De werkwijze ter bereiding van deze nieuwe acylderivaten heeft gewoonlijk plaats bij een temperatuur tussen 0°C en 80°C in een watervrije omgeving en bij aanwezigheid van een organisch op-20 losmiddel gekozen uit de groep bestaande uit trifluorazijnzuur en de onder formule 1 hiervoor vermelde lijst zuren (h) tot en met (i) overeenkomend met het gebruikte zuurchloride. Wanneer het zuurchloride vast is en niet gemakkelijk oplosbaar in het organische oplosmiddel, is het mogelijk de oplosbaarheid ervan op een zodanige 25 wijze te verbeteren, dat een homogene fase verkregen wordt door toevoeging van een kleine hoeveelheid van een gechloreerd oplosmiddel, zoals chloroform of watervrij dichloormethaan.
In het bijzonder dient er op gelet te worden, dat de omgeving watervrij wordt gehouden door het reactiesysteem te beschermen met 30 geschikte dehydrateringsmiddelen, bijvoorbeeld calciumchloride bevattende buizen.
Aan het einde van de reactie wordt het verkregen mengsel gekoeld en gewoonlijk met aceton behandeld; de eventueel afgescheiden vaste stof wordt verwijderd, terwijl het precipitaat, dat door 35 toevoeging van diethylether wordt gevormd, verzameld wordt.
Het precipitaat kan gezuiverd worden door kristallisatie met diethylether. In het algemeen zijn een of twee kristallisaties voldoende voor het verkrijgen van een produkt met grote zuiverheid, hetgeen gemakkelijk gecontroleerd kan worden door dunne-kO laag-chromatografie (D.L.C.) onder toepassing van siliciumoxide- 8101067 * % 3 , platen en verschillende elueermiddelen zoals CHCl^-MeOH-gecenc.

In het algemeen varieert de reactieopbrengst van 60 tot 85 % 5 met dien verstande, dat de eventuele opbrengst-verlaging, die plaats kan hebben tijdens de zuivering door kristallisatie, niet in overweging is genomen.
De volgende voorbeelden lichten naast de opstelling van de verschillende fysisch-chemische gegevens van enkele verbindin-10 gen van de onderhavige uitvinding, het syntheseproces zonder de uitvinding te beperken, toe.
Voorbeeld I
Bereiding van tert.butylacetylcarnitinechloride (verbinding met formule 2) 3 15 1*97 g (0,01 mol) carhitinechloride werden in 10 cnr tri- fluorazijnzuur opgelost. Aan de oplossing werden 1,½ cm (0,01 mol) tert.butylacetylchloride langzaam onder roeren toegevoegd. Het reactiemengsel werd ^8 uren bij kamertemperatuur geroerd. Aan het mengsel werd diethylether toegevoegd en het aldus verkregen neer-20 slag werd afgefiltreerd. Het onzuivere produkt werd gekristalliseerd uit isopropanol-diethylether, waarbij een zuiver produkt werd verkregen. Smeltpunt 16½ - 165°C.
Opbrengst 80 %.

(a) Bereiding van cyclohexylacetylchloride (verbinding met formule 3) 3 30 2,5 g (0,02 mol) cyclohexylazijnzuur werden met 3,7 cm (0,05.mol) S0C12 gemengd en de verkregen oplossing werd 90 minuten op 80°C gehouden. Het mengsel werd onder een verminderde druk geconcentreerd en enkele wasbehandelingen werden uitgevoerd met watervrije tolueen ter verwijdering van S0Cl2· Vervolgens werd 35 het mengsel onder een verminderde druk gedroogd en het bovengenoemde produkt werd verkregen als een grondstof, die als zodanig 8101067 3 *·' 5 k bij de volgende trap (b) werd gebruikt.
(b) Bereiding van cyclohexylacetylcarnitinechloride (verbinding met formule 4) 2*97 S (O»01 mol) carnitinechloride werden in 10 cm tri-5 fluorazijnzuur opgelost· Aan deze oplossing werd 0,01 mol cyclo-hexylacetylchloride, bereid zoals hiervoor beschreven, toegevoegd en het verkregen mengsel werd ij-8 uren geroerd. Daarna werd diethyl-ether toegevoegd en het verkregen mengsel werd 30 minuten bij 0°C geroerd. Een neerslag werd verkregen, dat werd afgefiltreerd en 10 onder een verminderde druk werd gedroogd.

