Description
Foreliggende oppfinnelse angår en forbedret fremgangsmåte ved fremstilling av fosfodiesterforbindelser, hvilken fremgangsmåte omfatter de trekk som er nevnt i krav 1. Spesielt angår oppfinnelsen en forbedret fremgangsmåte for å fremstille fosfodiesterforbindelser som er anvendelige som kontrastmidler for diagnostisk avbildning og mer spesielt for å fremstille dietylentriamin-pentaeddiksyre ("DTPA")-forbindelser omfattende fosfodiestere.
Bakgrunn for oppfinnelsen.
Mange viktige biologiske substanser innbefattende fosfoli-pider, oligonukleotider, deoksynukleotider, nukleotider og nukleosider, eksisterer som symmetriske og usymmetriske fosfodiestere. Anvendeligheten av slike fosfodiesterforbindelser i medisinsk bruk er velkjent. Se for eksempel Desseaux et al., "Synthesis of Phosphodiester and Triester Derivatives of AZT with Tethered N-Methyl Piperazine and N,N,N' trimetyletylendiamin," Bioorg. & Med. Chem. Letters, vol. 3, nr. 8, s. 1547-50 (1993); PCT-publikasjon nr. WO 96/27379. Nylig beskriver PCT-publikasjon nr. WO 96/23526 innbefattet heri pr. referanse fosfodiesterforbindelser som er anvendelige som kontrastmidler for diagnostisk avbildning .
Et antall metoder for å danne fosfodiesterforbindelser ba-sert på P(III) kjemi er kjent. Generelt spiller fosforylering en viktig rolle i syntesen av fosfodiesterforbindelser. Imidlertid lider de kjente syntetiske fosfodiester-metoder alle av et antall problemer innbefattende hvordan fosforylering blir oppnådd.

Slike fosforamidittmetoder lider imidlertid av det faktum at fosforamidittene typisk er ustabile forbindelser (både kjemisk og kinetisk) og kan ved opprenskning ved destilla-sjon antenne eller forårsake en eksplosjon. Videre er fosforamidittmetoder generelt ikke egnet for å fremstille fosfodiesterforbindelser på en kommersiell basis. Dette er fordi fosforamiditt-utgangsmaterialene er meget dyre og er ikke lett tilgjengelige, og fordi metoder som bruker fos-foramidittforbindelser har en tendens til å involvere ytterligere prosesstrinn (for eksempel ytterligere trinn ved spaltning av beskyttende grupper etter fosforylering) så vel som flere isolerings- og/eller opprenskningstrinn av intermediatene.
Metoder som involverer bruken av fosfodikloridater som fos-foryleringsmiddel lider av lignende problemer. En annen metode brukt for å danne fosfodiesterforbindelser, involverer bruken av PCI3 for å utvikle hydrogenfosfonatin-termediater. Se for eksempel Lindh et al., "A General Methos for the Synthesis of Glycerophospholipids and Their Analogues via H-Phosphonate Intermediates," J. Org. Chem., vol. 54, s. 1338-42 (1989); Garcia et al., "Synthesis of New Ether Glycerophospholipids Structurally Related to Mo-dulator," Tetrahedron, vol. 47, nr. 48, s. 10023-34 (1991); Garigapati et al., "Synthesis of Short Chain Phosphatidyli-nositols," Tetrahedron Letters, vol. 34, nr. 5, s. 769-72
(1993). Denne metode krever imidlertid bruk av et koplingsreagens som enten kan bli kjøpt eller uavhengig syntetisert og gjør således slike metoder dyre og mer sam-mensatte. I tillegg er flere isolerings- og opprenskningstrinn av intermediatene nødvendig, ofte med arbeidskrevende tørkebetingelser for H-fosfonatintermediatet.
Fra Y. Lu et al.: synthetic Communications 23 (4), 1943-1946 (1993) er det kjent en syntese av fosfodiesterforbindelser ved å benytte kloro-N,N-diisopropylaminoalkyloksy-fosfiner.
Følgelig finnes det et behov for en sikker, effektiv og billig prosess for fremstilling med høyt utbytte av fosfodiesterforbindelser med potensiale å ha et stort antall substituenter som ikke krever enten bruken av en beskyttende gruppe eller et koblingsmiddel. Spesielt eksisterer et behov for en prosess som kan bli utført i et reaksjonskammer og som ikke krever mange isolasjons- og opprenskningstrinn på grunn av dannelsen av flere intermediater.
