Description
Foreliggende oppfinnelse angår nye 1 -aza-bicykloalkylderivater, deres anvendelse som farmasøytika, farmasøytiske preparater og kombinasjoner omfattende disse.
WO 2004/022556 beskriver 1-aza-bicykloalkylderivater som er oc7-nikotinacetylkolin-reseptoragonister.
WO 2004/016608 beskriver kinuklidinderivater som er modulatorer for nikotin- og/eller monoamin-reseptorene.
Foreliggende oppfinnelse tilveiebringer i et første aspekt en forbindelse med formel I

b) Syreaddisjonssalter kan fremstilles fra de frie baser på i og for seg kjent måte og vice versa. Alternativt kan optisk rene utgangsmaterialer benyttes. Egnede
syreaddisjonssalter for bruk i henhold til oppfinnelsen inkluderer for eksempel hydroklorid.
c) Stereoisomere blandinger, for eksempel blandinger av diastereomerer, kan separeres i de tilsvarende isomerer på i og for seg kjent måte ved hjelp av egnede
separeringsmetoder. Diastereomere blandinger kan for eksempel separeres i de individuelle diastereomerer ved hjelp av fraksjonert krystallisering, kromatografering, oppløsningsmiddelfordeling og tilsvarende prosedyrer. Denne separering kan skje enten på nivå med utgangsforbindelser eller i en forbindelse med I per se. Enantiomerer kan separeres via dannelse av diastereomere salter, for eksempel ved saltdannelse med en enantiomerren kiralsyre eller ved hjelp av kromatografi, for eksempel HPLC, ved bruk av kromatografiske substrater med kirale ligander. Alternativt kan benyttes rene utgangsmaterialer.
d) Egnede diluenter for å gjennomføre det som er beskrevet ovenfor er spesielt inerte, organiske oppløsningsmidler. Disse inkluderer særlig alifatiske, alicykliske eller
aromatiske, eventuelt halogenerte hydrokarboner som eksempel, benzin, benzen, toluen, xylen, klorbenzen, diklorbenzen, petroleumeter, heksan, cykloheksan, diklormetan, klorform, karbontetraklorid; etere, slik som dietyleter, diisopropyleter, dioksan, tetrahydrofuran eller etylenglykoldimetyleter eller etylenglykoldietyleter; ketoner, slik som aceton, butanon eller metylisobutylketon; nitriler, slik som acetonitrilpropionitril eller butyronitril; amider, slik som N,N-dimetylformamid, N,N-dimetylacetamid, N-metyl-formanilid, N-metyl-pyrrolidon eller heksametylfosfortriamid; esteres, slik som metylacetat eller etylacetat, sulfoksider, slik som dimetylsulfoksid, alkoholer, slik som metanol, etanol, n- eller i-propanol, etylenglykolmonometyleter, etylenglykolmonoetyl-eter, dietyelenglykolmonometyleter, dietylenglykolmonoetyleter. Videre kan det benyttes blandinger av diluenter. Avhengig av utgangsmaterialene,
reaksjonsbetingelsene og hjelpestoffet kan vann eller diluenter inneholdende vann være egnet. Det er også mulig samtidig å benytte et utgangsmateriale som diluent.
e) Reaksjonstemperaturer kan varieres innen et relativt vidt område. Generelt gjennomføres prosessene ved temperaturer mellom 0°C og 150°C, og særlig mellom
10°C og 120°C. Deprotoneringsreaksjoner kan varieres innen et relativt vidt område. Generelt blir prosessene gjennomført ved temperaturer mellom -150°C og +50°C, og særlig mellom -75°C og 0°C.
f) Reaksjonen gjennomføres generelt under atmosfærisk trykk. Imidlertid er det også mulig å gjennomføre prosessene ifølge oppfinnelsen under forhøyede eller reduserte
trykk, generelt mellom 0,1 og 10 bar.
g) Utgangsmaterialene benyttes generelt i omtrent ekvimolare mengder. Imidlertid er det også mulig å benytte et relativt stort overskudd av en av komponentene. Reaksjonen
gjennomføres generelt i en egnet diluent i nærvær av et reaksjonshjelpestoff og reaksjonsblandingen blir generelt omrørt ved ønsket temperature et antall timer.
h) Opparbeiding av reaksjonsblandingene i henhold til prosessene ovenfor og rensing av forbindelsene som oppnås kan gjennomføres i henhold til i og for seg kjente prosedyrer
(se f.eks. fremstillingseksemplene).
