Abstract
본 발명은 금속단백질 분해효소 섬유소 용해제 (섬유소 분해효소 또는 NAT)를 함유하는 냉동 및 동결건조된 조성물, 상기 동결건조된 조성물을 제조하는 방법 및 동결건조된 조성물을 재구성하는 키트 및 방법에 관한 것이다.
Description
일반적으로, 폴리펩티드는 수성 상태에서 겨우 안정하며 가공 및 보관하는 동안 화학적·물리적으로 분해되어 생물학적 활성을 잃는다. 수용액에서 부딪히는 또 다른 문제점은 특히 탈아미드화 및 펩티드 결합 절단과 같은 가수분해이다. 이러한 효과들은 치료적으로 활성이 있는 폴리펩티드에 대하여 생물학적 활성에 근거한 특정 투여량으로 인체에 투여하기로 되어 있는 것에 심각한 문제점을 나타낸다.

발명의 요약
본 발명은 섬유소 분해효소 및 "신규의 대체 혈전용해제[novel acting thrombolytic (NAT)]" 의 안정한 약학적 조성물들에 관한 것으로, 이의 일부는 냉동 상태에서 저장하기 용이한 액체 조성물이고, 나머지 것은 동결건조에 적합한 것에 관한 것이다.

본 발명의 목적상, 용어 "NAT" 는 SEQ ID NO : 1 로 특징되는, 섬유소 용해 활성을 갖는 금속단백질 분해효소(metalloproteinase)를 지칭한다. NAT 폴리펩티드는, 동일한 폴리펩티드를 암호화하고 있는 상이한 서열의 임의의 DNA 분자가 하기 언급되는 방법에 따라서 발현 및 제조하는 데 사용될 수 있지만, SEQ ID NO : 2 의 cDNA 분자에 의해 암호화된다.

본 발명은 섬유소 용해제를 함유하는 신규한 약학적 조성물에 관한 것이다. 특히, 본 발명은 섬유소 분해효소(fibrolase) 및 "신규의 대체 혈전 용해제"(NAT)의 냉동액 및 동결 건조 조성물은 물론 이의 제조 및 사용 방법에 관한 것이다.

일단 상기 폴리펩티드(섬유소 분해효소 또는 NAT)가 제조되어 정제되고 활성에 대해 분석되면(당해 분야의 숙련자에게 공지된 섬유소 용해제에 대한 방법을 사용하여), 이는 본 발명에 따라서 약학적 조성물로 제제화될 수 있다.

아연 안정화제, 보다 상세하게는 수용성 아연염을 첨가하는 것은, 무기 또는 다른 형태의 유기화합물을 사용하여 응집 및/또는 폴리펩티드 분해를 방지하는 제제형(formulation)에 비하여, 각 형태의 조성물에서 금속단백질 분해효소의 안정성을 높인다. 특히, 0.01 밀리몰(mM) 이상의 아연 농도는 금속단백질 분해효소를 안정화시킬 것이지만, 1 mM 이상의 아연 농도는 섬유소 분해효소 또는 NAT 의 용해도를 상당히 제한한다.따라서, 약 0.01 mM 내지 약 1 mM 의 범위가 제시된다. 적절한 아연염의 예는 아세트산 아연, 황산 아연 및 염화 아연이다.
본 발명에 따른 냉동된 액체 조성물은, 특히 선택적으로(필수적이 아닌) 또한 추가적인 안정화제로서 수용성 칼슘염을 포함한다. 이의 예는 아세트산 칼슘, 황산 칼슘 또는 염화 칼슘이고, 이들은 바람직하게는 약 0.001 내지 약 0.02 mM 및 더욱 바람직하게는 약 0.01 ± 0.002 mM 의 농도로 존재한다.

