsende midler tilsettes, så som tverrbundet polyvinylpyrrolidon, agar eller algininsyre eller et salt derav, så som natriumalginat. Hvis ønsket kan faste doseringsformer være sukkerbelagte eller enterisk belagte ved å bruke standard teknikker.
For orale preparater så som f.eks. suspensjoner, eliksirer og løsninger inkluderer egnede bærere, eksipienser eller fortynningsmidler vann, glykoler, oljer, alkoholer, etc. I tillegg kan smaksmidler, preserverende midler, fargemidler og lignende tilsettes.
For munnhuleadministrering kan sammensetningen ha form av tabletter, sugetabletter, etc., formulert som vanlig.
For administrering ved inhalasjon blir β-hårnålpeptidhermere ifølge oppfinnelsen egnet levert i form av en aerosolspray fra trykksatte pakker eller en nebulisator, med anvendelse av et egnet drivmiddel, f.eks. diklordifluormetan, triklorfluormetan, karbondioksid eller en annen egnet gass. I tilfelle av en trykksatt aerosol kan doseringsenheten bestemmes ved å tilveiebringe en ventil for å levere en utmålt mengde. Kapsler og kassetter med f.eks. gelatin for anvendelse i en inhalator eller insufflator kan formuleres slik at det inneholder en pulverblanding av βhårnålpeptidhermere ifølge oppfinnelsen og en egnet pulverbase så som laktose eller stivelse.
Forbindelsene kan også formuleres i rektale eller vaginale sammensetninger så som stikkpiller sammen med egnet stikkpillebasis så som kakaosmør eller andre glyserider.
I tillegg til formuleringene beskrevet tidligere kan β-hårnålpeptidhermere ifølge oppfinnelsen også formuleres som depotpreparater. Slike langtidsvirkende formuleringer kan administreres ved implantasjon (f.eks. subkutant eller intramuskulært) eller ved intramuskulær injeksjon. For fremstilling av slike depotpreparater kan β-hårnålhermerne ifølge oppfinnelsen formuleres med egnet polymert eller hydrofobt materiale (f.eks. som en emulsjon i en akseptabel olje) eller ionebytteharpikser, eller som tungt løselige salter.
I tillegg kan andre farmasøytiske leveringssystemer anvendes så som liposomer og emulsjoner velkjent på området. Visse organiske løsningsmidler så som dimetylsulfoksid kan også anvendes. I tillegg kan β-hårnålpeptidhermerne ifølge oppfinnelsen leveres ved å bruke et forsinket frigjøringssystem, så som semipermeable matriser av faste polymerer som inneholder det terapeutiske middel. Forskjellige forsinkede frigjøringsmaterialer er blitt etablert og er velkjent for de med kunnskap på området. Forsinket frigjøringskapsler kan, avhengig av deres kjemiske natur, frigjøre forbindelsene for noen uker opptil over hundre dager.
Avhengig av den kjemiske natur og den biologiske stabilitet til det terapeutiske middel, kan ytterligere strategier for proteinstabilisering anvendes.
Idet β-hårnålpeptidhermerne ifølge oppfinnelsen inneholder ladede residuer, kan de inkluderes i enhver av de ovenfor beskrevne formuleringer som sådan eller som farmasøytisk akseptable salter. Farmasøytisk akseptable salter har en tendens til å være mer løselig i vandige og andre protiske løsningsmidler enn de er i de tilsvarende frie former. Særlig egnede farmasøytisk akseptable salter inkluderer salter med karboksylsyrer, fosfonsyrer, sulfonsyrer og sulfaminsyrer, f.eks. eddiksyre, propionsyre, oktansyre, dekansyre, dodekansyre, glykolsyre, melkesyre, fumarsyre, ravsyre, adipinsyre, pimelinsyre, suberinsyre, acelainsyre, malinsyre, tartarsyre, sitronsyre, aminosyrer, så som glutaminsyre eller asparaginsyre, maleinsyre, hydroksymaleinsyre, metylmaleinsyre, sykloheksankarboksylsyre, adamantankarboksylsyre, benzosyre, salisylsyre, 4-aminosalisylsyre, ftalsyre, fenyleddiksyre, mandelsyre, kanelsyre, metan- eller etansulfonsyre, 2-hydroksyetansulfonsyre, etan-1,2-disulfonsyre, benzensulfonsyre, 2-naftalensulfonsyre, 1,5-naftalen-disulfonsyre, 2-, 3- eller 4-metylbenzensulfonsyre, metylsvovelsyre, etylsvovelsyre, dodecylsvovelsyre, N-sykloheksylsulfamsyre, N-metyl-, N-etyl- eller N-propyl-sulfamsyre og andre organiske protoniske syrer, så som askorbinsyre. Egnede uorganiske syrer er f.eks. hydrohalogensyrer, så som saltsyre, svovelsyre og fosforsyre.
