Abstract

Настоящее изобретение охватывает способ регулирования распределения рекомбинантного белка между супернатантом и периплазмой в культурах клеток-хозяев Е coli, отличающийся тем, что экспрессия рекомбинантного белка в указанных клетках осуществляется под контролем индуцибельной системы, причём указанный способ включает: а) получение культуры клеток Е. coli; б) изменение скорости роста клеток-хозяев Е. coli; в) индуцирование экспрессии рекомбинантного белка, при этом стадии (б) и (в) можно осуществлять в любом порядке или одновременно; и затем г) определение выхода рекомбинантного белка в культуральном супернатанте и в периплазме клетки-хозяина Е. coli; д) сравнение выхода, определённого на стадии (г), с выходом, определённым в том случае, когда на стадии (б) используют по меньшей мере одну иную скорость роста; е) выбор в результате сравнения, сделанного на стадии (д), такой скорости роста, при которой распределение рекомбинантного белка между супернатантом и периплазмой наиболее соответствует первичной регенерации рекомбинантного белка.
