상기 화학식( I )로 표시되는 과불소화된 알카노일 L-카르니틴 에스테르는 약리학적 활성 화합물의 세포내 송달용 양이온성 지질로서 유용한 에스테르이다.
Description
"세포내 송달"이라 함은 치료기능을 갖고 있는 폴리뉴클레오티드가 세포에
침입하여 항바이러스성 약제 또는 면역원성 폴리펩티드를 세포에 전달하는 과정을 말한다.
약리학적 활성 물질들의 대부분은, 예를 들어 폴리펩티드와 단백질 또는 약제의 경우 일반적으로 세포아래(subcellular) 또는 분자 단계에서 세포기능에 영향을 주는 그들의 효과를 갖기 위해 세포를 투과하여야 한다. 이런 분자들에 대하여 세포막은 선택적으로 불침투성 장벽으로 이루어져 있다. 사실상 세포막은 보호 기능을 수행하며, 치료적인 작용과 함께 잠재적인 독성물질 전체를 막을 뿐만 아니라 화합물 전체도 막는다. 세포막의 복합 조성물은 인지질, 당지질과 단백질을 포함한다; 세포막의 기능은 Ca++와 다른 이온들, ATP, 미세섬유, 미세소관, 효소 및 Ca++와 결합한 단백질과 같은 세포질의 구성성분에 의해 영향을 받는다. 세포내의 구조적 성분과 세포질구성성분사이의 상호작용 및 외부신호에 대한 반응은 다양한 다른 세포 타입에 의해 보여지는 선택성에 영향을 미치게 된다. 막 장벽 효과는 복합체에서 물질과 자연적으로 발생하는 막지질 조성물을 재생하는 지질 산물을 결합함으로써 극복할 수 있다. 이런 지질들은 막에 용해될 수 있고, 그들을 세포에 결합시키는 물질을 방출할 수 있다. 지질 복합체들은 막 용해를 수단으로 세포내 이동을 촉진시킬수 있을 뿐만 아니라 막과 세포를 투과할 수 있는 분자 사이에 전하 반발력을 또한 감소시킬 수 있다. 막 인지질과 같은 양친매적 지질은 수성계에서 지질 소포 또는 리포좀을 형성한다.
리포좀은 통상적으로 인지질인 지질 분자로 이루어진 하나 또는 그이상의 막으로 전체적으로 닫혀진 수성체적인 소포이다. 친수성 머리와 탄소 체인(소수성 꼬리)의 쌍으로 이루어진 인지질은 생물학적 막의 주요 구성성분이다. 수성용액에서 소수성 꼬리는 물을 배제하기 위해서 자동응집하고, 반면에 친수성 머리는 수성용액과 상호작용을 하여 자발적으로 다양한 지름의 소포 집단을 형성한다. 지질은 일반적으로 쯔비터이온성, 중성 또는 음이온성이다. 이런 소포는 약제, 작은 분자, 단백질, 뉴클레오티드 및 플라스미드의 운반체로서 사용되어질 수 있다.
최근 몇 년 동안, 양이온성 리포좀, 즉 합성지질로부터 제조된 양성 하전된 소포의 부류는 세포에 유전 물질을 이동시키기 위하여 광범위하게 사용되어져 왔다. DNA의 음전하는 안정한 DNA-리포좀 복합체를 형성하기 위해 양이온성 지질의 양전하와 상호작용할 수 있다.
이런 기술의 단순성과 변환의 용이성(versatility)은 환자에게 유전자 치료를 위한 유전자 송달에 중요한 소포인 리포좀을 만들어왔다. 현재 미국 국립 위생 연구소 재조합 자문위원회(NIH recombinant advisory committee)에 의해 인정된 유전자 치료를 위한 벡터의 대부분은 바이러스계 및 합성계를 포함한다.
바이러스성 감염은 특정 세포를 공격할 수 있게 하고, 핵에 DNA를 운반할수 있게 하기위한 복합체 기작의 시리즈를 포함한다. 유전자 치료를 위해 바이러스성 벡터를 사용하는 합리성은 세포를 감염시키는 바이러스 입자의 능력을 제거하는 치료적 기능을 갖는 암호를 지닌 유전자를 바이러스 유전자로 대체할 수 있는 가능성에 기초를 두고 있다. 바이러스 치료의 한계는 면역원성, 세포변성 및 재조합성인 바이러스 요소들과 함께 작용해야 하는 점이다.
