Abstract
본 발명은 골형성 단백질; 다공성 입자 중합체 매트릭스; 골형성 단백질-격리양의 응혈; 및 황산칼슘 헤미히드레이트 - 함유 물질의 약학적으로 허용 가능한 부가 혼합물을 함유하는 조성물을 개시한다. 또한, 증진된 용해도 및/또는 안정성 특성을 갖는 뼈 형태발생 단백질의 제제가 개시된다.
Description
발명의 배경
본 발명은 골형성 단백질 및 그의 약학적 제제의 분야에 관한 것이다. 보다 특히, 본 발명은 단백질이 연골 및/또는 골 형성을 유도할 수 있는 충분한 시간을 위해 골형성 단백질을 그 자리에 격리되도록 고안된 약학적 제제를 포함한다.
골형성 단백질은 연골 및/또는 골 형성을 유도하거나, 또는 유도를 보조할 수 있는 단백질이다. 많은 골형성 단백질이 최근에 분리되어 특성화되어지고, 일부는 재조합 방법에 의해 생성되고 있다. 예를 들면, 소위 골 형태발생 단백질 (BMP)이 광물질 소실 골 조직으로 부터 분리되고 [Urist, 미합중국 특허 제 4,455,256 호]; 많은 BMP 단백질이 재조합 방법에 의해 생성되고 [Wang et al., 미합중국 특허 제 4,877,864호 및 Wang et al., 미합중국 특허 제 5,013,549 호]; 일련의 형질전환 성장 인자 (TGF-α 및 TGF-β) 가 골 질환 치료에 잠재적으로 유용한 것으로 밝혀지고 [Derynck et al., 유럽 특허 제 154,434 호]; Vgr-1 으로 명명된 단백질이 골 형성 세포에서 높은 수준으로 발현된다는 것이 알려지고 [Lyons et al., (1989) Proc. Nat'l. Acad. Sci. USA 86, 4554-4558]; 및 OP-1, COP-5 및 COP-7으로 명명된 단백질은 골 유도 활성을 나타낸다고 알려져 있다[Oppermann, et al., 미합중국 특허 제 5,001,691 호].
골 형성을 유도하고자 하는 위치로 골형성 단백질을 전달하도록 고안된 제재를 개발하기 위해 많은 시도가 있어왔다. 예를 들면, 아크릴화 에스테르 중합체 [Urist, 미합중국 특허 제 4,526,909호] 및 락트산 중합체 [Urist, 미합중국 특허 제 4,563,489 호] 와 같은 특정 중합 매트릭스가 이용되어지고 있으나, 상기 제제는 골 형성을 적정하게 유도하기 위한 충분한 시간을 위해 골형성 단백질을 격리시키지 못하고, 또는 적정 골 형성을 위해서는 너무 느리게 부식되는 것으로 밝혀졌다.
OP로 명명된 골형성 단백질의 송달을 위해 다공 입자의 생분해 가능한 매트릭스가 미합중국 특허 제 5,108,753 호 (Kuberasampath) 에 개시되어졌다. 미합중국 특허 제 5,108,753호에는 OP 용 성공적인 담체는 단백질에 결합되어져, 서방성 송달 시스템으로서 작용하고, 골 발달중 세포성 반응의 각 단계에 적응하고, 비특이적 단백분해로 부터 단백질을 보호하여야 한다고 개시되어 있지만, 골 형성이 기대되어 지는 위치에서 OP를 특이적으로 격리시키는 성분을 함유하는 제제는 제안되어 있지 않다.
미합중국 특허 제 5,171,579 호 [Ron et al.] 에는 다공성 입자당 평균표면적이 골 형성을 적정화하기 위해 중요하다고 개시되어 있다.
미합중국 특허 제 4,652,441 호, 미합중국 특허 제 4,711,782 호, 미합중국 특허 제 4,917,893 호 및 미합중국 특허 제 5,061,492 호 [Okada et al.] 및 미합중국 특허 제 4,954,298 호 [Yamamoto et al.] 에는, 외부 오일층내 중합체 벽 물질로 둘러싸인 내부 수성층으로 캡슐화된 폴리펩티드 약제 및 약제-보유 물질을 함유하는 지속적인 방출 마이크로캡슐이 개시되어 있다. 골 형태발생 단백질이 상기의 형성 가능한 폴리펩티드로서 기재되어 있다 할 지라도, 골형성 단백질의 마이크로캡슐화가 적정 골 형성을 위해 충분한 상기 단백질의 제어 방출을 방해한다.
문헌 [Yamazaki et al., Clin. Orthop. and Related Reserach, 234 : 240 - 249 (1988)] 에, 뼈로 부터 정제된 골 형태발생 단백질 1 mg 및 소석고 5 mg을 함유하는 이식물의 용도가 개시되어 있다. 미합중국 특허 제 4,645,503 호에는 골 이식 물질로서 히드록시아파타이트 및 소석고의 합성물이 개시되어 있다.
콜라겐 매트릭스는 또한 골형성 단백질용 송달 매개제로서 사용되고 있지만 [Jeffries, 미합중국 특허 제 4,394,370 호], 콜라겐은 환자에게서 바람직하지 않은 항원반응을 자주 유발한다. 그러므로, 골 형성 유도가 기대되는 위치에서 안전하고, 효과적으로 상기 골 형성을 유도할 수 있는 충분한 시간 동안 골형성 단백질을 격리시킬 수 있는 약학적 제제의 필요성이 남아있다.
발명의 요약
본 발명은 골형성 단백질의 약학적으로 허용 가능한 부가혼합물; 다공성 입자 중합체 매트릭스; 골형성 단백질 - 격리 양의 자가 이식 혈액; 및 허용량의 칼슘 술페이트 헤미히드레이트 -함유 물질 (CSHS)을 함유하는 조성물을 제공한다. 또 다른 구현예로는, 본 발명은 골형성 단백질의 약학적으로 허용 가능한 혼합물; 골형성 단백질 - 격리 양의 자가 이식 혈액; 및 허용량의 CSHS 을 함유한다. 반면 본 발명의 또 다른 구현예는 골형성 단백질 및 적정량의 CSHS 의 제제를 함유한다. 제제는 다른 단백질 격리제, 특히 셀룰로스성 물질을 임의로 포함할 수 있다.
