Abstract

본 발명은 발효 과정에서 형성딘 산성 및/또는 염기성 성분을 발효 육즙 추출물 또는 모액의 농축물의수-불혼화성 용액으로부터 수성 염기 또는 산 추출 과정을 사용하여 제거하고 비극성 성분을 선택적 용해도에 의해 마크롤라이드로부터 분리함으로써 마크롤라이드 항생물질, 특히 트리사이클릭 마크롤 라이드, 예를 들어 라파마이신을 상기 농축물로부터 회수하는 방법에 관한 것이다.

Claims (11)

  1. 마크롤라이드를 함유하는 발효 육즙 추출물 또는 모액의 농축물 중에 존재하는 산성, 염기성 및 비극성 중성 불순물로부터 마크롤라이드를 분리하는 방법에 있어서,
    a. 수-불혼화성 용매중의 상기 농축물의 용액을 수성 염기로 추출하여 모든 산성 불순물을 실질적으로 제거하는 방법;
    b. 수-불혼화성 용매중의 상기 농축물의 용액을 수성 산으로 추출하여 모든 염기성 불순물을 실질적으로 제거하는 방법;
    c. 상기 농축물의 용액을 비방향족 탄화수소 용매로 처리하여 비극성 중성 불순물을 제거하는 방법 중 1개 이상의 방법을 어떠한 순서로든 포함하는 방법.
  2. 제1항에 있어서, 마크롤라이드-함유 발효 육즙 추출 농축물 또는 모액 농축물 중의 비극성 중성성분이 제1용매중의 상기 농축물의 용액을, 제1용매와 불혼화성이고 마크롤라이드가 불용성인 제2비방향족 탄화수소 용매로 추출함으로써 제거되는 방법.
  3. 제1항에 있어서, 산성 및/또는 염기성 성분을 추출한 후 마크롤라이드를 함유하는 수-불혼화성 용액을 충분한 양의 혼화성 마크롤라이드 비용매로 처리하여 마크롤라이드가 생성된 용매 혼합물중에서 불용성이 되게 하고, 이렇게 하여 용액으로부터 분리되게 함으로써 마크롤라이드가 용매 혼합물로부터 분리되는 방법.
  4. 제1항에 있어서, 산성 및/또는 염기성 성분을 추출한 후에 마크롤라이드를 함유하는 수-불혼화성 용액을 농축시키고, 잔사를 결정화 용매 또는 용매 혼합물과 혼합하여 결정화하는 방법.
  5. 제1항에 있어서, 산성 및/또는 염기성 성분을 추출한 후에 마크롤라이드를 함유하는 수-불혼화성 용액을 농축시키고, 잔사를 제1용매에 용해시키고, 제1용매와는 불혼화성이고 마크롤라이드가 불용성인 제2비방향족 탄화수소 용매로 추출하여 중성 비극성 불순물을 제거하는 방법.
  6. 제1항에 있어서, 발효 육즙 추출 농축물 또는 모액 농축물을 우선 제1용매에 용해시키고, 제1용매와 불혼화성인 제2비방향족 탄화수소 용매로 추출하여 비극성 중성 불순물을 제거하고, 이어서 제1용매중의 용액을 농축시키고, 잔사를 수-불혼화성 용매중에 용해시키고 수성 염기 및/또는 산으로 추출하여 산성 및/또는 염기성 불순물을 제거하는 방법.
  7. 제1항에 있어서, 마크롤라이드가 트리사이클릭 마크롤라이드인 방법.
  8. 제7항에 있어서, 트리사이클릭 마크롤라이드가 라파마이신 또는 이의 자연발생 동족체 또는 유사체 또는 FK-506 또는 이의 자연발생 동족체 또는 유사체인 방법.
  9. 제8항에 있어서, 트리사이클릭 마크롤라이드가 라파마이신, 32-데스메틸라파마이신 또는 15-데옥소라파마이신인 방법.
  10. 제8항에 있어서, 트리사이클릭 마크롤라이드가 라파마이신인 방법.
  11. 제8항에 있어서, 트리사이클릭 마크롤라이드가 FK-506인 방법.