Forbindelser med formel (I) og deres farmasøytisk akseptable syreaddisjonssalter, heretter angitt som midler ifølge oppfinnelsen, viser verdifulle, farmakologiske egenskaper og er derfor nyttige som farmasøytika.
Forbindelsene ifølge oppfinnelsen og deres farmasøytisk akseptable syreaddisjonssalter, heretter angitt som forbindelser ifølge oppfinnelsen, viser verdifull, farmakologiske egenskaper når de testes in vitro og i dyr og er derfor nyttige som farmasøytika.
Således finnes forbindelsene ifølge oppfinnelsen å være kolinergiske ligander av nAChR. I tillegg viser foretrukne forbindelser ifølge oppfinnelsen selektiv oc7-nAChR aktivitet. Forbindelsene ifølge oppfinnelsen kan spesielt finnes å være agonister, partialagonister, antagonister eller allosteriske modulatorer av reseptoren.
På grunn av de farmakologiske profiler er forbindelsene ifølge oppfinnelsen ventet å være nyttige for behandling av sykdommer eller tilstander så forskjellige som CNS relaterte sykdommer, PNS relaterte sykdommer, sykdommer relatert til inflammasjon, smerte og awenningssymptomer forårsaket av misbruk av kjemiske substanser, sykdommer eller lidelser relatert til CNS inkludert generell angstforstyrrelser, kognitive forstyrrelser, lære- og hukommelsesdefiktiviteter og dysfunksjoner, Alzheimer" s sykdom, ADHD, Parkinsons sykdom, Huntington's sykdom, ALS, prioniske neurodegenerative forstyrrelser slik som Creutzfeld-Jacob sykdom og kuru sykdom, Gilles de la Tourette's syndrom, psykose, depresjon og depressive forstyrrelser, mani, manisk depresjon, schizofreni, kognitiv defektiviteter ved schizofreni, obsessive kompulsive forstyrrelser, paniske forstyrrelser, spiseforstyrrelser, narkolepsi, nocisepsjon, ATDS-demens, senildemens, mild kognitiv dysfunksjon i forbindelse med alder, autisme, dysleksi, tardiv dyskinesi, epilepsi og konvulsive forstyrrelser, post-traumatiske stress-forstyrrelser, transient anoksi, pseudodemensia, pre-menstruelt syndrom, sen lutealfasesyndrom, kronisk tetthetssyndrom og jet lag. Videre kan forbindelsene ifølge oppfinnelsen være nyttige for behandling av endokrine forstyrrelser som tyrotoksikose, feokromocytom, hypertensjon og arrytmier så vel som angina pektoris, hyperkinesi, prematur ejakulering og erektil vanskelighet. Videre kan forbindelsene ifølge oppfinnelsen være nyttige ved behandling av inflammatoriske forstyrrelser (Wang et al., Nature 2003, 421,384), forstyrrelser eller tilstander inkludert inflammatoriske hudforstyrrelser, Crohn's sykdom, inflammatoriske tarmsykdom, ulcerativ kolitt og diaré. Forbindelser ifølge oppfinnelsen kan videre være nyttige for terapi eller avvenning av symptomer forårsaket av avslutning av bruken av additive substanser som tobakk, nikotin, opioider, benzodiazepiner og alkohol. Videre kan forbindelsene ifølge oppfinnelsen være nyttige for behandling av smerte, for eksempel forårsaket av migrene, postoperativ smerte, fantomleddsmerte eller smerte assosiert med cancer. Smerten kan omfatte inflammatorisk eller neuropatisk smerte, sentralsmerte, kronisk hodepine, smerte relatert til diabetisk neuropati, postterapeutisk neuralgi og perifer nervesmerte.
