本願は、本願に援用されている、米国仮出願第62/444,660号(出願日:2017年1月10日)、米国仮出願第62/466,298号(出願日:2017年3月2日)、米国仮出願第62/500,203号(出願日:2017年5月2日)、米国仮出願第62/523,191号(出願日:2017年6月21日)、米国仮出願第62/523,200号(出願日:2017年6月21日)、米国仮出願第62/573,079号(出願日:2017年10月16日)、米国仮出願第62/580,783号(出願日:2017年11月2日)に対して優先権を主張する。
a)インテグリン結合性ポリペプチド−Fc融合体が発現する条件下で、請求項32に記載の宿主細胞を培養するステップと、
b)前記インテグリン結合性ポリペプチド−Fc融合体を回収するステップと、を含む、方法を、提供する。
I.前書き
インターロイキン−2(IL−2)は、T細胞およびNK細胞の増殖を活性化および誘導する多面性サイトカインである。IL−2はFDA承認の両方ではあるが、IL−2を用いる全身療法は著しい毒性を有し、患者の奏効率も25%未満に過ぎない。半減期を延長したIL−2と、腫瘍特異的抗原に対する抗体を組み合わせると、かなり有望な治療効果が期待できる。しかしながら、抗体を応用する療法は、ほとんどの腫瘍には既知の腫瘍関連抗原を持たないという弱点を抱えている。
請求の範囲および明細書で用いられる用語は、別途の明記がない限り、次のように定義される。原出願の仮出願書で用いられる用語と矛盾する場合は、本明細書の用語が優先するものとする。

本発明のインテグリン結合性ポリペプチド−Fc融合体は、IL−2および/または延長IL−2の非存在下で用いることができる。いくつかの実施形態では、本発明のインテグリン結合性ポリペプチド−Fc融合体はIL−2と組み合わせて用いることができ、半減期延長IL−2の使用を必要としない。いくつかの実施形態では、インテグリン結合性ポリペプチド−Fc融合体は、半減期延長IL−2と組み合わせて用いてもよい。
ある実施形態では、有効量のヒトIL−2を全身投与する。いくつかの実施形態では、有効量の拡張PK・IL−2を全身投与する。一実施形態では、IL−2は、プロロイキン(登録商標)(アルデスロイキン)などのヒト組換えIL−2である。プロロイキンは、大腸菌で産生されたヒト組換えインターロイキン−2製品である。プロロイキンは、(a)グリコシル化されていない、(b)N末端アラニンがない、(c)アミノ酸125位のシステインがセリンに置換されているという点で、天然インターロイキン−2とは異なる。プロロイキンは、生物学的活性を有し、非共有結合した微小凝集体として存在し、平均サイズは組換えインターロイキン−2分子27個に相当する。プロロイキン(アルデスロイキン)は、静脈内注入によって投与される。いくつかの態様では、拡張PK・IL−2のIL−2部分は野生型IL−2である(例えば、前駆体形のヒトIL−2(国際公開第WO2016/025642号に記載の配列番号33、本願に援用されている)、または成熟IL−2(国際公開第WO2016/025642号に記載の配列番号35、本願に援用されている)など)。
IL−2は、単独で、または本明細書に記載されるものなどのキメラポリペプチドの一部として、核酸分子の発現により得ることができる。従って、配列番号4、6、8、10、12、14、16、18、20、22、24、26、28、30、32、34の核酸配列を有するものなど、IL−2またはIL−2変異体を含むポリペプチドをコード化する核酸分子は、本発明の請求の範囲に属するものとして見なされる。野生型IL−2との同一性を以ってIL−2変異体を記述しているので、IL−2変異体をコードかする核酸分子も、必然的に野生型IL−2をコード化する核酸分子とある程度の同一性を有することになる。例えば、例えば、IL−2変異体をコード化する核酸分子は、全長の野生型IL−2(例えば、本願に援用されている国際公開第WO2016/025642号の配列番号32)、またはシグナルペプチドを含まない野生型IL−2(例えば、本願に援用されている国際公開第WO2016/025642号の配列番号34)をコード化する核酸に対して、少なくとも50%、少なくとも65%、好ましくは少なくとも75%、より好ましくは少なくとも85%、最も好ましくは少なくとも95%(例えば99%)の同一性を有する。
上記のように、IL−2または変異体IL−2は拡張PK基と融合して、循環半減期を延長することができる。拡張PK基の例としては以下のものが挙げられるが、これらに限定はされない。IL−2またはそのバリアントの循環半減期を延長する他のPK基も、拡張PK・IL−2に適用することができる。
