Description
Foreliggende oppfinnelse angår pyrazolo[1,5-b]pyirdazinderivater, fremgangsmåte for fremstilling og anvendelse derav, samt farmasøytiske preparater inneholdende dem.
Enzymet cyklooksygenase (COX) er nylig blitt oppdaget å eksistere i to isoformer, COX-1 og COX-2. COX-1 korresponderer med det opprinnelige identifiserte konstitu-tive enzymet, mens COX-2 er raskt og lett induserbar av flere midler omfattende mitogener, endotoxin, hormoner, cytokiner og vekstfaktorer. Prosta-glandiner dannet av virkningen av COX har både fysiologiske og patologiske roller. Det er generelt antatt at COX-1 er ansvarlig for viktige fysiologiske funksjoner slik som opprettholdelse av integriteten og blodstrømningen i nyrene. I motsetning til den induserbare formen,
er COX-2 antatt å våre ansvarlig for de patologiske effekter av prostaglandiner hvor rask induksjon av enzymet forekommer i respons til slike midler som inflammatoriske midler, hormoner, vekstfaktorer og cytokiner. En selektiv inhibitor av COX-2 vil derfor ha anti-inflammatoriske, anti-pyretiske og analgesiske egenskaper, uten potensielle bivirkninger assosiert med hemning av COX-1. Vi har nå funnet ny gruppe med forbindelser som er både kraftige og selektive inhibitorer av COX-2.
Oppfinnelsen tilveiebringer følgelig forbindelsene med formel (I)
og farmasøytisk godtagbare derivater derav hvor:
R° er halogen, C^alkoksy eller Ci^alkoksy substituert med én eller flere fluoratomer;
R<1> og R<2> er uavhengig valgt fra H, Ci^alkyl substituert med én eller flere fluoratomer, Ci.6alkylsulfonyl eller Ci.6alkoksy substituert med én eller flere fluoratomer; og
R3 er C^alkyl eller NH2.
Med farmasøytisk godtagbart derivat menes et hvilket som helst farmasøytisk godtagbart salt, solvat eller ester, eller salt eller solvat av slik ester, av forbindelsene med formel (I), eller hvilke som helst andre forbindelser som ved administrering til mottageren kan tilveiebringe (direkte eller indirekte) en forbindelse med formel (I) eller en aktiv metabolitt eller rest derav.
Det vil forstås at, for farmasøytisk anvendelse, vil salter referert til ovenfor være fysiologiske godtagbare salter, men andre salter kan finne anvendelse, for eksempel ved fremstilling av forbindelser med formel (I) og fysiologisk godtagbare salter derav.
Egnete farmasøytisk godtagbare salter av forbindelsene med formel (I) omfatter syreaddisjonssalter dannet med uorganiske eller organiske syrer, fortrinnsvis uorganiske syrer, f.eks. hydroklorider, hydrobromider og sulfater.
Betegnelsen halogen blir anvendt for å representere fluor, klor, brom eller jod.
Betegnelsen 'alkyl' som en gruppe eller del av en gruppe betyr en lineær eller forgre-net alkylgruppe, foreksempel en metyl, etyl, n-propyl, i-propyl, n-butyl, s-butyl ellert-butyl gruppe.
Fortrinnsvis er R° i 3- eller 4-stillingen av fenylringen, som definert i formel (I). Fortrinnsvis er R<1> i 6-stillingen i pyridazinringen, som definert i formel (I).
Fortrinnsvis er R° F, Ci.3alkoksy eller Ci^-alkoksy substituert med én eller mer fluoratomer.
Fortrinnsvis er R<1> Cnalkylsulfonyl, d^alkoksy substituert med én eller flere fluoratomer, eller, når R° er d-ealkoksy, kan den også være H. Mer foretrukket er R<1 >Ci^alkylsulfonyl, d^alkoksy substituert med én eller flere fluoratomer eller, når R° er Ci.6alkoksy eller d-ealkoksy substituert med én eller flere fluoratomer, kan også være H.
Fortrinnsvis er R<2> H.
Fortrinnsvis er R<3> metyl eller NH2.
I foreliggende oppfinnelse tilveiebringes én gruppe med forbindelser med formel (I)
(gruppe A) hvor: R° er F, daafkoksy eller Ci^alkoksy substituert med én eller flere fluoratomer, eller R<1> er Ci^alkylsulfonyl, C^alkoksy substituert med én eller flere fluoratomer, eller, når R° er C^alkoksy eller Ci_3alkoksy substituert med én eller flere fluoratomer, kan også være H; R2 er H og R3 er metyl eller NH2.
Innen gruppe A, tilveiebringes en annen gruppe med forbindelser (gruppe A1) hvor R° er F, Ci.2alkoksy eller OCHF2; R<1> er metylsulfonyl eller OCHF2, eller, når R° er Ci-2alkoksy eller OCHF2, kan også være H; R<2> er H og R<3> er metyl eller NH2.
Innen gruppe A, tilveiebringes en ytterligere gruppe med forbindelser (gruppe A2) hvor R° er F, Ci-3alkoksy eller Ci.3alkoksy substituert med én eller flere fluoratomer; R<1> er C^alkylsulfonyl, C^alkoksy substituert med én eller flere fluoratomer eller, når R° er Ci.3alkoksy eller d^alkoksy substituert med én eller flere fluoratomer, kan også være H; R<2> er H; og R<3> er metyl eller NH2.
Innen gruppene A, A1 og A2, er R° fortrinnsvis 3- eller 4-stillingen av fenylringen og R2 er fortrinnsvis i 6-stillingen av pyridazinringen.