Videre inkluderer degenerativ okkulærforstyrrelser som kan behandles okkulære sykdommer som direkte eller indirekte involverer degenerering av retinalceller inkludert iskemiske retinopatier generelt, anteriør iskemisk optisk neuropati, alle former for optisk neuritt, aldersrelatert makkulær regenerering (AMD), i sine tørrformer (tørr AMD) og våtformer (våt AMD), diabetisk retinopati, cystoid makkulær ødem (CME), retinal løsning, retinitt pigmentosa, Stargardt's sykdom, Best's vitelliform retinal degenerering, Leber's kongenitale amaurose og andre hereditære retinale degenerasjoner, patologisk myopia, retinopati og prematuritet, og Leber's heredititære optiske neuropati,
I et annet aspekt blir forbindelsen ifølge oppfinnelsen benyttet som diagnostiske midler og/eller PET-ligander, for eksempel for identifisering og lokalisering av nikotinreseptorer i forskjellige vev. Riktig isotopmerkede midler ifølge oppfinnelsen viser verdifulle egenskaper som histopatologiske merkemidler, billedgivende midler og/eller biomarkører, heretter kalt "markører", for selektiv merking av nAChR. Mer spesielt er midlene ifølge oppfinnelsen brukbare som markører for merking av a7 nAChR reseptorrer in vitro eller in vivo. Særlig er forbindelser ifølge oppfinnelsen som er merket riktig isotopisk brukbare som PET markører. Slike PET markører er merket med et eller flere atomer valgt fra gruppen beståo ende av 11 C, 13 N, 15 O,1<8>F.
Midlene ifølge oppfinnelsen er derfor brukbare for eksempel for å bestemme nivåene av reseptorokkupansen av et medikament som virker på nAChR, eller diagnostiske formål for sykdommer som skyldes en manglende balanse eller dysfunksjon når det gjelder nAChR, og for å styre effektiviteten av farmakoterapien for slike sykdommer.
I henhold til det ovenfor anførte beskrives et middel ifølge oppfinnelsen for bruk som markør for neuroavbildning.
Videre beskrives et preparat for å merke hjerne- og perifer nervesystemstrukturer som involverer nAChRiw vivo og in vitro omfattende et middel ifølge oppfinnelsen.
Det beskrives også en fremgangsmåte for merking av hjerne- og perifernervesystemstrukturer som involverer nAChRiw vitro eller in vivo, og omfattende å bringe hjernevevet i kontakt med et middel ifølge oppfinnelsen.
Den ovennevnte fremgangsmåten kan omfatte et ytterligere trinn som tar sikte på å bestemme hvorvidt midlet ifølge oppfinnelsen har merket målstrukturen. Nevnte ytterligere trinn kan effektures ved å observere målstrukturen ved bruk av positronemisjonstomografi (PET) eller enkel protonemisjonskomputert tomografi (SPECT), eller en hvilken som helst innretning som tillater detektering av radioaktiv stråling.
Særlig er midlene ifølge oppfinnelsen a7 nikotinisk acetylkolinreseptor (a7 nAChR) agonister.
I funksjonelle analyser viser midlene ifølge oppfinnelsen høy affinitet ved a7 nAChR som vist i de følgende tester: a) En funksjonell analyse for affinitet ved a7 nAChR gjennomføres med en rotte pituitær cellelinje som stabilt uttrykker a7 nAChR. Kort sagt ble GH3 celler som rekombinant uttrykker nAChR a7 sådd ut 72 timer før forsøket på sorte 96-brønners plater og inkubert ved 37°C i en fuktet atmosfære (5 % CO2/95 % luft). På forsøksdagen ble mediet fjernet ved å slå platene og det ble erstattet med 100 ul vekstmedium inneholdende fluoressens kalsiumsensitivt farvestoff i nærvær av 2,5 mM probenicid (Sigma). Cellene ble inkubert ved 37°C i en fuktet atmosfære (5 % C02/95 % luft) i 1 time. Platene ble slått for å fjerne overskytende Fluo-4, vasket to ganger med Hepes-bufret saltoppløsning (i mM: NaCl 130, KC1 5,4, CaCl22, MgS040,8, NaH2P040,9, glukose 25, Hepes 20, pH 7,4; HBS) og fylt igjen med 100 ul HBS inneholdende antagonister der dette var hensiktsmessig. Inkuberingen i nærvær av antagonisten var mellom 3 og 5 minutter. Platene ble så anbrakt i en billedplateleser og fluoressenssignalet notert i denne analyse og der forbindelsene ifølge oppfinnelsen vistePEC50verdier på rundt 5 til rundt 9. Partial- og potentagonister i denne teste er foretrukket.
