Abstract

본 발명은 혈액 샘플에서 특별한 헤모글로빈 유도체의 함량을 측정하는 방법에 관한 것이다. 특히, 본 발명은, 혈액중의 유도 헤모글로빈의 비율을 측정하기 위해, 전체 헤모글로빈 함량 및 헤모글로빈 유도체 함량의 분리측정을 필요로 하는, 글리세이트 헤모글로빈과 같은 헤모글로빈 유도체의 함량의 측정방법 및 이것을 위한 적합한 용혈시약에 관한 것이다.

Description

헤모글로빈 유도체의 면역학적 측정방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 A1에 대한 전체 헤모글로빈 농도의 보정곡선.
제2도는의 함량에 대한 흡광도차 △A=A3-k·A2의 보정곡선.
제3도는측정에서 피펫 계량 순서에 따른 감도를 나타낸 그래프.

Claims (17)

  1. (a) 혈액샘플을 5 내지 9.5의 pH를 갖은 이온성 청정제를 함유하는 용혈시약으로 처리하고, (b) 용혈된 혈액샘플에서 헤모글로빈 유도체를 면역학적으로 측정하는, 혈액샘플 중의 헤모글로빈 유도체의 함량의 측정방법.
  2. 제1항에 있어서, 혈액샘플을 4 내지 37℃에서 10분 이하 동안 용혈 시약으로 처리함을 특징으로 하는 방법.
  3. 제1항 또는 2항에 있어서, 용혈시약이 비-이온성 청정제를 더 함유함을 특징으로 하는 방법.
  4. 제1항 또는 2항에 있어서, 비이온성 또는 양쪽성 이온 청정제를 함유하는 반응 완충액이 용혈된 샘플에 첨가되고, 생성된 혼합물에서 헤모글로빈 유도체가 면역학적으로 측정됨을 특징으로 하는 방법.
  5. 제1항 또는 2항에 있어서, 헤모글로빈 유도체가임을 특징으로 하는 방법.
  6. (a) 혈액샘플을 5.0 내지 9.5의 ph를 갖는 이온성 청정제를 함유하는 용혈시약으로 처리하고, (b) 용혈된 샘플에서 전체 헤모글로빈 함량을 측정하고, (c) 용혈된 샘플에서 유도체를 측정하는, 혈액샘플중의 헤모글로빈 유도체 및 전체 헤모글로빈의 함량의 측정방법.
  7. 제6항에 있어서, 특이성 헤모글로빈 발색단의 특성 흡광도를 측정함으로써 전체 헤모글로빈 함량이 측정됨을 특징으로 하는 방법.
  8. 제6항 또는 7항에 있어서, 용혈 시약이 비-이온성 청정제를 더 함유함을 특징으로 하는 방법.
  9. 제6항 또는 7항에 있어서, 비-이온성 또는 양쪽성 이온 청정제를 함유하는 반응 완충액이 용혈된 샘플에 첨가되고, 생성된 혼합물에서 전체 헤모글로빈 및 헤모글로빈 유도체가 측정됨을 특징으로 하는 방법.
  10. 5 내지 9.5의 pH를 갖는 이온성 청정제를 함유하는 용혈시약.
  11. 제10항에 있어서, 비-이온성 청정제를 함유함을 특징으로 하는 용혈시약.
  12. 제10항 또는 11항에 있어서, 티올기의 산화성 공격에 근거한 작용 메카니즘을 갖는 안정한 산화제 또는 보존제가 포함됨을 특징으로 하는 용혈시약.
  13. 헤모글로빈 유도체의 면역학적 측정, 또는 샘플중의 헤모글로빈 유도체 및 전체 헤모글로빈 함량의 동시측정을 위한, 제10항에 따르는 용혈시약의 용도.
  14. 하나의 패키지가 제10항에 따르는 용혈시약을 함유하고, 나머지 팩키지가 헤모글로빈 유도체의 면역학적 검출을 위해 필요한 시약으로 이루어진 추가의 시약 혼합물을 함유하는, 2개 이상의 분리 팩키지로 구성되는, 헤모글로빈 유도체의 면역학적 검출을 위한 테스트 킷.
  15. 제14항에 있어서, 면역학적 시험에 필요한 시약이 시험 운반체에 도포됨을 특징으로 하는 테스트 킷.
  16. 완충액으로서 pH5.0 내지 8.0의 MES완충액이 사용되고 용액이 EDTA를 함유하는, 글리코실레이트화 단백질, 펩티드 또는 폴리헵텐의 용액의 안정화 방법.
  17. 제16항에 있어서, BSA가 첨가됨을 특징으로 하는, 용액의 안정화 방법.