Det er å bemerke at foreliggende oppfinnelse omfatter alle isomerer av forbindelsene med formel (I) og deres farmasøytisk godtagbare derivater, omfattende alle geomet-riske, tautomere og optiske former, og blandinger derav (f.eks. racemiske blandinger).
Spesielt foretrukne forbindelser ifølge foreliggende oppfinnelse er: 3-{4-metansulfonyl-fenyl)-2-(4-metoksy-fenyl)-pyrazolo[1,5-b]pyridazin; 6-difluorme-toksy-2-(4-fluor-fenyl)-3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin; 2-(4-etoksy-fenyl)-3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin; 2-(4-fluor-fenyl)-6-metansulfonyl-3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin; 2-{4-difluormetoksy-fenyl)-3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin; 4-[2-(4-etoksy-fenyl)-pyra-zolo[1,5-b]pyridazin-3-yl]-benzensulfonamid; 6-difluormetoksy-2-(3-fluor-fenyl)-3-{4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin; og farmasøytisk godtagbare derivater derav.
Forbindelser ifølge foreliggende oppfinnelse er sterke og selektive inhibitorer av COX-2. Denne aktiviteten er illustrert ved deres evne til selektivt å hemme COX-2 over COX-1.
I betraktning av deres selektive COX-2 hemmende aktivitet, er forbindelsene ifølge foreliggende oppfinnelse av interesse for anvendelse i human og veterinærmedisin, spesielt ved behandling av smerte (both kronisk og akutt), feber og inflammasjon ved en rekke tilstander og sykdommer. Slike tilstander og sykdommer er velkjente i fag-området og omfatter revmatisk feber; symptomer assosiert med influensa eller andre viral infeksjoner, slik som vanlig forkjølelse; smerter i lavere del av rygg og nakke; hodepine; tannpine; sprengninger og smerte; myosit; nevralgi; synovitt; artritt, omfattende revmatoid artritt; degenerative ledd sykdommer, omfattende osteoartritt; gikt og ankylosing spondylitt; tendinit; bursitt; hud-beslektede tilstander, slik som psoriasis, eksem, brannsår og dermatitt; skader, slik som sportsskader og de som oppstår fra kirurgiske og dentale prosedyrer.
Forbindelsene ifølge foreliggende oppfinnelse kan også være nyttige for behandlingen av andre tilstander mediert av selektiv hemning av COX-2.
For eksempel kan forbindelsene ifølge foreliggende oppfinnelse hemme cellulær og neoplastisk transformasjon og metastasisk tumorvekst og følgelig være nyttige ved behandlingen av visse cancersykdommer, slik som tarmkreft.
Forbindelser ifølge foreliggende oppfinnelse kan også forhindre neuronal skade ved å hemme dannelsen av neuronale frie rester (og følgelig oksydativ stress) og kan derfor anvendes ved behandling av slag; epilepsi; og epileptiske anfall (omfattende grand mal, petit mal, myoklonisk epilepsi og del-anfall).
Forbindelser ifølge foreliggende oppfinnelse hemmer også prostanoid-fremkalt glatt-muskelkontraksjon og kan følgelig anvendes ved behandling av dysmenorré og pre-matur fødsel.
Forbindelser ifølge foreliggende oppfinnelse hemmer inflammatoriske prosesser og kan derfor anvendes ved behandling av astma, allergisk rhinitt og åndenød syndrom; gastrointestinale tilstander slik som inflammatorisk tarmsykdom, Chron's sykdom, gastritt, irritabel tarmsyndrom og ulcerativ kolitt; og inflammasjon i slike sykdommer som vaskulær sykdom, migrene, periarterit nodosa, tyroidit, aplastisk anemi, Hodgkin's sykdom, sclerodoma, type I diabetes, myasthenia gravis, multippel skle-rose, sorcoidosis, nefrotisk syndrom, Bechefs syndrom, polymyositis, gingivitt, kon-junktivitt og myokardial ischemi.
Forbindelser ifølge foreliggende oppfinnelse kan også være nyttige ved behandling av oftalmiske sykdommer slik som retinit, retinopatier, uveitt og akutt skade av øye-vev.
Forbindelser ifølge foreliggende oppfinnelse kan også være nyttige for behandlingen av kognitive lidelser slik som demens, spesielt degenerativ demens (omfattende senil demens, Alzheimers sykdom, Pick's sykdom, Huntington's chorea, Parkinson's sykdom og Creutzfeldt-Jakob sykdom), og vaskulær demens (omfattende multiinfarkt demens), som demens assosiert med intracranial-rom okkuperende lesjoner, traume, infeksjoner og beslektede tilstander (omfattende HIV infeksjon), metabolisme, toksi-ner, anoksi og vitaminmangel; og mild kognitiv reduksjon ved aldring, spesielt assosiert med alder hukommelsessvikt.
I henhold til et annet aspekt ved oppfinnelsen, er det tilveiebrakt anvendelse av en forbindelse med formel (I) eller et farmasøytisk godtagbart derivat derav som definert i hvilket som helst av kravene 1 til 7 for fremstilling av et terapeutisk middel for behandling av en tilstand som blir mediert av selektiv hemning av COX-2, så som en inflammatorisk lidelse.
Det er å bemerke at referanse til behandling heri omfatter både behandling av eta-blerte symptomer og profylaktisk behandling, hvis det ikke er angitt eksplisitt på annen måte.