냉동될 적절한 액체 조성물은 pH 7.4 에서 용해된 섬유소 분해효소 또는 NAT 외에, 아세트산 아연을 약 0.08 mM 내지 약 0.12 mM 의 농도로, 아세트산 칼슘을 약 0.008 mM 내지 약 0.012 mM 의 농도로, 구연산(또는 구연산 나트륨)을 약 95 mM 내지 약 105 mM 의 농도로 함유할 것이다. 다른 바람직한 액체 조성물은, 약 pH 8.0 에서 섬유소 분해효소 또는 NAT, 아세트산 아연 약 0.08 mM 내지 약 0.12 mM, 구연산(또는 구연산 나트륨) 약 18 mM 내지 약 22 mM, 트리스 약 0.02 mM 내지 약 0.06 mM, 만니톨 약 3 % 내지 약 6 % (w/v) 및 Tween 80 약 0.008 % 내지 약 0.012 % (w/v)를 함유할 것이다.
바람직한 동결건조 가능한 조성물은, 약 pH 8.0 에서 섬유소 분해효소 또는 NAT 외에, 황산 아연 약 0.08 mM 내지 약 0.12 mM, 구연산(또는 구연산 나트륨) 약 18 mM 내지 약 22 mM, 트리스 약 3 mM 내지 약 6 mM, 만니톨 약 3 % 내지 약 6 %(w/v) 및 Tween 80 약 0.008 % 내지 약 0.012 % (w/v)를 함유할 것이다.
본 발명에 따른 모든 조성물에 대하여, 섬유소 분해효소 또는 NAT 는 약 0.1 mg/ml 내지 약 50 mg/ml 의 농도로 존재하며 약 5 mg/ml 내지 약 40 mg/ml 의 농도가 더욱 바람직하고, 약 10 mg/ml 내지 약 15 mg/ml 의 농도가 가장 바람직하다.
이러한 조성물의 부형제의 상대적 비율은 몇 가지 인자에 의존한다. 예를 들어, 금속단백질 분해효소 및 증량제(예를 들어, 만니톨)의 양은 조성물을 안정화시키는 데 필요한 아연(및 칼슘, 존재한다면)의 양에 영향을 끼친다. 상기 조성물에 사용되는 안정화제의 양은 동결건조 또는 다른 과정 중에 또는 저장시 섬유소 분해효소 또는 NAT 의 구조적 일체성을 유지하는 데 필요한 양에 좌우될 것이다.

상기 조성물들은 냉동 조성물의 경우 - 30 ℃ 에서 적어도 2 년간, 동결건조된 조성물의 경우 2 ℃ 내지 8 ℃ 에서 2 년간 안정적이라고 예상된다. 이러한 장기 안정성은 약학 제품의 수명을 연장시키고 멀리선적하는 것에 유익하다.
다른 양상에서, 본 발명은 또한 다음 단계를 포함하는 동결건조된 조성물을 제조하는 방법을 제공한다 :

섬유소 분해효소 또는 NAT 및 부형제의 유효량을, 재구성 매질, 예를 들어, 선택된 투여경로에 적합하고 금속단백질 분해효소에 부정적으로 간섭하지 않는 용매, 예를 들어, 멸균수, 생리적 염류 용액, 글루코스 용액 또는 다른 수성 용매(예를 들어, 에틸, n-프로필 또는 이소프로필, 부틸 알콜 또는 이의 혼합물) 및 선택적으로 항균제와 같은 다른 성분을 사용하여 재구성시, 건조된 섬유소 분해효소 또는 NAT 의 응집을 감소시키는 데 유효한 조건하에서 혼합한다.
상기 부형제는 동결건조 전에 적절한 시간에 상기 금속단백질 분해효소와 혼합될 수 있다. 부형제 및 금속단백질 분해효소를 혼합하는 데 걸리는 시간은 적절한 혼합물을 제조하는 데 충분한 기간이어야 한다 ; 바람직하게, 혼합은 약 1 내지 약 30 분간 수행될 것이다.