출원인은 또한, 놀랍게도, 증진된 용해도 및 안정성 특징을 갖는, BHPs, 특히 BMP-2의 개량된 제제가 pH 6.0 이하에서 글리신, 수크로스, 및 글루탐산 히드로클로라이드의 조성물을 사용하여 달성될 수 있다는 것을 발견하였다. 발명의 바람직한 구현예로는, 상기 제제는 pH 약 4.5 에서, 약 2.5 % 글리신 (g/100 ㎖ (w/v)), 약 0.5 % 수크로스(w/v), 약 5 mM 글루탐산 히드로클로라이드 (약 0.1 % w/v), 및 약 0.01% (w/v) 폴리소르베이트 80 을 함유한다.
본 발명의 조성물은, 다른 용도중, 조직 손상 및 골절의 치유를 위한 연골 및/또는 골 형성을 촉진하기 위해 사용될 수 있는 골형성 단백질 제제의 제조에 유용하다.
미합중국 특허 제 5,171,579 호에는, 다공성 입자화 중합체 매트릭스가 제제에 혼입된다면, 항섬유소 용해제를 사용하지 않고, 자가 이식 혈액을 사용하여 골 형성 유도 활성이 기대되는 위치에서 골형성 단백질이 격리될 수 있다는 것이 개시되었다. 제조 시간을 감소시키고 상기 제제의 조작 특성을 개량하기 위해서, 출원인은 놀랍게도 칼슘 술페이트 헤미히드레이트 - 함유 물질 (CSHS)을 첨가하는 것이 바람직하다는 것을 발견하였다. CSHS 는 소석고 (POP)로서 공지된, 정제 칼슘 술페이트 헤미히드레이트 (CaSO4·½H2O), 또는 POP 및 히드록시아파타이트(POP:HA)의 혼합물 중 어느 하나인 것이 바람직하다. CSHS의 첨가는 장치 시간을 감소시키고, 생성된 제제의 증진된 성형성(moldability) 및 농도를 제공한다.
본 발명의 실행에 있어서 유용한 골형성 단백질은 당 분야의 숙련가에게는잘 공지되어 있고, 상기 설명된 것들에 포함된다. 본 사용을 위해 바람직한 골형성 단백질은 미합중국 특허 제 4,877,864 호; 미합중국 특허 제 5,013,649 호; WO 90/11366 (October 4, 1990 발행); WO 91/18098(November 28, 1991 발행); WO 93/00432 (January 7, 1993 발행) 및 미합중국 출원 시리즈 (May 12, 1993 출원) 에서 BMP-1 내지 BMP-10 으로서 명시된 BMP 류의 것들이다. 상기 공보의 개시는 참고로서 포함된다.
649 특허에 상세하게 기재된 전 길이의 cDNA 서열인, BMP-2가 가장 바람직하다. 물론, 2개 이상의 상기 골형성 단백질의 조합이 사용될 수 있고, 또한 골형성성을 나타내는 상기 단백질의 단편일 수 있다.
상기 골형성 단백질은 호모이량체 종인 것으로 공지되어 있으나, 또한 혼합된 헤테로이량체로서 활성을 나타낸다. 골형성 단백질의 헤테로이량체 형태는 또한 본 발명의 실행에 있어서 사용될 수 있다. BMP 헤테로이량체는 WO 93/09229에 기재되어 있고, 이의 개시는 참고로서 포함된다. 재조합 단백질은 천연 발생의 단리된 단백질에 대해서 바람직하다. 여기에서 사용되는 골형성 단백질의 양은 선구 세포를 침윤시키는 증가된 골형성성을 자극하는 유효량이고, 이는 하기에 보다 상세하게 기재된 바와 같이 처리되는 결함의 크기 및 성질에 의존할 것이고, 상기 양은 사용된 다공성 입자화 중합체 매트릭스 보다 적은 규모순인, 일반적으로, 사용된 다공성 입자화 중합체 매트릭스 각 10 mg에 대해서 1 ∼ 50 ㎍의 단백질의 범위이고, 보다 바람직하게는 사용된 중합체 매트릭스 (0.2 g/cc의 밀도로 추정)의 각 밀리그램에 대해서 0.5 ∼ 10 ㎍의 범위이다.
골형성 단백질은 약학적으로 허용 가능한 용액의 형태 (동결건조된 형태로 부터 재조성을 포함)로 사용될 수 있다. 골형성 단백질을 약 1 mg/㎖ 이상, 바람직하게는 약2 ∼ 8 mg/㎖ 의 농도로 용해시키는 것이 적정하므로, 약학적 유효량의 단백질은 과도한 부피의 담체를 필요로 하지 않고 전달될 수 있다. 어떠한 이용에 대해서는, 상기 2 mg/㎖ 이상의 농도가 바람직할 수 있다. 총 양성 전하를 갖는 아미노산 (예: 아르기닌, 히스티딘, 리신 및 글리신과 베타-알라닌의 에틸 에스테르와 같은 총 1+ 종류), 바람직하게는 총 2+ 전하 (예: 히스티딘의 에틸 에스테르, 리신 및 아르기닌의 메틸 에스테르, 및 아그마틴)를 갖는 아미노산이 상기 관점에 대해서는 유용하다. 화합물의 양성 전하가 음성 전하 (예: 감마-아미노부티르산, 베타-아미노 프로피온산, 및 글리신-글리신 디펩티드와 같은 총 중성 종류) 를 중화시키기에 충분히 떨어져있다면 (적어도 2 ∼ 3 개의 CH2단위체 만큼 떨어진), 총 제로 전하를 갖는 아미노산은 상기 관점에 관하여 유용하다. 여기에서 사용된 다른 용해제로는 폴리(소르베이트), 덱스트란 술페이트, 구아니딘, 헤파린, 염화나트륨, 글루탐산 히드로클로라이드, 아세트산 및 숙신산이 포함된다. BMP-2, 3, 4, 5, 6, 7, 8, 9 및 10 과 같은 이량체 BMP, 및 BMP-2/6 및 BMP-2/7과 같은 BMPs의 헤테로이량체의 용해에 사용하기 위해, 바람직한 용해제에는 아르기닌 및 히스티딘 (그의 에스테르 포함) 및 글루탐산 히드로클로라이드가 포함된다.