ある実施形態では、拡張PK基は、血清アルブミンまたはその断片である。血清アルブミンをタンパク質に融合する方法は、例えば、本願にも援用されている米国公開第2010/0144599号、米国公開第US2007/0048282号、米国公開第2011/0020345号に開示されている。ある実施形態では、拡張PK基は、米国特許第5,876,969号、国際公開第2011/124718号、国際公開第WO2013/075066号、国際公開第WO2011/0514789号に記載されている、HSA、またはそのバリアントまたは断片である。
ある実施形態では、本明細書で用いられる拡張PK・IL−2は、ポリエチレングリコール(PEG)ドメインを含む。PEG化がタンパク質の循環半減期を増加させることは、当技術分野で周知の事実である。PEG化の方法については、例えば、本願に援用されている米国特許第7,610,156号、米国特許第7,847,062号に開示されている。
ある実施形態では、拡張PK基は、本願に援用されている米国特許第7176278号および米国特許第8158579号に開示されているように、トランスフェリンである。
ある実施形態では、国際公開第WO2013/177187号に記載されているように、拡張PK・IL−2はFcドメインを含む。Fcドメインには、抗原に結合する可変領域は含まれていない。本明細書に記載の拡張PK・IL−2の産生に有用に用いられるFcドメインは、様々な供給源から入手することができる。ある実施形態では、拡張PK・IL−2のFcドメインは、ヒト免疫グロブリンに由来する。ある実施形態では、Fcドメインは、ヒトIgG1定常領域に由来する(例えば、配列番号126)。ヒトIgG1のFcドメインは、配列番号126に記載されている。ある実施形態では、ヒトIgG1のFcドメインは、上側ヒンジ領域を有しない(本願に援用されている国際公開第WO2016/025642号の配列番号3)。しかしながら、Fcドメインは、例えばげっ歯類(例えば、マウス、ラット、ウサギ、モルモット)または非ヒト霊長類(例えば、チンパンジー、マカク)を含む、その他の哺乳類の免疫グロブリンに由来してもよい。さらに、Fcドメインまたはその一部は、IgM、IgG、IgD、IgA、IgEを含む免疫グロブリンクラス、並びに、IgG1(例えば、配列番号126)、IgG2(例えば、配列番号127)、IgG3(例えば、配列番号128)、IgG4(例えば、配列番号129)を含む免疫グロブリンアイソタイプに由来してもよい。
いくつかの実施形態では、IL−2をインターフェロン−α(INFα)で置き換えることができる。いくつかの実施形態では、INFαは天然ヒトINFαである。いくつかの実施形態では、INFαは、ヒト血清アルブミンと融合した米国公開第2006/0051859号に記載されているものなど、長時間作用型INFαである。いくつかの実施形態では、IFNαはUniProtKB参照番号P01562またはP01563の配列を有する。いくつかの実施形態では、IFNαはUniProtKB参照番号P01562またはP01563の配列の機能的バリアントを有する。いくつかの実施形態では、IFNαは、UniProtKB参照番号P01562またはP01563(配列番号148または154)の配列に対して80%、85%、90%、95%、または100%の同一性を有する配列を含む。いくつかの実施形態では、IFNαは、メルク社から市販されるイントロン−Aである(例えば、米国特許第6610830号およびhttps://www.merck.com/product/usa/pi_circulars/i/intron_a/intron_a_pi.pdfを参照)。いくつかの実施形態では、IFNαはPEG−IFNαである。いくつかの実施形態では、IFNαはペギントロンである(例えば、米国特許第6610830号および第6180096号を参照)。いくつかの実施形態では、IFNαはシラトロンである(例えば、米国特許第6610830号および第6180096号を参照)。
インテグリンは、充実性腫瘍の発生、進行、転移に重要な役割を担う各種の細胞機能を調節する細胞買いマトリックス接着受容体のファミリーである。腫瘍の進行においてあまりも重要であるため、インテグリンはがん療法の有効な目標として見なされ、様々な種類のがんの治療が可能となった。がん細胞に存在するインテグリンとしては、ανβ1、ανβ3、ανβ5、ανβ6、α5β1が挙げられる。