Det vil forstås at forbindelsene ifølge foreliggende oppfinnelse kan fordelaktig bli anvendt i sammenheng med én eller flere andre terapeutisk midler. Eksempler på egn-ede midler for adjunktiv terapi omfatter smerte-lindrende midler slik som en glycin-antagonist, en natriumkanal inhibitor (f.eks. lamotrigin), en forbindelse P antagonist (f.eks. en NKi antagonist), acetaminofen eller fenacetin; en matriks metallo-protein-ase inhibitor; en salpetersyre-oksyd-syntase (NOS) inhibitor (f.eks. en iNOS eller en nNOS inhibitor); en inhibitor for frigjøringen, eller virkning, av tumor nekrose faktor a; en antistoff-terapi (f.eks. en monoklonal antistoff-terapi); et stimuleringsmiddel omfattende kaffein; en H2-antagonist, slik som ranitidin; en proton pumpeinhibitor, slik som omeprazol; et antacid, slik som aluminium eller magnesium-hydroksyd; et anti-flatuleringsmiddel, slik som simetikon; et dekongestant, slik som fenylefrin, fenylpro-panolamin, pseudoefedrin, oksymetazolin, epinefrin, nafazolin, xylometazolin, propyl-heksedrin, eller levo-desoksyefedrin; et anti-tussiv, slik som kodein, hydrokodon, kar-mifen, karbetapentan, eller dekstrametorfan; et diuretisk middel; eller et beroligende middel eller ikke-beroligende antihistamin. Det er å bemerke at forbindelsene med formel (I) eller et farmasøytisk godtagbart derivat derav kan anvendes i kombinasjon med én eller flere andre terapeutiske midler.
Forbindelsene med formel (I) og deres farmasøytisk godtagbare derivater blir hensiktsmessig administrert i form av farmasøytiske preparater. Således, i et annet aspekt ved oppfinnelsen, er det tilveiebrakt et farmasøytisk preparat omfattende en forbindelse med formel (I) eller et farmasøytisk godtagbart derivat derav i blanding med én eller flere fysiologisk godtagbare bærere eller tilsetningsmidler. Slike preparater kan hensiktsmessig presenteres for anvendelse på konvensjonell måte i blanding med én eller flere fysiologisk godtagbare bærere eller tilsetningsmidler.
Forbindelsene med formel (I) og deres farmasøytiske godtagbare derivater kan være formulert for administrering på en hvilken som helst egnet måte. De kan, for eksempel, være formulert for topisk administrering eller administrering ved inhalering eller, mer foretrukket, for oral, transdermal eller parenteral administrering. Det farmasøy-tiske preparatet kan være i en form slik at det kan tilveiebringe kontrollert frigjøring av forbindelsene med formel (I) og deres farmasøytisk godtagbare derivater.
For oral administrering kan det farmasøytiske preparatet være i form av, for eksempel, tabletter (omfattende sub-linguale tabletter), kapsler, pulvere, løsninger, siruper eller suspensjoner fremstilt ved konvensjonelle metoder med godtagbare tilsetningsmidler.
For transdermal administrering kan det farmasøytiske preparat bli gitt i form av et transdermalt plaster, slik som et transdermalt iontoforetisk plaster.
For parenteral administrering kan det farmasøytiske preparat bli gitt som en injeksjon eller en kontinuerlig infusjon (f.eks. intravenøst, intravaskulært eller subkutant). Preparater kan være i slike former som suspensjoner, løsninger eller emulsjoner i oljeaktige eller vandige konstituenter og kan inneholde formulerings-midler slik som suspenderings-, stabilisings- og/eller dispergeringsmidler. For administrering ved injeksjon kan disse være i form av en enhetsdose presentasjon eller som en multidose presentasjon, fortrinnsvis med et tilsatt konserveringsmiddel. Alternativt kan for parenteral administrering den aktive bestanddel være i pulverform for rekonstituering med en egnet konstituent.
Forbindelsene ifølge foreliggende oppfinnelse kan også være formulert som et depot-preparat. Slike lenge virkende preparater kan administreres ved implantasjon (for
eksempel subkutant eller intramuskulært) eller ved intramuskulær injeksjon. Således kan for eksempel forbindelsene ifølge foreliggende oppfinnelse bli vær formulert med egnete polymerer eller hydrofobt materialer (for eksempel som en emulsjon i en god-tagbar olje) eller ione-bytte harpikser, eller som delvis oppløste derivater, for eksempel, som et delvis oppløst salt.
Som angitt ovenfor kan forbindelsene ifølge foreliggende oppfinnelse også bli anvendt i kombinasjon med andre terapeutisk midler.
Kombinasjoner referert til ovenfor kan hensiktsmessig presenteres for anvendelse i form av en farmasøytisk formulering. De individuell komponentene til slike kombinasjoner kan administreres enten sekvensielt eller samtidig i separate eller kombinerte farmasøytisk preparater.
Når en forbindelse med formel (I) eller et farmasøytisk godtagbart derivat derav blir anvendt i kombinasjon med et andre terapeutisk middel aktiv mot samme sykdom kan dosen av hver forbindelse være forskjellig fra den når forbindelsen blir anvendt alene. Hensiktsmessige doser vil være lett å bestemme av fagfolk på området. En foreslått daglig dose av en forbindelse med formel (I) for behandlingen av et menn-eske er 0,01 mg/kg til 500mg/kg, slik som 0,05mg/kg til 100mg/kg, f.eks.