글루탐산 히드로클로라이드 (HCl)는 바람직한 용해제이다. 약 1 ∼ 약 20 mM (약 0.02 ∼ 약 0.37 % w/v), 바람직하게는 약 5 ∼ 약 10 mM (약 0.1 ∼ 약 1.8 %w/v) 의 농도, 및 약 6.0 이하, 바람직하게는 약 3.5 ∼ 약 5.25, 바람직하게는 약 4.5의 pH에서 사용된다. 조성물은 또한 약 0.1 ∼ 약 5.0 % (w/v), 바람직하게는 약 0.5 ∼ 약 2.5 %의 당; 가장 바람직하게는 약 0.5 %의 수크로스; 및 약 0.5 ∼ 약 10.0 % (w/v), 바람직하게는 약 2.0 ∼ 약 2.5 %의 글리신을 포함한다. 입자의 형성을 방지하기 위해서, 글루탐산 히드로클로라이드 조성물은, 예를 들어, 폴리소르베이트 80, 폴리소르베이트 20 또는 플루로닉 F-68 인, 폴리옥시에스테르와 같은, 약 0.01 ∼ 약 0.1 % (w/v)의 비이온성 계면활성제를 임의로 포함할 수 있다. 수크로스/글리신의 비를 다양하게 하여 조성물에 대한 습기 함량 및 크기 특성을 제공할 수 있다. 다른 성분은 본 발명의 조성물에서 대체될 수 있다. 예를 들면, 글루탐산 대신에, 4.5에 가까운 pKa를 갖는 아세트산과 같은 모노-산이 사용될 수 있다. 숙신산과 같은 디-산은 낮은 농도 (약 5 mM 이하, 바람직하게는 약 1 mM) 에서 사용될 수 있다. 글루탐산 히드로클로라이드가 사용되는 경우, 글루탐산/히드로클로라이드 또는 글루탐산 및 HCl의 혼합물중 어느 하나가 사용될 수 있다. 글루탐산 히드로클로라이드는 또한 글루탐산 히드로클로라이드 모노히드레이트와 같은, 수화물 형태로 사용될 수 있다. 적은 변형으로서, 예를 들어, 글루탐산 나트륨과 같은, 글루탐산 또는 글루탐산염이 글루탐산 히드로클로라이드 대신에 사용될 수 있다. 글리신은 유사한 전하를 갖는 다른 아미노산에 의해 전체 또는 부분적으로 대체될 수 있다. 또한, 알부민, 글리세롤, 만니톨 및 다른 당과 같은 다른 선택적인 성분이 첨가될 수 있다. 따라서, 본 발명은 상기 기재된 바와같이, 예를 들어, 글루탐산 히드로클로라이드 대신에 글루탐산 또는 글루탐산 나트륨을 사용하는 조성물인, 대체 성분을 사용하는 조성물을 포함한다. 다른 변형은 당 분야의 숙련자에게 자명할 것이고, 또한 본 발명에 포함된다.
상기 제제는 저장, 운반 및 안정성에 있어서 유리하다면, 동결건조되고 물로 재조성될 수 있다. 본 발명의 동결건조된 제제는 하기의 조성 : 약 3.12 중량 % ∼ 약 24.38 중량 %의 BMP; 약 0.52 % ∼ 약 10.27 %, 바람직하게는 약 1.84 % ∼ 5.27 % (w/w)인 글루탐산 히드로클로라이드; 약 38.4 중량 % ∼ 약 75.7 중량 %인 글리신; 약 14.28 중량 % ∼ 47.15 중량 % 인 수크로스; 및 선택적으로 약 0.15 중량 % ∼ 2.94 중량 %인 폴리소르베이트 80 으로 이루어질 수 있다. 예를 들면, 동결 건조된 두가지 바람직한 조성물을 하기의 표 1에 기재한다 :

상기 제제는 40 시간 미만 후 잘 조절된 수준의 습기를 가지며, 재생 가능하게 동결 건조될 수 있다. 제제는 다양한 온도에서 우수한 저장 안정성을 제공한다. 상기 제제는 또한 BMP의 더 높은 투여량이 투여될 수 있도록, BMP의 더 높은 용해도를 제공하고, 상기 기재된 장치와의 우수한 상용성을 나타낸다. 본 발명내에서 최소한 변형 및 개량된다면, 상기 제제는 약4 mg/㎖ 이상의 더 높은 농도의 BMP-2 또는 다른 BMPs의 용액을 제조하는데 사용될 수 있다.
여기에서의 용도를 위해 골형성 단백질 및 용해제를 혼합하는 방법으로는, 초여과법, 투석, 겔 여과법, 및 소수성 반응 크로마토그래피법 등의 각종 잘 공지된 방법이 사용될 수 있으나, 여기에 국한되지는 않는다.
본 발명의 실행에 있어서 유용한 다공성 입자 중합체 매트릭스 성분은 하기 기재되는 바와 같이 그 자체가 골형성 단백질을 위한 비계 재료(scaffolding)를 제공하는 다공성 입자로 형성될 수 있는 중합성 물질이면서, 새로운 골 성장에 의해 대체될 수 있도록 하는 생분해성을 갖는다. 예로는 아미노산, 오르토에스테르, 무수화물, 프로필렌-공-푸마르산염의 중합체, 또는 하나 이상의 α-히드록시 카르복실산 단량체의 중합체이다(예: α-히드록시 아세트산 (글리콜산) 및/또는 α-히드록시 프로피온산 (락트산)). 후자는 그의 d-또는 1-형으로, 또는 라세미 혼합물로서 사용될 수 있고, 라세미 혼합물이 바람직하다. 락트산 및 글리콜산의 공중합체(PLGA)가 사용되는 경우, 단량체의 몰비는 임상적 지표가 나타내는 바에 따른 목적 생부식 수명 (bioerosion lifetime) 에 따라 1:99 ∼ 99:1의 범위일 수 있고, 이때 50 % 이상인 단량체 중 어느 하나가 더 오랜 생부식 수명을 나타낸다 (더 느린 생분해). 중합체의 분자량은 약 1,000 ∼ 100,000 (CHCl3중 폴리스티렌 기준)의 범위일 수 있고, 50 : 50의 공중합체가 사용되는 경우 30,000 ∼ 50,000가 바람직하다. 일반적으로, 분자량이 커질 수록, 생분해는 더 느려진다.
본 발명의 중합성 매트릭스 성분은 속이 빈 입자 (표면 다공질인)의 매우 다공성인 형태로 사용되고, 이하 총체적으로 "다공성 입자"로 칭한다. 상기 다공성 입자는 일반적으로 150 ∼ 850 마이크론, 바람직하제는 150 ∼ 500 마이크론, 가장 바람직하게는 150 ∼ 300 마이크론의 직경을 갖는 구형이다. 상기 입자 크기는 포유류 골형성 선구 세포가 여과될 수 있고 골형성 단백질에 의해 양성의 영향 (골형성성/골 성장 속도의 증가에 의해 증명)을 받도록 입자사이에 충분한 공간을 생성한다.