이후, 제형화된 금속단백질 분해효소는 표준 방법을 사용하여 동결건조, 저장 및 재구성될 수 있다[Pikal, 상기 문헌 참조]. 섬유소 분해효소 또는 NAT 가 냉동 - 건조 및 재구성되는 특수한 조건은, 금속단백질 분해효소를 분해하고 안정화제에 불리하지 않는 한 특별히 제한되지 않는다. 바람직한 동결건조 사이클은 - 40 ℃ 에서 조성물을 냉동하고, 냉동된 시료를 - 12 ℃ 에서 어닐링하고, - 30 ℃ 내지 - 35 ℃ 에서 20 내지 50 시간동안 일차 건조시키며 20 ℃ 에서 20 내지 40 시간동안 이차 건조시키는 것을 포함한다. 일반적으로, 재구성된 조성물은 재구성된 후 빨리 사용한다.
NAT 및 섬유소 분해효소 둘다 혈전의 위치로 국소적으로 최선적으로 수송되어 최대의 효과적인 치료를 나타내게 한다. 섬유소 분해효소와 같이, NAT 는 일반 순환에서 α2고분자글로불린에 의해 공유결합된다. α2고분자글로불린과 복합되어 있기는 하나, 섬유소 분해효소나 NAT 중 어느 것도 목표 기질(예를 들어, 피브린 또는 피브리노겐)에 접근할 수 없고 α2고분자글로불린의 최대 고유 농도가 넘지 않는다면 및 넘을 때까지 크게 유효하지 않다. 따라서, 본 발명의 조성물은 동맥내 또는 정맥내 카테터화를 통해 혈전으로 직접적으로 투여되는 것이 바람직하다.
특정 실시형태의 기술
하기 실시예는 본 발명을 보다 상세히 설명한다.
실시예 1 - 3 에 사용된 재조합 NAT (SEQ ID NO : 1)는 피. 파스토리스에서의 발현으로 생성되었다. 적절한 발현계 및 방법에 관한 자세한 사항은 상기 언급된 Stroman 등, Lair 등, Cregg 등 및 Cregg 특허 등에서 볼 수 있다. 모든 화학물질은 분석 또는 USP 등급이었다.
실시예 1
냉동 액체 조성물의 제조
구연산 100 mM, 아세트산 칼슘 0.01 mM, 황산 아연 0.1 mM 을 함유하는 수용액을, pH 를 7.4 로 유지시키면서 성분을 혼합하여 제조한다. NAT - 함유 용액을 투석으로(대안적으로는 한외여과를 사용할 수 있다) 상기 용액과 완충액 교환한다. 결과적으로 생성된 NAT 용액을 10 mg/ml 로 농축시키고 사용할 때까지 - 30 ℃ 의 온도로 냉동 저장한다.
실시예 2
동결건조된 조성물의 제조
동결건조 가능한 조성물의 제조.트리스 5 mM, 구연산 20 mM, 만니톨 5 % (w/v), 수크로스 0.5 % (w/v), 및 황산 아연 0.1 mM 을 함유하는 수용액을, pH 를 8.0 으로 조절하면서 성분들을 혼합하여 제조하였다. NAT 함유 용액을 투석으로(대안적으로는 한외여과법을 사용할 수 있다) 상기 용액과 완충액 교환하였다. 결과적으로 생성된 NAT 용액을 10 내지 12 mg/ml 로 농축시켰다. Tween 80 을 최종 농도 0.01 % (w/v)로 첨가하였다. 용액을 동결건조때 까지 2 - 8 ℃ 의 온도에서 저장하였다.
동결건조된 제품을 위한 냉동 - 건조 사이클.상기 제조된 조성물을 먼저 동결건조기에서 - 40 ℃ 의 온도로 냉동시켰다. 어닐링 온도는 - 12 ℃ 에서 정해졌다 ; 일차 건조 온도는 - 30 ℃ 에서 정해졌다 ; 이차 건조 온도는 20 ℃ 에서 정해졌다. 결과적으로 생성된 냉동 - 건조 케이크는 양호한 외관을 보여주었고, 칼 피셔 적정법으로측정한 결과 3 % 이하의 물을 함유하고 있었다. Fischer, Angew Chemie, 48 권, 394 쪽 (1935)를 참조하라. 상기 냉동 - 건조 과정을 마친 후, 동결건조된 케이크를 바이알에 넣고 상기 동결건조기에서 상부 금속 선반을 가압하여 누름으로써 진공하에서 고무 마개로 완전히 밀봉하였다. 13 - mm 플립 - 오프 알루미늄 마개로 틀을 잡고 상이한 온도로 정해진 항온기에 두었다.