β-hårnålpeptidhermere ifølge oppfinnelsen, i fri form eller i form av farmasøytisk akseptable salter eller sammensetninger derav, vil generelt anvendes i en mengde som er effektiv til å oppnå den tenkte hensikt. Det skal forstås at mengden som anvendes vil avhenge av den spesielle applikasjonen.
For topisk administrering for å behandle eller forhindre HIV-infeksjoner kan en terapeutisk effektiv dose bestemmes ved å bruke f.eks. in vitro analyser tilveiebrakt i eksemplene. Behandlingen kan appliseres mens HIV-infeksjonen er synlig, eller til og med når den ikke er synlig. En vanlig ekspert med kunnskap på området vil være i stand til å bestemme de terapeutisk effektive mengder til å behandle topiske HIV-infeksjoner uten unødvendig eksperimentering.
For systemisk administrering kan en terapeutisk effektiv dose estimeres initielt fra in vitro analyser. F.eks. kan en dose formuleres i dyremodeller for å oppnå et sirkulerende β-hårnålpeptidhermekonsentrasjonsområde som inkluderer IC50som bestemt i cellekulturen (dvs. konsentrasjonen av en testforbindelse som er letal for 50 % av cellekulturen). Slik informasjon kan anvendes for mer nøyaktig å bestemme nyttige doser hos mennesker.
Initielle doseringer kan også bestemmes fra in vivo data, f.eks. dyremodeller, ved å bruke teknikker som er velkjent på området. En som har vanlig kunnskap på området kan lett optimalisere administreringen til mennesker basert på dyredata.
Doseringsmengder for applikasjoner som anti-HIV-midler kan justeres individuelt for å tilveiebringe plasmanivåer av β-hårnålpeptidhermere ifølge oppfinnelsen som er tilstrekkelig til å opprettholde den terapeutiske virkning. Terapeutisk effektive serumnivåer kan oppnås ved å administrere et flertall av doser hver dag.
I tilfelle av lokal administrering eller selektivt opptak, kan den effektive lokale konsentrasjon av β-hårnålpeptidohermere i henhold til oppfinnelsen ikke være relatert til plasmakonsentrasjonen. En som har vanlig kunnskap på området vil være i stand til å optimalisere terapeutisk effektive lokale doseringer uten unødvendig eksperimentering.
Mengden av β-hårnålpeptidhermere administrert vil selvfølgelig være avhengig av individet som blir behandlet, av individets vekt, alvorligheten av lidelsen, administrasjonsmåten og bedøvelsen til den utstedende lege.
Anti-HIV-terapi kan gjentas med avbrudd mens infeksjoner er detekterbare eller til og med når de ikke er detekterbare. Terapien kan tilveiebringes alene i kombinasjon med andre medikamenter, så som f.eks. andre anti-HIV-midler eller antikreftmidler, eller andre antimikrobielle midler.
Normalt vil en terapeutisk effektiv dose av β-hårnålpeptidhermere beskrevet heri tilveiebringer terapeutisk gunstig virkning uten å forårsake hovedsakelig toksisitet.
Toksisitet til β-hårnålpeptidhermerne i oppfinnelsen kan bestemmes ved standard farmasøytiske prosedyrer i cellekulturer eller eksperimentdyr, f.eks. ved å bestemme LD50(dosen som er letal for 50 % av befolkningen) eller LD100(dosen som er letal for 100 % av befolkningen). Doseforholdet mellom toksisk og terapeutisk effekt er den terapeutiske indeks. Forbindelser som utviser høye terapeutiske indekser er foretrukket. Dataene oppnådd fra disse cellekulturanalysene og dyrestudiene kan anvendes til å formulere et doseringsområde som ikke er toksisk for anvendelse hos mennesker. Doseringen av β-hårnålpeptidhermerne ifølge oppfinnelsen ligger fortrinnsvis i et område av sirkulerende konsentrasjoner som inkluderer den effektive dosen med liten eller ingen toksisitet. Doseringen kan variere innenfor området avhengig av doseringsformen som anvendes og den benyttede administrasjonsrute. Den nøyaktige formuleringen, administrasjonsrute og dosering kan velges av den individuelle lege i lys av pasientens tilstand (se f.eks. Fingl et al., 1975, i: The Pharmacological Basis of Therapeutics, kapittel 1, side 1).