미합중국 특히 제 5,171,579 호에는, 다공성 입자당 평균 표면적이 골 형성을 적정화하는데 중요하다는 것이 개시되어 있다. 구체적으로, 본 발명에 따른 골 형성에 유용한 다공성 입자는 약 0.02 ∼ 4 ㎡/g의 평균 표면적을 가져야 한다. WO 93/06872 에는, 또한 다공성 입자를 생성하기 위해 사용되는 용액에 "포로시겐 (porosigen)" (입자 표면적을 증가시킴으로써 다공성을 부여할 수 있는 조성물)을 도입시킴으로써 목적 표면적을 갖는 다공성 입자를 생성하는 것이 가능하다고 개시되어 있다. 상기 공보의 개시는 여기에서 참고로서 포함된다. 다공성 입자를 γ-방사선을 조사하여 멸균시킴으로써 생부식 속도를 또한 조절하는 것이 가능하다. γ-방사량이 많을 수록, 생부식이 더 빨라진다. 여기에서 사용되는 입자는 상응하는크기의 비-다공성 입자에 비해 입자의 표면적이 약 2 ∼ 250 배로 증가하도록 하는 다공성을 갖는다.
발명에서 사용되는 다공성 입자의 바람직한 생성방법은, 일반적으로 말하자면, 중합체를 용해 (예: CH2Cl2중) 시키고, 고체 및/또는 액체 형태인 NaCl, 만니톨 또는 수크로스와 같은 포로시겐을 첨가하는 것으로 이루어지는 용매 증발 방법이다. 포로시겐을 고형으로 첨가하는 경우, 매트릭스-포로시겐 용액은 현탁액의 형태이다.
발명에서 사용되는 다공성 입자의 또 다른 바람직한 생성 방법은 용매 추출법으로, 균질화시킴과 동시에 포로시겐을 액체 형태로 첨가한다. 균질화시킴과 동시에 포로시겐을 액체 형태로 첨가하는 경우, 매트릭스 포로시겐 용액은 유액의 형태이다. 둘중 하나의 방법으로, 매트릭스-포로시겐 유액을 조절된 교반 및 온도에서, 폴리(비닐 알콜)과 같은 계면활성제를 함유하는 과량의 수용액에 첨가한다. 생성된 다공성 입자는 잔류 용매를 추출 또는 증발시키고, 건조시키므로서 경화된다. 포로시겐으로서 50 % NaCl을 사용하여 제조된, 본 발명에 사용된 PLGA 입자는 약 0.2 ∼ 0.6 ㎡/g의 표면적으로 가지며; 포로시겐으로서 수크로스를 사용하여 제조된 입자는 약 0.04 ∼ 0.09 ㎡/g의 표면적을 갖는다. 균질화와 동시에 액체 포로시겐을 사용하여 제조된, 본 발명에서 사용된 PLGA 입자는 약 0.02 ∼ 4 ㎡/g의 표면적을 갖는다.
본 발명에서 사용되는 입자의 다공성은 단백질 흡착에 충분한 표면적을 생성하고 생분해를 증가시키는데, 이의 바람직한 정도는 임상적 지표가 나타내는 것에 의존한다. 표면적은 통상적인 방법으로 측정될 수 있다. 예를 들면, BET 표면적 분석은 마이크로매리틱스 ASAP 2000 시스템을 사용하여 수행될 수 있고, 이는 표면에서의 크립톤(Krypton)가스의 흡착 및 분리를 기준으로 하여 고형 시료의 구멍내에서 표면적을 측정하는 것이다. 단위체는 하기와 같이 계산되어 표면적을 나타낸다:

특정 결함의 치료에 사용되는 다공성 입자의 양은, 물론, 치료될 결함의 크기, 및 골형성 단백질의 흡착에 필요한 유효량에 의존할 것이다.
본 발명의 실행에 사용되는 단백질 격리 물질은 약학적으로 허용 가능한 사람 혈액이고, 바람직하게는 자가 이식 혈액이다. 골형성 단백질/다공성 입자 혼합물에 첨가되는 경우, 응혈은 가단성 합성물을 형성하고 그경우 흡착 단백질은 충분한 시간 동안 매트릭스내에 격리되어 단백질이 이와는 다른 포유류 선구 세포에 스며드는 골형성성의 천연 속도를 증가시킬 수 있다. 상기 응혈이 없는 경우, 골형성 단백질은 단백질의 골형성 유도 효과가 임상적으로 중요하지 않은 속도로 그 자리에서 PLGA 입자로 부터 분리된다. 여기에서 사용되는 다공성 입자에 대한 혈액의 비는 1 : 0.5 ∼ 1 : 10 (v:v)이고, 바람직하게는 1 : 5 (v:v)이고, 보다 바람직하게는 1 : 2 (v:v)이며, 이 비율은 중합체 매트릭스로 부터 분리되는 것을 방지하기 위해 필요한 양을 나타내지만, 선구 세포가 매트릭스로 스며드는 것을 방지할 만큼 많지는 않고, 그에 의해 선구 세포의 골형성성을 보조하는 기회를 단백질에 제공한다. 각1 ㎖의 결함에 대해서, 필요한 혈액의 양은 일반적으로 약 0.5 ∼ 1.0 ㎖일 것이다. 많은 양의 골형성 단백질이 사용되는 경우에, 트롬빈과 같은 응집 촉진제가 골형성 단백질의 희석 효과를 상쇄하기 위해 사용될 수 있다.
본 발명의 조성물은 다른 단백질 - 격리제를 선택적으로 포함할 수 있다. 상기 단백질 -격리제는 바람직하게는 골형성 또는 골전도성을 갖는다. 적합한 제제로는 메틸셀룰로스, 에틸셀룰로스, 히드록시에틸셀룰로스, 히드록시프로필셀룰로스, 히드록시프로필-메틸셀룰로스, 및 카르복시메틸셀룰로스와 같은, 알킬셀룰로스 (히드록시알킬셀룰로스를 포함) 와 같은 셀룰로스성 물질이 포함된다. 다른 격리제로는 히알루론산, 알긴산 나트륨 또는 알긴산 칼슘과 같은 알긴산염, 폴리(에틸렌 글리콜), 산화 폴리옥시에틸렌, 카르복시비닐 중합체, 및 폴리(비닐 알콜)이 포함된다.