실시예 3
재구성된 동결건조 시료의 분석
시료의 시간대 분석.시료 바이알을 소정의 시간 간격으로 항온기로부터 꺼내어 시간대 분석을 실시한다. 먼저 동결건조된 시료 케이크를 멸균수, 예를 들어, "주사용 물"(Water - for - injection, 미국 캘리포니아 이르빈 소재 맥거우에서 입수)로 재구성하였다. 재구성된 시료 용액의 투명도를 시각적으로 조사하였다. 이러한 동결건조된 시료에 남아있는 용해된 NAT 를 정량하기 위해, 여과된 용액을 HPLC, UV-Vis 분광분석 및 효소 활성으로 분석하였다.
상기 분석에 근거하여, 동결건조된 제품의 재구성 후에 90 % 가 넘는 NAT 가 회복되었다.
UV/Vis 흡광도.NAT 용액 150 - 200 μl 를 석영 유리 수프라실 1 - cm 투과 길이 울트라 - 마이크로셀에 넣었다. UV/Vis 흡광도를 HP 8452A 다이오드 - 어레이 분광계(미국 델라웨어 윌밍턴 소재 휴렛-팩커드 컴파니에서 입수) 상에서 측정하였다. 아미노산 조성물로부터의 계산에 근거하여, NAT 농도를 280 nm 에서 A0.1 %= 1.05 를 사용하여 결정하였다[Edelhoch, Biochemistry, 6 권 1948 - 1954 쪽 (1967)]. 상기 동결건조된 생성물을 재수화한 후, 350 나노미터(nm)에서 흡광도 측정시 감지할 만한 혼탁도가 발견되지 않았다.
고성능 액체 크로마토그래피.데이터 수득용 HP 3D 을 구비한 HP 1050 액체 크로마토그래피 시스템(휴렛 - 팩커드 콤파니에서 입수)을 사용하여 NAT 시료의 HPLC 분석을 실행하였다. HP 다이오드 - 어레이 검출기를 사용하여 280 nm 및 214 nm 에서의 흡광도로 NAT 종을 검출하였다.
역상 HPLC (RP - HPLC)의 경우, 시료를 51.5 % 완충액 A (2 % 이소프로판올, 0.1 % TFA) 및 48.5 % 완충액 B (90 % 아세토니트릴, 2 % 이소프로판올, 0.1 % TFA)로 구성되고 0.6 ml/분의 유동 속도로 움직이는 이동상의 조르백스 300SB - C8 컬럼(4.6 x 250 mm)으로 주입하였다. 완충액B 는 6 분간 고정시킨 다음 20 분간에 걸쳐 51 % 까지 증가시켰다. 이 농도를 1 분간 고정시킨 후, 8 분간 증가시키고 90 % 에서 5 분간 유지하였다. 최종적으로, 완충액 B 는 3 분간에 걸쳐 48.5 % 로 되돌아 왔다. 이러한 방법으로 측정된 NAT 의 동결건조후 회복율은 92 % 이상이었다.
이온 교환 HPLC (IEX - HPLC)의 경우, 90 % 완충액 A (20 mM 트리스, pH 8.5) 및 10 % 완충액 B (20 mM 트리스, 250 mM NaCl, pH 8.5)로 구성되고 0.5 ml/분의 유동 속도로 움직이는 이동상의 토소하스 DEAE - 5PW 컬럼(7.5 x 75 mm)(미국 앨라배마 먼트거머리빌 소재 토소하스에서 입수)에 시료를 주입하였다. 그런 다음, 구배를 적용하여 10 % 완충액 B 에서 75 % 완충액 B 로 20 분내에 증가시킨 다음, 75 % 완충액 B 에서 90 % 완충액 B 로 1 분내에 증가시켰다. 그런 다음, 완충액 B 를 5 분간 유지한 후 4 분내에 10 % 완충액 B 로 내렸다. 이러한 방법으로 측정된 NAT 의 동결건조후 회복율은 90 % 이상이었다.