유전자 치료를 위해 양이온성 지질의 사용에 큰 희망을 갖고 있다. 이런 벡터는 더 안전하고, 독성이 적고, 또한 큰 크기의 유전자와 결합할 수 있다는 점에서 생물학적으로 비교해서 커다란 잠재력을 가지고 있다. 그러나 생물학적 타입의 벡터들과 비교해서 그들은 낮은 세포내 유전자 전사량을 가진다. 그러나, 그런 트랜스펙션 시스템의 사용은 아직 초기연구단계에 있다는 것을 명심해야한다. 양이온성 지질은 DNA-지질 복합체의 형성, 세포-복합체 상호작용, 막에 있어서 용해, 세포내부에서 DNA 방출 및 전사에 매우 중요한 역할을 한다.
양이온성 지질의 생체내 응용의 중요한 예가 있다. 유전자 치료에서 첫번째 임상 시도는 흑색종 치료를 위해 인간 리포좀-복합체화된 HLA-B7 유전자를 포함하는 표현 매개체를 도입함으로서 수행되어졌다. 또다른 중요한 응용은 폐를 통한 투약 또는 리포좀-복합체화된 표현 벡터 SV-40C-FTR의 나살 스프레이(nasal spray)의 방법을 통한 폐 담낭 섬유증의 치료와 관련되어 있다. 또다른 임상 시도는 암치료를 위한 유전자 치료에 리포좀 사용을 포함하는 것으로 현재 진행중이다.
다음 4가지 구성 요소는 일반적으로 양이온성 지질의 구조와 동일시되어진다; 양성 하전된 양이온성 머리, 스페이서, 앵커(anchor) 지질 및 링커 본드(linker bond).
양이온성 머리는 양이온성 리포좀과 DNA 사이와 DNA-리포좀 복합체와 세포막 사이와 세포의 다른 구성성분 사이의 상호작용에 대해 영향을 미치게 된다. 그것은 쉽게 치환될 수 있는 모노 또는 폴리양이온성 그룹(전하의 수에 의존)으로 구성된다.
스페이서는 소수성 꼬리로부터 양이온성 머리를 분리하는 분자의 부분이고 양이온성 머리와 DNA 포스페이트의 음전하 사이의 최적의 접촉을 갖는 것을 포함한다.

리포좀의 제조와 사용에 관한 과학문헌 및 특허자료는 풍부하나 오직 EP 0279887 A2특허만이 리포좀 제조를 위해서 카르니틴 유도체, 즉 기타 다른 인지질과 지질(콜레스테롤, 포스파티딜 콜린, 포스파티딜 세린)의 혼합물의 상태인 포스파티딜 카르니틴을 사용하는 것에 대해 개시하고 있다.
리포좀 제조에 관해 제공된 유일한 예로서, 포스파티딜 카르니틴의 리포좀은 프로판올올과 결합하여 생성되어지고, 약제는 항고혈합성(antihypertensive), 항협심성(anti-angina) 및 항부정맥성(antiarrhythmia) 물질로서 작용되어지는 것은 알려져 있다. 카르니틴 유도체는 카르니틴의 알려진 심근 친화성 때문에 여기에 사용되어진다. 이 친화성은 바람직한 목표자리에 도달하기전 간에 의해 변형된 리포좀을 피할 수 있게 한다.
포스파티딜 카르니틴의 존재는 그것이 장내 리파아제에 저항한다는 점에서 리포좀을 경구적으로 투여할 수 있게 한다.
본 발명은 막을 통한 이동을 용이하게 하는 약리학적 활성 화합물의 세포내 송달을 좋게 하거나 또는 이들의 특정세포막 자리(수용체)와 상호작용을 증진시키기위해 적합한 양이온성 지질로서, 과불소화된 알카노일 L-카르니틴 에스테르 종류와 이의 용도에 관한 것이다.
생물학적 활성 화합물의 세포내 송달을 순조롭게 하는 양이온성 지질의 주요작용은 다음 화학식( I )로 표시되는 과불소화된 알카노일 L-카르니틴 에스테르에 기인된 것이다.
여기에서;
R1은 탄소원자 2 ∼ 20, 바람직하게는 4 ∼ 12의 직쇄 또는 측쇄의 알카노일, 선택적으로 과불소화된 알카노일을 나타내고;
R2는 탄소원자 4 ∼ 20, 바람직하게는 5 ∼ 12의 직쇄 또는 측쇄의 과불소화된 알킬을 나타내며;
X-는 약리학적으로 허용가능한 산의 음이온을 나타낸다.