De følgende eksempler illustrerer oppfinnelsen er detaljert, men er ikke ment å begrense dets område på noen måte. De følgende forkortelser er brukt i disse eksemplene:

1. Peptidsyntese
Kobling av det første beskyttede aminosyreresidu til harpiksen
0,5 g 2-klortritylkloridharpiks (100-200 mesh, kopoly(styren-1 % DVB) polymermatriks, kat.nr. 01-64-0114, Novabiochem, Merck Biosciences Ltd.) (Barlos et al. Tetrahedron Lett. 1989, 30, 3943-3946) (1,4 mMol/g, 0,7 mmol) ble fylt i en tørr flaske. Harpiksen ble suspendert i CH2Cl2(2,5 ml) og latt svelle ved romtemperatur under konstant omrøring i 30 min. Harpiksen ble behandlet med 0,49 mMol (0,7 ekv.) av den første egnede beskyttede aminosyreresidu og 488 μl (4 ekv.) av diisopropyletylamin (DIEA) i CH2Cl2(2,5 ml), hvor blandingen ble ristet ved 25<o>C i 4 timer. Harpiksen ble ristet (CH2Cl2/MeOH/DIEA: 17/2/1), 30 ml i 30 min.; deretter vasket i følgende rekkefølge med CH2Cl2(1x), DMF (1x), CH2Cl2(1x), MeOH (1x), CH2Cl2(1x), MeOH (1x), CH2Cl2(2x), Et2O (2x) og tørket under vakuum i 6 timer. Oppastingen var typisk 0,6-0,9 mMol/g.
Den følgende foropplastede harpiks ble fremstilt: Fmoc-Pro-2-klortritylharpiks.
Syntese av det totalt beskyttede peptidfragment
Syntesen ble utført på en Syro-peptidsynteseanordning (MultiSynTech GmbH) ved å bruke 24-96 reaksjonskar. I hvert kar ble det plassert ca.60 mg (vekten av harpiksen før opplasting) av den overnevnte harpiks. De følgende reaksjonssykluser ble programmert og utført:
Trinnene 3 til 6 blir gjentatt for å tilsette hver aminosyre.
Analyttisk fremgangsmåte:
Analyttisk HPLC tilbakeholdelsestider (RT, i min.) ble bestemt ved å bruke en Jupiter Proteo 90 A kolonne, 150 x 2,0 mm (kode 00F-4396-B0-fenomeneks) med følgende løsningsmidler A (H2O 0,1 % TFA) og B (CH3CN 0,1 % TFA) og gradienten: 0 min.: 95 % A, 5 % B, 0,5 min.: 95 % A, 5 % B; 20 min.: 40 % A, 60 % B; 21 min.: 0 % A, 100 % B; 23 min.: 0 % A, 100 % B; 23,1 min.: 95 % A, 5 % B; 31 min.: 95 % A, 5 % B.
Dannelse av disulfid β-trådkobling
Etter avslutning av syntesen ble harpiksen oppsvellet i 3 ml tørr DMF i 1 time. Deretter ble 10 ekv. jodløsning i DMF (6 ml) tilsatt reaktoren, etterfulgt av omrøring i 1,5 timer. Harpiksen ble filtrert og en fersk løsning av jod (10 ekv.) i DMF (6 ml) ble tilsatt, etterfulgt av omrøring i ytterligere 3 timer. Harpiksen ble filtrert og vasket med DMF (3x) og CH2Cl2(3x).
Avspaltning, ryggradssyklisering, avbeskyttelse og rensing av peptidet
Etter dannelsen av disulfid β-trådkoblingen, ble harpiksen suspendert i 1 ml (0,14 mMol) 1 % TFA i CH2Cl2(vol/vol) i 3 min. og filtrert, og filtratet ble nøytralisert med 1 ml (1,15 mMol) 20 % DIEA i CH2Cl2(vol/vol). Denne prosedyre ble gjentatt to ganger for å sikre fullstendig avspalting. Harpiksen ble vasket tre ganger med 1 ml CH2Cl2. Deretter ble CH2Cl2-laget inndampet til tørrhet.
De flyktige stoffene ble fjernet og 8 ml tørr DMF ble tilsatt røret. Deretter ble 2 ekv. HATU i tørr DMF (1 ml) og 4 ekv. DIPEA i tørr DMF (1 ml) tilsatt til peptidet, etterfulgt av omrøring i 16 timer. De flyktige stoffene ble inndampet til tørrhet. Det rå sykliserte peptid ble oppløst i 7 ml CH2Cl2og ekstrahert med 10 % acetonitril i H2O (4,5 ml) tre ganger. CH2Cl2-laget ble inndampet til tørrhet. For å avbeskytte peptidet totalt, ble 3 ml avspaltingscocktail TFA:TIS:H2O (95:2,5:2,5) tilsatt, og blandingen ble holdt i 2,5 timer. De flyktige stoffene ble inndampet til tørrhet og råpeptidet ble oppløst i 20 % AcOH i vann (7 ml) og ekstrahert med isopropyleter (4 ml) tre ganger. Det vandige lag ble innsamlet og inndampet til tørrhet, og residuet ble renset ved preparerende revers fase HPLC.