b) For å bedømme antagonistaktiviteten for forbindelsene ifølge oppfinnelsen på den humane neuronale nAChR a4p2, ble en tilsvarende funksjonell analyse gjennomført
ved bruk av en human epitelcellelinje som stabilt uttrykker den humane a4p2 subtype (Michelmore et al., Naunyn-Schmiedeberg's Arch. Pharmacol. (2002) 366, 235). I denne analyse viste de foretrukne forbindelser ifølge oppfinnelsen selektivitet for a7 nAChR subtypene.
c) For å bedømme antagonistaktiviteten for forbindelser ifølge oppfinnelsen på den "ganglioniske subtype" (oc3p4), ble muskeltype nikotinisk reseptor (ocipiyS) og 5-HT3
reseptor, tilsvarende funksjonelle tester som beskrevet nettopp under a) gjennomført med en human epitelcellelinje som stabilt uttrykker den humane ganglioniske subtype, en cellelinje som endogent uttrykker humanmuskeltypen av nikotiniske reseptorer og en cellelinje som endogent uttrykker den murin 5-HT3reseptor (Michelmore et al., Naunyn-Schmiedeberg's Arch. Pharmacol. (2002) 366. 235). Forbindelser som viser liten eller ingen aktivitet på a3f}4 nAChR, muskelsubtypen av nikotinreseptor så vel som 5-HT3reseptoren, er spesielt foretrukket.
I musemodellen som viser sensoriske portingsdefektivitet (DBA/2-mus) beskrevet av S. Leonard et al. in Schizophrenia Bulletin 22, 431-445 (1996), induserer forbindelsene ifølge oppfinnelsen signifikant sensorisk gating ved konsentrasjoner på rundt 10 til rundt 40 (jM.
Forbindelsene ifølge oppfinnelsen kan påvises å øke oppmerksomheten i en test når det gjelder gnageroppmersomhet (Robbins, J. Neuropsychiatry Clin. Neurosci.
(2001) 13, 326-35), nemlig 5-valg seriereaksjonstidstesten (5-CSRTT). I denne test må rotter observeres en vegg med 5 hull. Når et lysglimt opptrer i et av disse må rottene respondere med en neseaktivitet i det korrekte hull innen 5 sek. For å motta en belønning form av en næringspellet, avgitt i en foringsanordning på den motsatte vegg.
Forbindelsene ifølge oppfinnelsen kan også vise lære-hukommelsesforbedrende effekter i sosial erkjennelsestesten hos mus og rotter (Ennaceur og Delacour, Behav. Brain Res.
(1988) 31,47-59).
Forbindelsene ifølge oppfinnelsen er derfor nyttige ved prevensjon og terapi (inkludert mitigrering og prevensjon) av forskjellige forstyrrelser, og særlig de som er nevnt ovenfor. Brukbarheten av oc7 nAChR agonister ved neurodegenerering er dokumenter I litteraturen, for eksempel hos Wang et al., J. Biol. Chem. 275, 5626-5632 (2000).
For behandling av de ovenfor angitte og andre forstyrrelser vil selvfølgelig den egnede dosering av en forbindelse (aktiv bestanddel) ifølge oppfinnelsen avhenge for eksempel av verten, administreringsmodus og alvoret og arten av tilstanden som skal behandles så vel som den relative potens for det spesielle middel ifølge oppfinnelsen som benyttes. For eksempel kan mengden av aktivt middel som kreves bestemmes på basis av kjente in vitro og in vivo teknikker, ved bestemmelse av hvor lenge en spesiell aktiv middelkonsentrasjon i blodplasma forblir på et akseptabelt nivå for en terapeutisk effekt Generelt indikeres tilfredsstillende resultater hos dyr til å kunne oppnås ved daglige doser fra rundt 0,01 til rundt 30,0 mg/kg p.o. Hos mennesker ligger en indikert daglig dose i området fra rundt 0,7 til rundt 1400 mg/dag p.o., for eksempel fra rundt 50 til 200 mg (70 kg personer), hensiktsmessig administrert en gang eller i oppdelte doser opptil 4 ganger pr. dag eller i en form for vedvarende frigivning. Oraldoseringsformer omfatter i henhold til dette fra rundt 1,75 eller 2,0 til rundt 700 eller 1400 mg av en forbindelse ifølge oppfinnelsen blandet med en egnet farmasøytisk akseptabel diluent eller bærer.
