제1도는 교차 결합되지 않은 Fab<TSH>(커브 1), 교차결합된 Fab<TSH>(커브 2), Fab<TSH>/β-Gal의 결합체(커브 3) 및 Fab<TSH>써모(thermo)-BSA(커브 4)를 사용함으로써 TSH를 측정하는 캘리브래이션(calibration)커브이다.
제2도는 고정화 스트랩타비딘 및 비오티닐화 AK<TSH>, 교차결합되지 않은 스트랩타비딘(커브 1), 교차결합된 스트랩타비딘(커브 2), 스트랩타비딘/β -gal결합체(커브 3), 스트랩타비딘/BSA 결합체(커브 4), 스트랩타비닌/써모 -BSA 결합체(커브 P/5) 및 스트랩타비딘/써모-BSA 결합체(커브6)FMF 사용함으로써 TSH을 측정하는 캘리브레이션 커브이다.
Claims (15)
- 특이적으로 결합가능한 물질보다 더 소수성이며 약 500,000이상의 분자량을 지니는 가용성 단백질이 특이적으로 결합 가능한 물질에 결합되고 반응 성분과 단백질의 결합제가 소수성 고체상에 흡착되고, 특히 면역 검정 원리에 따라 이종 분석 방법에 사용하는, 불용성 담체 물질에 결합된 특이적으로 결합 가능한 단백질 물질의 제조방법.
- 제1항에 있어서, 가용성 단백질로서 500,000-20밀리온의 분자량을 지니는 단백질이 사용되는 방법.
- 제2항에 있어서, 가용성 단백질이 10,000-70,000분자량의 단백질로부터 분자량을 500,000-20밀리온까지 증가시킴에 의해 제조되는 방법.
- 전기한 항의 어느 한 항에 있어서, 결합전에 단백질이 이기능 또는 다기능 단백질 시약으로 원하는 분자량을 얻을 때까지 교차 결합되는 방법.
- 전기한 항의 어느 한 항에 있어서, 소수성화(hydrophobed)단백질이 사용되는 방법.
- 제5항에 있어서, 결합전에 단백질이 열을 사용해 산, 변성제 또는 캐오트로픽 이온으로 처리함에 의해 및/또는 소수서 화합물과 화학적으로 결합함에 의해 소수성 화되는 방법.
- 제4항에 있어서, 다숙신이미딜 수버레이트가 이기능 화합물로서 사용되는 방법.
- 제3항 내지 제7항의 어느 한 항에 있어서, 소의 혈청 알부민, 리파제 및/또는 면역 r-글로블린이 단백질로서 사용되는 방법.
- 제1항 내지 제8항의 어느 한 항의 방법으로 제조된, 불용성 담체 물질에 결합된 특이적으로 결합가능한 단백질.
- 약 500,000이상의 분자량을 갖는 소수성 단백질 및 특이적으로 결합가능한 단백질 물질의 결합체가 흉착되는 소수성 고체상으로 구성되는 제1항 내지 제8항의 어느 한 항에 따라 제조된 고체상 면역검정에 사용되는 담체 물질.
- 제19항에 있어서, 소수성 고체상이 폴리스티렌, 폴리메트아크릴레이트, 폴리아미드, 폴리테트라플루오로에틸렌 또는 스트렌과 아크릴로니트릴의 공중합체로 구성되는 담체 물질.
- 제10항 또는 제11항에 있어서, 단백질이 소수성화된 소의 혈청 알부민(써모-BSA), 소수성화된 리파제 또는 소수성화된 면역 r-글로블린인 담체 물질.
- 제10항 내지 제12항의 어느 한 항에 있어서, 특이적으로 결합가능한 물질이 항원 또는 항체인 담체 물질.
- 제10항 내지 제12항의 어느 한 항에 있어서, 특이적으로 결합가능한 물질이 스트랩타비딘 또는 아비딘인 담체 물질.
- 제10항 내지 제14항의 어느 한 항에 있어서, 담체가 양모상 유리섬유 또는 셀룰로스-/셀룰로스 에스테르 섬유 및 중합체 섬유로부터의 양모상 섬유인 담체 물질.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