Etter lyofiliseringen ble produktene oppnådd som hvitt pulver og analysert av HPLC-ESI-MS analyttisk fremgangsmåte beskrevet over. De analyttiske dataene som omfatter renhet etter preparerende HPLC og ESI-MS er gitt.
Eksempel 1: Peptidet ble syntetisert ved å starte med aminosyren L-Pro som ble podet på harpiksen. Startharpiksen var Fmoc-Pro-2-klortritylharpiks, som ble fremstilt som beskrevet over. Det lineære peptid ble syntetisert på fast underlag i henhold til prosedyren beskrevet over i følgende sekvens: Harpiks-Pro-<D>Pro-Lys-Gln-Tyr-Cys-Tyr-Arg-Dab-<D>Pro-Ala-Ser-Cys-Ala-His-Tyr. En disulfid βtrådkobling ble innført som beskrevet over. Produktet ble spaltet fra harpiksen, syklisert, avbeskyttet og renset som angitt ved preparerende revers fase LC-MS. Etter lyofilisering ble produktet oppnådd som et hvitt pulver og analysert ved den HPLC-ESI-MS analyttiske fremgangsmåte beskrevet over ([M+2H]<2+>: 933,1; RT: 10,47; UV-renhet: 72 %).
Eksempel 2: Peptidet ble syntetisert ved å starte med aminosyren L-Pro som ble podet til harpiksen. Utgangsharpiks var Fmoc-Pro-2-klortritylharpiks, som ble fremstilt som beskrevet ovenfor. Det lineære peptid ble syntetisert på fast underlag i henhold til prosedyren beskrevet ovenfor med den følgende sekvens: harpiks-Pro-<D>Pro-Lys-Gln-Tyr-Cys-Tyr-Arg-Dab-<D>Pro-Ala-Ser-Cys-Tyr-His-Tyr. En disulfid βtrådkobling ble innført som beskrevet over. Produktet ble spaltet fra harpiksen, syklisert, avbeskyttet og renset som angitt ved preparativ revers fase LC-MS. Etter lyofilisering ble produktet oppnådd som et hvitt pulver og analysert med HPLC-ESI-MS analyttisk fremgangsmåte beskrevet over ([M+2H]<2+>: 978,6; RT: 10,95; UV-renhet: 82 %).
2. Biologiske fremgangsmåter
2.1. Fremstilling av peptidene
Lyofiliserte peptider ble veid på en mikrovekt (Mettler MT5) og løst i sterilt vann til en endelig konsentrasjon på 1 mM med mindre noe annet er angitt.
Stamløsninger ble holdt på 4<o>C, beskyttet mot lys.
2.2. Ca<2+>-analyse: CXCR4-antagoniserende aktivitet til peptidene Økninger i intracellulært kalsium ble overvåket ved å bruke en Flexistation 384 (Molecular Devices, Sunnyvale, CA) for å analysere peptidene for CXCR4-antagonisme i en pre-B-musecellelinje 300-19 stabilt overført med humant CXCR4 [se referanser 1, 2 og 3 nedenfor]. Cellene ble porsjonsoppladet med kalsium 3 analysesett (Molecular Devices) i analysebuffer (Hanks balanserte saltløsning, HBSS, 20 mM HEPES, pH 7,4, 0,1 % BSA) i 1 time ved romtemperatur og deretter ble 200 000 merkede celler fordelt i sorte 96-brønners analyseplater (Costar nr. 3603). En 20 ganger konsentrert løsning av peptid i analysebuffer ble tilsatt cellene og hele platen ble sentrifugert for å felle ut cellene til bunnen av brønnene.
Kalsiummobilisering indusert ved 10 nM stromaavledet faktor-1 (SDF-1) ble målt i Flexstation 384 (eksitasjon, 485 nM; emisjon 525 nM) i 90 sek. En maksimal forandring i fluorescensrespons over basislinjen ble brukt til å beregne antagonistaktivitet. Dataene for doseresponskurver (antagonistkonsentrasjon mot prosent maksimum respons) ble tilpasset en fire parameter logistisk ligning ved å bruke SoftmaxPro 4.6 (Molecular Devices), fra hvilke IC50-verdier ble beregnet.
2.3. FIGS-Analyse<TM>
Analysen ble utført i henhold til referanse 5 nedenfor. Stamløsninger av peptider (10 μm) ble fremstilt ved oppløsning i 10 μm Tris-HCl ved romtemperatur.
