Description
Oppfinnelsen vedrører nye amino-funksjonaliserte akryl-kopolymerer, oppnådd ved funksjonalisering med etylendiamin på R3-substituenten i de kopolymerer som er dannet ved å kopolymerisere bestanddelene I, II og III, hvor bestanddel I består av 2-50 vekt% av en monomer (bindemiddel) som tilsvarer
et N,N'-diakryloyldiaminoalkan med formel
hvor n2 = 1 eller 2.
De nye kopolymerer i henhold til oppfinnelsen stammer fra
dem som er beskrevet i patentsøknad nr. 82.3864 ved funksjonalisering ved hjelp av etylendiamin. Kopolymerene i henhold til den tidligere søknad - som man kan betegne A eller P-COR, hvor R = H eller -CH3 - oppnås ved kopolymerisering av de tre følgende monomerer:
I - En monomer som er kjede i bæreren og som består av et N-
akryloylpolymetylenimin med følgende formel:

Det karakteristiske ved kopolymerene i henhold til oppfinnelsen fremgår av patentkravet.
Funksjonaliseringen for oppnåelse av kopolymerene i henhold til oppfinnelsen foregår ved å skape en arm ved amidifisering med etylendiamin, slik at B dannes:
Denne amidifisering kan utføres på en av følgende måter:
a) ut fra syrefunksjonen ( A, R = H) , ved kobling med etylendiamin ved hjelp av et av de reagenser som det er kjent å anvende i henhold til litteraturen: dicykloheksylkarbodiimid (DCC) i nærvær av et aktiveringsmiddel, f.eks. 1-hydroksy-benzotriazol (HOBt), eller benzotriazol-l-yl-oksy-tris(dime-tylamino)fosfonium-heksafluorfosfat (Bop); b) ut fra esterfunksjonen (A, R = CH3), ved røring av harpiksen ved omgivelsestemperatur sammen mtd et overskudd av vannfritt etylendiamin.
Med denne sistnevnte metode har et systematisk studium av de eksperimentelle betingelser gjort det mulig å begrense de sekundære reaksjoner med hensyn til brodannelse og bevare 90 % av den innledende funksjonaliseringsgrad.
Peptidsyntesemetoden i fast fase, som er utviklet siden 1962 ved hovedsakelig å anvende polystyrenbærere (harpiks fra Merrifield, harpiks med benzhydrylamin, alkoksybenzylharpiks, harpiks (PAM,...) er velkjent og har vært gjenstand for tallrike arbeider (f.eks. Erickson & Merrifield i Neurath og Hill, The Proteins, vol. 2, Academic Press, 1976, s.255; Birr, Aspects of the Merrifield peptide synthesis, Springer 1978; Barany og Merrifield i Gross og Meienhofer. The peptides : analysis, synthesis, biology, Vol. 2, del A, Academic Press, 1980, s.l). Harpiksen, passende funksjonalisert, forestres eller amidifise-res.med N-beskyttet aminosyre som er opptatt i peptidet for å syntetisere den C-'endesté ende posisjon. Etter avbeskyttelse av aminfunksjonen utføres koblingen, ved anvendelse av en av de tallrike metoder som er beskrevet i litteraturen,- med den N-beskyttede aminosyre som opptar den nest siste posisjon i sekven-sen for syntetisering. Disse operasjoner gjentas til det på harpiksen oppnås den ønskede peptid-kjededannelse. I siste trinn spaltes peptidet fra sin bærer.
Denne fremgangsmåte presenterer visse fordeler i forhold til syntese i løsning: spesielt enkelheten og hurtigheten ved prosessen (det er ikke lenger nødvendig å isolere og rense peptidet etter hver kobling) , lett fjerning av reagensene som er i overskudd og forurensninger ved filtrering og vasking av bæreren, fravær eller sterk reduksjon av racemisering. Det er ellers nødvendig, for hver kobling, å nå frem.til et utbytte som er praktisk talt kvantitativt; hver ufullstendig reaksjon fører i virkeligheten til en akkumulasjon av avstumpede peptid-kjeder som kontaminerer sluttproduktet. Derav følger nødvendig-heten for'å anvende betydelige overskudd av reagenser og å gjenta hver koblingsoperasjon helt til en passende analysetest (f.eks. med ninhydrin) indikerer at enhver fri NH2~funksjon er forsvunnet på harpiksen-. Slike forsøksbetingelser er imidlertid å fraråde når syntesen innbefatter anvendelse av ikke-na-turlige aminosyrer, eventuelt sjeldne og kostbare sådanne.