Farmasøytiske preparater inneholder for eksempel, fra rundt 0,1 % til rundt 99,9 %, og særlig fra rundt 20 % til rundt 60 %, av den eller de aktive bestanddeler.
Eksempler på preparater omfattende en forbindelse ifølge oppfinnelsen inkluderer for eksempel en fast dispersjon, en vandig oppløsning, for eksempel inneholdende et oppløseliggjørende middel, en mikroemulsjon og en suspensjon av for eksempel et salt eller en forbindelse med formel I eller en fri forbindelse med formel I i området fra 0,1 til 1 %, for eksempel 0,5 %. Preparatet kan være bufret til en pH-verdi i området for eksempel fra 3,5 til 9,5, særlig pH 4,5, ved hjelp av en egnet buffer.
Forbindelsene ifølge oppfinnelsen er også kommersielt nyttige som forsøkskjemikalier.
For bruk ifølge oppfinnelsen kan en forbindelse med formel I og/eller et farmasøytisk akseptabelt salt derav administreres som enkelt aktivt middel eller i kombinasjon med et eller flere aktive midler med formel I og/eller et farmasøytisk akseptabelt salt derav eller særlig andre aktive midler som vanligvis benyttes, særlig for behandling av forstyrrelser som nevnt her eller andre ytterligere forstyrrelser, på vanlig måte, for eksempel oralt, for eksempel i form av tabletter, kapsler eller som nasalspray, eller parenteralt, for eksempel i form avinjeksjonsoppløsninger eller suspensjoner. Slike andre aktive midler som benyttes i slike kombinasjoner er fortrinnsvis valgt blant gruppen bestående av benzodiazepiner, selektive serotoningjenopptaksinhibitorer (SSRFer), selektive serotonin- og norepinefrin gjenopptaksinhibitorer (SNRFer), konvensjonelle antipsykotika, atypiske antipsykotika, buspiron, karbamazepin, okscarbazepin, gabapentin og pregabalin.
En SSRI som er egnet ifølge oppfinnelsen er særlig valgt blant fluoksetin, fuvoksamin, sertralin, paroksdetin, citalopram og escitalopram. En SNRI som er egnet ifølge oppfinnelsen er særlig valgt blant venlafaksin og duloksetin. Uttrykket "benzodiazepiner" som benyttet her inkluderer, men er ikke begrenset til klonazepam, diazepam og lorazepam. Uttrykket "konvensjonelle antipsykotika" som benyttet her inkluderer, men er ikke begrenset til haloperidol, flufenazin, tiotiksen og flupentiksol. Uttrykket "atypiske antipsykotika" som benyttet her gjelder klozaril, risperidon, olanzapin, quetiapin, ziprasidon og aripiprazol.
Buspiron kan administreres i fri form eller som et salt, for eksempel som hydroklorid, for eksempel i form som markedsført, for eksempel under varemerket Buspar™ eller Bespar™. Det kan fremstilles og administreres, for eksempel, som beskrevet i US 3,717,634. Fluoksetin kan administreres, for eksempel, i form avhydroklorid som markedsført, for eksempel under varemerket Prozac™. Det kan fremstilles og administreres, for eksempel, som beskrevet i CA 2002182. Paroxetine ((3S,4R)-3-[(l,3-benzodioksol-5-yloksy)metyl]-4-(4-fluorofenyl)piperidin) kan administreres, for eksempel, i form som markedsført, for eksempel under varemerket Paxil™. Når det gjelder en kombinasjon kan de farmasøytiske preparater for separat administrering av kombinasjonspartnere og/eller de for administrering av en fast kombinasjon, dvs. et enkelt, galenisk preparat omfattende minst to kombinasjonspartnere, i henhold til oppfinnelsen, fremstilles på i og for seg kjent måte og er således egnet for enteral som oral eller rektal, eller parenteral administrering til pattedyr inkludert mennesker, omfattende en terapeutisk effektiv mengde av minst en farmakologisk aktiv kombinasjonspartner alene eller i kombinasjon med en eller flere farmakologisk akseptable bærere, særlig egnet for enteral eller parenteral applikering. Når kombinasjonspartnerne som benyttes anvendes i form som markedført som ankeltmedikamenter, kan deres doserings- og administreringsmodus skje i henhold til den informasjon som tilveiebringes i emballasjeinformasjonen for det respektive markedsførte medikament for å gi en fordelaktig effekt som beskrevet her, hvis ikke annet er sagt.