본 발명의 바람직한 구현예에서, 상기 셀룰로스성 단백질 격리제는 희석제로서 수용성 글리세롤을 사용하여 희석될 수 있다. 상기 구현을 위해 바람직한 격리제로는 카르복시메틸셀룰로스가 포함된다. 셀룰로스성 단백질 격리제는 약 2 ∼ 약 10 % (w/v), 바람직하게는 약 2.75 ∼ 5.5 % (w/v)의 농도로 존재하는 것이 바람직하고; 수용성 글리세롤은 약 20 ∼약 80 % (w/v), 바람직하게는 악 60 ∼ 80 % (w/v) 의 농도로 존재하며; 다공성 마이크로중합체 입자에 대한 격리제 액화 용액의 비 (v/v) 는 약 0.1 ∼ 약 0.9, 바람직하게는 약 0.2 ∼ 0.5 이다.
상처 부위에서 제제의 골전도성 및 제제의 보유력을 증진시키고, 제제의 장치 시간을 감소시키고 조작 특성을 개선하기 위해서, 황산칼슘 헤미히드레이트 함유 물질 (CSHS)을 제제에 첨가한다. 장치의 증진된 형성성 및 조작과 더불어, CSHS의 첨가는 장치에 필요한 제조 시간을 감소시켰다. 또한, 불량하거나 또는 느린 천액 응집을 갖는 환자에게 있어서 우수한 조작성 및 형성성을 갖는 장치를 제조할 수 있다. 바람직한 CSHS는 또한 소석고로서 공지된 정제 황산칼슘 헤미히드레이트 및 소석고와 히드록시아파타이트의 35 : 65 혼합물을 포함한다. rhBMP-2/응혈/BEP 장치 중 2 ㎖ 의 다공성 입자당 약 0.2 ∼ 2.0 g 이상, 바람하게는 약 0.8 ∼ 2.0 g 이상의 CSHS 를 첨가하는 것이 바람직할 수 있다.
본 발명의 또 다른 구현예에서, 제제는 다공성 중합성 입자 없이, 골형성 단백질, 자가 이식 혈액 및 황산칼슘 헤미히드레이트 함유 물질(CSHS)을 함유한다. 상기 제제의 바람직한 조성물은 약 0.5 ㎖의 골형성 단백질, 약 5.5 ㎖의 자가 이식 혈액 및 약 1 ∼ 10 g의 CSHS의 상대적인 양인 성분들을 함유한다. 상기 구현에서, CSHS는 구조 매트릭스 기능, 골전도성 매트릭스, 및 단백질 격리 기능, 더불어 자가 이식혈액의 것도 제공한다. 적정 제제는 혈액 및 골형성 단백질의 혼합물에 대해 1 g 증가에 있어서 CSHS의 양을 다양화시켜 첨가함으로써 결정된다. 하기의 제조 이후 즉시 및 제조 이후 1 ∼ 2시간 후 장치를 관찰한다. 제조 이후 즉시 및 1 ∼ 2시간 후 모두 최상의 조작성을 제공하는 CSHS의 양이 적정할 것이다.
본 발명의 또 다른 구현예에서, 제제는 골형성 단백질 및 황산칼슘 헤미히드레이트 함유 물질의 적절한 혼합물을 함유한다. 상기 제제의 바람직한 조성물은 약 3 ㎖의 물중에 10 ∼ 20 그램, 바람직하게는 12 그램의 CSHS 당 약 0.5 ∼ 2 그램, 바람직하게는 1 그램의 골형성 단백질을 함유한다. 예를 들면, 골형성 단백질은 2 mg/㎖ BMP-2 용액 중 0.5 ㎖, 또는 4 mg/㎖ BMP-2 용액 중 0.25 ㎖을 사용하여 송달될 수 있다. 상기 구현에서, CSHS는 구조 매트릭스 기능, 골전도성 매트릭스, 및 단백질 격리 기능을 제공한다.
상기 제제는 상기 기재된 바와 같이, 단백질 -격리제를 선택적으로 포함할 수 있다. 본 발명의 실행에서 사용되는 부가적인 선택적 성분들로는, 예를 들어, 만니톨, 수크로스, 락토스, 글루코스, 또는 글리신과 같은 (동결 건조중 분해로 부터 제제를 보호하기 위한)저온에 적합한 보호제, 메틸 및 프로필 파라벤 및 벤질 알콜과 같은 항미생물 보존제, EDTA, 시트르산염, 및 BHT (부틸화 히드록시톨루엔)과 같은 항산화제, 및 폴리(소르베이트) 및 폴리(옥시에틸렌) 과 같은 계면활성제 등이 포함된다. 물론, 약학적으로 허용 가능한 형태인 제제의 통상적인 제조 (예: 피로겐무, 적절한 pH 및 등장성, 멸균성 등) 는 당분야에서 잘 알려져 있고 본 발명의 제제에 이용될 수 있다.
골형성 단백질 및 제제의 다공성 입자는 용액 또는 동결 건조된 형태중 하나 로서, 단일 바이알 제제로서 임상에 제공될 수 있고, 또는 제제는 다성분 키트로서 제공될 수 있는데, 예를 들어, 골형성 단백질이 하나의 바이알로 제공되고, 다공성 입자 및 황산칼슘 헤미히드레이트 함유 물질이 각각 별개의 바이알로 제공된다. 제제에 사용되는 혈액은 응집될 수 있도록 사용되기 전, 일반적으로 사용하기 l5 ∼ 180 분전에 충분히 혼합되어, 장치의 형성성을 촉진화시키는 CSHS의 활성 뿐아니라, 응집시 공지된 환자-대-환자 다양성을 설명한다.
본 발명의 제제는 치료학적 유효량의 골형성 단백질이 연골 핀/또는 골 형성이 요구되는 상처 부위로 송닥될 수 있도록 하는 가단성 이식물을 제공한다. 상기 이식물은 신선한 비결합 골절, 척추 융합, 및 정형외과 분야의 골 손상 치유에 있어서; 두개/상악골안면 재구성에 있어서; 자가이식 골 이식편을 위해; 특히 인공삽입물로 도포된 히드록시아파타이트와 같은 보철 이식물의 고정을 증진시키기 위한 표면 피복으로서 인공삽입물 융합을 위해; 골수염에 있어서 골 재생을 위해; 및 치과에서 치조 융선및 치주위 질병의 첨가물 및 발치 소케트로서의 치환물로서 사용될 수 있다. 골수염의 치료 또는 골 손상에 감염을 최소화하여 사용되는 경우, 골형성 단백질은 수용성 글리세롤을 사용하여 희석된, 알긴산염, 셀룰로스, 특히 카르복시메틸셀룰로스와 같은 단백질 격리제를 첨가하면서, 다공성 미세인자 및 항생제와 혼합되어 사용될 수 있다. 항생제는 골 형성에 대한 최소한의 역효과를 가지며 감염을 감소시키는 성능을 위해 선택된다. 본 발명의 장치에서 사용하기 위한바람직한 항생제로는 반코마이신 및 젠타마이신이 포함된다. 항생제는 반코마이신 HCl 또는 젠타마이신 황산염과 같은, 임의의 약학적으로 허용 가능한 형태일 수 있다. 항생제는 약 0.1 mg/㎖ ∼약 10.0 mg/㎖의 농도로 존재하는 것이 바람직하다.
