- 前記対象に投与される前記オルソゴナルヒトCAR-T細胞の用量が、該対象の体重1kg当たり100,000~1,000,000個のオルソゴナルヒトCAR-T細胞であるように使用される、請求項32~39のいずれか一項に記載の薬学的組成物。
- 前記対象の体重1kg当たり10,000~1,000,000個の活性化オルソゴナルヒトCAR-T細胞のレベルを維持するために、前記hoIL2が、該対象に少なくとも1か月の期間にわたって定期的に投与されるように使用される、請求項32~39のいずれか一項に記載の薬学的組成物。
- 請求項32~39のいずれか一項に記載の薬学的組成物による処置の後の新生物疾患の再発を処置するための薬学的組成物であって、
hoIL2の医薬製剤の治療有効量を含み、かつ、
追加の量のオルソゴナルヒトCAR-T細胞を投与することなく、該hoIL2の投与が患者において過去に投与されたオルソゴナルヒトCAR-T細胞の活性化および/または増殖を誘導するように、該患者に投与される、
前記薬学的組成物。 - ヒト対象における新生物疾患を処置する方法において使用するための薬学的組成物であって、該薬学的組成物がオルソゴナルリガンドを含み、該方法が以下の工程を含む、前記薬学的組成物:
(i)該対象に、該オルソゴナルリガンドの治療有効用量を投与する工程、
(ii)次いで、該対象に、請求項1~31のいずれか一項に記載のオルソゴナルヒトCAR-T細胞を、1日から2週間の期間内に投与する工程であって、該オルソゴナルリガンドが、オルソゴナルCAR-T細胞のオルソゴナルCD122受容体の同種hIL2リガンドである、投与する工程、
(iii)新生物疾患の証拠の欠如が観察されるまで、(i)のオルソゴナルリガンドおよび(ii)のオルソゴナルヒトCAR-T細胞を該対象に投与する工程、ならびに
(iv)新生細胞の免疫監視を維持するのに十分な循環オルソゴナルCAR-T細胞のレベルを維持するのに十分な用量で、該対象に前記オルソゴナルリガンドの治療有効量を投与する工程。 - ヒト対象における新生物疾患を処置する方法において使用するための薬学的組成物であって、該薬学的組成物が請求項1~31のいずれか一項に記載のオルソゴナルヒトCAR-T細胞を含み、該方法が以下の工程を含む、前記薬学的組成物:
(i)該対象に、オルソゴナルリガンドの治療有効用量を投与する工程、
(ii)次いで、該対象に、該オルソゴナルヒトCAR-T細胞を、1日から2週間の期間内に投与する工程であって、該オルソゴナルリガンドが、オルソゴナルCAR-T細胞のオルソゴナルCD122受容体の同種hIL2リガンドである、投与する工程、
(iii)新生物疾患の証拠の欠如が観察されるまで、(i)のオルソゴナルリガンドおよび(ii)のオルソゴナルヒトCAR-T細胞を該対象に投与する工程、ならびに
(iv)新生細胞の免疫監視を維持するのに十分な循環オルソゴナルCAR-T細胞のレベルを維持するのに十分な用量で、該対象に前記オルソゴナルリガンドの治療有効量を投与する工程。 - 5’から3’にかけて、第1の核酸配列、第2の核酸配列、および第3の核酸配列を含む核酸配列を含む組換え発現ベクターであって、
(A)該第1の核酸配列が、細胞表面発現のためのシグナルペプチドおよびキメラ抗原受容体(CAR)を含むポリペプチドをコードし、該CARが抗原結合ドメイン(ABD)、任意のヒンジ領域、膜貫通ドメイン、および細胞内シグナル伝達ドメイン(ISD)を含み、
(i)前記CARの抗原結合ドメインが、CD123、CD19、CD20、BCMA、CD22、CD30、CD70、GPC3、Lewis Y、GD3、GD3、メソテリン、ROR CD44、CD171、EGP2、EphA2、ErbB2、ErbB3/4、FAP、FAR IL11Ra、PSCA、PSMA、NCAM、HER2、NY-ESO-1、MUC1、CD123、FLT3、B7-H3、CD33、IL1RAP、CLL1(CLEC12A)、PSA、CEA、VEGF、VEGF-R2、CD22、ROR1、メソテリン、c-Met、糖脂質F77、FAP、EGFRvIII、MAGE A3、5T4、WT1、KG2Dリガンド、葉酸受容体(FRa)、およびWnt1抗原からなる群より選択される腫瘍抗原に特異的に結合するポリペプチドを含み、かつ
(ii)前記CARの細胞内シグナル伝達ドメインが、ヒトCD3 ζ鎖、FcεR1γ鎖、FcεR1β鎖、MB1(Igα)鎖、B29(Igβ)鎖、CD3δポリペプチド、CD3Δポリペプチド、およびCD3εポリペプチドからなる群より選択されるシグナル伝達ドメインを含み、任意で、前記CARの細胞内シグナル伝達ドメインが、CD28、CD137(4-1BB)、CD134(OX40)、Dap10、CD27、CD2、CD5、ICAM-1、LFA-1(CD11a/CD18)、Lck、TNFR-I、TNFR-II、Fas、CD30、およびCD40共刺激ドメインからなる群より選択される共刺激ドメインをさらに含み、