Farmasøytiske preparater for kombinasjonsterapi for enteral eller parenteral administrering er, for eksempel, de i enhetsdoseformer, slik som sukkerbelagte tabletter,
tabletter, kapsler eller suppositorier, eller også ampuller. Hvis ikke annet er sagt fremstilles disse på i og for seg kjent måte, for eksempel ved hjelp av konvensjonell blanding, granulering, sukkerbelegning, oppløsning eller lyofilisering. Det vil erkjennes at enhetsinnholdet for en kombinasjonspartner inneholdt i den individuelle dose av hver doseringsform ikke i seg selv må utgjøre noen effektiv mengde fordi den nødvendige, effektive mengde også i stedet kan oppnås ved en enkelt doseringsenhet ved administrering av to eller flere doseringsenheter.
Særlig kan en terapeutisk effektiv mengde av hver av kombinasjonspartnerne administreres simultant eller sekvensielt eller i en hvilken som helst rekkefølge, og komponentene kan administreres separat (f. eks. sekvensielt etter faste eller variable tidsrom), eller som en fast kombinasjon. For eksempel kan fremgangsmåten for behandling (inkludert mitigering) av en forstyrrelse ifølge oppfinnelsen omfatte (i) administrering av kombinasjonspartene (a) (en forbindelse ifølge oppfinnelsen) i fri eller farmasøytisk akseptabel saltform, og (ii) administrering av en kombinasjonspartner (b) (for eksempel en annen forbindelse ifølge oppfinnelsen eller en aktiv bestanddel med en annen formel) i fri eller farmasøytisk akseptabel saltform, samtidig eller sekvensielt i en hvilken som helst rekkefølge, i felles terapeutisk effektive mengder, fortrinnsvis i synergistisk effektive mengder, f.eks. i daglige doser som tilsvarer de her beskrevne metoder. De individuelle kombinasjonspartnere kan administreres separat ved forskjellige tidsrom under terapiforløpet eller samtidig i oppdelte eller enkeltkombinasjonsformer. Videre omfatter uttrykket "administrering" også bruken av en pro-drug av en kombinasjonspartner som konverteres in vivo til kombinasjonspartneren per se. Oppfinnelsen skal derfor forstås til å omfatte alle slike skiver av samtidig og/eller alternerende behandling og uttrykket "administering" skal tolkes tilsvarende.
Den effektive dose av kombinasjonspartnere som benyttes kan for eksempel variere avhengig av den spesielle forbindelse eller de farmasøytiske preparater som benyttes, administreringsmodus, forstyrrelse som behandles og eller alvoret av denne. Således velges doseringsregimet i henhold til et antall faktorer inkludert administreringsvei, metabolismen hos og renal og hepatisk funksjon hos pasienten. En lege, veterinær eller tilsvarende med vanlige fagkunnskaper kan bestemme eller forskrive en effektiv mengde av enkeltaktive bestanddeler som kreves for å forhindre, mitigatere, motvirke eller stanse forstyrrelsen. Optimal nøyaktighet for å oppnå konsentrasjon av aktive bestanddeler innen området som gir effektivitet uten toksisitet krever et regime basert på kinetikken hos de aktive bestanddeler når det gjelder tilgjengelighet for målsetene.