0,1 mg/kg til 50mg/kg, som kan bli hensiktsmessig administrert i 1 til 4 doser. Den nøyaktige anvendte dosen vil avhenge av alderen og tilstanden til pasienten og ad-ministreringsvei. Således kan for eksempel en daglig dose på 0,25mg/kg til 10mg/kg være egnet for systemisk administrering.
Forbindelser med formel (I) og farmasøytisk godtagbare derivater derav kan fremstilles ved hvilken som helst kjent metode for fremstilling av forbindelser med analog struktur.
Egnet metoder for fremstilling av forbindelser med formel (I) og farmasøytisk godtagbare derivater derav er beskrevet nedenfor. I formlene som følger er R° til R<3> som definert i formel (I) ovenfor hvis ikke annet er angitt; Hal er et halogen, slik som Br eller I; X er et motion, slik som I ; og alkyl er som tidligere definert.
Foreliggende oppfinnelse vedrører følgelig videre fremgangsmåte for fremstilling av forbindelse med formel (I) og farmasøytisk godtagbare derivater derav som definert i hvilket som helst av kravene 1 tit 7, kjennetegnet ved at den omfatter:
(A) omsetning av en forbindelse med formel (II)
eller et beskyttet derivat derav, med en forbindelse med formel (III)
eller et beskyttet derivat derav; eller
(B) hvor R3 representerer Ci.6alkyl, omsetning av en forbindelse med formel (IV)
eller et beskyttet derivat derav med et oksyderingsmiddei; eller
(C) hvor R<1> er Ci.6alkylsulfonyl, oksydering av en forbindelse med formel (V)
eller et beskyttet derivat.
(D) hvor R<1> er Ci.ealkoksy substituert med én eller flere fluoratomer, omsetning av en alkohol med formel (VI)
eller et beskyttet derivat derav med et halogenfluoralkan. (E) innbyrdes omdannelse av en forbindelse med formel (l) til en annen forbindelse med formel (I); eller (F) avbeskyttelse av et beskyttet derivat av forbindelse med formel (I); og eventuelt omdannelse av forbindelser med formel (I) fremstilt ved hvilken som helst av fremgangsmåtene (A) til (F) til farmasøytisk godtagbare derivater derav. Således i henhold til første fremgangsmåte (A), kan forbindelser med formel (I) fremstilles ved omsetning av en forbindelse med formel (II)
eller et beskyttet derivat derav med en boronsyre med formel (III)
eller et egnet derivat derav i nærvær av en egnet overgangsmetaJIkatalysator. Egnete derivater med formel (III) omfatter boronsyreestere, slik som de beskrevet i R. Miyaura et al, J. Org. Chem., 1995, 60, 7508-7510. Hensiktsmessig blir reaksjonen utført i et oppløsningsmiddel, slik som en eter (f.eks. 1,2 dimetoksyetan); i nærvær av en base, slik som en uorganisk base (f.eks. natriumkarbonat); og ved anvendelse av en palladiumkatalysator, slik som tetrakis(trifenylfosfin)palladium(0).
I henhold til en annen fremgangsmåte (B), kan forbindelser med formel (I) hvor R<3> er d-ealkyl fremstilles ved oksydering av en forbindelse med formel (IV) eller et beskyttet derivat derav under konvensjonelle betingelser. Hensiktsmessig blir oksydasjonen oppnådd ved anvendelse av en monopersulfatforbindelse, slik som kaliumperoksymonosulfat (kjent som Oksone™) og reaksjonen blir utført i et oppløs-ningsmiddel, slik som en vandig alkohol, (f.eks. vandig metanol), og ved mellom
-78°C og omgivelsestemperatur.
I henhold til en annen fremgangsmåte (C), kan forbindelser med formel (I) hvor R<1> er Ci.6alkylsulfonyl fremstilles ved oksydering av en forbindelse med formel (V)
eller et beskyttet derivat derav under konvensjonelle betingelser. Hensiktsmessig blir oksydasjonen oppnådd som beskrevet ovenfor for fremgangsmåte (B).
I henhold til en annen fremgangsmåte (D), kan forbindelser med formel (I) hvor R<1> er C-i.6alkoksy substituert med én eller flere fluoratomer fremstilles ved omsetning av en alkohol med formel (VI) eller et beskyttet derivat derav med en halogenfluoralkan under konvensjonelle betingelser. Hensiktsmessig reaksjon blir oppnådd i et oppløsningsmiddel, slik som et polart oppløsningsmiddel (f.eks. N,N-dimetylformamid), i nærvær av en sterk base, slik som et uorganisk hyd rid (f.eks. natriumhydrid), ved omtrent omgivelsestemperatur og ved anvendelse av hensiktsmessig bromfluoralkan hvilket gir de ønskede forbindelsene med formel (I).
I henhold til en annen fremgangsmåte (E) kan forbindelser med formel (I) fremstilles ved innbyrdes omdannelse, ved å anvende andre forbindelser med formel (I) som forløpere. De følgende prosedyrer illustrerer egnet innbyrdes omdannelser. Forbindelser med formel (I) hvor R<1> eller R2 representerer Ci^alkyl substituert med én eller flere fluoratomer kan fremstilles fra hensiktsmessig forbindelse med formel (I) hvor R<1> eller R2 er d-ehydroksyalkyl, C(0)H eller C(0)Ci^alkyl, ved behandling med en egnet fluor. Egnete kilder av fluor omfatter, for eksempel, dietyiaminosvovel-trifluorid. Hensiktsmessig reaksjon blir oppnådd i nærvær av et oppløsningsmiddel, slik som et halogenert hydrokarbon (f.eks. diklormethan), og ved redusert temperatur, slik som - 78°C.
