Abstract
La presente invención refiere a la preparación de péptidos insulinotrópicos que se sintetizan utilizando una técnica en fase sólida y en solución ("híbrida") Generalmente la técnica incluye la síntesis de tres fragmentos intermediarios de péptido diferentes utilizando la química en fase sólida. Luego se utiliza la química en fase de solución para añadir material aminoácido adicional al tercer fragmento que entonces se acopla al segundo fragmento y luego al primer fragmento en solución. Alternativamente, un segundo fragmento diferente se acopla al primer fragmento en la fase sólida. Luego, se utiliza la química en fase de solución para añadir material aminoácido adicional a un tercer fragmento diferente. A continuación, este tercer fragmento diferente se acopla al primero acoplado y al segundo fragmento diferente en la fase de solución. La utilización de una pseudoprolina en uno de los fragmentos facilita la síntesis en fase sólida de ese fragmento y también facilita el subsiguiente acoplamiento en fase de solución de este fragmento a otros fragmentos. La presente invención es muy útil en la obtención de péptidos insulinotrópicos tal como el GLP-1 (7-36) y sus homólogos naturales y no naturales.