Stamløsninger ble holdt ved 4<o>C, beskyttet mot lys. Arbeidsfortynninger ble fremstilt etter bestilling ved seriefortynning i fosfatbufret saltløsning (PBS) og tilsatt i et endelig volum på 10 μl direkte til cellekulturene. Etter 48 timers samdyrking ble kulturene renset med PBS og deretter eksponert til glutaraldehyd/formaldehyd (0,2 %/2 %) i PBS i 5 min. For fotometrisk kvantifisering ble de fikserte kulturene deretter inkubert med orto-nitro-fenylgalaktopyranosid (ONPG) som et β-galaktosidasesubstrat, som ble enzymatisk konvertert til det kromofore ortonitrofenol (ONP). Utlesningen blir direkte oppnådd ved å måle optisk tetthet av brønnene ved 405 nm i en iEMS 96-brønners plateleser.
2.4. Cytotoksisk analyse
Cytotoksisiteten til peptidene for HELA-celler (Acc57) og COS-7-celler (CRL-1651) ble bestemt ved å bruke MTT reduksjonsanalyse [se referanse 6 og 7 nedenfor]. Kortfattet var fremgangsmåten som følger: HELA-celler og COS-7-celler ble sådd med henholdsvis 7,0 x 10<3>og 4,5 x 10<3>celler pr. brønn og dyrket i 96-brønners mikrotiterplater i 24 timer ved 37<o>C med 5 % CO2. På dette tidspunkt, ble tid null (Tz) bestemt med MTT-reduksjon (se nedenfor). Supernatanten i de gjenværende brønner ble kastet og ferskt medium og peptider i seriefortynninger på 12,5, 25 og 50 μm ble pipettert inn i brønnene. Hver peptidkonsentrasjon ble analysen i triplikat. Inkubasjon av cellene ble fortsatt i 48 timer ved 37<o>C med 5 % CO2. Brønnene ble deretter vasket én gang med PBS og deretter 100 μl MTT-reagens (0,5 mg/ml i medium RPMI1640 og, henholdsvis, DMEM) ble tilsatt brønnene. Dette ble inkubert ved 37<o>C i 2 timer og deretter ble mediet aspirert og 100 μl isopropanol ble tilsatt hver brønn. Absorbansen ved 595 nm av det løseliggjorte produkt ble målt (OD595peptid). For hver konsentrasjon ble gjennomsnitt beregnet fra triplikater. Prosentvis vekst ble beregnet som følger: (OD595peptid-OD595Tz-OD595tom brønn)/(OD595Tz-OD595tom brønn) x 100 % og ble plottet for hver peptidkonsentrasjon.
LC50-verdiene (letal konsentrasjon, definert som den konsentrasjonen som dreper 50 % av cellene) ble bestemt for hvert peptid ved å bruke tendenslinjefunksjonen til EXCEL (Microsoft Office 2000) for konsentrasjonene (50, 25, 12,5 og 0 μM), de tilsvarende vekstprosentandeler og verdien -50, (=TREND(C50:C0,%50:%0,-50)). GI 50 (vekstinhibisjon) konsentrasjoner ble beregnet for hvert peptid ved å bruke en tendenslinjefunksjon for konsentrasjonene (50, 25, 12,5 og 9 μg/ml), de tilsvarende prosentandeler og verdien 50, (=TREND(C50:C0,%50:%0,50).
2.5. Cellekultur
”CCR5”-celler ble dyrket i DMEM-medium med 4500 mg/ml glukose, 10 % føtalt bovint serum (FBS), tilsatt 50 U/ml penicillin og 50 μg/ml streptomycin (Pen/Strept.). Hut/4-3-celler ble opprettholdt i RPMI-medium, 10 % FBS, tilsatt Pen/Strept. og 10 mM HEPES. HELA-celler og CCRF-CEM-celler ble opprettholdt i RPMI1640 5 % FBS, Pen/Strept. og 2 mM L-glutamin. Cos-7-celler ble dyrket i DMEM-medium med 4500 mg/ml glukose tilsatt 10 % FCS, Pen/Strept. og 2 mM L-glutamin. Alle cellelinjer ble dyrket ved 37<o>C med 5 % CO2. Cellemedia, mediumtilsetning, PBS-buffer, HEPES, Pen/Strept., L-glutamin og sera ble kjøpt fra Gibco (Pailsey, GB). Alle fine kjemikalier kom fra Merck (Darmstadt, Tyskland).
2.6. Hemolyse
Peptidene ble testet for deres hemolytiske aktivitet mot humane røde blodceller (hRBC). Ferske hRBC ble vasket tre ganger med fosfatbufret saltløsning (PBS) ved sentrifugering i 10 min. på 2000 x g. Peptider ble med en konsentrasjon på 100 μm inkubert med 20 % vol/vol hRBC i 1 time ved 37<o>C. Den endelige erytrocyttkonsentrasjonen var ca. 0,9 x 10<9>celler/ml. En verdi på 0 %, henholdsvis 100 % cellelyse ble bestemt ved inkubasjon av hRBC i nærvær av PBS alene og henholdsvis 0,1 % Triton X-100 i H2O. Prøvene ble sentrifugert og supernatanten ble fortynnet 20 ganger i PBS-buffer og den optiske tetthet (OD) av prøven ved 540 nm ble målt. 100 % lyseverdi (OD540H2O) ga en OD540på ca.1,3-1,8. Prosentvis hemolyse ble beregnet som følger: (OD540peptid/OD540H2O) x 100 %.
