실시예

실시예 1 - TLR7 효능제의 합성
본 실시예 및 도 1은 본 발명의 화합물의 합성에 관한 것이다.
MeOH (30 mL) 및 THF (15 mL) 중 10-메틸-10H-페노티아진-3,7-디카르브알데히드 1 (문헌 [Synthesis, 1998, 1107]에 따라 제조) (2.00 g, 7.43 mmol)의 현탁액을 0℃에서 NaBH4 (0.562 g, 14.85 mmol)로 조금씩 처리한 다음, 냉각을 계속하면서 45분 동안 교반하였다. LCMS은 반응이 완결되었음을 나타내었다. 반응물을 물로 켄칭하고, EtOAc (3x100 mL)로 추출하였다. 유기 추출물을 Na2SO4 상에서 건조시키고, 여과하고, 농축시켜 디메탄올 2 (2.03 g, 94% 수율)를 수득하였다. LCMS ESI: C15H15NO2S에 대한 계산치 = 274.1(M+H+), 실측치 274.0(M+H+).
디메탄올 2 (2.43 g, 8.89 mmol)를 N.N-디메틸포름아미드 (DMF, 30 mL) 중에 용해시켰다. 이미다졸 (2.421 g, 35.6 mmol)을 첨가하고, 이어서 t-부틸디메틸실릴 클로라이드 (TBS-Cl, 5 ml DMF 중 0.804 g, 5.33 mmol)을 0℃에서 적가하였다. 0℃에서 3시간 동안 교반한 후, LCMS에 따르면 반응이 완결되었다. 반응물을 물로 켄칭하고, EtOAc (4x50 mL)로 추출하였다. 유기 추출물을 Na2SO4 상에서 건조시키고, 여과하고, 농축시켰다. 조 생성물을 80 g 실리카 칼럼에 의해 EtOAc;헥산 (0-100% 구배)로 용리시키면서 정제하여 화합물 3 (1.57g, 45.6% 수율)을 수득하였다. LCMS ESI: C21H29NO2SSi에 대한 계산치 = 388.2(M+H+), 실측치 388.0(M+H+).
화합물 3 (510 mg, 1.316 mmol)을 디클로로메탄 (DCM, 7.5 mL) 중에 용해시키고, 이어서 디-이소프로필에틸아민 (DIPEA, 0.483 mL, 2.76 mmol)을 첨가하고, 이어서 메탄술포닐 클로라이드 (MsCl, 0.133 mL, 1.710 mmol)를 0℃에서 첨가하였다. 0℃에서 1시간 동안 교반한 후, LCMS에 따르면 반응이 완결되었다. DCM (10 mL) 및 물 (10 mL)을 첨가하고 혼합하였다. 물을 제거하고, 반응 혼합물을 물로 1회 더 세척하였다. 유기 층을 Na2SO4 상에서 건조시키고, 여과하고, 농축시켜 조 클로로메틸 화합물 4 (582 mg)를 수득하였으며, 이는 처음에 형성된 메실레이트가 클로로메틸 화합물로 계내 전환된 것이다.
클로로메틸 화합물 4를 탄산세슘의 존재 하에 화합물 5 (CAS 등록 번호 866268-31-7)와 반응시켜 화합물 6을 수득하였다.
화합물 6을 HCl로 처리하여 히드록시메탄올 Ia-01를 수득하였고, 이어서 이를 티오닐 클로라이드로 처리하여 클로로메틸 화합물 Ia-09로 전환시켰다.
마지막으로, 화합물 Ia-09를 시클로부탄아민과 반응시켜 화합물 Ia-06을 수득하였다. LCMS ESI: C28H33N7O2S에 대한 계산치 = 532.2(M+H+), 실측치 532.0(M+H+). 1H NMR (DMSO-d6) 7.16-7.07 (m, 4 H), 6.86 (dd, J = 12.5, 8.5 2 H), 6.56 (s, 2 H), 4.76 (s, 2 H), 4.18 (t, J = 6.5 Hz, 2 H), 3.51 (s, 2 H), 3.12 (p, J = 7.5 Hz, 1 H), 2.07-2.02 (m, 2 H), 1.70-1.50 (m, 6 H), 1.40 (h, J = 7.5 Hz, 2 H), 0.93 (t, J = 7.5 Hz, 3 H). 페노티아진 고리의 N-Me에 상응하는 신호는 물 억제로 인해 존재하지 않았다.
일반적으로 상기 절차에 따라, 화학식 (I)에 따른 추가의 화합물을 하기 표 B에 제시된 바와 같이 대안적 아민을 사용하여 제조하였다.
관련 기술분야의 기술자는 본 명세서의 다른 화합물이 변경을 가하여 상기 절차에 따라 제조될 수 있다는 것을 알 것이다. 예를 들어, 화합물

실시예 2 - TLR7 효능제 활성 검정
본 실시예는 본 명세서에 개시된 화합물의 TLR7 효능제 활성을 검정하는 방법을 기재한다.
