Abstract
Foreliggende oppfinnelse tilveiebringer en fremgangsmåte for kontroll av områdeinndelingen av et rekombinant protein mellom supernatanten og periplasmaet i E Coli-vertscellekulturer, hvori ekspresjon av det rekombinante protein i nevnte celler er under kontroll av et induserbart system, hvorved fremgangsmåten omfatter a) tilveiebringelse av en E. Coli-vertscellekultur b) forandring av vekstraten til E. Coli-vertscellene c) induksjon av ekspresjon av det rekombinante protein hvori trinn (b) og (c) kan utføres i enhver rekkefølge eller samtidig og etter hverandre d) bestemmelse av utbytte av rekombinant protein i kultursupernatanten og E. Coli-vertscelleperiplasma e) sammenligning av utbyttet bestemt i trinn (d) med utbyttet bestemt når minst én annen vekstrate har blitt anvendt i trinn (b) f) utvelgelse av en vekstrate fra sammenligningen utført i trinn (e) hvorved områdeinndelingen av det rekombinante protein mellom supernatanten og periplasmaet er mest egnet for den primære utvinning av det rekombinante protein.