(B)該第2の核酸配列が、T2Aポリペプチドをコードし、
(C)該第3の核酸配列が、細胞表面発現のためのシグナルペプチドおよびオルソゴナル受容体ポリペプチドをコードし、該オルソゴナル受容体ポリペプチドが、細胞外ドメイン、膜貫通ドメイン、および細胞内ドメインを含み、

該核酸配列が、ヒト細胞中で機能的な1つまたは複数の発現制御配列に機能的に連結されている、前記組換え発現ベクター。 - 前記オルソログ受容体ポリペプチドの細胞内ドメインおよび膜貫通ドメインが、ヒトCD122の細胞内ドメインおよび膜貫通ドメイン、ヒトIL4受容体II型受容体サブユニットa(hIL4Ra)の細胞内ドメインおよび膜貫通ドメイン(SEQ ID NO:9および8)、ヒトIL-7受容体サブユニットa(hIL7Ra)の細胞内ドメインおよび膜貫通ドメイン(SEQ ID NO:11および10)、ヒトIL9受容体(hIL9R)の細胞内ドメインおよび膜貫通ドメイン(SEQ ID NO:13および12)、ならびにヒトIL21受容体(hIL21R)の細胞内ドメインおよび膜貫通ドメイン(SEQ ID NO:15および14)からなる群より選択される、請求項45または46に記載の組換えベクター。
- 前記オルソゴナル受容体ポリペプチドの細胞内ドメインおよび膜貫通ドメインが、ヒトCD122の細胞内ドメインおよび膜貫通ドメインである、請求項47に記載の組換えベクター。
- 前記オルソゴナル受容体ポリペプチドの細胞内ドメインが、少なくとも1つのSTAT3シグナル伝達モチーフをさらに含み、該STAT3シグナル伝達モチーフが、式YX 1 X 2 Qのポリペプチドを含み、式中、

- 以下の工程を含む、オルソゴナルヒトCAR-T細胞が実質的に濃縮された細胞製品を調製する方法:
(a)ヒト対象から単離されたある量のT細胞に、請求項45~59のいずれか一項に記載の組換えベクターで形質導入する工程、および
(b)工程(a)からの単離された前記量のT細胞を、エクスビボで、工程(a)の接触によって形質導入された細胞の増殖を誘導するのに十分な量のhoIL2と接触させる工程であって、該形質導入された細胞が集団の細胞の少なくとも20%を構成するように一定期間適用される、接触させる工程。 - (a)細胞表面発現のためのシグナルペプチドおよびオルソゴナルCD122受容体ポリペプチドをコードする第1の核酸配列であって、該オルソゴナルCD122受容体ポリペプチドが以下を含む、第1の核酸配列:
ならびに
(b)細胞表面発現のためのシグナルペプチドおよびCD19 CARを含むポリペプチドをコードする第2の核酸配列であって、該ポリペプチドが以下を含む、第2の核酸配列:
を含む、ヒトオルソゴナルCD19 CAR-T細胞であって、
前記オルソゴナルCD122受容体ポリペプチドおよび前記CD19 CARが、ヒトオルソゴナルCD19 CAR-T細胞により発現され、かつオルソゴナルCD122受容体ポリペプチドおよびCD19 CARの細胞外ドメインが細胞表面に提示されるように、第1の核酸配列および第2の核酸配列が、ヒト免疫細胞において機能的な1つまたは複数の発現制御エレメントに機能的に連結されている、
前記ヒトオルソゴナルCD19 CAR-T細胞。 - 治療有効量の補助作用物質と組み合わせて使用される、請求項62に記載の薬学的組成物。
- 前記補助作用物質が抗PD1または抗PDL1チェックポイント阻害抗体である、請求項63に記載の薬学的組成物。
- 前記対象が、ヒトオルソゴナルCD19 CAR-T細胞の投与の前にリンパ球枯渇レジメンで処置されている、請求項62~64のいずれか一項に記載の薬学的組成物。
- 先行リンパ球枯渇の非存在下で使用される、請求項62~64のいずれか一項に記載の薬学的組成物。
- 約10,000~約1,000,000個のヒトオルソゴナルCD19 CAR-T細胞が前記対象において少なくとも2か月の期間にわたって維持されるように、該対象を、hIL2オルソログを含む医薬製剤の治療有効量と定期的に接触させる、請求項62~66のいずれか一項に記載の薬学的組成物。
- 前記新生物疾患が血液悪性腫瘍である、請求項62~67のいずれか一項に記載の薬学的組成物。
- 血液悪性腫瘍が、再発したもしくは難治性の非ホジキンリンパ腫、再発したもしくは難治性の骨髄腫、再発したもしくは難治性の大細胞型B細胞リンパ腫、または再発したもしくは難治性のマントル細胞リンパ腫である、請求項68に記載の薬学的組成物。
- 前記がんが肝臓がんである、請求項70に記載の薬学的組成物。
- 前記肝臓がんが肝細胞がんである、請求項71に記載の薬学的組成物。
