Abstract
La presente invención se refiere a un ensayo altamente sensible que combina enriquecimiento inmunomagnético con citometría multiparamétrica o citometría de imágenes para detectar, enumerar y caracterizar células carcinomas en la sangre; la presente invención incorpora la conjugación de distintos anticuerpos al mismo ferrofluido; esto tiene el efecto de hacer al ferrofluido poliespecífico con respecto a los antígenos que el ferrofluido va a unir; los múltiples anticuerpos presentes en el mismo ferrofluido no parecen bloquearse o, de cualquiera otra forma, obstaculizarse entre sí; estos ferrofluidos tienen el muy deseable efecto de poder unirse específicamente a más de un tipo de célula; el ensayo es especialmente útil para permitir la captura de las CTC que tienen baja expresión de EpCAM, pero alta expresión de otros marcadores de tumores; por consiguiente, el ensayo facilita la caracterización biológica y la estadificación de células carcinomas. Figura 1B.