For å dra nytte av de -fordeler som er assosiert med de reaksjoner som intervenerer i homogen fase, er det foreslått å anvende løselige polymerer (kfr. Mutter og Bayer, i Gross og Meienhofer, The peptides: analysis, synthesis, biology, vol. 2, del A, Academic Press, 1980, s.285), og spesielt polyetylengly-kol. Denne teknikk er imidlertid begrenset anvendbar, sannsyn-lig på grunn av eksperimentelle problemer som skyldes: - nedsettelse av løseligheten som kan ledsage forlengelsen av peptidkjeden, - isoleringen av bæreren etter hver kobling, hvilket nød-vendiggjør ultrafiltrering (Mutter, Hagenmaier og Bayer, Angew. Chem. Int. Ed., 1971, 10, 811; Bayer, Gatfield, Mutter og Mutter, Tetrahedron, 1978 , _3_4» 1829), eller en krystallisa-sjon ved tilsetning av eter, etter eventuell fjerning av løs-ningsmidlet (Bayer og Mutter, Chem. Ber., 1974 , 1Q7 , 1344 ; .Mutter, Uhmann og Bayer, Ann. Chem., 1975, s.90l; Corring og Jung, Ann. Chem.-, 1975 , s.1765 og 1776); - spaltning av peptidet fra bæreren (Tjoeng, Tong og Hodges, J.Org. Chem.,1978 , 4_3, 4190; Tjoeng og Hodges, Tetrahedron Letters, 1979, s.1273; Mutter, Bayer og Gatfield, J.Org. Chem. , 1980, 4_5, 5364) .
De samme vanskeligheter påtreffes i det tilfelle hvor man har med løselige bærere å gjøre på basis av polystyren, som er svakt eller ikke tverrbundet, hvilket innebærer gelfiltrering (Birr, i "Peptides 19.72", North Holland Publ. , 1973 , s.72; Andreatta og Rink, Heiv.Chim.Acta, 1973 , 56_, 1205) eller en utfelling med alkohol (Narita, Bull.Chem.Soc. Japan, 1978 , 51 , 1477).
Endelig er teknikken med alternerende væskefase/fast fase (Frank, Meyer og Hagenmaier, Chem.Ze.it., 1977, 101, 188) like-ledes lite utviklet.
Ganske nylig er overlegenheten ved polyakrylbærerne som Pap de Walter • (J.Amer.Chem.Soc., 1979, 101, 5383) og harpiksen til Sheppard (JCS Perkin 1, 1981, s.529 og 538), som har svel-leegenskap i et meget større område av løsningsmidler enn sty-renpolymerene, sannsynliggjort (Atherton, Gait, Sheppard og Williams, Bioorg.Chem., 1979, 8, 774). Det finnes spesielt: en meget lett spaltning ved slutten av syntesen, mellom poly-peptidet og dets bærer; en bedre forlikelighet mellom solvate-ringsegenskapene til bæreren og peptiddelen som er i ferd med å vokse, hvilket er av naturen å begrense avstumpingene og de avvikende koblinger. Den gunstige innflytelse av solvateringen hos harpiks/peptidsystemet på utbyttet av koblingene er spesielt vist (Atherton, Woolney og Sheppard, Chem.Comm., 1980, s. 970). Forøvrig gjelder fortsatt de kommentarer som vi allerede har formulert for polystyrenharpiksenes vedkommende, hva angår anvendelsen av et stort overskudd av reagenser (av størrelses-orden 2,5 ganger) og nødvendigheten for generelt å gjenta hver koblingsoperasjon flere ganger. I denne henseende er det signi-fikant at de nylig foretatte synteser av polypeptider med lang kjede atter benytter løsningsmetoden (f.eks. Kenner, Rakage og Sheppard, Tetrahedron, 1979 , 35.' 2^67; Nagaray og Balaran, Tetrahedron, 1981, 37, 1263; Fujii og Yajima, JCS Perkin 1, 1981, s.789 og 804).
