Abstract
본 발명은 연속 공정을 사용하여 단일 반응 용기내에서 뇨 단백질 및 크레아티닌을 측정하는 방법에 관한 것이다. 이 방법은 단백질에 대한 면역 검정을 수행하기에 적합한 수준으로 pH를 조정하여 면역 응집 기술로써 단백질을 측정하는 것을 포함한다. 크레아티닌을 측정하기에 적합한 수준으노 pH를 상승시킴으로써 응집 반응으로 유발된 혼탁도가 소산되므로, 크레아티닌 측정을 지체없이 수행할 수 있다.

제4도는 단백질/크레아티닌 검정을 수행하기 위한 배열(A)의 반응용기를 도시한 것이다.
제5도는 단백질/크레아티닌 검정을 수행하기 위한 배열(B)의 반응 용기를 도시한 것이다,
제6도는 크레아티닌 검측 시약 및 알칼리성 시약의 용해후 후면으로 위치를 회전시킨 단백질/크레아티닌 검정용 반응 용기를 도시한 것이다,
제7도는 단백질 검정이 완료된 후, 다시 우측으로 135°회전시킨 단백질/크레아티닌 검정용 반응 용기를 도시한 것이다,
제8도는 반응 용기내에서 통상적으로 반응 채널(21)에 위치하고 있는 제1반응 영역(24)를 도시한 것이다.
Claims (15)
- a) 단백질 및 크레아티닌을 함유하는 용액을 제공하는 단계; 및 어느 순서로든지, b) 용액의 pH를, 단백질 농도에 대한 면역 검정을 수행하기에 적합한 수준으로 조정한 후 면역 검정기술로써 단백질 농도를 측정하는 단계; 및 7) 용액의 pH를, 크레아티닌에 대한 검정을 수행하기에 적합한 수준으로 조정한 후, 크레아티닌의 비색계 측정을 위한 시약을 첨가함으로써 뇨샘플중의 크레아티닌 농도를 측정하는 단계들을 포함하는, 수용액중의 단백질 및 크레아티닌을 측정하는 방법.
- 제1항에 있어서, 면역 검정을 수행하기 전에 pH를 약 7내지 9의 수준으로 조정하는 방법.
- 제1항에 있어서, 크레아티닌 검정을 수행하기 전에 PH를 약 11.5 내지 12.5의 수준으로 조정하는 방법.
- 제1항에 있어서, 크레아티닌의 비색계 측정용 시약이 피크르산 또는 3, 5-디니트로벤조산인 방법.
- 제1항에 있어서, 수성 용매가 뇨인 방법.
- 제1항에 있어서, 면역 검정이 응집 유형인 방법.
- 제6항에 있어서, 고려되는 단백질에 대해 특이적인 항체가 수현탁성 입자에 결합되는 방법.
- 제1항에 있어서, 단백질 농도를 먼저 적합한 pH에서 측정한 후, pH를 크레아티닌 측정에 적합한 수준으로 상승시키는 방법.
- 제8항에 있어서, 크레아티닌의 측정을, pH가 상승되는 시점으로부터 약 30분 이내에 개시하는 방법.
- 제9항에 있어서, 크레아티닌 시험을 약 1내지 5초 이내에 개시하는 방법.
- a) 뇨 단백질 및 크레아티닌을 함유하는 뇨 샘플을 제공하는 단계; b) 뇨의 pH를 약 7내지 9의 수준으로 조정한 후, 면역 검정 기술로써 단백질 농도를 측정하는 단계', c) 뇨의 pH를 약 11.5 내지 12.5의 수준으로 상승시키는 단계; 및 d) 크레아티닌의 비색계 측정용 시약을 첨가함으로써 뇨 샘플중의 크레아티닌 농도를 측정하는 단계들을 포함하여, 단일 반응 용기내에서 뇨 단백질 및 크레아티닌을 측정하는 방법.
- a) 뇨 단백질에 대해 특이적인 무수 면역시약과 혼입된 제1반응 영역과 반응 채널내로 도입된 반응 용액의 pH를 크레아티닌 측정에 적합한 수준으로 상승시킬 수 있는 무수 염기성 시약과 혼입된 제2반응 영역을 포함하는 분석 시약 반응 채널 및 실질적으로 수평의 회전축을 지닌 반응 용기를 제공하는 단계; b) 뇨 단백질 및 크레아티닌을 함유하는 것으로 여겨지는 뇨 샘플 및 뇨 단백질 농도를 측정하기에 적합한 pH로 완충된 반응 유액을 면역 혼탁계 수단으로써 반응 용기내에 도입하여 이를 건조된 면역 시약과 접촉시킴으로써 면역 시약을 용해시키고 면역 시약 및 뇨 단백질의 상호작용에 의해 반응 유액의 혼탁도가 증가되도록 하는 단계; c) 시간의 함수로서 혼탁도에 있어서의 변화를 기준으로 한 뇨 단백질의 농도를 측정하는 단계; d) 반응 유액 및 크레아티닌 측정용 시약을 제2반응 영역내에서 무수 염기성 시약과 접촉시킴으로써 이러한 염기성 시약을 용해시키고 반응 유액의 pH를 크레아티닌의 비색계 측정에 필요한 수준까지 상승시키는 단계; 및 e) 분광 광도계 수단으로써 크레아티닌의 농도를 측정하는 단계들을 포함하는, 단일 반응 용기내에서 뇨 단백질 및 크레아티닌을 측정하는 방법.
- 제12항에 있어서, 뇨 단백질 농도의 측정 이전에 pH를 약 7내지 9의 수준으로 조정하고 크레아티닌 측정 이전에 pH를 약 11.5 지 12.5의 수준으로 상승시키는 방법.
- 제12항에 있어서, 크레아티닌의 측정을, pH가 약 11.5 내지 12.5의 수준으로 상승되는 시점으로부터 약 30분 이내에 개시하는 방법.
- 제14항에 있어서, 크레아티닌의 측정을 약 1내지 5초 이내에 개시하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