Forbindelser med formel (I) hvor R<1> eller R2 representerer C(0)H kan fremstilles fra de tilsvarende forbindelser med formel (I) hvor R<1> eller R2 representerer CH2OH ved oksydasjon. Egnete oksyderingsmidler omfatter, for eksempel, mangan (IV) oxid. Hensiktsmessig blir oksydasjonene oppnådd i nærvær av et oppløsningsmiddel, slik som et halogenert hydrokarbon (f.eks. kloroform), og ved forhøyet temperatur (f.eks. tilbakeløp).
Forbindelser med formel (I) hvor R<1> eller R2 representerer Ci-ghydroksyalkyl, og hvor hydroksygruppen er tilknyttet til karbonet bundet til pyridazinringen, kan fremstilles ved reduksjon av forbindelsen med formel (I) hvor R<1> eller R<2> representerer tilsvarende aldehyd eller keton. Egnete reduksjonsmidler omfatter hydrid reduksjons-midler, slik som diisobutylaluminiumhydrid. Hensiktsmessig blir reduksjonen oppnådd i nærvær av et oppløsningsmiddel, slik som et halogenert hydrokarbon (f.eks. diklormethan), og ved redusert temperatur, slik som -78°C.
Som kjent for fagfolk på området kan det være nødvendig eller ønskelig ved et hvilket som helst trinn i syntesen av forbindelser med formel (I) å beskytte én eller flere sensitive grupper i molekylet for å forhindre uønskede side reaksjoner.
En annen fremgangsmåte (F) for fremstilling av forbindelser med formel (I) omfatter følgelig avbeskyttelse av beskyttede derivater av forbindelser med formel (I).
Beskyttelsesgruppene anvendt ved fremstilling av forbindelser med formel (I) kan bli anvendt på konvensjonell måte. Se for eksempel, de som er beskrevet i 'Protective Groups in Organic Synthesis' by Theodora W. Green og Peter G. M. Wuts, second edition, (John Wiley og Sons, 1991), som også beskriver metoder for fjernelse av slik grupper.
Forbindelser med formel (II) kan fremstilles ved halogenering av forbindelser med formel (VII)
ved konvensjonelle metoder.
Således blir estere med formel (VI) først hydrolysen til deres tilsvarende syrer, for eksempel ved behandling med en sterk base (f.eks. natriumhydroxid), i nærvær av et oppløsningsmiddel (f.eks. etanol) og ved forhøyet temperatur. Den tilsvarende syren blir deretter behandlet med et halogeneringsmiddel, hensiktsmessig ved omgivelsestemperatur og i et oppløsningsmiddel (f.eks. klorert hydrokarbon), under noen betingelser gjennomgår syren både halogenering og dekarboksylering. Det er hensiktsmessig at halogeneringsmiddelet er et bromineringsmiddel, slik som brom i nærvær av en sterk syre (f.eks. bromhydrogensyre i eddiksyre) eller N-brom-succinimid, hvilket gir
de tilsvarende forbindelse med formel (II) hvor Hal er brom.
Estere med formel (VII) kan fremstilles ved omsetning av en forbindelse med formel
(VIII)
med et aminopyridaziniumkompleks med formel (IX)
under konvensjonelle betingelser. Hensiktsmessig blir reaksjonen utført i nærvær av en base, slik som kaliumkarbonat, et oppløsningsmiddel, slik som N,N-dimetyl-forma-mid og ved omgivelsestemperatur.
Boronsyrer med formel (III) er enten kjente forbindelser eller kan fremstilles ved metoder kjent i litteraturen slik som de beskrevet i, for eksempel, EPA publikasjon nr. 533268.
Forbindelser med formlene (IV), (V) og (VI) kan fremstilles ved metoder analoge til de beskrevet for fremstilling av forbindelsen med formel (I) fra forbindelser med formel (II).
Forbindelser med formel (VIII) er enten kjente forbindelser eller kan fremstilles ved metoder kjent i litteraturen slik som de beskrevet i, for eksempel, D. H. Wadsworth et al., J Org Chem, (1987), 52(16), 3662-8 og J. Morris og D. G. Wishka, Synthesis
(1994), (1), 43-6.
Forbindelser med formel (IX) er enten kjente forbindelser eller kan fremstilles ved metoder kjent i litteraturen slik som de beskrevet i, for eksempel, Y. Kobayashi et ai, Chem Pharm Bull, (1971), 19(10), 2106-15; T. Tsuchiya, J. Kurita og K. Takayama, Chem Pharm Bull, 28(9), 2676-2681, (1980) og K. Novitskii et al., Khim Geterotskil Soedin, 1970 2,57-62.
Visse mellomprodukter beskrevet ovenfor er nye forbindelser, og det er å bemerke at alle nye mellomprodukter utgjør aspekter ifølge foreliggende oppfinnelse. Forbindelser med formel (II) er hoved mellomprodukter og representerer et spesiell aspekt ifølge foreliggende oppfinnelse.
Hensiktsmessig blir forbindelser ifølge foreliggende oppfinnelse isolert etter opparbei-delse i form av den frie basen. Farmasøytisk godtagbare syreaddisjonssalter av forbindelsene ifølge foreliggende oppfinnelse kan fremstilles ved anvendelse av konvensjonelle metoder.
Solvater (f.eks. hydrater) av en forbindelse ifølge foreliggende oppfinnelse kan bli dannet i løpet av opparbeidelsesprosedyrene i én av ovennevnte prosesstrinn.