크기 배제(size - exclusion) 크로마토그래피의 경우, 시료를 토소하스 G-2000 SWXL 컬럼(300 x 7.8 mm)에 적하하였다. 15 mM 인산 나트륨, pH 7.0 및 0.140 M 염화 나트륨을 함유하는 완충액을 사용하여 등용매용리를 0.8 ml/분의 유동 속도로 적용하였다. 이러한 방법으로 검출된NAT 의 동결건조후 회복율은 95 % 이상이었다.
생분석.피브린 응고물에 대한 활성을 분석하였다. 0.01 내지 1.0 mg/ml 범위의 NAT 연속 희석물의 작은 분획물들은 96-웰 플레이트상에 미리 형성된 피브린 응고물에 적하하였다. 상기 시료들을 18 시간 동안 항온처리하고 500 nm 에서의 흡광도로 혈전 용해를 정량화하였다. 다양한 제제형에 대하여 흡광도 대 NAT 농도의 그래프를 준비된 NAT 표준 그래프와 비교하여 상대적인 활성을 측정하였다. 대조 시료(비 동결건조)에 비하여 동결건조후 NAT 의 섬유소 용해 활성에서 측정할 만한 차이가 없었다.
냉동된 액체 조성물을 4 ℃ 에서 해동하고 동일한 방식으로 실험한 결과 유사한 결과를 얻었다.
상기 발명을 명확히 이해시키기 위해 상세히 설명하였다. 또한 청구의 범위에서 정의된 본 발명의 범위를 벗어나지 않는 한도내에서 기타 다른 조합이 이루어질 수 있음은 명백할 것이다.
실시예 4
NAT 대신 재조합 섬유소 분해효소를 사용하여 실시예 1 및 2 의 과정을 반복하여 유사한 냉동 액체 및 동결건조된 약학적 조성물을 생성하였다.
Claims (21)
- 섬유소 분해효소 및 신규의 대체 혈전용해제(NAT)로 이루어진 군으로부터 선택한 금속단백질 분해효소 섬유소 용해제, 아연 안정화제 및 선택적으로 증량제를 약학적으로 용인가능한 완충액에 함유하는 약학적 조성물.
- 제 1 항에 있어서, 아연 안정화제는 황산 아연, 아세트산 아연 및 염화 아연으로 이루어진 군으로부터 선택한 수용성 아연염임을 특징으로 하는 약학적 조성물.
- 제 1 항에 있어서, 상기 완충액은 구연산 또는 수용성 구연산염임을 특징으로 하는 약학적 조성물.
- 제 1 항에 있어서, 상기 증량제는 만니톨임을 특징으로 하는 약학적 조성물.
- 제 1 항에 있어서, pH 가 약 6.5 내지 약 8.5 의 범위임을 특징으로 하는 약학적 조성물.
- 제 1 항에 있어서, 냉동 액체 형태임을 특징으로 하는 약학적 조성물.
- 제 6 항에 있어서, 선택적으로 수용성 칼슘염을 함유함을 특징으로 하는 약학적 조성물.
- 제 7 항에 있어서, 상기 수용성 칼슘염은 아세트산 칼슘, 황산 칼슘 및 염화 칼슘으로 이루어진 군으로부터 선택함을 특징으로 하는 약학적 조성물.
- 제 1 항에 있어서, 동결건조시킴을 특징으로 하는 약학적 조성물.
- 제 1 항에 있어서, 상기 금속단백질 분해효소는 SEQ ID NO : 1 의 아미노산 서열을 가짐을 특징으로 하는 약학적 조성물.
