Abstract
La presente invención se refiere a métodos para producir proteínas heterólogas. En particular, la presente divulgación provee métodos mejorados para producir proteínas deseadas, que incluyen proteínas con múltiples subunidades tal como anticuerpos, con un rendimiento mayor y pureza mejorada. En formas de realización ejemplificativas, las células transformadas son una levadura, por ejemplo, levadura metilotrófica tal como Pichia pastoris. Un método para producir un complejo recombinante de múltiples subunidades en células de levadura, caracterizado porque comprende: (a) cultivar a una primera temperatura las células de lavadura que comprenden uno o más genes que codifican el complejo de múltiples subunidades; y (b) cultivar las células de levadura a una segunda temperatura y permitir que las células de levadura produzcan el complejo de múltiples subunidades.