De følgende eksempler illustrerer oppfinnelsen. Alle temperaturer er i °C. Flash kolonnekromatografi ble utført ved anvendelse av Merck 9385 silika. Tynnsjikts-kromatografi (Tic) ble utført på silikaplater. NMR ble utført på et Brucker 250MHz spectro-meter. Kjemiske skift er gitt, med hensyn til tetrametylsilan som indre kjemiske shift referanse, i □ ppm. De følgende forkortelser blir anvendt: Me = metyl, s = singlett,
d = dublett, t = triplett og m = multippelt.
EKSEMPEL 1
6-difluormetoksy-2-(4-fluor-fenyl)-3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]-pyridazin (i) 6-metoksy-2-{4-fluor-fenyl-pyrazolo[1,5-b]pyridazin-3-karboksylsyre-metylester.
1,8-diazabicyklo[5,4,0]undec-7-en (3,39 ml) ble satt til en blanding av 3-(4-fluorfenyl)-prop-2-ynsyre-metylester (3,36 g) og 1-amino-3-metoksy-pyirdazin-1-iummesitylen-sulfonate<1> (6,1419 g) i acetonitril (125 ml) og blandingen ble omrørt ved omgivelsestemperatur i 48 timer. I løpet av de første 2 timene ble en strøm av luft ført gjennom reaksjonen. Blandingen ble konsentrert;" vakuum, oppløst i etylacetat (150 ml), vasket med vann (3 x 25 ml), tørket (MgSCM, filtrert og inndampet /' vakuum, hvilket gir tittelforbindelsen som et brunt, fast stoff (4,77 g).

(iii) 2-(4-tluor-fenyl)-3-(4-metansulfonyl-fenyl)-6-metoksy-pyrazolo[1,5-bJ-pyridazin
En blanding av 6-metoksy-2-(4-fluor-fenyl-pyrazolo[1,5-b]pyridazini3-karboksylsyre (869 mg) og natriumbikarbonat (756 mg) i dimetylformamid (10 ml) ble behandlet med N-bromsuccinimid (587 mg) og omrørt ved omgivelsestemperatur i 1 time, deretter satt til vann (50 ml) og ekstrahert med etylacetat (3x50 ml), tørket (MgS04) og inndampet / vakuum. Det resulterende brune fast stoff (1,612 g) ble oppløst i 1,2 dimetoksyetan (20 ml). 2N vandig natriumkarbonat-løsning (10 ml) ble tilsatt sammen med 4-(metansulfonyl)fenylboronsyre (660 mg) og tetrakis(trifenylfosfin)-palladium (0) (100 mg) og blandingen ble oppvarmet ved tilbakeløp i 20 timer. Reaksjonsblandingen ble hellet i vann(50 ml), ekstrahert med diklormetan (3x100 ml). De samlede organiske ekstrakter ble tørket (MgS04) og inndampet / vakuum, hvilket gir et brunt, fast stoff (1,116 g) som ble renset ved flash kolonnekromatografi på silika, under eluering med cykloheksan/etylacetat (4:1 deretter 2:1), hvilket gir tittelforbindelsen som et gult, fast stoff (390 mg).

MH<+> 398 (iv) 2-(4-fluor-fenyl)-3-{4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin-6-ol En blanding av 2-(4-fluor-fenyl)-3-(4-metansulfonyl-fenyl)-6-metoksy-pyra-zolo[1,5-b]pyridazin (321 mg) og pyridinhydroklorid (1,4 g) ble oppvarmet til og ved 200°C i et forseglet kar (Reactivial™) i 3 timer. Den avkjølte reaksjon ble hellet i vann (20 ml) og ekstrahert med etylacetat (3x30 ml). De samlede organiske ekstrakter ble tørket (MgS04), filtrert og inndampet / vakuum, hvilket gir et fast stoff som ble utgnidd med dietyleter, hvilket gir tittelforbindelsen som et beige, fast stoff (119 mg).

En løsning av 2-(4-fluor-fenyl)-3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin-6-ol (0,2 g) i vannfri dimetylformamid (5 ml) ble behandlet med natriumhydrid ( 0,046 g, 60% dispersjon i mineralolje), etter at brusingen opphørte ble en strøm av brom-diflu-ormetangass ført gjennom blandingen ved omgivelsestemperatur i 30 minutter. Reaksjonsblandingen ble deretter hellet i vann (50 ml) og ekstrahert med etylacetat (50 ml), den organiske ekstrakten ble vasket med vann (3X 50 ml), tørket og konsentrert / vakuum. Residuet ble renset ved kromatografi, hvilket gir tittel-forbindelsen som et hvitt, fast stoff (0,17 g).

3-(4-metansulfonyl-fenyl)-2-(4-metoksy-fenyl)-pyrazolo[1,5-b]pyridazin
(i) 2-(4-metoksy-fenyl)-pyrazolo[1,5-b]pyridazin-3-karboksylsyre-metylester Diazabicyklo[5,4,0]undec-7-en (22,76 ml, 2ekv) ble satt dråpevis til en oppløsning av metyl 3-(4-metoksy-fenyl)-prop-2-ynsyre1 (14,46 g, 76mM) og 1-aminopyridazinium-jodid2 (2ekv) i acetonitril under nitrogen eig omrørt i 6 timer. Rensning ved kromatografi på silikagel under eluering med toluen, deretter toluen:etyiacetat (9:1) ga tittel-forbindelsen (2,76 g) som et brunt, fast stoff.