인간 TLR7-분비 배아 알칼리성 포스파타제 (SEAP) 리포터 트랜스진을 보유하는 조작된 인간 배아 신장 블루 세포 (HEK-블루™ TLR 세포; 인비보젠(Invivogen))를 비-선택적인 배양 배지 (10% 소 태아 혈청 (시그마(Sigma))가 보충된 DMEM 고-글루코스 (인비트로겐(Invitrogen))) 중에 현탁시켰다. HEK-블루™ TLR7 세포를 384-웰 조직-배양 플레이트의 각 웰에 첨가하고 (웰 당 15,000개 세포), 16-18시간 동안 37℃, 5% CO2에서 인큐베이션하였다. 화합물 (100 nl)을 HEK-블루™ TLR 세포를 함유하는 웰에 분배하고, 처리된 세포를 37℃, 5% CO2에서 인큐베이션하였다. 18시간 처리 후, 10 마이크로리터의 새로 제조된 퀀티-블루™ 시약 (인비보젠)을 각각의 웰에 첨가하고, 30분 동안 인큐베이션하고 (37℃, 5% CO2), SEAP 수준을 엔비전 플레이트 판독기 (OD = 620 nm)를 사용하여 측정하였다. 절반 최대 유효 농도 값 (EC50; 검정 기준선과 최대값 사이의 반응 중간값을 유도하는 화합물 농도)을 계산하였다.
도 4는 화합물 Ia-04에 대해 수득된 데이터를 보여주는 대표적인 그래프이다.
실시예 3 -트랜스글루타미나제-매개 접합
하기 절차는 효능제-링커 화합물의 트랜스글루타미나제 매개 접합에 사용될 수 있으며, 여기서 링커는 아민 공여자로서 작용할 수 있는 아민 기를 갖는다. 항체는 트랜스글루타미나제-반응성 글루타민을 갖는 것, 예를 들어 N297A 또는 N297Q 치환을 갖는 것일 수 있다. 접합을 5:1의 항체:효소의 몰비를 갖는 재조합 박테리아 트랜스글루타미나제에 의해 수행한다. 접합을 50 mM 트리스 완충제, pH 8.0 중에서 표준 프로토콜을 사용하여 수행하고, 37℃에서 밤새 인큐베이션한다. 생성된 접합체를 50 mM 트리스, pH 8.0으로 예비 평형화된 단백질 A 칼럼 상에서 정제한다. 접합체를 0.1 M 시트르산나트륨 완충제, pH 3.5로 용리시킨다. 용리된 분획을 1M 트리스 pH 9.0으로 중화시킨다. 접합체를 20 mg/mL 소르비톨, 10 mg/mL 글리신, pH 5.0 중에 제제화할 수 있다.
실시예 4 - 인터류킨 6 유도 검정
본 실시예는 본 명세서에 개시된 화합물에 의한 인터류킨 6 유도를 검정하는 방법을 기재한다.
DMSO 중에 희석된 화합물을 ECHO 음향 액체 취급 기술(acoustic liquid handling technology)을 사용하여 매트릭스 테크놀로지스(Matrix Technologies) 투명 V-바닥 384-웰 플레이트의 개별 웰로 옮겼다 (웰 당 25 nL). 인간 전혈 샘플 (25 uL)을 사이바이오 펠릭스(CyBio FeliX) 액체 취급 기기를 사용하여 각각의 웰에 첨가하였다. 플레이트를 플레이트 진탕기 상에서 3분 동안 진탕시킨 후, 반응 혼합물을 37℃에서 20시간 동안 인큐베이션하였다. 이어서, 원심분리 (450 x g, 5분, 주위 온도)에 의해 각각의 샘플로부터 혈장을 유리시키기 전에, 바젤(Basel) RPMI 1640 배지 (L-글루타민으로 보충됨)를 각 웰에 첨가하였다 (웰 당 25 uL). 이어서 처리된 혈장 샘플 (3 uL)을 백색 얕은 384-웰 프록시플레이트 (퍼킨 엘머(Perkin Elmer))의 개별 웰로 펠릭스 액체 취급 기기를 사용하여 옮기고, 그의 인터류킨 6 수준을 제조업체 퍼킨엘머에 의해 기재된 바와 같이 알파리사 (AlphaLlSA) 기술을 사용하여 측정하였다. 데이터 분석 소프트웨어를 사용하여 화합물 EC50 값을 결정하였고, 여기서 기준선은 평균 DMSO 값을 사용하여 확립하고, 100% 유도는 시험된 최고 농도에서의 참조 화합물 값을 사용하여 확립하였다. 그래프패드 프리즘™과 같은 소프트웨어를 사용하여, EC50을 결정할 수 있다.