Bereiding van oyclopentaanpropionylcarnitinechloride 1>97 g (0,01 mol) carnitinechloride werden in 10 cm^ tri-fluorazijnzuur opgelost, aan de verkregen oplossing werden 1,60 g (0,01 mol) 3-cyclopentaanpropionylchloride langzaam onder roeren 20 toegevoegd. Het verkregen mengsel werd een nacht bij 45°C geroerd. Vervolgens werd het mengsel gekoeld, werden i*0 ml aceton toegevoegd en werd het mengsel 2 uren in ijs geroerd. Het aldus gevormde neerslag werd afgefiltreerd en aan het filtraat werd diethyl-ether toegevoegd. Bereiding van vinylacetylcarnitinechloride (verbinding met formule 5)
Aan een oplossing van 2 g (0,01 mol) carnitinechloride in 5 6 ml trifluorazijnzuur werden 1,8 ml (0,02 mol) vinylacetylchlori- de onder roeren bij kamertemperatuur toegevoegd. Het reactiemeng-sel werd vervolgens op 50°C gebracht en een nacht onder reactie-omstandigheden gehouden· Vervolgens werd het mengsel tot kamertemperatuur gekoeld, in 200 ml diethylether gegoten en 20 minuten ge-10 roerd. De etherfase werd gedecanteerd en het aldus verkregen neerslag werd opgenomen met acetonitrile en de verkregen oplossing werd 2 uren bewaard· Het niet-omgezette carnitine, dat werd afgescheiden in de vorm van een kristallijne vaste stof, werd afgefiltreerd en aan het filtraat werd diethylether toegevoegd· Het aldus 15 gevormde neerslag was een witte kristallijne vaste stof.

Bereiding van ethoxyacetylcarnitineisobutylester (a) Bereiding van carnitineisobutylester.
30 10 g (0,05 mol) carnitinechloride werden in 100 ml isobutanol gesuspendeerd. De suspensie werd met een ijsbad gekoeld en gasvormig HCl werd erdoor geborreld tot volledige verzadiging bereikt was· Het verkregen mengsel werd 2 uren onder terugvloeiing verhit.
Het mengsel werd vervolgens geconcentreerd om de alcohol te verwij-35 deren, het concentraat werd in gedestilleerd water opgelost en de oplossing werd geneutraliseerd met IE-45 hars. Het verkregen produkt werd gelyofiliseerd, waarbij 12 g carnitineisobutylester wer- 8101067 _ ............*’ 6 den verkregen.
(b) Bereiding van ethoxyazijnzuurchloride.
1.1 en? (0,0125 mol) thionylchloride werden aan 1,5 cm^ (0,012 mol) ethoxyazijnzuur toegevoegd. Het verkregen mengsel werd 5 12 uren op kamertemperatuur gehouden· Het reactiemengsel werd driemaal gewassen met een mengsel van chloroform en watervrije diethylether en vervolgens onder een verminderde druk van 11,1 kPa bij 50°C geconcentreerd· 1,15 g ethoxyazijnzuurchloride werden verkregen.
10 (c) Reactie tussen carnitineisobutylester en ethoxyazijnzuur chloride.
1.1 g (0,043 mol) carnitineisobutylester werden in watervrije aceton opgelost en. aan de verkregen oplossing werden 1,15 g (0,009 mol) ethoxyazijnzuurchloride toegevoegd. Het reactiemengsel 15 werd gedroogd en het residu werd in een atmosfeer van inert gas (argon) twee dagen bij kamertemperatuur gehouden. Het residu werd vervolgens gekristalliseerd uit isopropanol-diethylether. Het beoogde produkt werd verkregen in een opbrengst van 65 %· D.L.C. 3° Bereiding van 3-cyclohexen.ylcarboxylcarnitinechloride (a) Bereiding van 3-cyclohexeencarboxylchloride 3 3 1.2 cnr (0,01 mol) cyclohexeencarbonzuur werden met 0,9 cnr 8101067 η 7 (0,01 mol) oxalylchloride gemengd en het verkregen mengsel werd 3,5 uren bij kamertemperatuur geroerd·
Vervolgens werd de Oplossing onder een verminderde druk van 13*3 kPa bij 70°C geconcentreerd. Het aldus verkregen onzuivere 5 produkt werd als zodanig bij de volgende trap gebruikt.
(b) Reactie van 3-cyclohexeencarbonzuurchloride met carnitine-chloride.