Oppsummering av oppfinnelsen.
Foreliggende oppfinnelse angår en sikrere, mer effektiv og mindre kostbar prosess for å fremstille fosfodiesterforbindelser, og mer spesielt fosfodiestere som har formel
hvor de enkelte substituenter har den betydning som er angitt i krav 1, og hvor fremgangsmåten omfatter trinnene a)
- d) som er angitt i krav 1.
I samsvar med fremgangsmåten generelt utføres trinnene å: (a) koble PCI3 med en alkohol for å danne et substituert diklorfosfin; (b) kobling av nevnte diklorfosfin med en aminbase for å danne en bis(amino)fosfinoforbindelse; (c) kobling av nevnte bis(amino)fosfinoforbindelse med en andre alkohol som kan være lik eller forskjellig fra alkoholen brukt i trinn (a) for å danne en disubstituert (amino)fosfinoforbindelse; (d) å omsette nevnte (amino)fosfinoforbindelse med vann og et oksidasjonsmiddel for å danne den ønskede fosfodiesterforbindelse.
Fremgangsmåten ifølge foreliggende oppfinnelse unngår bruken av ustabile fosforyleringsmidler så vel som behovet for å bruke en beskyttende gruppe eller et koblingsmiddel. Således unngår foreliggende fremgangsmåte unødvendige prosesstrinn så som deblokkering og koblingsreagenssyntese. I en foretrukket utførelsesform av foreliggende oppfinnelse finner den syntetiske fosfodiesterprosess sted i et reaksjonskammer, noe som unngår behovet for flere isolasjons-og/eller opprenskningstrinn.
Detaljert beskrivelse av oppfinnelsen.
For at oppfinnelsen heri beskrevet skal bli mer fullstendig forstått er den følgende detaljerte beskrivelse angitt.
Foreliggende oppfinnelse fremskaffer en forbedret fremgangsmåte for å fremstille fosfodiesterforbindelser av ge-nerell formel:
hvor R og R<1> kan være like eller forskjellige, og er valgt fra gruppen omfattende lineære, forgrenede eller cykliske alifatiske, aryl, heterocykliske, peptidiske, peptoide, de-oksyribo- eller ribo-nukleotide eller nukleoside, eller cykliske eller acykliske organiske gelateringsmiddelgrupper hvorav alle eventuelt kan være substituert med en eller flere nitrogen, oksygen, svovel, halogen, alifatiske, ami-der, ester, sulfonamider, aryl, acyl, sulfonater, fosfater, hydroksyl eller organometalliske substituenter, hvor fremgangsmåten omfatter trinnene å a) omsette en alkohol ROH med PCI3 i nærvær av et oppløs-ningsmiddel for å danne en diklorfosfinforbindelse av formel :
b) kople diklorfosfinforbindelsen dannet i trinn (a) med en aminbase i nærvær av et oppløsningsmiddel for å danne en
bis(amino)fosfinforbindelse som har formel:
c) kople bis(amino)fosfinforbindelsen dannet i trinn (b) med en andre alkohol R1 OE i nærvær av et oppløsningsmiddel hvor den andre alkohol kan være lik eller forskjellig fra den i trinn (a) for å danne en (amino)fosfinoforbindelse av formel: d) utsette (amino)fosfinoforbindelsen dannet i trinn c) for hydrolyse og oksidasjon.
I et foretrukket aspekt av oppfinnelsen blir alle syntetiske trinn utført i et reaksjonskammer, noe som unngår behovet for flere isolasjons- og/eller opprenskningstrinn. Foreliggende oppfinnelse viser en effektiv prosess med høyt utbytte for å fremstille fosfodiesterforbindelser som ikke avhenger av dyre eller ustabile utgangsmaterialer og ikke krever bruken av enten beskyttende gruppe eller koblings-midler. Videre er nevnte fremgangsmåte effektiv til fremstilling av fosfodiesterbindinger mellom en mengde substituenter.
Prosesskjerna.