Fcドメインには、抗原に結合する可変領域は含まれていない。本明細書に記載のインテグリン結合性ポリペプチド−Fc融合体有用に用いられるFcドメインは、様々な供給源から入手することができる。ある実施形態では、拡張PK・IL−2のFcドメインは、ヒト免疫グロブリンに由来する。ある実施形態では、Fcドメインは、ヒトIgG1定常領域に由来する(図1、配列番号126)。ヒトIgG1のFcドメインの例は、配列番号126に記載されている(図1)。ある実施形態では、ヒトIgG1のFcドメインは上側ヒンジ領域を有しない(図1および表1)。しかしながら、Fcドメインは、例えばげっ歯類(例えば、マウス、ラット、ウサギ、モルモット)または非ヒト霊長類(例えば、チンパンジー、マカク)を含む、その他の哺乳類の免疫グロブリンに由来してもよい。さらに、Fcドメインまたはその一部は、IgM、IgG、IgD、IgA、およびIgEを含む免疫グロブリンクラス、およびIgG1、IgG2、IgG3、およびIgG4を含む免疫グロブリンアイソタイプに由来してもよい。Fcドメインはマウスであってもよく、ヒトであってもよい。
ノッチンポリペプチド足場は、好ましくはループの形でインテグリン結合性配列を挿入して、特定のインテグリン結合をもたらすために用いられる。インテグリン結合は、好ましくは、RGDペプチドなどのインテグリン結合ペプチド配列を挿入することにより、ノッチンポリペプチド足場に設計される。いくつかの実施形態では、インテグリン結合性ペプチド配列の挿入により、天然ノッチンタンパク質の一部が置換される。例えば、一実施形態では、ループの全部または一部を、1または複数のインテグリンと結合するために選択された含RGDペプチド配列(例えば、5〜12個のアミノ酸配列)で置換することにより、RGDペプチド配列を天然溶媒曝露ループに挿入する。溶媒曝露ループ(つまり、表面上)は一般的に、天然ノッチンタンパク質配列におけるジスルフィド連結システイン残基によって固定される。インテグリン結合性置換アミノ酸配列は、ループ部分のコドンをランダム化し、改変ペプチドを発現させ、所定の配位子との結合度が最も高い変異体を選択することにより取得できる。上記の選択ステップを数回繰り返し、前のステップで最も強く結合したタンパク質を取り、ループを再ランダム化する。
Fc融合体に用いられるインテグリン結合性ポリペプチドには、インテグリン結合性ループ(例えば、RGDペプチド配列)およびノッチンポリペプチド足場が含まれる。上記のインテグリン結合性ポリペプチドは、本願に援用されている米国特許第8536301号に記載されている。米国特許第8536301号に記載されているように、インテグリン結合性ポリペプチドは、結合特異性および効力に影響を与えない範囲で、非RGD残基がある程度変化していてもよい。たとえば、11個の残基のうち3つが変化した場合、2.5Dに対して約70%の同一性を有することになる。表1は、本発明の範囲内のインテグリン結合性ポリペプチドの例、およびそれらの特定のノッチンポリペプチド足場(例えば、EETI−IIまたはAgRP)を示す。いくつかの実施形態では、Fc融合体に用いられるインテグリン結合性ポリペプチドは、本明細書に記載のGCPRPRGDNPPLTCSQDSDCLAGCVCGPNGFCG(2.5F、配列番号130)およびGCPRPRGDNPPLTCKQDSDCLAGCVCGPNGFCG(2.5FmodK、配列番号131)、並びにGCPRPRGDNPPLTCSQDSDCLAGCVCGPNGFCGGGGGS(配列番号132)、GCPRPRGDNPPLTCKQDSDCLAGCVCGPNGFCGGGGGS(配列番号133)、GCPRPRGDNPPLTCSQDSDCLAGCVCGPNGFCGGGGGSGGGGSGGGGS(配列番号134)、および/またはGCPRPRGDNPPLTCKQDSDCLAGCVCGPNGFCGGGGGSGGGGSGGGGS(配列番号135)である、ペプチド2.5Fおよび2.5FmodKである。
本明細書、並びに本願に援用されている米国公開第2014/0073518号に記載されたインテグリン結合性ポリペプチド−Fc融合体(ノッチン−Fc融合体)は、改変インテグリン結合性ポリペプチド(ノッチン足場内)と、FcyRと結合してADCCを誘導することのできる、Fcドメインまたは抗体様構築体と、を組み合わせる。