For å motvirke solvatiseringsforskjellene som allerede er nevnt foran, mellom matriksen og polypeptidkjeden som er i ferd
■ med å vokse, er en ny polyakrylamidgel nylig foreslått (R.Epton og A.Williams, Inst.J.Biol.Måcromol., 1981, 3, 334). På denne bærer er den steriske hindring slik at denne harpikstype bare er blitt anvendt for syntese av korte peptider. Kort sagt nød-vendiggjør den endelige avbeskyttelse meget lang behandlings-tid med bortrifluorid (flere dager).
Som man vil se i det følgende, tillater de bærere som anvendes ved foreliggende oppfinnelse betydelig forenkling og forbedring av betingelsene ved peptidsyntesen.
For å kunne foreta en peptidsyntese er det nødvendig å tilveiebringe en modifikasjon av harpiksen i henhold til oppfinnelsen ved å innføre et reversibelt forankringssystem..
Som ikke begrensende eksempel nevnes at det er mulig å anvende et reagens av type C (Mitchell, Erickspn , Ryabtsev, Hodges og Merrifield, J.Amer.Chem.Soc., 1976; Stahl, Walter og Smith, J.Amer.Chem.Soc., 1979, 101, 5383; Atherton, Logan og Sheppard, JCS Perkin I, 1981, s.538; Atherton, Hubscher, Sheppard og Wooley, 2, Physiol.Chem. 1981, 362, 833).

Reaksjonen mellom B og C for å gi D:
kan utføres praktisk talt kvantitativt ved hjelp av et av de ovenfor nevnte koblingsmidlereller ved innvirkning av II på en aktivert ester (fremstilt med 2 ,4,5-triklorfenol, pentaklor-fenol,....) av III, eventuelt i nærvær av en katalysator
(HOBT, BOP),
Et annet reversibelt forankringssystem er realisert ved anvendelse av et reagens av type E (R = H eller CH^) som er kon-densert med B sl"ik at man får F:
Denne type forankring, allerede anvendt på polystyrenhar-pikser (Mitzoguchi, Shigezan og Takamura, Chem.Pharm.Bull.,
1970, 18, 1465; Wang, J.Org.Chem., 1976, 41, 3258), er original når det gjelder polyakrylbærer.
Fikseringen av den endestående aminosyre C i første trinn, samt den endelige spaltning av peptidet fra dets bærer utføres
under de samme betingelser som angitt ovenfor.
De således funksjonaliserte og anvendte harpikser i peptidsyntese har en funksjonaliseringsgrad som ligger mellom 0,2 og 5 mekv. pr. gram, fortrinnsvis mellom 1,0 og 1,2 mekv. pr. gram tørr bærer.
Fikseringen på harpiksen av den endestående aminosyre C, hvis NH2 er beskyttet f.eks. med den acido-labile gruppe t-bu-tyloksykarbonyl (Boe) eller med den base-labile gruppe fluorenyl-metyloksykarbonyl (Fmoc) kan utføres på forskjellige måter:
- ved mellomproduktet cesiumsalt for X = Cl eller Br,
- ved innvirkning av dicykloheksylkarbodiimid (DCC) i nærvær av 4-dimetylaminopyridin (DMAP) som katalysator, for
X' = 0H,
- eller, hvis X = 0H, ved ekstemporært å fremstille det symmetriske anhydrid av den N-beskyttede aminosyre, og ved å omsette'D i nærvær av DMAP. Den endelige spaltning av peptidet fra dets bærer utføres: - ved en mineralbase i fortynnet løsning når Y er en mety-lengruppe eller når man anvender F; eller NH^/CH^OH er det mulig å oppnå et endestående amid C; - med en organisk syre, f.eks. triflucreddiksyre i fortynnet løsning, når Y representerer en gruppe -CH2_0-.
