Description
Foreliggende oppfinnelse er rettet mot nye taksanderivater som har anvendbarhet som antileukemi- og antitumormidler, samt farmasøytiske preparater som inneholder slike.
Taksanfamilien av terpener, hvor taksol er et medlem, har tiltrukket seg betydelig interesse både innen biologien og kjemien. Taksol er et lovende kjemoterapeutisk middel mot kreft med et bredt spektrum av antileukemisk og tumorinhiber-ende aktivitet. Taksol har en 2'R,3'S-konfigurasjon og den følgende strukturformel:
hvor Ac er acetyl. På grunn av denne lovende aktivitet gjennomgår taksol for tiden kliniske forsøk både i Frankrike og USA.
Colin et al. rapporterte i US patentskrift nr. 4.814.470 at taksolderivater med strukturformel (2) nedenunder har en aktivitet som er betydelig større enn aktiviteten til taksol (1).

Selv om taksol og taksoter er lovende kjemoterapeutiske midler, er de ikke effektive i alle tilfeller. Følgelig består det et behov for ytterligere kjemoterapeutiske midler.
Blant formålene ved foreliggende oppfinnelse er derfor tilveiebringelsen av nye taksanderivater som er verdifulle antileukemi- og antitumormidler.
Foreliggende oppfinnelse er derfor i korte trekk rettet mot taksanderivater med formelen:

I overensstemmelse med foreliggende oppfinnelse er det blitt oppdaget at forbindelser med strukturformel (3) generelt, og strukturformlene (4) og (5) spesielt, oppviser bemerkelsesverdige egenskaper in vitro og er verdifulle antileukemi- og antitumormidler. Deres biologiske aktivitet er blitt bestemt in vitro ved å anvende tubulinanalyser i henhold til metoden til Parness et al., J. Cell. Biology, 91, s. 479-487 (1981), og humane kreftcellelinjer, og er sammenlignbar med det som oppvises av taksol og taksoter.
Taksaner med formlene (4) og (5), som har 2'R,3'S-konfigurasjonen, kan fås ved å omsette et p-laktam med metallalkoksider som har den tetrasykliske kjerne til taksan og en C-13 metallisk oksidsubstituent, hvorved man får forbindelser med en (3-amidoestersubstituent i C-13. p-laktamene har følgende strukturformel:

p-laktamer (6) kan fremstilles fra lett tilgjengelige utgangsmaterialer, som vist ved hjelp av det følgende reaksjonsskjema:
reagenser:
(a) LDA, THF, -78 °C til -50 °C; (b) LHMDS, THF, -78 °C til 0 °C; (c) THF, -78 °C til 25 °C (2 t); og (d) trietylamin og et alkylklorformiat (R3=
alkoksykarbonyl).
3-hydroksylbeskyttelsesgruppen vist i reaksjons-skjemaet ovenfor er -SiR5, hvor R5er trialkyl eller triaryl, slik som trietyl. 3-hydroksylgruppen kan beskyttes med andre standard beskyttelsesgrupper, slik som 1-etoksyetyl eller 2,2,2-trikloretoksymetyl. Ytterligere hydroksybeskyttelses-grupper og syntesen derav kan finnes i "Protective groups in Organic Synthesis" av T.W. Greene, John Wiley & Sons, 1981.
De racemiske p-laktamer kan oppløses i de rene enantiomerene før beskyttelse ved rekrystallisering av de til-svarende 2-metoksy-2- (trif luormetyl) -f enyleddiksyreestere. Reaksjonen beskrevet nedenunder, hvor p-amidoestersidekjeden tilknyttes, har imidlertid fordelen ved å være svært dia-stereoselektiv og muliggjør derved bruken av en racemisk blanding av sidekjedeforløper.
Metallalkoksidene med den tetrasykliske kjerne til taksan og en C-13 metallisk oksidsubstituent har følgende strukturformel:

T3er hydrogen eller en hydroksybeskyttelsesgruppe; og M er et metall, fortrinnsvis valgt fra gruppen omfat-tende gruppe Ia, gruppe IIA og overgangsmetaller, helst Li, Mg, Na, K eller Ti.
Metallalkoksidene fremstilles fortrinnsvis ved å om sette en alkohol som har den tetrasykliske kjerne til taksan og en C-13-hydroksylgruppe med en organometallisk forbindelse i et egnet oppløsningsmiddel. Alkoholen er fortrinnsvis et beskyttet baccatin III, særlig 7-0-trietylsilyl-baccatin III (som kan fås som beskrevet av Greene et al. i JACS, 110, s. 5917 (1988), eller ad andre veier) eller 7,10-bis-O-trietylsilyl-baccatin III.
Som rapportert i Greene et al., omdannes 10-deacetyl-baccatin III til 7-0-trietylsilyl-lO-deacetyl-baccatin III i henhold til det følgende reaksjonsskjerna:
Under det som rapporteres å være forsiktig optimali-serte betingelser, omsettes 10-deacetyl-baccatin III med 20 ekvivalenter (C2H5)3SiCl ved 23 °C under argona tmos f ære i 20 timer i nærvær av 50 ml pyridin/mmol 10-deacetyl-baccatin III, hvorved man får 7-trietylsilyl-10-deacetyl-baccatin III (9a) som et reaksjonsprodukt i 84-86 % utbytte etter rensing. Reaksjonsproduktet kan så eventuelt acetyleres med 5 ekvivalenter CH3COCI og 25 ml pyridin/mmol av 9a ved 0 °C under argona tmos f ære i 48 timer, hvorved man får 86 % utbytte av 7-O-trietylsilyl-baccatin III (9b), Greene et al. i JACS, 110, s. 5917-5918 (1988).
Det erholdte 7-0-trietylsilyl-baccatin III (9b) omsettes med en organometallisk forbindelse, slik som n-butyl-litium, i et oppløsningsmiddel, slik som tetrahydrofuran (THF), hvorved metallalkoksidet 13-0-litium-7-0-trietylsilyl-baccatin III (10) fås, som vist i det følgende reaksjons-
Som vist i det etterfølgende reaksjonsskjema, reagerer 13-0-litium-7-0-trietylsilyl-baccatin III (10) med p-laktam (6), hvor R2er trietylsilyl, hvorved man får et mellom-produkt hvor C-7- og C-2'-hydroksylgruppene er beskyttet med en trietylsilylgruppe. Trietylsilylgruppene hydrolyseres så under milde betingelser for ikke å forstyrre esterbindingen eller taksansubstituentene.

Både omdannelsen av alkoholen til metallalkoksidet og den endelige syntese av taksanderivatet kan finne sted i den samme reaksjonsbeholder. p-laktamet tilsettes fortrinnsvis til reaksjonsbeholderen etter dannelse av metallalkoksidet i denne.
Foreliggende oppfinnelse tilveiebringer også farma-søytiske preparater som inneholder en forbindelse med formel (3) generelt, og særlig forbindelsene med formlene (4) og (5), i kombinasjon med ett eller flere farmakologisk akseptable, inerte eller fysiologisk aktive fortynningsmidler eller adjuvanser.
Disse preparatene kan presenteres i hvilken som helst form som er passende for den påtenkte administrasjonsvei. Den parenterale vei, og spesielt den intravenøse vei, er den fore-trukne administrasjonsvei.
Preparatene ifølge oppfinnelsen for parenteral administrering kan være vandige eller ikke-vandige, sterile oppløsninger, suspensjoner eller emulsjoner. Propylenglykol, vegetabilske oljer, spesielt olivenolje, og injiserbare organiske estere, f.eks. etyloleat, kan anvendes som oppløs-ningsmiddel eller bærer. Disse preparatene kan også inneholde adjuvanser, spesielt fuktemidler, emulgeringsmidler eller dis-pergeringsmidler. Steriliseringen kan utføres på flere måter, f.eks. ved å bruke et bakteriologisk filter, ved å inkorporere steriliseringsmidler i preparatet, ved bestråling eller ved oppvarming. De kan også være i form av sterile, faste preparater som kan oppløses eller dispergeres i sterilt vann eller i ethvert annet injiserbart, sterilt medium.
Produktene med generell formel (3) anvendes nærmere bestemt i behandlingen av akutte leukemier og faste tumorer, ved daglige doser som generelt er mellom 1 og 2 mg/kg via den intravenøse vei (perfusjon) for et voksent menneske.
De følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
Fremstilling av N- debenzovl- N- metoksykarbonyl- taksol
Til en oppløsning av 7-trietylsilyl-baccatin III (100 mg, 0,143 mmol) i 1 ml THF ved -45 °C ble det dråpevis tilsatt 0,087 ml av en 1,63 M oppløsning av nBuLi i heksan. Etter 0,5 time ved -45 °C ble en oppløsning av cis-l-metoksykarbonyl-3-trietylsilyloksy-4-fenylazetidin-2-on (252 mg, 0,715 mmol) i 1 ml THF tilsatt dråpevis til blandingen. Opp-løsningen ble varmet opp til 0 °C og holdt ved den temperaturen i 1 time før 1 ml av en 10 % oppløsning av AcOH i THF ble tilsatt. Blandingen ble fordelt mellom mettet, vandig NaHC03og 60/40 etylacetat/heksan. Inndamping av det organiske lag gav en rest som ble renset ved filtrering gjennom silikagel, hvorved man fikk 129 mg av en blanding inneholdende (2' R, 3' S) -2 ' , 7- (bis) trietylsilyl-N-debenzoyl-N-metoksykarbonyl-taksol og en liten mengde av (2'S,3'R)-isomeren.
Til en oppløsning av 129 mg (0,112 mmol) av blandingen erholdt fra den tidligere reaksjon i 6 ml acetonitril og 0,3 ml pyridin ved 0 °C ble det tilsatt 0,9 ml 48 % vandig HF. Blandingen ble omrørt ved 0 °C i 3 timer, så ved 25 "C i 13 timer og fordelt mellom mettet, vandig natriumbikarbonat og etylacetat. Inndamping av etylacetatoppløsningen gav 108 mg materiale som ble renset ved hurtigkromatografi, hvorved man fikk 90 mg (76 %) N-debenzoyl-N-metoksykarbonyl-taksol, som ble rekrystallisert fra metanol/vann.