様々なFcドメイン遺伝子配列(例えば、マウスおよびヒト定常領域遺伝子配列)は、公的保存機関に寄託された資源として入手することができる。Fcドメイン配列を含む定常領域ドメインは、特定のエフェクター機能を欠くもの、および/または免疫原性を低下させる特定の修飾を有するものを選択することができる。抗体および抗体をコード化する遺伝子における多くの配列は既に公開されており、よって、当技術分野で認定されている技術を用いてこれらの配列から適切なFcドメイン配列(例えば、ヒンジ、CH2、および/またはCH3配列、またはその一部)を得ることができる。次いで、前述の方法のいずれかを用いて取得した遺伝物質を変更または合成すると、本明細書で用いられるポリペプチドが得られる。さらに、定常領域DNA配列の対立遺伝子、バリアント、突然変異を、本明細書に開示の方法に適用することもできる。

ある実施形態では、免疫チェックポイント調節物質(阻害物質または刺激物質)は、本明細書に記載の他の治療薬(例えば、IL−2、拡張PK・IL−2、INFα、および/またはインテグリン結合性Fc融合タンパク質)と組み合わせて用いられる。T細胞の活性化とエフェクター機能は、「免疫チェックポイント」と呼ばれる共刺激および阻害シグナルによりバランスが取れている。T細胞エフェクター機能を調節する阻害性配位子および受容体は、腫瘍細胞で過剰に発現する。次いで、共刺激受容体のアゴニストまたは阻害シグナルのアンタゴニストにより、抗原特異的T細胞応答が増幅される。
ある実施形態では、免疫チェックポイント調節物質は免疫チェックポイント阻害物質である。腫瘍細胞を直接標的とする治療用抗体とは異なり、免疫チェックポイント阻害物質は内因性の抗腫瘍活性を高める。ある実施形態では、本明細書に開示の方法での使用に適する免疫チェックポイント阻害物質は、阻害シグナルのアンタゴニスト、例えば、PD−1、PD−L1、CTLA−4、B7−H3、B7−H4を標的とする抗体である。これらの配位子と受容体については、Pardoll, D., Nature. 12: 252−264, 2012で検討されている。
ある実施形態では、免疫チェックポイント調節物質は免疫チェックポイント刺激物質である。腫瘍細胞を直接標的とする抗体医薬とは異なり、免疫チェックポイント刺激物質は、内在性免疫系の活性化増進および/または内在性免疫系の抑制活性の低減を行う。ある実施形態では、本明細書で開示される方法において好適に用いられる免疫チェックポイント刺激物質は、刺激性シグナルのアゴニストまたは抑制性シグナルのアンタゴニストであり、例えば、抗4−1BB/CD137抗体、抗IFNα抗体、抗GITR抗体、および抗OX40抗体などである。これらのリガンドおよび受容体については、Peggs, K.S., et al., Clin Exp Immunol., 157(1): 9-19 (2009)にて概説されている。
ある実施形態では、血管内皮増殖因子(VEGF)のアンタゴニストが、免疫チェックポイント阻害物質の代わりに使用される。最近、VEGFは免疫抑制に関与することが示されている(Liang, W.-C. et al. J. Biol. Chem. (2006) Vol 281: 951-961; Voron, T. et al. Front Oncol (2014) Vol. 4: Article 70; Terme, M. et al, Clin Dev Immunol (2012) Vol. 2012: Article ID 492920; Kandalaft, E. et &\.,Curr Top Microbiol Immunol (201 1) Vol 344: 129-48)。そのため、VEGFの活性を遮断することによって、免疫チェックポイント阻害物質の場合と同様に、免疫応答が増強されると考えられる。「VEGFアンタゴニスト」とは、1または複数のVEGF受容体への結合性を包含するVEGF活性を、中和、遮断、阻害、抑制、低減または干渉可能な分子を指す。VEGFアンタゴニストの非限定的な例としては、抗VEGF抗体およびその抗原結合性断片、VEGFに特異的に結合することで1もしくは複数の受容体(例えば、VEGF受容体)に対するVEGFの結合性を封鎖する受容体分子および誘導体、抗VEGF受容体抗体、VEGFRチロシンキナーゼの小分子阻害剤などのVEGF受容体アンタゴニスト、並びにドミナントネガティブVEGFが挙げられる。