Metodikk ved peptidsvntesen
De bærere som anvendes ved foreliggende oppfinnelse har den egenskap at de viser god permeabilitet og at de sveller særlig i et stort område av protiske og aprotiske løsningsmidler, såvel som i vandige puffere av forskjellig slag.
Under disse betingelser under en kobling, oppfører bærer-harpiksen av N-Boc eller N-Fmoc, endestående aminosyrer C (eller av peptidet under utarbeidelsen og passende beskyttet) som en "pseudo-løsning", og den således sammensatte gel er ufiltrer-bar. Dette skaper betingelser som er spesielt gunstige, da de nærmer seg betingelsene for homogen fase, for dannelse av pep-tidvinningen. Når reaksjonen er avsluttet, fremkaller tilset-■ ning av et tredje løsningsmiddel (eter, THF, dioksan, ...) ut-drivning av løsningsmidlet som gjennomtrenger harpiksen., og utfelling av denne i en form som kan filtreres lett og hurtig,
i motsetning til hva som skjer i henhold til teknikkens stand.
Disse- spesielle og _spesifikke egenskaper ved harpiksene i henhold til oppfinnelsen fører til betydelig økning av hastig-heten (multiplisert med minst tre ganger i forhold til hva som er kjent) og utbyttet ved koblingene. Av dette blir resulta-tet for hver enkelt kobling at man får mulighet til særlig å redusere verdien av forholdet mellom antall mol N-beskyttet aminosyre og funksjonaliseringsgrad for bæreren, nemlig 1,1-1,2, mens de vanlige verdier er av størrelsesorden 2,5. Forøvrig er det aldri tilstopping av kolonnene under intermediære filt-reringer .
Alterneringen mellom kobling i kvasi-homogen fase og utfelling av bæreren utgjør således en ny metodikk, forenklet og forbedret peptidsyntese i fast fase.
Fremgangsmåten for kobling kan f.eks. utføres på følgende måte: 1. Tilsetning av et inert løsningsmiddel, f.eks. CHCl^, CH2Cl2 DMF for å svelle harpiksen som er bærer for N-Boc, endestående aminosyre C (eller det passende beskyttede peptid); 2. En vasking ved hjelp av en lineær eller cyklisk eter, f. eks. etylest-er, isopropyleter, TH F eller dioksan; 3. Tilsetning av trifluoreddiksyre i løsningsmidlet for å be-virke avbeskyttelse av NH2; 4. Utfelling av harpiksen ved tilsetning av eter som under punkt 2; 5. Filtrering fulgt av fire vaskinger med eter som under punkt 2; 6. Nøytralisering ved tilsetning av diisopropyletylamin i et inert 'løsningsmiddel; 7. Fire vaskinger med eter, som under punkt 2, deretter tørk-ing i en nitrogenstrøm; 8. Kobling ved agitering med en løsning, i klorert løsnings-middel, av det symmetriske anhydrid (fremstilt ekstemporært) av en N-Boc aminosyre; 9. Utfelling ved tilsetning av eter som under punkt 2, og filtrering, samt
10. fire vaskinger med eter som under punkt 2.
Denne nye metodikk er testet med hell i syntesen av pep-t^der som har biologisk interesse, f.eks. et undekapeptid Ac-Gly-Gly-Lys-Gly-Gly-Hla-Arg-Lys-Val-Leu-Homo-Ser.
Eksempel
Syntese av peptidet Ac- Gly- Glv- Lys- Gly- Gly- Ala- Arq- Lys-Val- Leu- Homo- Ser.
Peptidet syntetiseres på en akrylharpiks oppnådd ved å kopolymerisere N-akrylylpyrrolidin, etylenbisakrylamidet og N-akrylderivatet av metylesteren av a-aminokapronsyre. Bæreren funksjonaliseres deretter på følgende måte: Til 1 g harpiks (funksjonalisert med CC^CH^) tilsettes 32 ml redestillert etylendiamin.. og dette røres én natt ved omgivelsestemperatur. Man filtrerer, vasker med avmineralisert vann til nøytralitet og deretter med etanol og eter. Harpiksen tørkes til slutt i 24 h på dampbad under vakuum; den inneholder da 1 mekv. av NH2 pr. gram tørr polymer.