점도가 더 낮은 제제는 또한 근접한 골절의 치료를 촉진시키기 위한 경피 주사제로서 사용될 수 있다. 상기 용도중 어떤 경우에 있어서, 본 발명의 조성물은 폴리(프로필렌-공-푸마르산염) 및 특정 히드록시아파타이트 접착제와 같은 부식 가능한 골 접착제를 포함하는, 각종 골 접착제와 결합되어 사용될 수 있다. 또한, 상기의 어떠한 용도는 부식 가능한 플레이트, 스크류 등과 같은 생부식될 수 있는 하드웨어를 이용할 것이다. 상기에서 나타난 바와 같이, 투여량은 다양한 환자 유형 (예: 체중, 연령, 성별) 및 임상적 표현 (예: 상처 정도, 상처 부위 등)뿐만 아니라, 나타난 임상적 지표에 의해서 결정될 것이다. 일반적으로, 골형성 단백질의 투여량은 약 10 ∼ 1000 ㎍, 바람직하게는 약 10 ∼ 100 ㎍의 범위일 것이다.
하기의 실시예에서 본 발명을 설명하고 있고, 이에 국한되는 것은 아니다. 변형, 다양화 및 최소 증진이 시도되며 본 발명에 속한다.
실시예 1
글루탐산 HCl 을 사용한 BMP 의 제조
재조합으로 생성된 BMP-2 를 2.5 % (w/v) 글리신, 0.5 % (w/v) 수크로스, 0.01 % (w/v) 폴리소르베이트 80 및 5 mM (약 0.092 % w/v) 글루탐산 HCl 의 pH=4.5 인 조성물중에 용해시킨다 (2 mg/㎖).
실시예 2
용해제로서 글루탐산 HCl 을 사용하는 BMP 제제의 용해성 및 안정성
실시예 1의 상기 조성물을 동결건조시켜 -80, 4, 30 및 40 ℃에 저장한다. 동결건조된 시료는 재구성에 앞서 외형을 세밀하게 검사한다. 모두 양호한 백색 케이크로서 나타나고 시간 또는 온도에 따라 변화하지 않는다.
조성물은 동결건조후 2 주, 1 개월, 3 개월 및 4.5 개월째 재구성되어, 재구성 시간, 입자 형성, 재구성후 pH, 습기 함량 %, 재구성후 BMP-2의 농축, 응집 형성 % 및 BMP 활성에 대한 W-20 생물화학적 검정에서의 특이성에 관하여 연구한다.
상기 조성물들은 4.5 개월 보관 후에도 약 10 ∼ 30초 내에 재구성된다. 재구성에 대해서, 조성물은 가시적 입자 없이, 습기 함량에 어떠한 변화도 없고, 단백질의 손실도 없으며, 낮은 수준의 응집을 나타내고, W20 생물화학적 검정에서 활성을 갖는다.
실시예 3
황산칼슘 헤미히드레이트 함유 장치
0.8, 1.2, 1.6 및 2.0 g 의 황산칼슘 헤미히드레이트 [Calset?, Lifecore Biomedical 사 제], 또는 35 : 65인 황산칼슘 헤미히드레이트 : 히드록시아파타이트 [Hapset?, Lifecore Biomedical 사 제] 의 혼합물중 어느 하나를 2 ㎖의 rhBMP-2/자가 이식 혈액/다공성 미세입자 혼합물에 첨가한다. 제조시 점도에 대해서 제제를 관찰하고, 제조 5 분후 특성을 다루기 위해 10 포인트 스코어 규모 (5의 스코어가 적정하다)로 측정한다.
표 2 는 상기 측정의 결과를 나타낸다. 단지 2 ㎖의 혼합물을 사용하였기 때문에, 제제는 10 ㎖ 크기로 사용된 실제 이식물 보다 더 빠르게 건조되는 경향이 있다.

실시예 4
황산칼슘 혜미히드레이트 함유 물질을 사용한 이식물의 제조
30 ∼ 40 kD의 평균 분자량, 약 20 kD의 수평균 분자량 (폴리스티렌 표준 기준, 겔 투과 크로마토그래피에 의함) 및 0.35 ∼ 0.45 ㎗/g의 고유 점도를 갖는 락트산 및 글리콜산 (PLGA)의 50 : 50 랜덤 공중합체를 CH2Cl2(15 % w/v)중에 용해시키고, 10 g의 포로시겐 (7.5 % w/v)을 상기 용액에 현탁시킨다. 생성된 용액을 과량의 폴리(비닐 알콜)수용액 (0.1 % w/v)에 첨가한다. 몇 시간의 부분 진공 (24 인치 Hg)하 교반 후, 입자를 과량의 냉 에탄올 (95 %) 중에서 경화시킨다. 생성된 입자를 주사용수로 세척하고, 진공하 건조시켜 비-유동성생 성물을 수득한다. BET 표면적 분석은 상기 기재된 바와 같이 마이크로미터 ASAP 2000 시스템을 사용하여 수행되고, 입자는 약0.2 ∼ 1.0 ㎡/g의 표면적을 갖는다 : 포로시겐으로서 수크로스를 사용하여 제조된 입자는 약 0.04 ∼ 0.09 ㎡/g의 표면적을 갖는다. 다공성 입자는 사용전에 γ조사에 의해 멸균된다. 사람에게 사용되는 이식물 각각에 대해서, 10 ㎖의 다공성 입자를 멸균 컵에 둔다. 목적량 (4 ∼ 10 g)의 황산칼슘 헤미히드레이트 함유 물질을 멸균 컵중 다공성 입자에 첨가하고, 컵을 흔들어 혼합한다.
동결 건조된 재조합 사람 BMP-2 (rhBMP-2) (완충용액중 2 mg) 을 1 ㎖의 멸균된 주사용수 (WFI)로 재구성한다 0.5 ㎖의 rhBMP-2 용액 (1 mg) 을 22 g 바늘을 사용하는 3 ㎖ 주사기로 빨아들인 다음 제거시킨다.