(ii) 3-(4-metansulfonyl-fenyl)-2-(4-metoksy-fenyl)-pyrazolo[1,5-b]pyridazin En blanding av 2-(4-metoksy-fenyl)-pyrazolo[1,5-b]pyridazin-3-karboksylsyremetyl-ester (2,76 g) og vandig natriumhydroksyd (2N, 30 ml) i etanol (30 ml) ble tilbake-løpskokt under nitrogen i 2 timer. Den avkjølte blandingen ble surgjort med saltsyre (2N) og resulterende hvitt fast stoff (2,53 g) isolert ved filtrering. Dette faste stoffet ble oppløst i DMF og natriumbikarbonat (2,67 g, 3,3ekv) tilsatt, fulgt av N-brom-succinimid (1,88 g, 1,1ekv) porsjonsvis. Etter omrøring i 1 time under nitrogen, ble vann tilsatt og ekstrahert med etylacetat (2x 25 ml). Den tørkede organiske fasen ble konsentrert og residuet tatt opp i DME (60 ml). Vandig natriumkarbonat (2N, 15 ml) ble
tilsatt, fulgt av 4-metansulfonyl-fenylboronsyre (3,12 g) og tetrakis(trifenylfosfin)-palla-dium(0) (250 mg). Blandingen ble oppvarmet ved tilbakeløp under nitrogen i 18 timer, avkjølt, hellet i vann og ekstrahert med etylacetat (2 x 25 ml). De samlede organiske faser ble tørket og konsentrert på silikagel. Kromatografi på silikagel under eluering med toluen:etylacetat (8:1) ga, ved konsentrasjon, tittelforbindelsen (3,58 g) som et kremaktig fast stoff.

2-(4-etoksy-fenyl)-3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin
(i) 4-[3-(4-metansulfonyl-fenyl)-pyrazolo[1,5-b]pyridazin-2-yl]-fenol Bortribromid (1M løsning i CH2CI2, 2,1 ekv) ble satt til 3-(4-metansulfonyl-fenyl)-2-(4-metoksy-fenyl)-pyrazolo[1,5-b]pyridazin (3,58 g) i CH2CI2 ved -70°. Blandingen ble omrørt i 10min deretter oppvarmet til 0° og omrørt ved 0° natten over. Reaksjonsblandingen ble gjort alkalisk med kaliumkarbonat deretter surgjort med saltsyre (2M), hellet i vann og ekstrahert inn i CH2CI2. Den organiske fasen ble tørket, filtrert og konsentrert, hvilket gir tittelforbindelsen (1,87 g) som et gult, fast stoff. MH<+> 366

2-(4-fluor-fenyl)-6-metansulfonyl-3-(4-metansulfonyl-fenyl)-pyrazoio[1,5-b]-pyri-dazin
(i) 2-(4-fluor-fenyl)-6-metylsulfanyl-pyrazolo[1,5-b]pyridazin-3-karboksyl-syremetylester
Fast stoff av t-butoksykarbonyl-O-mesitylenesulfonylhydroksylamine<1> (7,8 g) ble tilsatt porsjonsvis med omrøring til TFA (25 ml) over 10min deretter omrørt i ytterligere 20 minutter. Løsningen ble hellet i is (-200 ml) og latt stå inntil isen smeltet. Resulterende hvitt fast stoff ble filtrert fra, vasket med vann og oppløst i DME (100 ml). Løsningen ble tørket over 4A mol.sikter i 1,5 timer, filtrert, deretter satt til en oppløs-ning av 3-metyltio-pyridazine<2> (2,6 g) i diklormetan (35 ml) og reaksjonen omrørt ved romtemperatur i 20 timer. Mellomproduktet, saltet, ble isolert ved filtrering som lyse-brune krystaller (3,87 g), suspendert i acetonitril (100 ml) og tilsatt metyl 3-(4-fluor-fenyl)-prop-2-ynsyre (2,02 g). 1,8-diazabicyklo[5,4,0]undec-7-en (2,1 ml) ble tilsatt dråpevis og reaksjonsblandingen ble omrørt ved romtemperatur i 20 timer. Det resulterende krystallinske presipitat ble filtrert fra, vasket og tørket (770 mg). Konsentrasjonen på filtratet ga en andre avling (430 mg). Bromdifluormetangass ble forsiktig boblet gjennom

(i) 2-{4-etoksy-fenyI)-pyrazolo[1,5-b]pyridazin-3-karboksylsyre-mety lester Diazabicyklo[5,4,0]undec-7-en (1,47 ml, 2ekv) ble satt dråpevis til en oppløsning av metyl 3-(4-etoksy-fenyl)-prop-2-ynsyre (1,0 g) og 1-aminopyridaziniumjodide<2>(2,19 g) i acetonitril (10 ml) under nitrogen og omrørt i 5 timer. Konsentrasjon og vandig opparbeiding ga tittelforbindelsen (1,2 g) som et klebrig brunt fast stoff.