따라서, 본 발명의 목적은 화학식( I )로 표시되는 과불소화된 L-카르니틴 에스테르로 구성되는 양이온성 리포좀을 제공하는 것이다. 따라서, 본 발명의 다른 목적은 신규한 상기화학식( I )로 표시되는 에스테르로 이루어지는 양이온성 리포좀의 제조에 있어서 이들을 함께 사용하는 것이다.
본 발명의 또 다른 목적은 상기에 언급한 약리학적 활성 화합물의 세포내 송달을 위해 유용하고, 또한 세포막 수용체와 약리학적 활성 화합물의 상호작용을 증진시키는데 유용한 약제의 제조로서 양이온성 리포좀의 용도를 제공하는 것이다. 특히, 본 발명에 따르면 약리학적 활성 화합물은 유전자이고, 기타 적합한 벡터도 포함되어 있다. 그러므로, 본 발명에 따른 약제는 유전자 치료에 유용하며, 이때 상기 유전자의 예를 들면 β-갈이다.
상기 R1의 예는 다음과 같으며 이에 한정되는 것은 아니다; 아세틸, 프로피오닐, 부티릴, 발레릴, 이소발레릴, 운데카노일, 라우로일, 트리데카플루오로헵타노일, 헵타데카플루오로노나노일, 헵타코사플루오로미리스토일, 펜타데카플루오로-옥타노일 및 5H-옥타플루오로펜타노일 등이다.
상기 과불소화된 R2는 수소원자의 적어도 40%가 불소원자로 치환된 알킬을 의미하는 것이다.
이러한 알킬의 예는 다음과 같고, 이에 한정되는 것은 아니다;
1,1H-2,2H-트리데카플루오로-옥틸;
1,1H-2,2H-3,3H-펜타플루오로펜틸;
1,1H-2,2H-노나플루오로헥실;
1,1H-2,2H-3,3H-4,4H-5,5H-6,6H-노나플루오로데실;
1,1H-2,2H-헵타데카플루오로데실;
1,1H-2,2H-헤인이코사플루오로도데실; 및
1,1H-트리코사플루오로도데실 등이다.
약리학적으로 허용가능한 산은 원치않는 독성 또는 부작용을 일으키지 않는 산의 음이온을 말한다.
이런 산은 약리학자와 통상의 당업자들에게 잘 알려져 있다.
이런 음이온의 예는 다음과 같고, 이에 한정되는 것은 아니다; 클로라이드; 브로마이드; 이오다이드; 아스파테이트; 에시드 아스파테이트; 시트레이트; 에시드 시트레이트; 타르트레이트; 무케이트; 포스페이트; 에시드 포스페이트; 푸마레이트; 에시드 푸마레이트; 글리세로포스페이트; 글루코스 포스페이트; 락테이트; 말리에이트; 에시드 말리에이트; 오로테이트; 옥살레이트; 에시드 옥살레이트; 설페이트; 에시드 설페이트; 트리클로로아세테이트; 트리플루오로아세테이트 및 메탄 설포네이트 등이다.
이하, 본 발명을 다음의 실시예에 의거하여 상세히 설명하겠는바, 본 발명이 이에 한정되는 것은 아니다.
실시예 1
운데카노일-L-카르니틴 클로라이드 1,1H-2,2H-헤파트데카플루오로데실 에스테르
(ST 1223)의 제조
먼저 40℃, 진공하에서 건조된 운데카노일-L-카르니틴 클로라이드(6.6g; 0.018몰)를 20mL 무수 CH2Cl2에 용해시켰다. 상기용액을 교반하면서 0℃에서 티오닐 클로라이드(2.2mL; 0.03몰)를 한방울씩 떨어뜨렸다.
혼합용액을 상온에서 3시간 동안 교반시켰다. 그런다음, 반응용액을 진공하에서 농축하고 CH2Cl2(50mL 3번)를 사용하여 회수하였고, 용액을 다시 진공에서 농축시켰다.
얻어진 운데카노일-L-카르니틴 에시드 클로라이드는 20mL 무수 메틸렌클로라이드에 용해시키고, 0℃에서 1,1H-2,2H-헤파트데카플루오로데칸올-1(13.9g; 0.03몰)으로 상기용액을 한방울씩 첨가하였다.
