Abstract
La presente invención se relaciona con métodos para desarrollar células T diseñadas para inmunoterapia y más específicamente a métodos para modificar células T al inactivarse en genes de punto de control inmune, de preferencia al menos dos seleccionados de diferentes rutas para incrementar la actividad inmune de las células T. este método implica el uso de endonucleasas específicas de corte raro, en particular TALE-nucleasas (endonucleasas efectoras de TAL) y polinucleótidos que codifican tales polipéptidos para dirigir de forma precisa una selección de genes clave en células T, las cuales se encuentran disponibles a partir de donadores o del cultivo de células primarias. La invención abre la forma para estrategias de inmunoterapia adoptiva altamente eficientes para tratar cáncer e infecciones virales.