2.7. Kjemotaktisk analyse (cellemigreringsanalyse)
Den kjemotaktiske respons til CCRF-CEM-celler til en gradient av stromacelleavledet faktor 1 α (SDF-1) ble målt ved å bruke engangsanalyseplater fra Neuroprobe (5 μ porestørrelse) (Gaithersburg, MD), i henhold til produsentens retningslinjer og referanser deri [spesielt referanse 8 nedenfor]. Kortfattet ble én 175 cm<3>flaske vasket én gang med Dulbeccos fosfatbufret saltløsning (DPBS), og trypsinert i 10 min. eller inntil cellene hadde løftet seg. Trypsinet ble nøytralisert ved tilsetting av ferskt medium som inneholdt serum og cellene ble pelletert, vasket én gang i DPBS, og resuspendert med 1-0,5 x 10<7>celle/ml i RPMI 0,5 % bovint serumalbumin (BSA). 45 μl cellesuspensjon ble blandet med 5 μl av 10 ganger konsentrert PEM peptid fortynnet i det samme analysemedium. 35 μl av denne blandingen ble applisert på toppen av analysefilteret. Cellene ble tillatt å migrere (ved 37<o>C) inn i bunnkammeret i analyseplaten som inneholdt 1 nM SDF-1. Etter 4 timer ble filteret fjernet og MTT ble tilsatt de migrerte celler til en endelig konsentrasjon på 0,5 mg/ml, og inkubert for ytterligere 4 timer. Etter merking med MTT ble alt mediet fjernet og 100 μl av isopropanol pluss 10 mM HCl ble tilsatt cellene. Den optiske absorbansen ved 595 nm (ABS595) ble lest ved å bruke en Tecan Genios plateleser med Magellan programvare. Antall migrerte celler ble bestemt ved å sammenligne ABS 595-verdiene mot en standardkurve dannet med et kjent antall celler i analyseplaten og ble plottet mot SDF-1-konsentrasjonen for å oppnå en sigmoid kurve og for å bestemme IC50-verdiene. Verdiene for IC50ble bestemt ved å bruke tendenslinjefunksjonen i Microsoft Excel ved å tilpasse en logaritmisk kurve til de gjennomsnittsberegnede datapunkter.
2.8. Plasmastabilitet
405 μl av plasma/albuminløsning ble plassert i et polypropylen (PP) rør og spritet opp med 45 μl av en forbindelse fra en 100 μM løsning B, avledet fra 135 μl av PBS og 15 μl av 1 mM peptid i PBS, pH 7,4.150 μl porsjoner ble overført til individuelle brønner på den 10 kDa filterplaten (Millipore MAPPB 1010 Biomax membran). For ”0 min. kontroller”: 270 μl PBS ble plassert i et PP-rør og 30 μl stamløsning B ble tilsatt og spunnet. 150 μl kontrolløsning ble plassert i én brønn av filterplaten og tjener som ”filtrert kontroll”.
Ytterligere 150 μl av kontrolløsning ble plassert direkte i en mottakerbrønn (reservert for filtrat) og tjener som ”ikke-filtrert kontroll”. Hele platen inkludert inndampingslokk ble inkubert i 60 min. ved 37<o>C. Plasmaprøvene (rotteplasma: Harlan Sera lab GB, human plasma: Blutspendenzentrum Zürich) ble sentrifugert minst 2 timer ved 4300 rpm (3500 g) og 15<o>C for å gi 100 μl filtrat. For ”serumalbumin”-prøver (ferskt fremstilt human albumin: Sigma A-4327, rottealbumin: Sigma A.6272, alle ved 40 mg/ml konsentrasjon i PBS) er ca. 1 time sentrifugering tilstrekkelig. Filtratene i mottaker-PP-platen ble analysert med LC/MS som følger: Kolonne: Jupiter C18 (Phenomenex), mobile faser: (A) 0,1 % maursyre i vann og (B) acetonitril, gradient: 5 %-100 % (B) i 2 min., elektrosprayionisering, MRM-deteksjon (trippel kvadrupol). Topparealene ble bestemt og gjennomsnitt ble beregnet av triplikate verdier. Bindingen blir uttrykt i prosent av (filtrert og ikke-filtrert tidspunkt 0 min.) kontroll 1 og 2 med: 100-(100 * T60/T0). Gjennomsnittet fra disse verdiene blir deretter beregnet (se referanse 9 nedenfor).