I samsvar med foreliggende oppfinnelse blir en alkohol ROH, hvor R har samme betydning som angitt ovenfor omsatt med PCI3, fortrinnsvis ved et molart forhold på 1:1 for å danne et diklorfosfin reaksjonsprodukt (I):
Denne reaksjonen finner sted i nærvær av et eterisk eller hydrokarbon oppløsningsmiddel og blir utført ved en temperatur på fra omkring -50°C til omkring 15°C, fortrinnsvis fra omkring -10°C til omkring -5°C over en periode fra omkring 30 minutter til omkring 3 timer, fortrinnsvis fra omkring 1 til omkring 1,5 timer. Oppløsningsmidlet kan være ethvert eterisk eller hydrokarbon oppløsningsmiddel, og kan fortrinnsvis være valgt fra gruppen omfattende heptaner, metyl-t-butyletere, dioksaner, tetrahydrofuraner, dietyletere og etylenglykoldialkyletere. Mer foretrukket er opp-løsningsmidlet tetrahydrofuran.
Diklorfosfinet (I) blir så omsatt med fra omkring 5 til omkring 6 ekvivalentdeler av en aminbase for å danne et bis(amino)fosfino reaksjonsprodukt(II)
Denne reaksjonen finner sted i nærvær av et eterisk eller hydrokarbonoppløsningsmiddel som beskrevet ovenfor, og blir utført ved en temperatur på fra omkring -50°C til omkring 15°C, fortrinnsvis fra omkring -10°C til omkring -5°C, over en periode fra omkring 30 minutter til omkring 3 timer, fortrinnsvis fra omkring 15 til omkring 30 minutter. Basen som benyttes for å danne reaksjonsprodukt (II) kan være enhver aminbase, fortrinnsvis en base som har en pKa-verdi på fra omkring 5 til 11, og mer foretrukket valgt fra gruppen omfattende imidazol, 2,4-dimetylimidazol, lH-tetrazol, dia-lakylaminer (metyl, etyl, butyl), pyridin, piperazin, piperidin, pyrrol, 1H-1,2,4-triazol og 1,2,4-triazol. I en mer foretrukket utførelsesform er basen imidazol.
Bis(amino)fosfinoforbindelsen (II) blir så omsatt med fra omkring 0,75 til omkring 1,0 ekvivalentdeler av en andre alkohol I^OH, hvor R<1> har samme betydning som angitt ovenfor, for å danne et (amino)fosfino reaksjonsprodukt (III):
Denne reaksjonen finner sted i nærvær av et eterisk eller hydrokarbonoppløsningsmiddel og utføres ved en temperatur på fra omkring -50°C til omkring 15°C, fortrinnsvis fra omkring -10°C til omkring -5°C over en periode fra omkring 30 minutter til omkring 3 timer, fortrinnsvis fra omkring 1,0 til omkring 1,5 timer. Oppløsningsmidlet kan være ethvert eterisk eller hydrokarbonoppløsningsmiddel og kan foretrukket være valgt fra gruppen omfattende heptaner, metyl-t-butyletere, dioksaner, tetrahydrofuraner, 1,3-dioksolaner, diglymer, dietyletere, dialkyletere, og etylenglykoldialkyletere. Mer foretrukket er oppløsningsmidlet tetrahydrofuran.
Endelig blir (amino)fosfinoforbindelsen (III) omsatt med omkring en ekvivalentdel surt vann, fortrinnsvis ved en pH på omkring 2,5 til omkring 5, og omkring 1 eller mer ekvivalentdeler av et oksidasjonsmiddel for å danne den ønskede fosfodiesterforbindelse (IV):
Oksidasjonsmidlet kan være enhver peroksidtype oksidasjonsmiddel og er fortrinnsvis valgt fra gruppen omfattende pe-riodater. Mer foretrukket er oksidasjonsmidlet natriumperiodat.
Den ovennevnte hydrolyse og oksidasjon utføres i en oppløs-ningsblanding ved en temperatur fra omkring -15°C til omkring 25°C, fortrinnsvis fra omkring 0°C til omkring 2°C, over en periode fra omkring 10 til omkring 24 timer, fortrinnsvis fra omkring 10 til omkring 15 timer. Oppløs-ningsmiddelblandingen omfatter enhver kombinasjon av opp-løsningsmidler valgt fra gruppen omfattende eteriske eller hydrokarbonoppløsningsmidler. Fortrinnsvis omfatter opp-løsningsmiddelblandingen tetrahydrofuran, heptan og toluen i et volumforhold på 10:10:1.