ある実施形態では、前記長期PK基は、所望によりリンカーを介して、IL−2に融合されていてもよい。ある実施形態では、インテグリン結合性ポリペプチドは、リンカーを介してFc断片に融合されている。好適なリンカーが当該技術分野においては周知であり、例えば、その全体が参照により本明細書に援用される米国特許出願公開第2010/0210511号、同第2010/0179094号、および同第2012/0094909号に開示されているリンカーなどがある。代表的なリンカーとして、グリシン―セリンポリペプチドリンカー、グリシン−プロリンポリペプチドリンカー、およびプロリン−アラニンポリペプチドリンカーが挙げられる。ある実施形態では、リンカーはグリシン―セリンポリペプチドリンカー、すなわち、グリシン残基とセリン残基とからなるペプチドである。
本明細書で開示される方法において好適に使用されるインテグリン結合性Fc融合タンパク質は、1または複数の治療薬と併用することができる。一実施形態では、治療薬は抗体医薬である。別の実施形態では、治療薬は治療用タンパク質である。別の実施形態では、治療薬は小分子である。別の実施形態では、治療薬は抗原である。別の実施形態では、治療薬は細胞集団である。
また、本発明は、2つ以上のIL−2と、本明細書に記載の抗体(例えば、抗体医薬、免疫チェックポイント阻害物質、またはVEGFと拮抗する抗体)または抗体断片とを含んでなる、改変分子を提供する。そのような改変分子は、本明細書に記載の方法において対象(例えば、がん患者)に投与されることとなる成分の数を、効果的に減らすことができる。いくつかの実施形態では、前記抗体または抗体断片は、他の構成成分(例えば、IL−2)と結合する際の足場として機能する。
いくつかの態様において、本明細書に記載のポリペプチド(例えば、長期PK性IL−2などのIL−2、ノッチンFc、インテグリン結合性タンパク質Fc融合体)は、組換えDNA技術を用いて、形質転換した宿主細胞において作製される。そのために、前記ペプチドをコードする組換えDNA分子が作製される。そのようなDNA分子の作製法は当該技術分野において周知である。例えば、好適な制限酵素を用いて、前記ペプチドをコードする配列をDNAから切り出してもよい。あるいは、前記DNA分子を、ホスホルアミダート法などの化学合成手法を用いて合成してもよい。また、これら手法を組み合わせたものを用いてもよい。
上記の核酸分子を、例えばベクターを導入された細胞内で自身の発現を指示できるベクター内に、含有させることができる。従って、長期PK性IL−2変異体およびノッチン−Fc変異体に加えて、長期PK性IL−2変異体またはノッチン−Fc変異体をコードする核酸分子を含有する発現ベクター、およびこれらのベクターをトランスフェクトした細胞も、実施形態に含まれる。

これは、例えばCancer Research 50. 2009-2017, ’90に記載されており、その表4が以下に記載される。
IL−2は、皮下注射により0.5または1.0MU/m2の用量で投与した。活性は、注射の0.5、1、2、3、4、6、8、および24時間後に採取した血清において測定したものである。中央値用量の正規化されたピークレベル(10.7ユニット/ml)およびピーク時間(180分)は、筋肉内投与後に観察された対応するそれぞれの値(14.0ユニット/mlおよび150分)と類似している(表4)。筋肉内での試験と比較して、この試験では服用レベルが低くなり、意味のあるAUC値が得られなかった。
本明細書に記載されるような、インテグリン結合性ポリペプチド−Fc融合体および/またはそれを発現する核酸は、異常なアポトーシスまたは分化過程と関連した障害(例えば、がんなどの細胞増殖障害または細胞分化障害)の治療に有用である。さらに、本明細書に記載されるような、IL−2または長期PK性IL−2、インテグリン結合性ポリペプチド−Fc融合体、および所望による、阻害物質または刺激物質を含む免疫チェックポイント調節物質(またはVEGFのアンタゴニスト)、および/またはそれらを発現する核酸も、異常なアポトーシスまたは分化過程と関連した障害(例えば、がんなどの細胞増殖障害または細胞分化障害)の治療に有用である。本発明の方法で治療可能ながんの非限定的な例を以下に記載する。

MC38− NODU−E202 材料および方法
