Abstract

(구성)
이하의 (1)∼(4)의 성질을 갖는 모노클론성 항체, 이 모노클론성 항체를 생산하는 하이브리도마, 이 모노클론성 항체를 사용하는 체액으로부터의 호염기구의 분리방법, 호염기구로부터의 케미칼 메디에이터의 유리 방법 및 호염기구로부터의 케미칼 메디에이터의 유리 시험관.
(1)호염기구와 반응하며, (2)담체 고상화후도 항체활성을 유지하고, (3)이 호염기구의 IgE를 통한 특이적인 케미칼 메디에이터 유리를 저해하지 않으며 (4)이 호염기구의 비특이적인 케미칼 메디에이터 유리를 유발하지 않는다.
(효과)
본 발명의 모노클론 항체는 IgE를 통한 특이적인 케미칼 메디에이터 유리시험에 적합한 호염기구의 분리에 이용할수가 있다.

호염기구에 결합하는 모노클론성 항체, 호염기구의 분리방법, 호염기구로부터의 케미칼 메디에이터의 유리방법, 및 호염기구 유래 케미칼 메디에이터의 유리 시험법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 자성 구슬에 고상화시킨 본 발명의 모노클론성 항체를 사용하여 분 리한 호염기구로부터의 총 히스타민 양(백색 막대 그래프), IgE를 통한 특이적인 히스타민 유리양(사선 막대그래프), 및 비특이적인 히스타민 유리양(흑색 막대 그래프)을 나타낸 그래프이며,
제2도는 전혈 및 BA312 고상화 자성구슬에 의한 분리세포를 사용한 히스타민 유리 반응의 모노클론성 항인체 IgE항체 농도 의존 곡선이며,
제3도는 BA312 고상화 자성 구슬에 의한 소아 아토피성 피부염 환자 혈액 유래 분리세포를 사용한 히스타민 유리반응의 난백 알레르겐 농도 의존 곡선이며,
제4도는 방사선 면역 검정법에 의해 얻어진 히스타민 농도의 표준 곡선이며, B/Bo는 표지된 결합 피검체의 %를 나타낸다.

Claims (12)

  1. 하기의 성질을 갖는 모노클론성 항체. (1)호염기구와 반응하며, (2)담체 고상화후도 항체활성을 유지하고, (3)이 호염기구의 IgE를 통한 특이적인 케미칼 메디에이터 유리를 저해하지 않으며 (4)이 호염기구의 비특이적인 케미칼 메디에이터 유리를 유발하지 않는다.
  2. 제1항에 있어서, 모노클론성 항체 BA2O, BA135, 및 BA312로 이루어진 군으로부터 선택된 모노클론성 항체.
  3. 제1항 또는 2항의 모노클론성 항체를 담체에 고상화시킨 고상화 모노클론성 항체.
  4. 제3항에 있어서, 이 담체가 자성입자인 고상화 모노클론성 항체.
  5. 제1항 또는 2항의 모노클론성 항체를 생산하는 하이브리도마.
  6. 제5항에 있어서, 하이브리도마 BA2O 시오노기, BA101 시오노기, BA135 시오노기, 및 BA312 시오노기로 이루어지는 군으로부터 선택된 하이브리도마.
  7. 제3항 또는 4항의 고상화 모노클론성 항체와 체액을 반응시켜, 이 체액중의 호염기구를 이 고상화 모노클론성 항체에 결합시켜서 포착하는 것을 특징으로 하는 호염기구의 분리 방법.
  8. 제7항의 방법에 의해 분리한 호염기구를 알레르겐 또는 항 IgE항체와 반응시키는 것을 특징으로 하는 이 호염기구로부터 케미칼 메디에이터의 유리방법.
  9. 하기의 공정을 갖는 호염기구로부터의 케미칼 메디에이터의 유리방법. (1)청구항 3 또는 4의 고상화 모노클론성 항체와 체액을 반응시켜, 이 체액중의 호염기구를 이 고상화 모노클론성 항체에 결합시켜서 포착하고, 이 호염기구를 분리하는 공정. (2)분리한 호영기구를 알레르겐 또는 항 IgE항체와 반응시켜서 호염기구로부터 케미칼 메디에이터를 유리시키는 공정. (3)유리한 케미칼 메디에이터의 양을 측정하는 공정.
  10. 제9항에 있어서, 케미칼 메디에이터가 히스타민인 케미칼 메디에이터의 유리 시험법.
  11. 제9항에 있어서, 케미칼 메디에이터가 로이코트리엔인 케미칼 메디에이터의 유리 시험법.
  12. 제9항에 있어서, 케미칼 메디에이터가 혈소판 활성화 인자(PAF)인 케미칼 메디에이터의 유리 시험법.