1 ε (0,05 mol) carnitinechloride werd in 2 cm^ trifluorazijn-zuur opgelost. Aan de verkregen oplossing werd langzaam onder roe-10 ren bij kamertemperatuur 0,01 mol van het bij de voorafgaande trap verkregen chloride toegevoegd. Het verkregen reactiemengsel werd 13'uren geroerd. Diethylether werd vervolgens aan het reactiemengsel toegevoegd en een neerslag werd verkregen, dat vervolgens tweemaal met diethylether werd gewassen om de overmaat zuurchloride te 15 verwijderen. Het neerslag werd afgefiltreerd en onder een verminderde druk gedroogd. Opbrengst: 90 Sé.

Bereiding van methoxyacetylcarnitinechloride
Een mengsel van 3 ml (0,Öif mol) methoxyazijnzuur en 3 ml 25 (0,03 mol) methoxyacetylchloride werd 2 uren bij 30°C geroerd.
Aan dit mengsel werden if g (0,02 mol) carnitine chloride, dat vooraf was gedroogd, toegevoegd· Het verkregen mengsel werd op ifO°C gebracht en 7 dagen geroerd. Vervolgens werd het mengsel opgenomen met acetonitrile en het niet-omgezette carnitinechloride werd af-30 gefiltreerd. Na toevoeging van tert.butylmethylether sloeg een olie neer. De olieprecipitatie werd viermaal herhaald met acetonitrile/ tert.butylmethylether. Bereiding van ethoxyacetylcarnitinechloride 5 Een mengsel van 5 ml (0,07 mol) ethoxyazijnzuur en 1,8 ml (0,015 mol) ethoxyacetylchloride, dat bereid was zoals beschreven in trap (b) van voorbeeld V, werd 2 uren bij 80°C geroerd· Aan dit mengsel werden 2 g (0,01 mol) carnitinechloride, dat vooraf was gedroogd, toegevoegd· Het verkregen mengsel werd eerst op 80°C ge-10 houden tot volledige oplossing van carnitine had plaats gevonden, gevolgd door 8 dagen op 40°C. Een verdere hoeveelheid ethoxyacetylchloride (0,9 ml, 0,007 mol) werd toegevoegd en het verkregen mengsel werd 7 dagen op 40° C gehouden· Na toevoeging aan het mengsel van tert.butylmethylether sloeg een olie neer· De olieprecipitatie 15 werd viermaal herhaald met acetonitrile/tert.butylmethylether tot volledige verwijdering van de overmaat ethoxyazijnzuur. 1,5 g van de beoogde verbinding werden op deze wijze verkregen#

De kenmerken van de farmacologische activiteit van de verbindingen met de algemene formule 1 zijn hierna toegelicht.
De acute toxiciteit van de verbindingen met de algemene for-25 mule 1 werd onderzocht bij muizen onder toepassing van de methode van Weil (Weil C*S.» Biometr. J. 8, (1952) 249· De in tabel A hierna gegeven LD^Q-waarden laten zien, dat de verbindingen een goede tolerantie hebben.
Het cardiokinetische effect werd onderzocht bij konijne-harten, 30 die geïsoleerd waren volgens de methode van Langendorff. Volgens deze methode geïsoleerde konijne-harten werden bij 38,2°C overgoten met een met zuurstof behandelde Ringer oplossing. De isometri-sche contracties, het elektro-cardiogram en de coronaire stroming werden geregistreerd onder toepassing van een "Battaglia-Rangoni” 8101067 10 polygraaf. Door verwijdering van de zuurstof uit de perfusie-vloeistof werd een metabolische beschadiging teweeg gebracht in de hartspier tot een reductie van 80 % in de cardiale contractie-kracht.
5 Onder deze omstandigheden van voortgezette anoxie wordt de aerobe glycolyse van het myocardium vertraagd met de begeleidende vorming van zure catabolieten tengevolge van zowel de accumulatie van pyrodruivezuur en de omzetting ervan tot melkzuur, die niet nuttig gebruikt kunnen worden vanwege de verlaging van pyridine- 10 enzymen» zoals LDH (lactodehydrogenase). Dit heeft repercussies „ een op de anaerobe glycolyse onder beïnvloeding vanysteeds toenemend aantal enzymen, vergezeld van een progressieve en toenemend kritische uitputting van het myocardium. Derhalve komt een gehele \ reeks hartspier-vermoeidheidsniveaus voor, die kunnen worden waar-15 genomen door het gedrag van de onderzochte parameters, namelijk de contractiekracht, de coronaire stroming, het harttempo en het hartritme. Zodra de contractiekracht met 80 % verminderd was, werd de perfusie-vloeistof opnieuw met zuurstof behandeld hetzij zonder toevoeging van andere verbindingen (controle) hetzij met 20 toevoeging van de onderzochte verbindingen.
