Claims (17)
- ⅰ) 인간을 제외한 포유류 유래의 난자의 핵을 염색하지 않고 제 1 극체와 10 내지 20%의 세포질을 흡입하여 난자의 핵을 제거하는 단계;ⅱ) 제 1 극체와 핵을 제거한 상기 난자의 위란강에 인간을 제외한 포유류 유래의 체세포를 주입하여 난자-체세포 복합체를 제조하는 단계; 및ⅲ) 0.5 내지 2 ㎸/㎝, 6 내지 150 ㎲로 1 내지 3회 시행하는 전기적 자극만으로 상기 난자-체세포 복합체를 세포융합 및 활성화시키는 단계를 포함하는, 동물의 복제란 제조방법.
- 제 1항에 있어서, 난자는 i)난소를 채취하는 단계; ii) 난소의 직경 2 내지 6mm 난포에서 난포액을 흡입하여 채취하는 단계; 및 iii) 난자는 배양액에서 배양하여 성숙시키는 것을 특징으로 하는 복제란의 제조방법.
- 제 2항에 있어서, 배양액은 TCM 199에 0.1% 폴리비닐알콜(polyvinylalcohol), 3.05 mM 포도당(D-glucose), 0.91 mM 소듐피루베이트(sodium pyruvate), 0.57 mM 시스테인(cysteine), 0.5 ㎍/㎖ LH, 0.5 ㎍/㎖ FSH, 10 ng/㎖ 표피성장인자, 75 ㎍/㎖ 페니실린 G 및 50 ㎍/㎖ 스트렙토마이신을 첨가하여 제조하는 것을 특징으로 하는 복제란의 제조방법.
- 제 1항에 있어서, 체세포는 i) 돼지 태아의 체세포를 채취하여 배양시키는 단계; ii) 원심분리하여 상등액을 제거한 후 배양시키는 단계; 및 iii) 세포가 바닥의 80% 정도를 덮을 때까지 배양시킨 후 트립신 처리 후 계대배양하는 단계를 포함하는 것을 특징으로 하는 복제란의 제조방법.
- 제 4항에 있어서, 단계 i)의 배양액은 0.05% 트립신 및 0.5mM EDTA가 첨가된 PBS(Phosphate Buffered Saline)인 것을 특징으로 하는 복제란의 제조방법.
- 제 4항에 있어서, 단계 ii)의 배양액은 10% FBS(Fetal Bovine Serum)이 포함된 DMEM(Dulbecco's Modified Eagle Medium)인 것을 특징으로 하는 복제란의 제조방법.
- 제 1항에 있어서, 난자와 체세포는 동일한 미세조작용 배양액에 포함되어 있는 것을 특징으로 하는 복제란의 제조방법.
- 제 7항에 있어서, 난자를 먼저 미세조작용 배양액에서 5-10분간 배양한 후, 체세포를 가하여 제조되는 것을 특징으로 하는 복제란의 제조방법.
- 제 8항에 있어서, 미세조작용 배양액은 TCM 199에 0.3% BSA 및 7.5 ㎍/㎖ CB(cytochalasin B)가 첨가된 것임을 특징으로 하는 복제란의 제조방법.
- 제 1항에 있어서, 흡입은 직경 20-40 ㎛, 30-60°의 선단 구조인 유리관인 것을 특징으로 하는 복제란의 제조방법.
- 제 1항에 있어서, 흡입은 유압차를 이용하는 것을 특징으로 하는 복제란의 제조방법.
- 제 1항에 있어서, 단계 i)은 난자의 극체를 1시 방향으로 고정시키고, 미세유리관을 극체가 있는 방향으로 찔러 제1극체와 10~20%의 세포질을 흡입하여 핵을 제거하는 것을 특징을 하는 복제란의 제조방법.
- 제 1항에 있어서, 단계 ii)는 미세유리관을 이용하여 체세포를 흡입하고 상기 미세관을 핵이 제거된 난자의 위란강에 위치시킨 후 유압을 이용하여 체세포를 난자의 위란강에 주입하는 것을 특징을 하는 복제란의 제조방법.
- 제 1 항에 있어서, 인간을 제외한 포유동물은 돼지, 소, 산양, 생쥐, 큰쥐, 토끼, 개, 고양이로 구성되는 군으로부터 선택되는 한 종인 것을 특징으로 하는 복제란의 제조방법.
- 제 14항에 있어서, 상기 포유동물은 돼지인 것을 특징으로 하는 제조방법.
- 제 1 항에 있어서, 상기 i) 단계 이후에 1분 내에 상기 체세포를 주입하는 것을 특징으로 하는 복제란의 제조방법.
- 제 1항에 있어서, 1.0 내지 2.0 ㎸/㎝, 30 ㎲로 2회 전기적 자극을 시행하는 것을 특징으로 하는 복제란의 제조방법.