3つの有望な治療候補物質を設計し、検証した。これらバリアントは抗体Fcドメイン(ヒトIgG1)に融合された腫瘍標的化ペプチド(2.5F)を含有した。NOD201:リンカー無し、NOD203:短い[Gly4Ser]リンカー、そしてNOD204:長い[Gly4Ser3]リンカー。これら3つの構築体は、アズロシジン(Azurocidin)プレプロタンパク質由来のシグナルペプチドとともに発現された(注記:このシグナルペプチドは、細胞内でのタンパク質発現を誘導する配列である)。これら構築体のオープンリーディングフレームをコードする遺伝子は、哺乳類細胞発現にコドン最適化された。タンパク質構築体は、ヒト胎児腎細胞において一過性発現により産生された。100mLの培養液をMabSelect SuRe樹脂により精製し、UV/Vis吸光度により定量して、SDS−PAGEおよびサイズ排除クロマトグラフィーにより解析した。サイズ排除クロマトグラフィーで解析したところ、タンパク質の98%超が単量体であった(すなわち凝集していない)。
MC38−NOD−E202材料および方法
マウス
試験1日目、メスのC57BL/6マウス(C57BL/6/NCrl、チャールズリバー社)は7週齢であり、15.5〜21.8gの範囲のBWであった。動物は水(逆浸透、1ppm Cl)ならびに、18.0%の粗タンパク質、5.0%の粗脂肪、および5.0%の粗繊維からなるNIH 31 Modified and Irradiated Lab Diet(登録商標)を不断給餌された。マウスは照射済みのEnrich−o’cobs(商標)で飼育され、20〜22℃(68〜72°F)および40〜60%の湿度、12時間の明暗サイクルで、据え付けのマイクロアイソレーター中で眠った。CR Discovery Servicesは、拘束、管理、外科的手順、給餌および給水管理ならびに獣医師によるケアに関し、実験動物の管理と使用に関する指針の推奨を特に遵守している。CR Discovery Servicesでの動物の世話および使用のプログラムは、国際実験動物管理公認協会により認定されており、実験動物の管理と使用に関し承認された標準を遵守していることを保証する。
Nodus Therapeutics,Inc.から提供されたMC38齧歯類大腸癌細胞を、10%ウシ胎仔血清ならびに2mMグルタミン、100単位/mLペニシリンGナトリウム、100μg/mLストレプトマイシン硫酸塩、および25μg/mLゲンタマイシンを補充したダルベッコ改変イーグル培地(DMEM)中で増殖させた。細胞培養は、37℃の加湿インキュベーター中、5%CO2と95%空気の雰囲気下、組織培養フラスコ中で維持された。
移植に使用されたMC38齧歯類大腸細胞は対数増殖期に回収され、冷却されたRPMI培地中に再懸濁された。マウスはイソフルランで麻酔され、その後移植された。各マウスは右わき腹に1x106個の腫瘍細胞(0.1mLの細胞懸濁液)を皮下注射され、腫瘍体積が60〜180mm3の標的範囲に達するまで腫瘍を監視した。腫瘍移植7日後、試験の1日目で、63〜172mm3の範囲の個体腫瘍体積を有する動物を11群(n=10)に分けた。群の平均腫瘍体積は109〜112mm3の範囲であった。ノギスを使用して週に2回、二次元で腫瘍を測定した。腫瘍サイズは以下の式を使用して算出された:

インテグリン結合性ポリペプチド−Fc融合体のNOD201M(コード名称 KW2、ロット番号 BP−046−016−2)、プロロイキン(Proleukin)(ロット番号 502519AA)および抗PD−1(ロット番号 614616J2)。NOD201M、プロロイキンおよび抗PD−1は遮光され、4℃で保存された。すべての剤は、説明書に従い調製された。
11群のC57BL/6マウス(n=10)に、図13のMC38−NODU−e202プロトコールに従いD1に投与を開始した。ビヒクル(PBS)およびNOD201M(500または1000μg/動物で投与された)を、0.24mL/マウスの投与量で静脈内(i.v.)投与した。プロロイキン(4または40μg/動物で投与された)は、0.1mL/マウスの投与量で静脈内投与または皮下(s.c.)投与した。抗PD−1(200μg/動物で投与された)は、0.1mL/マウスの投与量で静脈内投与された。全投与量とも、個々の動物の体重に対して調製されず投与された。
腫瘍は、週に2回、ノギスを使用して測定された。各動物は、その腫瘍が1000m3のエンドポイント体積に達したとき、または試験が終了したとき(85日目)のいずれか早いときに安楽死処理された。腫瘍体積エンドポイントで本試験を離脱した動物は、安楽死の日付とともに腫瘍進行(TP:tumor progression)のために安楽死されたと記録された。解析用のエンドポイントまでの時間(TTE:The time to endpoint)は、各マウスに対し、以下の式により算出された:

治療有効性は、最終日に本試験に残った動物の腫瘍体積から決定され得る。MTV(n)は、腫瘍がエンドポイント体積に達しなかった残留動物数(n)における、本試験の最終日のメジアン腫瘍体積として規定された。