Anvendelse av prosessproduktene.
Det har blitt funnet at den ovennevnte prosess er spesielt anvendelig ved fremstilling av kontrastmidler for diagnostisk avbildning. Eksempler på fosfodiesterkontrastmidler som kan bli fremstilt ved denne forbedrede fremgangsmåte innbefatter forbindelsene vist nedenfor så vel som andre beskrevet i PCT publikasjon nr. WO 96/23526. I slike tilfeller er det påtenkt at minst en av de to alko-holer (ROH, R^H) som definert heri ytterligere omfatter en cyklisk eller acyklisk organisk chelaterende ligand med enhver sensitiv funksjonell gruppe (for eksempel karboksyla-ter) på et slikt chelat beskyttet med passende grupper (for eksempel t-butylgrupper). Egnede chelaterende ligander er velkjent innen faget. For eksempel hvor fosfodiesterfor-bindelsen skal bli brukt som kontrastmiddel for magnetisk resonansavbildning innbefatter foretrukne chelaterende ligander Fjerning av enhver beskyttende gruppe på chelatet så vel som kompleksbindingen av chelatet med det ønskede metall kan bli utført etter utføring av den fosfodiester syntetiske prosess i følgende oppfinnelse ved metoder velkjent innen faget. Se for eksempel Grote et al., "Stereocon-trolled synthesis of DTPA Analogues Branged in the Ethylene Unit", J. Org. Chem. 60 :6987-97 (1995); Kang et al., "Synthesis, Characterization, and Crystal Structure of the Gadolinium (III) Chelate of (IR,4R,7R)- a, a', a''-Trimethyl-1,4,7,10-tetraazacyclododecane-l,4,7-triacetic Acid(D03MA), " Inorg. Chem.., 32:2912-18 (1993) samt referanser angitt deri.
Det er også påtenkt at for slike fosfodiesterkontrastmidler kan alkoholen (ROH eller R^H) omfatte en enhet utformet
for å lette lokalisering av det resulterende middel til ve-vet, cellen, proteinet, reseptoren eller området som ønskes å bli avbildet. Eksempler på slike enheter innbefatter li-pofile eller amfifile substanser, reseptorligander, anti-stoff eller antistoffragmenter, peptider eller andre bimo-lekyler som er kjent for å konsentrere i den spesielle biologiske komponent som ønskes å bli avbildet.
For at foreliggende oppfinnelse skal bli bedre forstått er det følgende eksempel gitt. Dette eksempel er for illu-strasjonsformål alene og er ikke ment å begrense omfanget av foreliggende oppfinnelse på noen måte.
Eksempel.
Fremstilling av [(4,4-difenylcykloheksyl)fosfooksyme-tyl]dietylen-triaminpenta-eddiksyre som vist nedenfor i skjema I: I et enkelt reaksjonskammer som inneholdt en oppløsning av fosfortriklorid (13,2 mL, 0,15 mol) i tetrahydrofuran (202 ml) ble det tilsatt en oppløsning av 4,4-difenyl-cyklohek-sanol(I) (38, 34 g, 0,152 mol) i tetrahydrofuran (243 ml) under omrøring og mens det opprettholdes en innvendig temperatur på -6,2°C til -5,3°C i 1,5 timer. Blandingen ble så omrørt i ytterligere 34 minutter, noe som ga et diklorfosfin reaksjonsprodukt (2) ved et <31>P NMR kjemisk skift på 174,28 ppm.
Til denne oppløsning ble det tilsatt imidazol (51,34 g, 0,753 mol) i tetrahydrofuran (243 ml) under omrøring og
mens det ble opprettholdt en innvendig temperatur på -7,8°C til -3,6°C i 37 minutter. Den resulterende blanding ble så omrørt i ytterligere 20 minutter, noe som ga en oppløsning av et bis(amino)fosfino reaksjonsprodukt (3) med et <31>P NMR kjemisk skift på 106,36 ppm.
Til denne blanding ble det tilsatt en oppløsning bestående av 2-(R)-hydroksymetyldietylentriamin pentaeddiksyre, penta-t-butylester( 4)(160,0 g, 0,128 mol, renhet: 56,32 vekt%) i heptan (114 ml) under omrøring og ved å opprett-holde en innvendig temperatur på -6,8°C til -4,8°C i 1 time og 6 minutter. Denne blanding ble så omrørt i ytterligere 23 minutter, noe som ga en oppløsning på ( 5) som har et 31P NMR kjemisk skift på 123,8 ppm.
