Abstract

La presente invención se refiere a un método para la producción de una proteína recombinada que comprende (a) la fermentación de una célula huésped procariota, donde dicha célula huésped procariota ha sido transformada con un ácido nucleico que codifica dicha proteína recombinada, y (b) la cosecha de dicha proteína recombinada en condiciones donde los niveles de dO2 son superiores a 0%, y (c) la purificación de dicha proteína recombinada hasta un volumen filtrado, donde dicho volumen filtrado no contiene una cantidad detectable de aducto de DHNA-proteína recombinada, medida por un ensayo de IEC a 310 nm. La presente invención además contempla un método para la producción de una proteína recombinada que comprende (a) la fermentación de una célula huésped procariota con deleción del gen menE, donde dicha célula huésped procariota ha sido transformada con un ácido nucleico que codifica dicha proteína recombinada, (b) la cosecha de dicha proteína recombinada; y (c) la purificación de dicha proteína recombinada hasta un volumen filtrado, donde dicho volumen filtrado no contiene una cantidad detectable de aducto de DHNA-proteína recombinada, medida por un ensayo de IEC a 310 nm, y donde el rendimiento de proteína recombinada se incrementa alrededor de 20% o más.
