Claims (34)
- α-1,2-만노시다제 또는 이의 기능적 부분을 코딩하는 뉴클레오타이드 서열을 포함하고, 메틸요구성 효모 균주에서 α-1,2-만노시다제 또는 상기 기능적 부분을 발현시킬 수 있는 벡터.
- 제 1 항에 있어서, 상기 α-1,2-만노시다제가 진균류로부터 유래하는 단백질인 벡터.
- 제 2 항에 있어서, 상기 진균이 트리코데르마 레세이(Trichoderma reesei)인 벡터.
- 제 1 항에 있어서, 상기 α-1,2-만노시다제가 포유류로부터 유래하는 단백질인 벡터.
- 제 4 항에 있어서, 상기 α-1,2-만노시다제가 쥐의 α-1,2-만노시다제 IA 또는 IB인 벡터.
- 제 1 항에 있어서, 상기 α-1,2-만노시다제 또는 상기 기능적 부분이 ER-체류 신호(ER-retention signal)로 표지된 벡터.
- 제 6 항에 있어서 상기 ER-체류 신호가 펩타이드 HDEL을 포함하는 벡터.
- 제 1 항에 있어서, 상기 α-1,2-만노시다제 또는 상기 기능적 부분을 코딩하는 뉴클레오타이드 서열이 프로모터 및 3' 종결 서열에 조작가능하게 연결된 벡터.

- 글루코시다제 II 또는 이의 기능적 부분을 코딩하는 뉴클레오타이드 서열을 포함하고, 메틸요구성 효모 균주에서 글루코시다제 II 또는 이의 기능적 부분을 발현시킬 수 있는 벡터.
- 제 11 항에 있어서, 상기 글루코시다제 II가 진균류로부터 유래한 단백질인 벡터.
- 제 12 항에 있어서, 상기 진균류가 사카로마이세스 세레비시애(Saccharomyces cerevisiae)인 벡터.
- 제 11 항에 있어서, 상기 글루코시다제 II가 포유류로부터 유래하는 단백질인 벡터.
- 제 11 항에 있어서, 상기 글루코시다제 II 또는 이의 기능적 부분이 ER-체류 신호로 표지된 벡터.
- 제 15 항에 있어서, 상기 ER-체류 신호가 펩타이드 HDEL을 포함하는 벡터.
- 제 11 항에 있어서, 상기 α-1,2-만노시다제 또는 상기 기능적 부분을 코딩하는 뉴클레오타이드 서열이 프로모터 및 3' 종결 서열과 조작가능하게 연결되어 있는 벡터.

- Och1 유전자 부분과 선별 마커 유전자를 포함하고, 상기 Och1 유전자 부분과 상기 선별 마커 유전자가 메틸요구성 효모 균주에서 게놈 Och1 유전자의 파괴를 초래하도록 연결되어 있는, 메틸요구성 효모 균주 내 Och1 유전자를 파괴하기 위한 벡터.
- pBLURA5'PpOCHl을 포함하는 벡터.
- 제 1 항 내지 제 21 항의 벡터 중 어느 한 가지를 사용하여 메틸요구성 효모를 형질전환시키는 것을 포함하는, 메틸요구성 효모로부터 생산되는 단백질 상에서의 글리코실화를 감소시키는 방법.

- 제 23 항에 있어서, 상기 효모가 GS115(NRRL Y-15851), GS190(NRRL Y-18014), PPF1(NRRL Y-18017), PPY12-OH, yGC4, 또는 이들의 유도체로부터 선택되는 피히아 파스토리스(Pichia pastoris)인 방법.
- 제 22 항 내지 제 25 항 중 어느 한 항에 있어서, 상기 효모가 이종 단백질을 발현하도록 유전적으로 조작되어 있는 방법.
- 제 1 항 내지 제 21 항의 벡터 중 적어도 하나를 사용하여 형질전환된, 메틸요구성 효모 균주의 유전적으로 조작된 균주.
- 제 1 항 내지 제 21 항의 벡터 중 적어도 하나를 사용하여 메틸요구성 효모 세포를 형질전환시키고, 형질전환시킨 세포로부터 당단백질을 생산하는 것을 포함하는, 메틸요구성 효모로부터 발현되는 이종 당단백질의 글리코실화를 감소시키는 방법.
- 제 1 항 내지 제 21 항의 벡터 중 한 가지 이상 및 메틸요구성 효모 내에서 당단백질을 발현시킬 수 있는 뉴클레오타이드 서열로 메틸요구성 효모 세포를 형질전환시키고, 형질전환시킨 세포로부터 상기 당단백질을 생산하는 것을 포함하는, 메틸요구성 효모 내에서 글리코실화가 감소된 당단백질을 생산하는 방법.
- 제 27 항 또는 제 28 항의 방법에 의하여 생산된 당단백질.
- 제 29 항에 있어서, 메틸요구성 효모의 야생형 균주로부터 생산된 당단백질과 비교하여 감소된 면역원성을 갖는 당단백질.
- 제 29 항에 있어서, 인간 치료에의 사용에 적합한 당단백질.
- 제 1 항 내지 제 21 항의 벡터 중 어느 하나를 포함하는 키트.
- 제 32 항에 있어서, 메틸요구성 효모 균주를 추가적으로 포함하는 키트.
- 제 26 항의 메틸요구성 효모 균주를 포함하는 키트.