Til slutt ble vann (202 ml) tilsatt over en periode på omkring 1 minutt mens det ble opprettholdt en innvendig temperatur på -6,5°C til 6,5°C. Blandingen ble omrørt i 5 minutter etterfulgt av tilsetning av heptan (620 ml), toluen (70 ml) og 5N vandig saltsyre (202 ml) over 5 minutter mens det ble opprettholdt en innvendig temperatur på 1,0°C til
12,1°C. Natriumperiodat (22,6 g, 0,106 mol) ble så tilsatt over en periode på 3 minutter mens det ble opprettholdt en innvendig temperatur på 10,5°C. Reaksjonsblandingen ble oppvarmet til romtemperatur over 35 minutter og omrørt ytterligere 2,5 timer, noe som ga en oppløsning ( 6) med et
<31>P NMR kjemisk skift på 4,27 ppm. Lagene ble atskilt og det organiske lag ble vasket med 10% vandig natrium tiosul-fat (2 x 809 mL).
Til det ovennevnte organiske lag ble det tilsatt tetraokty-lammoniumbromid (8,21 g, 0,015 mol). Konsentrert saltsyre (11,51 M, 405 mL) ble så tilsatt over en periode på 22 minutter mens det ble opprettholdt en innvendig temperatur på 22,8°C til 25,0°C. Blandingen ble omrørt i 16,0 timer, noe som ga en forbindelse ( 7) med et <31>P NMR kjemisk skift på 7,78 ppm. Lagene ble atskilt og det organiske lag kastet.
Til det ovennevnte lag ble det tilsatt 8M vandig natrium-hydroksid (630 mL) inntil en pH på 6,56 ble målt. Oppløs-ningen ble konsentrert under redusert trykk (50°C til 55°C, vakuum 85 mm Hg) inntil 400 mL oppløsningsmiddel ble samlet opp (omtrent 1 time). Oppløsningen ble avkjølt til romtemperatur og amberlite XAD-4 resin (92,0 g) ble tilsatt. Suspensjonen ble omrørt i 50 minutter ved romtemperatur og filtrert for å gi en lyst gul vandig oppløsning (1,1 L).
Den ovennevnte oppløsning ble satt på en C-18 reversfase silikagel (271 g, pakket våt i metanol og så vasket med 800 mL metanol, 800 mL metanol/vann, 1:1 og 800 mL vann) og eluert med vann. Den første 1,0 L av oppsamlet eluat ble kastet og de neste 1,3 L som ble samlet opp ble spart. Til den oppsparte oppløsning ble det tilsatt 6N vandig saltsyre (60 mL til an pH = 2,15) og 3N vandig saltsyre (30 mL til en pH = 1,63). Oppslemmingen ble omrørt i 1,25 timer og filtrert. Faststoffet ble vasket med pH 1,67 vandig opp-løsning (500 mL) og tørket (48-50°C, 4-6 mm Hg) til en kon-stant vekt (18,0 timer) for å gi en off-white fast forbindelse med formel:
(65,5 g, utbytte: 68,89 renhet:99,45 vekt%, 98,95% pr. are-al, 3,02% vann og 9,81 chelaterbart materiale.
Claims (16)
1. Fremgangsmåte ved fremstilling av fosfodiesterforbindelser av formel:
hvor R og R' kan være like eller forskjellige og er valgt fra gruppen omfattende lineære, forgrenede, eller cykliske alifatiske, aryl, heterocykliske, peptidiske, peptoide, de-oksyribo- eller ribo-nukleotide eller nukleoside, eller cykliske eller acykliske organsike chelateringsmiddelgrup-per, alle eventuelt substituert med en eller flere nitrogen, oksygen, svovel, halogen, alifatiske, amid, ester, sulfonamid, acyl, sulfonat, fosfat, hydroksyl eller organometalliske substituenter hvor fremgangsmåten utføres i et reaksjonskammer og omfatter trinnene å: a) omsette en alkohol ROH med PCI3 i nærvær av et oppløs-ningsmiddel for å danne en diklorfosfinforbindelse av formel : b) kople diklorfosfinforbindelsen dannet i trinn (a) med en aminbase i nærvær av et oppløsningsmiddel for å danne en bis(amino)fosfinforbindelse som har formel: c) kople bis(amino)fosfinforbindelsen dannet i trinn (b) med en andre alkohol R1 OE i nærvær av et oppløsningsmiddel hvor den andre alkohol kan være lik eller forskjellig fra den i trinn (a) for å danne en (amino)fosfinoforbindelse av formel: d) utsette (amino)fosfinoforbindelsen dannet i trinn c) for hydrolyse og oksidasjon.