動物は本試験の最初の5日間は毎日、そしてその後は週に2回体重を測定された。マウスは健康状態を頻繁に観察され、すべての有害な治療関連(TR)副作用の明白な兆候が観察された。注目すべき臨床観察結果は記録された。個々の体重の減少はプロトコールに沿って監視され、1回の測定で30%を越える体重の減少があった動物、または3回の測定で25%を越える体重減少があった動物はすべてTR死として健康状態を理由に安楽死されるものとした。群の平均体重が回復した場合、当該群の投与は再開されたが、投与量を少なく、または投与スケジュールの頻度を少なくした。受容可能な毒性は、本試験期間中、20%未満の群平均BW減少、かつ10匹の治療動物で1匹以下、または10%以下のTR死として規定された。高い毒性を生じさせた投与レジメンはすべて、最大耐用量(MTD:maximum tolerated dose)を上回るとみなされる。死は、臨床兆候により明らかな、および/または解剖により明らかな治療副作用に起因した場合、TRとして分類されるものとした。または投与期間中もしくは最後の投与から14日以内の未知の原因に起因した場合も、TRとして分類され得る。死亡が治療関連ではなくその腫瘍モデルに関連したことが明白であった場合、その死亡はNTRと分類された。NTR死はさらにNTRa(アクシデントまたはヒューマンエラーが原因)、NTRm(解剖により確認される浸潤または転移による腫瘍播種が原因)、およびNTRu(未知の原因)としてカテゴライズされる。
ウィンドウズ7.01用のPrism(GraphPad)を図形提示および統計解析に使用した。生存率はカプランマイヤー法により解析した。ログランク検定(マンテル−コックス)およびゲーハン−ブレスロウ−ウィルコクソン検定が、TTE値に基づき、2群の全生存経験(生存曲線)の間の差異の有意性を決定した。すべての検定結果を添付のBに示す。両側統計解析をP=0.05の有意レベルで実施した。解析は、多重比較に対し補正されなかった。Prismは検定結果を、P>0.05で有意ではない(ns)、0.01<P≦0.05で有意(*の記号)、0.001<P≦0.01でとても有意(**)、およびP≦0.001で非常に有意として要約する。統計的有意性の検定は、群間の差異の程度の推定を提供しないため、有意性のすべてのレベルを、本報告書の文章内では有意か有意でないかのいずれかとして記載した。受容可能な毒性の限界を超えたレジメンの群は、統計的評価を受けなかった。
B16F10実験の説明
メスのC57BL/6マウス(C57BL/6/NCrl、チャールズリバー)は、試験開始時、8〜12週齢であった。試験開始は、腫瘍移植の日である(1日目)。動物(1群当たりn=10)は、1日目の体重に基づき治療群に無作為化された。移植に使用されたB16F10メラノーマ細胞はチャールズリバーラボラトリーから提供され、対数増殖期の間に回収され、培地中に再懸濁された。マウスは移植前にイソフルランで麻酔された。
腫瘍体積(mm3)=w2 xl/2
式中、wは腫瘍の幅、lは長さであり、単位はmmである。腫瘍の重量は、腫瘍体積1mm3は1mgに等しいという仮定で推定され得る。
1、7日目に1000μg/動物のNOD201Mを単独静脈内投与された動物、または1、7日目に200μg/動物の抗PD−1の静脈内投与、および/または2〜4、8〜9日目に4μg/動物のプロロイキンの皮下投与を併用された動物。9日目、プロロイキン注射の24時間後、腫瘍を採取し、保存し、単一細胞懸濁液に処理した。細胞懸濁液は、以下に記載される抗体を使用して細胞表面マーカーに対して染色し、フローサイトメトリーにより解析した。データは、腫瘍中のCD45+細胞の%として表される。
目的:NOD201M単独の有効性と、抗PD−1、抗PD−L1、抗CTLA−4、抗LAG3、抗TIM3、抗TIGIT、および抗CD137との併用の有効性を、メスC57BL/6マウスを使用してMC38同系大腸モデルにおいて決定する。データを図23〜24に示す。
〇 細胞移植のためにイソフルランでマウスを麻酔し、潰瘍を低減させる。
〇 240匹のCRメスC57BL/6マウスを設定し、0%Matrigel中、1x106個のMC38腫瘍細胞を用いてわき腹に皮下注射する。
〇 細胞注射体積は、0.1mL/マウスであった。
〇 開始日の齢:8〜12週齢。
〇 腫瘍が60〜180mm3の平均サイズに達したとき、ペアマッチを実施し、治療を開始する。
・約100mm3(細胞移植後約6〜7日)を標的とする。