Det ble anvendt 2 g harpiks, f.eks. 2 mekv. av NH2• Ope-ras jonssyklusen som er nødvendig for å tilveiebringe en aminosyre på harpiksen er som følger:
Bekreftelse på utførelse av koblingen ved hjelp av Kaiser-testen med ninhydrin.
Som den eksperimentelle prosess foreskriver, er det systematisk utført to koblinger for hver innført aminert syre. Koblingsreaksjonen utføres ved hjelp av symmetriske anhydrider som fremstilles på følgende måte: 1 mmol av den beskyttede aminerte syre oppløses i 20 ml vannfritt CH Cl2/ løsningen avkjøles til 0° og tilsettes 0,5 mmol dicykloheksylkarbodiimid i løsning i 10 ml CH2C12. Etter 20 minutters reaksjon ved 0°C filtreres utfellingen av dicyklohek-sylurinstoff som skylles med 20 ml CH2C12, løsningene forenes og bringes i kontakt med harpiksen i reaktoren.
Ved denne metode har man kunnet fremstille de symmetriske anhydrider ut fra Boe Gly OH, NE Z Na Boe Lys OH Boe Val OH,
Boe Leu OH, Boe Met OH,\og Ac- Gly t0H. Når,, det gjelder N N0o ,

N Boe Arg OH, oppløses produktet i minst mulig DMF, og volumet fylles opp til 20 med tørt metylenklorid. Så utføres reaksjo-
nen som beskrevet ovenfor.
Etter kobling av den siste aminosyre tørkes harpiksen under vakuum i nærvær av P2<~>5 i ^ timer og bringes så i suspensjon i 50 ml trifluoreddiksyre. Man lar i 2 timer en svak strøm av tørt HBr gjennpmperle. Man filtrerer og skyller rikelig med CH2C12 og Et20. Denne acidolyse avbeskytter sidekjedene til lysinene.
Det oppnådde rød-oransje pulver bringes deretter i suspensjon i 100 ml 2 M trifluoreddiksyre, og man tilsetter 5 g sink-pulver. Etter 12 timers agitering ved omgivelsestemperatur vasker man rikelig med 2N HC1, med vann, etanol og til slutt med vannfri etyleter. Denne hydrogenolyse spalter nitrogengruppen i guanidinogruppen i argininet.
Etter tørking av harpiksen under vakuum og over P205 i 12 timer fremkommer 1 gram polymer, som behandles med 2,5 g cyanogen-bromid i 70 ml propionsyre. Etter 24 timers reaksjon ved omgivelsestemperatur -lyofyliserer man og oppnår det ønskede peptid med et utbytte på 4 5 %.
Claims (1)
- Amino-funksjonaliserte akryl-kopolymerer som er oppnådd ved funksjonalisering med etylendiamin på R3-substituenten i de kopolymerer som er dannet ved å kopolymerisere bestanddelene I, II og III, hvor bestanddel I består av 2-50 vekt% av en monomer (bindemiddel) som tilsvarer et N .t^-diakryloyldiaminoalkan med formelhvor n2 = 1 eller 2,karakterisert ved at det som bestanddeler II og III er anvendt - II) 30-90 vekt% av en monomer (matriks) som tilsvarer etN-akryloylpolymetylenimin med formelhvor R x = H eller -CH3Z = —(CH?) — hvor ni = 4, 5 eller 6 eller —(CH2)2—X—(CH2)2 med X = 0 eller N—CH3 eller et N-akryloyldialkylamid med formelhvor Ri har samme betydning som ovenfor, R2 = —CH3 eller —C2H5» 0<? -III) 2-65 vekt% av en monomer (funksjonaliseringsmiddel) som tilsvarer en akryloylaminosyre eller ester eller racemat med formelhvor Rx = H eller -CH3R3 = H eller -CH3n3 = 1, 2, 3 eller 5eller en asymmetrisk N-akryloylaminosyre (eller ester) (L-serie) med formel:hvor R^ og R3 har samme betydning som ovenfor,eller N-akryloyl(L)prolin eller dets metylester med formelhvor Ri og R3 har samme betydning som ovenfor.