2. Fremgangsmåte ifølge krav 1 hvor alkoksydiklorfosfin-forbindelsen dannet i trinn (a) omsettes med fra omkring 5 til omkring 6 ekvivalentdeler av aminbasen.
3. Fremgangsmåte ifølge ethvert av kravene 1 eller 2 hvor aminbasen har en pKa-verdi på fra omkring 5,0 til omkring 11,0.
4. Fremgangsmåte ifølge krav 3, hvor basen valgt fra gruppen omfattende imidazol, 2,4-dimetylimidazol, lH-tetrazol, dialkylaminer(metyl, etyl, butyl), pyridin, piperazin, piperidin, pyrrol, 1H,1,2,3-triazol og 1,2,4-triazol.
5. Fremgangsmåte ifølge krav 4 hvor basen er imidazol.
6. Fremgangsmåte ifølge krav 1 hvor omkring en ekvivalentdel ROH omsettes med omkring 1 ekvivalentdel PCI3.
7. Fremgangsmåte ifølge krav 1 hvor oppløsningsmidlet benyttet i trinn (a), (b) og (c) kan være likt eller forskjellig og er valgt fra gruppen omfattende eteriske og hydrokarbonholdige oppløsningsmidler.
8. Fremgangsmåte ifølge krav 7 hvor oppløsningsmidlet er valgt fra gruppen omfattende heptaner, metyl-t-butyletere, dioksaner, tetrahydrofuraner, 1,3-dioksolan, diglymmer, dietyletere, dialkyletere og etylenglykoldialkyletere.
9. Fremgangsmåte ifølge krav 8 hvor oppløsningsmidlet er tetrahydrofuran.
10. Fremgangsmåte ifølge krav 1 hvor alkoksy(amino)fos-finoforbindelsen dannet i trinn (b) blir koplet med omkring en ekvivalentdel av I^OH.
11. Fremgangsmåte ifølge krav 1, hvor hydrolyse og oksi-dering av dialkoksy(amino)fosfinoforbindelsen dannet i trinn (c) utføres med vann og et oksidasjonsmiddel i et oppløsningsmiddel i et temperaturområde fra omkring -15°C til omkring 25°C over en periode på 10 til 24 timer.
12. Fremgangsmåte ifølge krav 11 hvor oksidasjonsmidlet omfatter natriumperiodat.
13. Fremgangsmåte ifølge krav 11 hvor oppløsningsmidlet omfatter en blanding av tetrahydrofuran, heptan og toluen.
14. Fremgangsmåte ved fremstilling av fosfodiesterforbindelser ifølge krav 1 omfattende trinnene å: a) omsette en 4,4-difenylcykloheksanolforbindelse med PCI3 for å danne 4, 4-difenylcykloheksyloksy-diklorfosfin av formel: b) kople 4,4-difenylcykloheksyloksy-diklorfosfinforbindelsen dannet i trinn (a) med en aminbase for å danne 4,4-difenylcykloheksyloksy-diaminofosfin av formel: c) kople 4,4-difenylcykloheksyloksy-diaminofosfinforbind-elsen dannet i trinn (b) med hydroksymetyl-DTPA penta tert-butylester for å danne 4,4-difenylcykloheksyloksy(hydroksymetyl-DTPA-oksy, penta-tert-butylester)amino-fosfino av formel: d) hydrolyse og oksidasjon av 4,4-difenylcykloheksyloksy-(hydroksymetyl-DTPA oksy, penta tertbutylester)aminofos-finoforbindelsen dannet i trinn c) med fortynnet HC1 og et oksideringsmiddel for å danne [(4,4-difenylcyklohek-syl)fosfonooksymetyl]dietylentriamin, penta t-butylester av formel:
15.
