실험 균주에 대한 소목 열수추출물의 최소성장억제농도(MIC) 측정을 위해 5% 소목 열수추출물을 멸균된 TSB broth 로 1/2씩 최소 농도가 0.005%가 될 때까지 희석하고 96 well plate 에 100 ㎕ 씩 넣은 후 각각의 균주를 1 x 105 cfu/㎖ 100 ㎕를 첨가하여 well 당 200 ㎕ 씩 맞춘 후 37 ℃ 에서 24시간 동안 배양한 후 흡광광도계(Multickan™ GO, Thermofisher, Waltham, MA, USA)를 통해 595nm 파장에서 흡광도를 측정하여 접종 균주가 성장하지 않은 소목열수추출물 농도를 MIC로 하였다.To measure the minimum growth inhibitory concentration (MIC) of the woody extract for the test strain, dilute 5% of the hotwater extract of the woody wood in sterilized TSB broth until the minimum concentration is 0.005% and add 100 ㎕ to a 96 well plate. 소목 열수추출물의 항균 활성은 디스크 확산법으로 확인하였다. 멸균된 8mm paper disc에 소목 열수추출물을 농도별로 80 ㎕씩 접종하여 준비하였다. 균주 접종을 위해 고온 고압 멸균기를 통해 멸균된 배양배지 3㎖에 배양균을 이식하고 37 ℃에서 24시간 동안 진탕 배양 하였다. 배양된 균주를 1 x 105 cfu/㎖로 희석하여 agar plate에 고루 퍼지도록 도말한 후 준비한 paper disc를 배지 표면에 가볍게 놓은 후 37 ℃에서 24시간 동안 배양한 후 생육저해환(clear zone) 생성 유무를 3회 반복 측정 확인 하였다.