- 섬유소 분해효소 및 신규의 대체 혈전용해제(NAT)로 이루어진 군으로부터 선택한 금속단백질 분해효소 섬유소 용해제 약 0.1 내지 약 50 mg/ml, 황산 아연 약 0.08 내지 약 0.12 mM, 아세트산 칼슘 약 0.008 mM 내지 약 0.012 mM 및 구연산 또는 구연산 나트륨 약 95 내지 110mM 을 함유하며 pH 약 7.4 인 수성 약학적 조성물.
- 제 11 항에 있어서, 구연산 100 mM, 아세트산 칼슘 0.01 mM 및 황산 아연 0.1 mM 을 함유하는 수용액에 금속단백질 분해효소 10 mg/ml 를 함유하는 약학적 조성물.
- 섬유소 분해효소 및 신규의 대체 혈전용해제(NAT)로 이루어진 군으로부터 선택한 금속단백질 분해효소 섬유소 용해제 약 0.1 내지 약 50 mg/ml, 아세트산 아연 약 0.08 내지 약 0.12 mM, 구연산 또는 구연산 나트륨 약 18 내지 약 22 mM, 트리스 약 0.02 내지 약 0.06 mM, 만니톨 약 3 내지 약 6 % (w/v) 및 Tween 80 약 0.008 내지 약 0.012 % (w/v)를 함유하며, pH 약 8.0 인 수성 약학적 조성물.
- 섬유소 분해효소 및 신규의 대체 혈전용해제(NAT)로 이루어진 군으로부터 선택한 금속단백질 분해효소 섬유소 용해제 약 0.1 내지 약 50 mg/ml, 황산 아연 약 0.08 내지 약 0.12 mM, 구연산 또는 구연산 나트륨 약 18 내지 약 22 mM, 트리스 약 3 내지 약 6 mM, 만니톨 약 3 내지 약 6 % (w/v) 및 Tween 80 약 0.008 내지 약 0.012 % (w/v) 및 선택적으로 수크로스 약 0.1 내지 약 0.5 % (w/v)를 함유하며, pH 약 8.0 인 수성 약학적 조성물.
- 제 14 항에 있어서, 금속단백질 분해효소 12 mg/ml, 트리스 5 mM, 구연산 20 mM, 만니톨 5 % (w/v), 수크로스 0.5 % (w/v), Tween 80 0.01 % (w/v) 및 황산 아연 0.1 mM 을 함유하며, pH 약 8.0 임을 특징으로 하는 약학적 조성물.
- 다음 단계를 포함하는 동결건조된 조성물을 제조하는 방법 :(a) 섬유소 분해효소 및 신규의 대체 혈전용해제(NAT)로 이루어진 군으로부터 선택한 금속단백질 분해효소, 아연염, 증량제, 안정화 이당체 및 계면활성제를 완충액에서 혼합물로 형성하는 단계, 및(b) 단계 (a) 의 혼합물을 동결건조하는 단계.
- 제 16 항에 있어서, 상기 조성물의 pH 는 동결건조 전에 약 7.8 내지 약 8.2 로 조절하는 방법.
- 제 16 항에 있어서, 다음 단계를 포함함을 특징으로 하는 방법 :(a) 아연염, 증량제 및 안정화 이당체를 함유하는 용액의 pH 를 pH 7.6 내지 8.2 로 조절하는 단계,(b) 금속단백질 분해효소 함유 용액을 단계 (a) 의 용액과 완충액 교환한 후, 상기 계면활성제의 유효량을 가하는 단계, 및(c) 단계 (b) 의 혼합물을 동결건조하는 단계.
- 제 18 항의 방법으로 제조한 동결건조된 약학적 조성물.
- 동결건조된 제 1 항의 수성 약학적 조성물을 재구성하고, 재구성된 조성물을 혈전 용해가 필요한 환자에게 투여하는 단계를 포함하는 방법.
- 섬유소 분해효소 및 신규의 대체 혈전용해제(NAT)로 이루어진 군으로부터 선택한 금속단백질 분해효소 섬유소 용해제의 동결건조된 조성물을 갖는 제 1 용기 및 상기 동결건조된 조성물에 대해 생리적으로 용인가능한 용매를 갖는 제 2 용기를 포함하는, 수성 약학적 조성물 제조용 키트.