결과 혼합물을 상온에서 하룻밤 교반하였다. 그후 페트롤리움 에테르를 첨가하여 오일 생성물의 침전을 얻었다. 반응생성물은 CHCl3:CHCl-MeOH 95:5로 용리한 실리콘 겔 크로마토그래피에 의해 정제되었다.
적정 생성물 4g이 얻어졌다. 수율:27%.

라우로일-L-카르니틴 클로라이드 1,1H-2,2H-3,3H-펜타플루오로펜틸 에스테르(ST 1245);
운데카노일-L-카르니틴 클로라이드 1,1H-2,2H-노나플루오로헥실 에스테르(ST 1246);
이소발레릴-L-카르니틴 클로라이드 1,1H-2,2H-3,3H-4,4H-5,5H-6,6H-노나플루오로데실 에스테르(ST 1192); 및
운데카노일-L-카르니틴 클로라이드 1,1H-2,2H-3,3H-4,4H-5,5H-6,6H-노나플루오로데실 에스테르(ST 1193)를 제조하였다.
이러한 화합물들에 대한 중요한 물리화학적 데이타는 다음표에 나타내었다.
실시예 7
트랜스펙션 분석에 사용된 ST1223 리포좀의 제조
상기 실시예 1과 같은 방법에 따라 합성되고, 50mL 클로로포름(용해상)에 용해된 ST1223 200mg을 출발물질로 하여 리포좀을 제조하였다. 그리고나서 진공하(30 ∼ 40℃; 400 ∼ 700mm Hg)에서 하루동안 용매를 제거하였다. 샘플은 최종농도가 5mM이 되도록 증류수로 수화되었다(수화상). 그런다음 초음파조를 사용하여 1시간 동안 10초 간격으로 리포좀을 소니케이션시켰다. 이렇게 제조된 리포좀은 결과적으로 단층소포(unilaminar vesicles)를 형성하였다.
실시예 8
ST1223-DNA 리포좀 복합체의 형성
리포좀-DNA 복합체는 아래에 기술한 A, B 두 용액를 혼합하여 제조되었다.
용액 A: 플라스미드 DNA 2.5㎍(pCMV/β-갈, 7200 염기쌍, 효소 β-갈락토시다제의 표현을 위해 사용됨)은 멸균된 HBS(HEPES 20mM; NaCl 150mM, pH 7.4) 50㎕로 희석시켰다.
용액 B: 실시예 7에 의해 제조된 리포좀은 1.29mM농도로 증류수에서 희석되었고, 이 용액 15μL은 증류수 50μL로 희석되었다.
두 용액을 서서히 교반하여 혼합하였고, 상온에서 10 ∼ 30분 동안 배양되었다.
이 과정은 양하전된 리포좀에 대하여 순 음전하를 가진 DNA를 컴플렉스시켜 리포좀-DNA 복합체를 형성하게 하였다.
실시예 9
트랜스펙션 프로토콜
양이온성 리포좀에 의한 간접적인 트랜스펙션 효율은 배양액에 혈청의 존재 또는 부재, 셀 라인, 밀도 같은 다양한 인자들에 의해 영향받는다.
DNA 트랜스펙션은 다음 인자들에 따라 수행되었다:
혈청 존재:
배양배지에 혈청의 존재는 양이온성 리포좀에 의한 간접적인 트랜스펙션을 억제할것이라는 것은 문헌에 나타나 있다. 본 발명에 따른 실험은 갓 태어난 소의 혈청(fetal calf serum)하에서 행하여졌으나, 그럼에도 불구하고 활성을 나타내었다.
셀 라인의 선택:
pCMV-β-갈 플라스미드 DNA 트랜스펙션은 다음의 4가지 다른 셀 라인에서 행해졌다:
HeLa 사람의 자궁암
MCF-7 사람의 유방 선암
Caco2 사람의 결장 선암
T98-G 사람의 교아종
세포 밀도:
HeLa, MCF-7, Caco2 및 T98-G 같은 다양한 셀 라인에서 양이온성 리포좀에 의한 간접적인 pCMV-β-갈 DNA 트랜스펙션은 다양한 세포 밀도에서 행해졌다: 100,000 세포/디쉬, 200,000 세포/디쉬, 300,000 세포/디쉬.
가장 큰 트랜스펙션 효율은 Caco2가 100,000 세포/디쉬, HeLa와 MCF-7가 200,000/디쉬, T98-G는 300,000 세포/디쉬 밀도에서 관찰되었다.