Den avkjølte reaksjonsblandingen ble konsentrert / vakuum og residuet suspendert i 1,2 dimetoksyetan (10 ml), 2-(4-etoksy-fenyl)-3-jod-pyrazolo[1,5-bjpyridazin (0,4 g) ble tilsatt sammen med 2N natriumkarbonat (4 ml) og tetrakis(trifenylfosfin)palladium (0) (20 mg) og blandingen oppvarmet ved tilbakeløp under nitrogen i 18 timer. (i) 1 -<2,2-dibrom-vinyl)-3-fluor-benzen
Til en omrørt avkjølt (is/salt, 0°) løsning av karbontetrabromid (48,82 g) i vannfri CH2CI2 (200 ml) ble tilsatt porsjonsvis over 3 minutter, trifenylfosfin (77,1 g), med opprettholdelse av temperaturen under 10°. Den resulterende oransje suspensjon ble omrørt ved 0° i 1 time før tilsetning av 3-fluorbenzaldehyd (7,8 ml). Etter at tilset-ningen var fullstendig, ble suspensjonen omrørt ved 0° i 1 time og deretter stoppet ved tilsetning av vann (75 ml). Den organiske fasen ble separert og vasket med saltvann (75 ml), tørket (Na2S04) og inndampet til tørrhet. Gjenværende gummi ble hellet i cykloheksan (1L) og omrørt i 30 minutter. Den organiske fasen ble dekantert og residuet tatt opp i CH2Cb og hellet i cykloheksan (1L). Den resulterende blanding ble ekstrahert med etylacetat (3x100 ml) og de samlede organiske ekstrakter vasket med vann (7x75 ml), tørket (Na2S04) og inndampet, hvilket gir des-brom fenol som et brunt, fast stoff (1,0 g, MH<+> 230). Dette faste stoffet ble oppløst i vannfri DMF (10 ml) og natriumhydrid (60% dispersjon i mineralolje, 200 mg) tilsatt porsjonsyis. BIOLOGISK DATA
Hemmende aktivitet mot human COX-1 og COX-2 ble bedømt i COS celler som var blitt stabilt transfektert med cDNA for human COX-1 og human COX-2. 24 timer før forsøk, ble COS celler overført fra 175cm<2> kolber hvor de ble dyrket, på 24-brønn cel-lekulturplater ved anvendelse av følgende prosedyre. Inkuberingsmedium (Dulbecco's modifisert eagles medium (DMEM) supplert med varme-inaktivert føtalt kalveserum (10%v/v), penicillin (100 IU/ml), streptomycin (100>g/ml) og geneticin (600ng/ml)) ble fjernet fra en kolbe av konfluente celler (1 kolbe ved sammenflyting inneholder omtrent 1x10<7> celler). 10 ml fosfatbufret saltvann (PBS) ble satt til kolben for å vaske cellene. Etter kasting av PBS, ble cellene deretter skyllet i 10 ml trypsin i 20 sekunder, hvoretter trypsin ble fjernet og kolben plassert i en inkubator (37°) i 1-2 minutter inntil celler ble løsnet fra kolben. Kolben ble deretter fjernet fra inkubato-ren og celler resuspendert i 10 ml frisk inkubering medium. Innholdet av kolben ble overført til en 250 ml steril beholder og volumet av inkuberingsmediumet ble deretter dannet opptil 100 ml. 1 ml cellesuspensjon ble pipeteret inni hver brønn av 4x24-brønn cellekultur plater. Platene ble deretter plassert i en inkubator (37°C, 95% luft / 5% CO2) natten over. Hvis mer enn 1 kolbe celler var nødvendig, ble cellene fra de individuelle kolbene samlet før applisert på 24-brønnplater.
Etter inkubering over natten, ble inkuberingesmedium fullstendig fjernet fra he 24-brønn cellekultur plater og erstattet med 250^1 frisk DMEM (37<0>C). Testforbindelsene ble dannet opptil 250x av nødvendig test konsentrasjon i DMSO og ble satt til brøn-nene i et volum på 1 (d. Plater ble deretter blandet forsiktig ved omdreining og deretter plassert i en inkubator i 1 time (37°C, 95% luft/5% CO2). Etter inkuberings-perio-den, ble 10^1 arachidonsyre (750|iM) satt til hver brønn, hvilket gir en endelig arachidonsyre konsentrasjon på 30uM. Plater ble deretter inkubert i ytterligere 15 minutter, etter som inkuberingsmediumet ble fjernet fra hver brønn av platene og lagret ved -20°C, før bestemmelse av prostaglandin E2 (PGE2) nivåer ved anvendelse av en-zym immunoanalyse. 8. Fremgangsmåte for fremstilling av forbindelse med formel (I) og farmasøytisk godtagbare derivater derav som definert i hvilket som helst av kravene 1 til 7, karakterisert ved at den omfatter: (A) omsetning av en forbindelse med formel (II)
eller et beskyttet derivat derav, med en forbindelse med formel (III)
eller et beskyttet derivat derav; eller (B) hvor R<3> representerer Ci.6alkyl, omsetning av en forbindelse med formel (IV)
eller et beskyttet derivat derav med et oksyderingsmiddel; eller (C) hvor R<1> er Ci^alkylsulfonyl, oksydering av en forbindelse med formel (V)
eller et beskyttet derivat. (D) hvor R<1> er Ci-6alkoksy substituert med én eller flere fluoratomer, omsetning av en alkohol med formel (VI)
eller et beskyttet derivat derav med et halogenfluoralkan. (E) innbyrdes omdannelse av en forbindelse med formel (I) til en annen forbindelse med formel (I); eller (F) avbeskyttelse av et beskyttet derivat av forbindelse med formel (I);
og eventuelt omdannelse av forbindelser med formel (I) fremstilt ved hvilken som helst av fremgangsmåtene (A) til (F) til farmasøytisk godtagbare derivater derav.
9. Farmasøytisk preparat, karakterisert ved at det omfatter en forbindelse med formel (I) eller et farmasøytisk godtagbart derivat derav som definert i hvilket som helst av kravene 1 til 7 i blanding med én eller flere fysiologisk godtagbare bærere eller tilsetningsmidler.