3.0. In vivo undersøkelser
3.1. Maksimal tolerert dose hos mus
a) Forbindelsen i eksempel 1, dispergert i vann for injeksjon eller 0,9 % fysiologisk saltløsning) ble administrert i den preliminære undersøkelsen med i.v. injeksjon med dosenivåer på 35, 50, 70, 85, 100, 150, 250 eller 500 mg/kg til grupper som bestod av én hannmus og én hunnmus (Crl:CD1(ICR)). I tillegg mottok to grupper som omfattet to hannmus og to hunnmus dosenivåer på henholdsvis 90 og 100 mg/kg, og 50 mg/kg dosenivået ble gjentatt i en gruppe som omfatter én hannmus og én hunnmus.
b) Maksimalt tolererte doseundersøkelser (MTD) utført med forbindelsen i eksempel 2 ved å bruke CD1-mus (3 mus/gruppe) ble utført ved å bruke i.v., ip og sc administrering.
3.2. Gjentatte toksisitetsundersøkelser hos mus
Toksisiteten og toksikokinetikken til forbindelsen i eksempel 1 ble undersøkt etter daglig i.v. injeksjon i musen over minst 14 dager. Grupper på 12 hannmus og 12 hunnmus Crl:CD1(ICR) mottok dosepreparater som inneholdt kontrollartikkel (50 mM natriumdihydrogenortofosfatbuffer som inneholdt 0,9 % vekt/vol natriumklorid) eller 8, 24 eller 30 mg/kg/dag POL6326 med et dosevolum på 5 ml/kg. Satellittgrupper på 24 dyr pr. kjønn pr. gruppe ble inkludert på hvert dosenivå. Vurdering av toksisitet ble basert på mortalitet, kliniske tegn, kroppsvekt, matinntak, øyeundersøkelser, klinisk og anatomisk patologi, og toksokinetiske evalueringer.
3.3. Stamcellemobilisering
a) Musemodell:
Målet for undersøkelsen var å evaluere evnen til forbindelsen i eksempel 1 og eksempel 2 til å mobilisere hematopoietiske forløpere fra musbenmarg til perifert blod ved å bruke in vitro hematopoietiske kolonianalyser. Hos mennesker er nøyaktig informasjon om forløpercellemobilisering tilveiebrakt ved de kolonidannende enhetsgranulocyttmonocytt (CFU-GM) analysen eller ved å bestemme hyppigheten av CD34(+) celler med FACS-analyse (se referanse 10 nedenfor). Hos mus er CD34 ikke en nyttig markør for stamceller; isteden er CFU-GM mer vanlig brukt (se referanse 11 nedenfor).
For å vurdere evnen til forbindelsene i eksempel 1 og 2 til å mobilisere murine stamceller (CFU-GM) ble C3H/HeJ hunnmus (Jackson Laboratory) injisert subkutant med 5 mg/k, av forbindelsene i eksempel 1 og eksempel 2 og som referanse AMD3100 (Broxmeyer et all., J Exp Med 201, 1307-1318), (som for tiden gjennomgår fase III kliniske forsøk) for stamcellemobilisering. Perifere blodprøver fra 5 dyr pr. testgruppe ble innsamlet på hvert tidspunkt og tellinger av celler med kjerne ble utført som standard analyser.
b) Apemodell:
En vurdering av mobilisering av hematopoietiske stamceller fra perifert blod i cynomolgusaper (Macaca fascicularis) ble utført. Forbindelsen i eksempel 1 ble administrert til 4 aper (2 hannlige og 2 hunnlige) som en langsom bolus i.v. injeksjon over to min. og CD34(+) celler ble bestemt med FACS-analyser.
Toksikokinetisk blodprøvetaking ble også utført.
4.0. Resultater
Resultatene av eksperimentene beskrevet under 2.2.-2.8. ovenfor er angitt i de følgende tabeller 1 og 2.

4.1: MTD-undersøkelse i mus
a) MTD-undersøkelse, forbindelse i eksempel 1.
Det akutte minimum letale intravenøse dosenivå i eksempel 1 i musen ble funnet å overskride 90 mg/kg.
b) MTD-undersøkelse, forbindelse i eksempel 2.
Den høyeste dose testet for alle tre administrasjonsrutene var 120 mg/kg bolus. Ved denne dosen overlevde alle dyrene og bare milde symptomer ble observert.
Symptomene som ble utvist var lett adferdsdepresjon, lett cyanose, en økning i respirasjonsdybde og muskelavslapning.
4.2: 14 dagers intravenøs injeksjonstoksisitet og toksikokinetiske undersøkelser NOAEL-nivå for forbindelsen i eksempel 1 etter i.v. dosering i musen var 40 mg/kg/dag.