5.5 ㎖의 환자의 정맥 혈액을 10 ㎖ 주사기로 (헤파린화 라인을 통하지 않고) 빨아들여, 그의 바늘을 제거한다. 혈액이 컵중 다공성 입자/황산칼슘 혼합물상에 균일하게 나타난다. rhBMP-2 용액을 멸균 컵중 성분에 첨가한다. 필요에 따라, 혼합물 전체를 스테인레스 스틸 스패튤라로 부드럽게 교반시켜 균일한 점도를 달성한다. 컵에 덮개를 하여 실온에서 약 15 분간 방치시킨다. 이식 혼합물은 이후 2 시간내에 사용해야 한다.
가단성 이식물의 목적한 점도가 수득되는 경우, 상처 부위에서의 외과 절개에 의해 골 손상을 노출시킨다. 다공성 입자/CSHS/rhBMP-2/응혈의 가단성 합성물을 외과의사에 의해 치료하고자 하는 골 손상을 둘러싸도록 성형시킴으로써 이식물이 사용된다. 이어서 통상적인 방법을 사용하여 절개를 밀봉시킨다. 치료는 손상 부위의 X-선 분석에 의해 확인한다.
실시예 5
W-20 생물화학적 검정
A. W-20 세포의 설명
표식 세포라인으로서 W-20 골수 기질 세포의 사용은 BMP 단백질로 치료후 상기 세포를 골아세포-유사 세포로 전환시키는 것을 바탕으로 한다 [Thies et al,Journal of Bone and Mineral Research, 5 : 305 (1990); and Thies et al, Endocrinology, 130 : 1118 (1992)]. 구체적으로, W-20 세포는 연구실 [Dr D. Nathan, Children's Hospital, Boston, MA] 의 연구원들에 의해 성인쥐로 부터 유래된 클론성 골수 기질세포라인이다. 특정 BMP 단백질을 갖는 W-20 세포의 처리는, (1) 알칼린 포스파타제의 생성 증가, (2) PTU 유도된 cAMP의 유도, 및 (3) 세포에 의한 오스테오칼신 합성의 유도를 일으킨다. (1) 및 (2)는 골아세포 표현형과 연관된 특성을 나타내는 반면, 오스테오칼신을 합성하는 능력은 단지 성숙 골아세포에 의해서 나타나는 표현성 성질이다. 더구나, 지금까지는 BMPs 처리에 대해서만 W-20 기질 세포의 골아세포-유사 세포로의 전환이 관찰되었다. 상기 방법에 있어서, BMP-처리된 W-20 세포에 의해 나타나는 시험관내 활성은 BMPs로 공지된 체내 골 형성성과 상호관련된다.
공지된 BMPs의 활성을 갖는 BMPs 제제의 BMP 활성의 비교에 유용한 2가지의 시험관내 분석법을 하기에 기재한다.
B. W-20 알칼린 포스파타제 분석 방법
W-20 세포를 96 홈 조직 배양 플레이트에 200 ㎕ 의 배지 [10 % 가열 불활성화된 소 태아 혈청을 갖는 DME, 2 mM 글루타민 및 100 유니트/㎖ 페니실린 + 100 ㎍/㎖ 스트렙토마이신] 중 10,000 세포의 농도로 플레이트한다. 세포를 37℃ 에서 95 % 공기, 5 % CO2인 항온배양기에서 하룻밤동안 부착시킨다.
200 ㎕의 배지를 각 홈으로 부터 멀티채널 피펫으로 제거하고 10 % 가열 불활성화된 소 태아 혈청, 2 mM 글루탐산 및 1 % 페니실린-스트렙토마이신을 갖는 DME로 전달된 동 부피의 시험 시료로 대체시킨다. 시험 물질을 3 차례에 걸쳐 분석한다.
시험 시료 및 표준 시료를 W-20 표식 세포와 함께 24 시간 항온배양 시킨다. 24 시간 후, 플레이트를 37 ℃ 항온배양기로 부터 제거하고 시험 배지를 세포로 부터 제거한다.
W-20 세포층을 홈당 200 ㎕의 칼슘/마그네슘이 없는 인산염 완충식염수로 3회 세척하고, 상기 세척액을 버린다.
50 ㎕의 글래스 멸균수를 각 홈에 첨가한 다음, 분석 플레이트를 신속한 동결을 위해 드라이 아이스/에탄올 욕상에 위치시킨다. 일단 동결되면, 분석 플레이트를 드라이 아이스/에탄올 욕으로 부터 제거하고, 37℃에서 해빙시킨다. 상기 단계를 2회 더 반복하여, 총 3회 동결-해빙과정을 거친다. 일단 완료되면, 막 결합된 알칼린 포스파타제를 측정에 이용할 수 있다.
50 ㎕의 분석 믹스 (50 mM 글리신, 0.05 % 트리톤 X-100, 4 mM MgCl2, 5 mM p-니트로페놀 포스페이트, pH=10.3) 을 각 분석 홈에 첨가한 다음 분석 플레이트를 분당 60 진동되는 진탕 수욕에서 30 분간 37 ℃로 항온 배양시킨다.
30 분의 항온 배양후, 각 홈에 100 ㎕의 0.2 N NaOH를 첨가하고 얼음위에 분석 플레이트를 위치시켜 반응을 중단시킨다.
각 홈에 대한 분광 광도 흡수를 405 나노미터의 파장에서 판독한다. 이어서상기 수치를 공지된 표준과 비교하여 각 시료에서의 알칼린포스파타제의 활성의 측정을 수득한다. 예를 들면, 공지된 양의 p-니트로페놀 포스페이트를 사용하여, 흡수치를 수득한다. 상기를 표 3에 명시한다.

C. 오스테오칼신 RIA 방법
W-20 세포를 10 % 가열 불활성화된 소 태아 혈청, 2 mM 글루타민을 함유하는 DME의 2 ㎖중 24 홈 멀티홈 조직 배양 접시의 홈당 106세포가 되도록 플레이트한다. 세포가 37 ℃에서 95 % 공기 5 % CO2의 대기하에서 하룻밤동안 부착되도록 한다.
다음날 배지를 총 부피가 2 ㎖인 10 % 소 태아 혈청, 2 mM 글루타민 및 시험 물질을 함유하는 DME 로 교환한다. 각 시험 물질을, 홈이 3 배 되도록 투여한다. 시험 물질을 48 시간 되었을 때 동일 시험 배지로 교환하고 총 96 시간 동안 W-20 세포와 항온배양시킨다.
