Claims (24)

  1. 브로모크시닐을 기질로 하여 최소한 약 0.1μmol NH3/분/mg 단백질의 비활성을 갖는 것을 특징으로 하는 약 34kd(정제 상태)의 실질적으로 정제된 세균성 니트릴라제.
  2. 3,5-디할로겐화-p-히드록시벤조니트릴에 특이적인 니트릴라제를 형질 발현시키는 외부 유전자를 갖는 것을 특징으로 하는 세균 숙주.
  3. 브로모크시닐을 기질로 하여 최소한 약 0.1μmol NH3/분/mg 단백질의 비활성을 갖는 것을 특징으로 하는 약 34 KDal의 니트릴라제를 함유하는 조성물.
  4. 제 3 항에 있어서, 니트릴라제가 세균성 니트릴라제임을 특징으로 하는 조성물.
  5. 제 4 항에 있어서, 세균성 니트릴라제가 클레브시엘라로 부터 얻은 것임을 특징으로 하는 조성물.
  6. 제 3 항에 있어서, 최소한 약 0.5μmol NH3/분/mg 단백질의 비활성을 가짐을 특징으로 하는 조성물.
  7. 3,5-디할로겐화-P-히드록시벤조니트릴에 특이적인 니트릴라제를 형질 발현시키는 외부 유전자를 갖는 것을 특징으로 하는 세균 숙주.
  8. 제 7 항에 있어서, 세균 숙주가 이ㆍ콜리임을 특징으로 하는 세균 숙주.
  9. 식물 세포내에서 작용하는 조절 구역의 전사 및 번역 조절 조정하에 있는 브로모크시닐 및/또는 이소크시닐에 특이적인 니트릴라제를 코딩하는 구조 유전자를 함유함을 특징으로 하는 형질 발현 카세트.
  10. 제 9 항에 있어서, 니트릴라제가 세균성 니트리라제임을 특징으로 하는 형질 발현 카세트.
  11. 제 9 항 또는 10항에 있어서, 카세트가 최소한 하나의 T-DNA 가장 자리를 갖는 것임을 특징으로 하는 형질 발현 카세트.
  12. 제 9 항 내지 11항에 있어서, 전사 개시 구역이 오파인을 코딩하는 유전자의 것임을 특징으로 하는 형질 발현 카세트.
  13. 제 9 항에 따른 형질 발현 카세트를 포함하는 이ㆍ콜리 및 에이, 투메파시엔스 중 최소한 하나에서 복재될 수 있는 플라스미드.
  14. 제13항에 있어서, 형질 발현 카세트가 최소한 하나의 T-DAN 가장자리를 가짐을 특징으로 하는 플라스미드.
  15. 클레브시엘라에서 발견되는 브로모크시닐 및/또는 이소크시닐에 특이적인 세균성 니트릴라제에 대한 야생형 전사 개시 구역이외의 전사 조절 구역의 조정하에 있는, 명세서에 나타낸 DNA 서열.
  16. 제15항에 있어서, 전사 조절 구역이 식물내에서 작용성을 갖는 것임을 특징으로 하는 DNA 서열.
  17. 제15항에 있어서, 전사 조절 구역이 세균 내에서 작용성을 갖는 것임을 특징으로 하는 DNA 서열.
  18. 야생형 전사 개시 구역 이외의 5'-말단에 3,5-디할로겐화-P-히드록시벤조니트릴에 특이적인 니트릴라제 효소를 코딩하는 오픈 리딩 프레임(open reading frame)을 갖는 DNA 서열.
  19. 제18항에 있어서, 니트릴라제 효소가 세균성 니트릴라제 효소임을 특징으로 하는 DNA 서열.
  20. 제19항에 있어서, 클레브시엘라의 DNA 서열과 실질적으로 일치하는 것임을 특징으로 하는 DNA 서열.
  21. 제 9 항 내지 12항에 따른 형질 발현 카세트를 포함하는 식물 세포.
  22. 제21항에 따른 식물 세포를 갖는 식물.
  23. 3,5-디할로겐화-P-히드록시벤조니트릴에 특이적인 니트릴라제를 생산하는 케이ㆍ오자에나에를 분리하고;케이ㆍ오자에나에를 적절한 영양 배지에서 생장시키고;상기 케이ㆍ오자에나에를 분해시키고 상기 니트릴라제를 분리함을 특징으로 하는 3,5-디할로겐화-P-히드록시벤조니트릴에 특이적인 니트릴라제를 제조하는 방법.
  24. 상술한 선택적인 특성을 갖는 세균을 스크리닝하고 상기 선택적인 특성을 갖는 상기 세균을 선택하고;상기 세균의 게놈을 절단하여 원하는 크기 범위의 단편을 수득하고;세균내에서 적절한 벡터에 상기 단편을 클로닝시키고 상술한 선택적 특성을 갖는 효소를 선택하고;상기 선택적 특성을 갖는 상기 효소를 형질 발현 하는 구조 유전자를 갖는 DNA 서열을 분리함을 특징으로 하는 선택적 특성을 갖는 효소를 수득하는 방법.