Det var ingen merkbar virkning av behandlingen på kroppsvekt, kroppsvektsendring, eller matinntak, eller på øyeobservasjoner under den avsluttende uke i doseringsfasen.
Administrering av forbindelsen i eksempel 1 ble assosiert med litt høyere hvite blodceller og absolutt lymfocyttellinger for hunnmus som gir 40 mg/kg/dag.
Hannmus som ble gitt 40 mg/kg/dag ble ikke påvirket på samme måte, og disse små virkningene ble ikke vurdert å være negative. Klinisk kjemiske resultater var upåvirket ved administrering av forbindelsen i eksempel 1. Økninger i organvekter (nyrer hos hannmus gitt 8 mg/kg/dag og seminale vesikler hos hannmus gitt 24 eller 40 mg/kg/dag) ble vurdert tilfeldig og ikke relatert til behandlingen. Ingen store lesjoner relatert til testartikkelen ble registrert. Tre dyr (1 kontrollgruppe hunnmus, 1 hannmus med 8 mg/kg/dag og 1 hunnmus med 24 mg/kg/dag) hadde et fokalt, rødt, skorpet område på injeksjonsstedet (halen), som var resultat av stikk fra underhudsnåler. Ingen mikroskopiske lesjoner relatert til testartikkelen ble observert.
4.3: Stamcellemobilisering
a) Stamcellemobilisering i mus, forbindelse i eksempel 1:
Administrering av 5 mg/kg av forbindelsen i eksempel 1 øket CFU-GM blodcelleantall med en maksimal effekt på 120 min. og returnerte til basislinjenivåer 6 timer etter administrering (fig. 1). I samme analyse ble AMD3100 (som for tiden gjennomgår fase III kliniske utprøvinger for stamcellemobilisering) ble brukt som en sammenlignende forbindelse. I en oppfyllingsundersøkelse ble doseresponsen til eksempel 1 ved frigjøring av CFU-GM bestemt (fig. 1). Det var en klar doseresponseffekt for eksempel 1 med hensyn på frigjøring av CFU-GM hos mus med en toppnivåøkning på 5 mg/kg.

b) Stamcellemobilisering i mus, forbindelsen i eksempel 2:
Administrering av 5 mg/kg av forbindelsen i eksempel 2 øket CFU-GM blodcelleantall opp til 6 timer etter administrering med en maksimal virkning på 240 min., mens administrering av AMD3100 er assosiert med en økning i frekvens og antall av forløpere ved 30 og 60 min. sammenlignet med kontrollmus (fig. 2).

c) Stamcellemobilisering hos aper, forbindelse i eksempel 1:
Administrering av forbindelsen i eksempel 1 induserte mobilisering av CD34(+) hematopoietiske celler i cynomolgusaper. Som observert hos mus var igangsettelsen av mobiliseringen hurtig med et toppnivå etter to timer. Mobiliseringen var også forbigående og antall stamceller i perifert blod returnerte til basislinjenivå med minkende plasmanivåer av forbindelsen i eksempel 1 (fig. 3).

(e) kobling av produktet således oppnådd med et egnet N-beskyttet derivat av aminosyren som i det ønskede endeprodukt er i posisjon 14, dvs. Lys, hvor aminogruppen som er til stede i dens sidekjede likeledes er egnet beskyttet;
(f) fjerning av den N-beskyttende gruppen fra produktet således oppnådd; (g) utførelse av trinnene som hovedsakelig tilsvarer trinn (e) og (f) men ved å bruke egnede N-beskyttede derivater av aminosyrene som i det ønskede endeprodukt er i posisjoner 13 til 1, dvs. Gln, Tyr, Cys, Tyr, Arg, Dab,<D>Pro, Ala, Ser, Cys, Ala eller Tyr, His og Tyr, enhver funksjonell gruppe som kan være til stede i nevnte N-beskyttede aminosyrederivat er likeledes egnet beskyttet;
(h) dannelse av disulfid β-trådkoblingen mellom sidekjedene til Cysresiduene i posisjon 4 og 11;
(i) løsning av produktet som således er oppnådd fra den faste understøttelse; (j) syklisering av produktet avspaltet fra den faste understøttelsen;
(k) fjerning av enhver beskyttende gruppe som er til stede på funksjonelle grupper til ethvert ledd i kjeden av aminosyreresiduer; og
(l) hvis ønskelig, konvertering av produktet således oppnådd til et farmasøytisk akseptabelt salt eller konvertering av et farmasøytisk akseptabelt, eller uakseptabelt, salt som således er oppnådd til den tilsvarende frie forbindelse eller til et annet, farmasøytisk akseptabelt salt.