96 시간후, 50 ㎕의 시험 배지를 각 홈으로 부터 제거하고, 쥐 오스테오칼신에 대한 방사능면역분석법을 사용하여 오스테오칼신 생성을 분석한다. 분석의 상세한 것은 키트 [Biomedical Technologies Inc. 사 제; 378 Page Street, Stoughton, MA 02072]에 기재되어 있다. 분석용 시약은 BT-431 (쥐 오스테오칼신 표준), BT-432 (염소 항-쥐 오스테오칼신), BT-431R (요오드화 쥐 오스테오칼신), BT-415 (정상 염소 혈청) 및 BT-414 (당나귀 항-염소 IgG) 와 같이 제품 번호로 나타낸다. BMP 처리에 대한 반응으로써, W-20 세포에 의해 합성된 오스테오칼신에 대한 RIA를, 제조자에 의해 제공된 설명서에 기재되어 있는 바와 같이 수행한다.
실시예 6
항생제를 포함하는 장치의 이식
0.55 g의 카르복시메틸셀룰로스 (CMC) 건조 분말을 반코마이신 HCl 또는 젠타마이신 황산염 항생제의 표 5에 나타낸 양과 함께 20 ㎖의 글리세롤/H2O (75 : 25 v/v) (또는 60 : 40 v/v) 에 첨가한다.

본 발명의 장치의 효과는 하기의 분석법으로 평가될 수 있다.
개 두개골 손상 이식 분석
2 마리의 수컷 및 2 마리의 암컷 개를 일반적으로 마취시키고, 각 두개골에 아크라-컷트 DG II 두개골용 드릴을 사용하여 각각 약 12 mm의 4 개 천공 구멍을 낸다. rhBMP-2, PLGA 다공성 입자, 황산칼슘 헤미히드레이트 - 함유 물질, 및 응집되어 성형가능 이식물을 형성할 수 있는 자가이식 혈액으로 각 구멍을 충전시킨다. rhBMP-2 의 투여량은 각 이식물에 대해 약 200 ㎍/㎖이다. 수컷 1 마리 및 암컷 1 마리의 동물은 4 주만에 희생되고, 나머지 동물들은 외과시술후 8 주만에 희생되었다.

쥐 전위성 연구
24 마리의 롱-에반스 쥐를 6 개 시험군으로 나눈다. 각각은 투여량이 0 또는 20 ㎍/100 ㎕ 중 어느 하나인 rhBMP-2/PLGA 다공성 입자/응혈 및 황산칼슘 헤미히드레이트 - 함유 물질이 없는; 200 mg 칼셋 (POP); 또는 200 mg 하프셋 (35 : 65 POP : HA)를 갖는, 200 ㎕의 크기의 피하이식을 받았다.
14 일 후, 쥐를 희생시켜 각 동물을 골 형성에 대해 평가한다.
쥐 대퇴골 손상 이식 분석
미리-천공된 폴리에틸렌 플레이트를 대퇴골의 전방부에 부착시키고 카바이드 치과용 드릴로 골 절편을 절단시킴으로써, 56 마리의 각 수컷 롱에반스 퇴화 번식 쥐 (450 ∼ 550 그램)의 좌측 대퇴골의 중심-골간에 결정적 크기의 손상 (5 mm)을 외과적으로 낸다. rhBMP-2 (다양한 양으로), PLGA 다공성 입자, 황산칼슘 헤미히드레이트 - 함유 물질 및 쥐 정맥혈액을 혼합하고, 혈액을 응집시켜, 성형가능한 이식물을 제조함으로써 생분해 가능한 이식물을 제조한다. 각 7 마리의 동물로 된 8 개 군에 하기와 같이 이식한다 : 0 ㎍ rhBMP-2; 0.93 ㎍ rhBMP-2; 3.1 ㎍ rhBMP-2; 및 9.3 ㎍ rhBMP-2.
0 일 및 3 주, 6 주 및 9 주에 동물을 방사선촬영한다. 동물을 9 주에 희생시켜, 조직학적 조사를 위해 폴리에틸렌 플레이트 주위의 조직을 제거하고, 이식되고 대측성 비-이식된 대퇴골을 수확한다. 각 군에서 2 개의 대퇴골을 조직학적으로 조사하고, 남아있는 대퇴부는 생물역학적 시험을 위해 사용한다.
Claims (7)
- 골형성 단백질, 0.1 ∼ 5.0 % (w/v)의 당, 1.0 ∼ 10.0 % (w/v)의 글리신, 및 1 ∼ 20 mM의 글루탐산 히드로클로라이드를 함유하고, 3.5 내지 6.0의 pH 를 갖는 제제인 조성물.
- 제 1 항에 있어서, 0.01 ∼ 0.1 % (w/v)의 비이온성 계면활성제를 더 함유하는 제제인 조성물.
- 골형성 단백질, 1 ∼ 10% (w/v)의 글리신, 0.1 ∼ 5.0 % (w/v) 의 수크로스, 0.01 ∼ 0.1 % (w/v) 의 비이온성 계면활성제 및 5 ∼ 10 mM 의 글루탐산 히드로클로라이드를 함유하고, 3.5 내지 6 의 pH를 갖는 제제인 조성물.
- 골형성 단백질, 2.0 ∼ 2.5 % (w/v) 의 글리신, 0.5 ∼ 2.5 % (w/v) 의 수크로스, 5 ∼ 7.5 mM 의 글루탐산 히드로클로라이드, 및 0.01 ∼ 0.05 % (w/v)의 폴리소르베이트 80을 함유하고, 3.5 ∼ 5.25의 pH 를 갖는 제제인 조성물.
- 3.12 % ∼ 24.38 % (w/w) 의 BMP; 0.52 % ∼ 10.27 % (w/w) 의 글루탐산 히드로클로라이드; 38.4 % ∼ 75.7 % (w/w) 의 글리신; 14.28 ∼ 47.15 % (w/w)의 수크로스, 및 선택적으로 0.15 % ∼ 2.94 % (w/w)의 폴리소르베이트 80를 함유한 동결건조 제제를 함유하는 조성물.
- 제 5 항에 있어서, 제제가 약 4.0 mg/㎖의 BMP-2; 약 0.918 mg/㎖의 글루탐산 히드로클로라이드; 약 25 mg/㎖의 글리신; 약 5 mg/㎖의 수크로스; 및 선택적으로 약 0.1 mg/㎖의 폴리소르베이트 80의 상대적인 양을 함유하는 조성물.
- 제 5 항에 있어서, 제제가 약 2.0 mg/㎖의 BMP-2; 약 0.918 mg/㎖의 글루탐산 히드로클로라이드; 약 25 mg/㎖의 글리신; 약 5 mg/㎖의 수크로스; 및 선택적으로 약 0.1 mg/㎖의 폴리소르베이트 80의 상대적인 양을 함유하는 조성물.