I henhold til det ovenfor anførte tilveiebringer oppfinnelsen også:
(1) En forbindelse med formel I, og/eller et salt derav, for bruk ved terapeutisk behandling av pattedyr, særlig mennesker; særlig for bruk som en a-7 reseptoragonist, for eksempel for bruk ved behandling av (inkludert mitigrering) en hvilken som helst av en eller flere forstyrrelser, særlig en eller flere forstyrrelser av de spesielle som er angitt ovenfor og nedenfor; (2) Et farmasøytisk preparat omfattende en forbindelse med formel I, og/eller et farmasøytisk akseptabelt salt derav, som aktiv bestanddel sammen med en farmasøytisk akseptabel diluent eller bærer.
(2') Et farmasøytisk preparat for terapi eller prevensjon av en forstyrrelse ved terapi der oc-7 reseptoraktivering spiller en rolle og er involvert og/eller der a -7 reseptor aktivt er involvert, særlig en hvilken som helst eller flere av de forstyrrelser som er nevnt ovenfor eller nedenfor, omfattende en forbindelse med formel I, og/eller et farmasøytisk akseptabelt salt derav, og en farmasøytisk akseptabel diluent eller bærer.
(
(4) Anvendelse av en forbindelse med formel I, og/eller et farmasøytisk akseptabelt salt derav, for fremstilling av et medikament for terapi eller prevensjon av en sykdom eller tilstand der behandling med a-7 reseptoraktivering spiller en rolle, eller er involvert og/eller der a-7 reseptor aktivt er involvert, særlig en eller flere av forstyrrelser som nevnt ovenfor. (6) En kombinasjon omfattende en terapeutisk effektiv mengde av en a-7 agonist med formel I, og/eller et farmasøytisk akseptabelt salt derav, og en andre, farmasøytisk aktiv forbindelse og/eller et farmasøytisk akseptabelt salt derav, idet den andre farmasøytisk aktive forbindelse særlig er for bruk eller anvendelse ved terapi av en hvilken som helst av de spesielle forstyrrelser som er gitt ovenfor. (7) Et produkt ifølge det som er beskrevet ovenfor og som karakteriseres ved at (+)-trans-2-metyl-l-aza-bicyklo[2.2.2]oktan-3-ol benyttes som utgangsmateriale og Y betyr pyrimidinyl eller pyridazinyl.
(7") Et produkt oppnådd ved prosessen ovenfor,karakterisert vedat (-)-trans-2-metyl-l-aza-bicyklo[2.2.2]oktan-3-ol benyttes som utgangsmateriale og Y betyr pyridinyl.

Temperaturene måles i grader celsius. Hvis ikke annet er sagt gjennomføres reaksjonene ved romtemperatur. Strukturen for sluttprodukter, mellomprodukter og utgangsstoffer bekreftes ved analytiske standardmetoder, for eksempel mikroanalyse og spektroskopisk karakterisering (for eksempel MS, IR, NMR).
Referanseeksempel: Fremstilling av Rac.- trans- 3-[ 6-( 2- Fluoro- 4- metyl- fenyl)-pyridazin- 3- yloksyl- 2- metyl- 1- aza- bicyklo [ 2. 2. 21 oktan
En oppløsning av rac.-trans-2-metyl-l-aza-bicyklo[2.2.2]oktan-3-ol (1,3 mmol) i DMF (5 ml) behandles med natriumhydrid (60% i mineralolje; 1,3 mmol). Etter 1 time ved romtemperatur, blir en oppløsning av 3-klor-6-(2-fluoro-4-metyl-fenyl)-pyridazin (1,5 mmol) i DMF (30 ml) tilsatt, og reaksjonsblandingen varmet opp til 50°C i 16 timer. Etter avkjøling til romtemperatur blir DMF oppløsningen kvensjet med en 10 % NaCl oppløsning, ekstrahert med metylendiklorid (2X15 ml), fulgt av natriumkloridoppløsning (20 ml). Det organiske sjikt tørkes over vannfri magnesiumsulfat, filtreres og fordampes til tørr tilstand, hvoretter restoljen renses ved silikagelkolonnekromatografi (eluent: CH2Cl2:CH30H:NH3; 95:5:0,) for å gi rac.-trans-3-[6-(2-Fluoro-4-metyl-fenyl)-pyridazin-3-yloksy]-2-metyl-l-aza-bicyklo[2.2.2]oktan som et farveløst faststoff.