Tabel B geeft de percentage-waarden van de contractiekracht van het hart, waarbij een positief inotroop effect wordt getoond, berekend na 10 minuten vanaf de onderbreking van de anoxie periode (myocardisch herstel).
25 Het anti-aritmische effect van de verbindingen werd eveneens bij muizen onderzocht volgens de methode van P.W. Nwangwu en T. Holcslow (P.W. Nwangwu, T.L. Holcslow; Arch. Int. Pharmacodyn. 229, (1977) 219* Onder toepassing van aconitine (5 //ml) als aritmogeen middel, werden de veranderingen in het hartritme van de 30 dieren geregistreerd en de aanvangstijd van initiële aritmie en/of ventriculaire tachycardie werd (en) als eindpunt gebruikt. De resultaten zijn in tabel C samengevat.
Het effect van de verbindingen op de modificatie van het lipoproteïne-patroon alsmede op de niveaus van plasma cholesterol 35 en triglyceriden, gewijzigd door orale toediening van olijfolie, werd onderzocht bij normaal gevoederde ratten, oraal behandeld met _Ί 15 ml kg olijfolie 1 uur voordat de verbinding oraal werd toege -diend en met de verbindingen bij verschillende concentraties.
Het meest belangrijke effect, 2 uren na de toediening van ifO olijfolie, dat een toename in triglyceriden en cholesterol met zich 8101067 11
bracht met vermindering van α-lipoproteïnen en toename van de β- en voor-β-fractieSj werd goed geantagoneerd door sommige verbindingen, die» zoals aangegeven in tabel D, bewezen in staat te zijn de voorgaande parameters» die in ruime zin de grenzen van de 5 statistische betekenis bereiken, tot normaal terug te brengen. FARMACEUTISCHE PREPARATEN
1. Oplossingen en steriele waterige oplossingen, die acyl-carnitinen met formule 1'in concentraties van 25 mg tot 500 mg per ml bevatten.
5 a) Het versnijdingsmiddel voor injecteerbare ampullen/flesjes wordt bereid volgens de volgende niet-beperkende samenstelling: natriumcarboxymethylcellulose 10 mg/ml (met lage viscositeit) 10 polysorbaat 80 ^ mg/ml propylparaben 0, if mg/ml water voor injecties voldoende voor 1 ml, 2 ml, 5 ml en 10 ml ampullen/flesjes.
b) Het versnijdingsmiddel voor fleboclyse flessen, die 50 ml, 15 100 ml, 250 ml, 500 ml en 1000 ml bevatten, wordt bereid vol gens de volgende niet-beperkende samenstelling:
NaCl 8,6 g/1 KC1 0,3 g/1
CaCl2 0,33 g/1 20 water voor injecties voldoende voor 1 liter.
c) Het versnijdingsmiddel voor flessen voor oraal gebruikt, die 5 ml tot 100 ml bevatten, wordt bereid volgens de volgende niet-beperkende samenstelling: mannitol . 11 mg/ml 25 sorbitol 6OO mg/ml natriumbenzoaat 3 mg/ml sinaasappelextract 200 mg/ml vitamine B^ 3 mcg/ml voldoende gezuiverd water.
* 30 2. Tabletten, die 20 mg tot 500 mg acyl-carnitine met formule 1 bevatten. Het versnijdingsmiddel wordt bereid volgens de volgende niet-beperkende samenstelling: zetmeel k5 % avicel if5 % 35 talk 10 % 3. Capsules, die 20 mg tot 500 mg acyl-carnitine met formule 1 zonder versnijdingsmiddel bevatten.
k· Aerosol en spuitpreparaten met 500 mg tot 10 g van een acyl-carnitine met formule 1. Het versnijdingsmiddel wordt bereid ifO volgens de volgende niet-beperkende samenstelling: 8101067 17 ethanol 30 % gezuiverd water 30 % voldoende freon 12/11 if (gelijke delen) i 8101067
Claims (13)
1· Acylderivaten van carnitine met de algemene formule 1, waarin X“ een anion is» bij voorkeur een halogenide of sulfaatanion, en 5 R het radikaal is van de volgende organische zuren: (a) onverzadigde organische zuren» (b) verzadigde organische zuren gesubstitueerd met een tertiaire alkylgroep, (c) verzadigde organische zuren gesubstitueerd met een cycloalkyl- 10 groep, (d) verzadigde organische zuren gesubstitueerd met een cycloalke-nylgroep, (e) verzadigde organische zuren gesubstitueerd met een alkoxy-groep,
15 Cf) verzadigde organische zuren gesubstitueerd met een carbalko-xygroep, (g) verzadigde, met hydroxy gesubstitueerde organische zuren, (h) verzadigde organische zuren gesubstitueerd met een aldehyd-groep en 20 (i) verzadigde organische zuren gesubstitueerd met een hetero cyclische groep.
2. Acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat het zuur (a) gekozen is uit de groep bestaande uit acrylzuur, vinylazijnzuur en allylazijnzuur. 25 3· Acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat het zuur (b) gekozen is uit de groep bestaande uit tert.butylazijnzuur en tert.butylpropionzuur. k· Acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat het zuur (c) gekozen is uit de groep 30 bestaande uit cyclopentaancarbonzuur, cyclopentaanazijnzuur, cyelopentaanpropionzuur, cyclohexylazijnzuur en cyclohexylboter-zuur.
5· Acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat het zuur (d) gekozen is uit de groep 35 bestaande uit 3-cyclohexeen-carbonzuur en 2-cyclopenteenazijnzuur.
6. Acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat het zuur (e) gekozen is uit de groep bestaande uit methoxyazijnzuur en ethoxyazijnzuur.
7· Acylderivaten van carnitine volgens conclusie 1, met ifO het kenmerk, dat het zuur (f) gekozen is uit de groep 8101067 19 . bestaande uit 3-carbometh.oxypropi onzuur ea 4-carbomethoxyboter-zuur.
8. Acylderivaten van carnitine volgens conclusie 1, met bet kenmerk» dat het zuur (g) gekozen is uit de groep 5 bestaande uit 2-hydroxy-isoboterzuur, 2-hydroxy-isovaleriaanzuur, 2- hydroxy-isocapronzuur* 2-hydroxy-2-methylboterzuur, 2-methyl- 3- hydroxypropionzuur, 2-hydroxy-tert.butylazijnzuur, 3-hydroxy- 3-methylglutaarzuur (monoester), 3-hydroxy-2-methylglutaarzuur (monoester) en 3-hydroxypropionzuur. 10 9· Acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat het zuur (h) gekozen is uit de groep bestaande uit 2-formylpropionzuur en formyl-isoboterzuur.
10. Acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat het zuur (i) gekozen is uit de groep 13 bestaande uit 1,2-dithiolan-3-pentaanzuur, 2-thiofeencarbonzuur en 2-thiofeenazijnzuur.
11. Acylderivaten van carnitine volgens conclusies 1 tot 10, met het kenmerk, dat de derivaten in hun optisch actieve vormen zijn.
12. Acylderivaten van carnitine volgens conclusies 1 tot 10, met het kenmerk, dat de derivaten in hun racemische vorm zijn. 13* Farmaceutisch aanvaardbaar zout van de acylderivaten van conclusies 1 tot 12. 25 14· Farmaceutisch preparaat voor de behandeling van hart ziekten, hyperlipoproteïnemieën en hyperlipidemieën, met het kenmerk, dat het preparaat een therapeutisch werkzame hoeveelheid van een of meer acylderivaten van carnitine volgens conclusies 1 tot 12, bevat. 30 15· Farmaceutisch preparaat voor de behandeling van hart ziekten, hyperlipoproteïnemieën en hyperlipidemieën, met het kenmerk, dat het preparaat een therapeutisch werkzame hoeveelheid van een farmaceutisch aanvaardbaar zout volgens conclusie 13 bevat.
16. Werkwijze ter bereiding van acylderivaten van carnitine volgens conclusie 1, met het kenmerk, dat men (1) volgens op zichzelf bekende methoden het chloride van het overeenkomstige zuur (a) tot en met (i) bereidt, en (2) het chloride van trap (1) omzet met carnitine-chloride bij 40 aanwezigheid van een organisch oplosmiddel bij een tempera- 8101067 tuur tussen ongeveer kamertemperatuur en ongeveer 80°C.
17· Werkwijze volgens conclusie 16* met het kenmerk, dat men als organisch oplosmiddel trifluorazijnzuur en de zuren (a) tot en met (i) overeenkomend met het zuurchloride •5 van trap (1) toepast.
18.