트랜스펙션 과정은 다음과 같다:
HeLa, MCF-7, Caco2 및 T98-G 세포는 다음 배양액에서 48시간동안 성장되었다(37℃, 5% CO2):
HeLa 세포; RPMI 1640(HyQ 상품), 10% 갓 태어난 소의 혈청, 1% L-글루타민, 1% 스트렙토마이신, 1% 페니실린

T98-G 세포; EMEM(에센셜 미니멈 이글 미디움, SIGMA 상품), 10% 갓 태 난 소의 혈청, 1% L-글루타민, 1% 스트렙토마이신, 1% 페니실린
세포는 직경이 약 3cm인 페트리 디쉬(코닝 또는 팔콘)에 다양한 밀도(100,000세포/디쉬, 200,000세포/디쉬, 300,000세포/디쉬)로 치료되기 전 18시간 동안 2mL 배양배지에서 배양되었다.
새로운 배양배지 2mL로 배지를 갈아준 다음, 실시예 8에 서술한것과 같이 제조된 리포좀-DNA 복합체 100μL는 각 디쉬에 첨가되었다; 천천히 교반시킨 후, 디쉬는 5시간동안 온도조절장치(37℃, 5% CO2)에 놓여졌다. 그 후, 세포는 5mL PBS 버퍼(Gibco 상품)에 3번 워싱된후, 16시간동안(37℃, 5% CO2) 배양되었다. 그 후, 세포는 5mL PBS 버퍼(Gibco 상품)에 3번 워싱된후, 전체 단백질은 추출되었고, 이것의 50㎍은 면역측정법(β-갈 ELISA 키트, 보링거)에 의한 생성물 β-갈락토시다제를 검출하기 위해 각 웰마다 넣어졌다.
실시예 10

- 다음 화학식 ( I )로 표시되는 것임을 특징으로 하는 과불소화된 알카노일-L-카르니틴 에스테르.여기에서;R1은 탄소원자 2 ∼ 20의 직쇄 또는 측쇄의 알카노일, 선택적으로 과불소화된 알카노일을 나타내고;R2는 탄소원자 4 ∼ 20의 직쇄 또는 측쇄의 과불소화된 알킬을 나타내며;X-는 약리학적으로 허용가능한 산의 음이온을 나타낸다.
- 제 1 항에 있어서, 상기 R1은 아세틸, 프로피오닐, 부티릴, 발레릴, 이소발레릴, 운데카노일, 라우로일, 트리데카플루오로헵타노일, 헵타데카플루오로노나노일, 헵타코사플루오로미리스토일, 펜타데카플루오로옥타노일, 5H-옥타플루오로펜타노일 중에서 선택되는 것임을 특징으로 하는 에스테르.

- 제 1 항 또는 2 항에 있어서, 상기 X-는 클로라이드; 브로마이드; 이오다이드; 아스파테이트; 에시드 아스파테이트; 시트레이트; 에시드 시트레이트; 타르트레이트; 무케이트; 포스페이트; 에시드 포스페이트; 푸마레이트; 에시드 푸마레이트; 글리세로포스페이트; 글루코스 포스페이트; 락테이트; 말리에이트; 에시드 말리에이트; 오로테이트; 옥살레이트; 에시드 옥살레이트; 설페이트; 에시드 설페이트; 트리클로로아세테이트; 트리플루오로아세테이트; 메탄 설포네이트 중에서 선택된 것임을 특징으로 하는 에스테르.
- 운데카노일-L-카르니틴 클로라이드 1,1H-2,2H-헵타데카플루오로-데실 에스테르.
- 라우로일-L-카르니틴 클로라이드 1,1H-2,2H-트리데카플루오로-옥틸 에스테르.
- 라우로일-L-카르니틴 클로라이드 1,1H-2,2H-3,3H-펜타플루오로펜틸 에스테르.
- 운데카노일-L-카르니틴 클로라이드 1,1H-2,2H-노나플루오로헥실 에스테르.

- 다음 화학식 ( 1 )로 표시되는 과불소화된 알카노일-L-카르니틴 에스테르가 포함된 것임을 특징으로 하는 리포좀:여기에서;R1은 탄소원자 2 ∼ 20의 직쇄 또는 측쇄의 알카노일, 선택적으로 과불소화된 알카노일을 나타내고;R2는 탄소원자 4 ∼ 20의 직쇄 또는 측쇄의 과불소화된 알킬을 나타내며;X-는 약리학적으로 허용가능한 산의 음이온을 나타낸다.