Eksempel 2
Fremstilling av N- debenzoyl- N- etoksykarbonyl- taksol
Til en oppløsning av 7-trietylsilyl-baccatin III (100 mg, 0,221 mmol) i 2 ml THF ved -45 °C ble det dråpevis tilsatt 0,136 ml av en 1,63 M oppløsning av nBuLi i heksan. Etter 0,5 time ved -45 °C ble en oppløsning av cis-l-etoksykarbonyl-3-trietylsilyloksy-4-fenylazetidin-2-on (386 mg,
1,11 mmol) i 2 ml THF tilsatt dråpevis til blandingen. Oppløs-ningen ble varmet opp til 0 "C og holdt ved den temperaturen i 1 time før 1 ml av en 10 % oppløsning av AcOH i THF ble tilsatt. Blandingen ble fordelt mellom mettet, vandig NaHC03og 60/40 etylacetat/heksan. Inndamping av det organiske lag gav en rest som ble renset ved filtrering gjennom silikagel, hvorved man fikk 252 mg av en blanding inneholdende (2'R,3'S)-2',7-(bis)trietylsilyl-N-debenzoyl-N-etoksykarbonyl-taksol og en liten mengde av (2'S,3'R)-isomeren.
Til en oppløsning av 252 mg (0,112 mmol) av blandingen erholdt fra den tidligere reaksjon i 12 ml acetonitril og 0,6 ml pyridin ved 0 °C ble det tilsatt 1,8 ml 48 % vandig HF. Blandingen ble omrørt ved 0 °C i 3 timer, så ved 25 °C i 13 timer og fordelt mellom mettet, vandig natriumbikarbonat og etylacetat. Inndamping av etylacetatoppløsningen gav 216 mg materiale som ble renset ved hurtigkromatografi, hvorved man fikk 155 mg (85 %) N-debenzoyl-N-etoksykarbonyl-taksol, som ble rekrystallisert fra metanol/vann.
Sm.p. 161,5-162,5 °C; [a]<25>Na-62,2° (c 0,51, CHC13).

Eksempel 3
Tubulinbindingsanalyser ble utført ved å bruke forbindelsene (4) og (5) i det vesentlige som angitt i Parness et al., J. Cell. Biology, 91, s. 479-487 (1981), og det ble sammenlignet med taksol og taksoter. Resultatene er vist i tabell 1.
Eksempel 4
IC50-data ble erholdt in vitro på en human kreft-cellelinje (HCT 116), som er tilgjengelig fra National Cancer
Institute, og en multilegemiddelresistent cellelinje (HCT/VM46), som er resistent overfor mange forskjellige hydro-fobe midler, inkludert taksol. Cytotoksisitet ble fastslått i HCT 116 og HCT VM46 humane tykktarmkarsinomceller ved hjelp av analyse med XTT (2,3-bis(2-metoksy-4-nitro-5-sulfofenyl)-5-[ ( fenylamino)karbonyl]-2H-tetrazolium-hydroksid) (Scudiero et al., "Evaluation of a soluble tetrazolium/formazan assay for cell growth and drug sensitivity in culture using human and other tumor cell lines", Cancer Res., 48, s. 4827-4833, 1988). Celler ble platet ut ved 4000 celler/brønn i 96-brønners mikrotiterplater, og 24 timer senere ble legemidler tilsatt og seriefortynnet. Cellene ble inkubert ved 37 °C i 72 timer, og på dette tidspunkt ble tetrazoliumfargestoffet XTT tilsatt. Et dehydrogenaseenzym i levende celler reduserer XTT til en form som absorberer lys ved 450 nm, som kan kvantifiseres spektro-fotometrisk. Dess større absorbansen er, dess høyere er antal-let levende celler. Resultatene er uttrykt som en IC50-verdi, som er den legemiddelkonsentrasjon som kreves for å inhibere celleproliferasjon (dvs. absorbans ved 450 nm) til 50 % av proliferasjonen til ubehandlede kontrollceller. Resultatene er vist i tabell 2. Lavere tall indikerer høyere aktivitet.