〇 体重:5/2、その後、週に2回で最後まで。
〇 ノギス測定:週に2回で最後まで。
〇 30%を越える体重減少が1回観察された個々の動物、または25%を越える体重減少が3回連続的に測定された個々の動物はすべて安楽死された。
〇 20%を越える平均体重減少、または10%を越える死亡率があった群はすべて投与を停止する。
・ 群は安楽死されず、回復が許可される。20%を超える体重減少のあった群内で、個体体重減少エンドポイントに合致した個体は安楽死される。
・ 群の治療関連体重減少が、元の体重の10%以内に回復した場合、投与は投与量を減少させて、または投与スケジュールの頻度を低くして再開され得る。
・ 非治療体重%回復に対する除外は、個別的に許可され得る。
〇 エンドポイントTGD。動物は個々に監視された。実験のエンドポイントは、1000mm3の腫瘍体積か、55日のいずれか早い方であった。反応個体はより長く追跡され得る。エンドポイントに達したとき、動物はSOPに準じて安楽死されるものとした。

〇 投与レジメン2を最初に、次いでレジメン1。
MC38−NODU−p010
目的:NOD201M単独で治療され、および抗PD−1で併用治療され、MC38同系大腸腫瘍を担持するメスC57BL/6マウスからフローサイトメトリー用のサンプルを採取する。データを図25に示す。
〇 細胞移植のためにイソフルランでマウスを麻酔し、潰瘍を低減させる。
〇 CRメスC57BL/6マウスを設定し、0%Matrigel中、1x106個のMC38−NODU腫瘍細胞を用いてわき腹に皮下注射する。
・9日目のサンプリングが、当該週の前半に当たるように細胞移植を予定する。
〇 細胞注射体積は、0.1mL/マウスである。
〇 開始日の齢:8〜12週齢。
〇 腫瘍が60〜180mm3の平均サイズに達したとき、ペアマッチを実施し、治療を開始する。
・約100mm3(細胞移植後約6〜7日)を標的とする。
〇 体重:qdで最後まで。
〇 ノギス測定:1、3、5、7、9日目。
〇 30%を越える体重減少が1回観察された個々の動物、または25%を越える体重減少が3回連続的に測定された個々の動物はすべて安楽死された。
〇 20%を越える平均体重減少、または10%を越える死亡率があった群はすべて投与を停止した。群は安楽死されず、回復が許可された。
・ 20%を超える体重減少のあった群内で、個体体重減少エンドポイントに合致した個体は安楽死される。群の治療関連体重減少が、元の体重の10%以内に回復した場合、投与は投与量を減少させて、または投与スケジュールの頻度を低くして再開され得た。
・ 非治療体重%回復に対する除外は、個別的に許可され得た。
〇 エンドポイントTGI。動物は群として監視されるものとする。
・ 実験のエンドポイントは、対照群における1000mm3の平均腫瘍体積か、9日のいずれか早い方であった。
・ エンドポイントに達したとき、全ての動物は安楽死されるものとした。
〇 塩型の化合物:なし
〇 投与用溶液の調製:
・抗PD−1−NODU、KW2、プロロイキン−NODU、4℃で保存。遮光
・抗PD−1−NODU=抗PD−1−NODUのPBS溶液
・NOD201MのPBS溶液(内部でKW2と呼称された)
・凍結禁止。前もって製剤化されて提供され、すぐに使用できた。

・腫瘍(サンプルの重量測定−mg):単一細胞懸濁液に処理、輸送条件−室温。フローサイトメトリーのためにCRL−NCに送付。インビトロ実験室と予定。以下のパネルを参照。
併用療法
NOD201と、IFNα、抗TIM3、抗LAG3、抗TIGIT、抗PD1、抗PDL1および抗CTLA4の併用療法。図24および40のデータを参照のこと。注記:NOD201Mとは、同系モデルに必要とされる齧歯類Fcドメインを指す。
腫瘍体積(mm3)=(w2 xl)/2
式中、wは腫瘍の幅、lは長さであり、単位はmmである。
試験1日目、確立されたMC38腫瘍を担持するC57BL/6マウスを10群、n=12/群に分けた。すべての剤は静脈内(i.v.)投与された。すべての抗体およびIFN−α治療剤が最初に投与され、次いでNOD201M治療が行われた。
〇 1群マウスは対照とされ、1、7、および13日目にビヒクル(PBS)を静脈内投与された。
〇 2群は1、7、13、および19日目に500μg/動物のNOD201Mを静脈内投与された。
〇 3群は2群の投与量およびスケジュールでNOD201Mを静脈内投与され、1、7、13、および19日目に200μg/動物の抗PD−1の静脈内投与を併用された。
〇 4群は1および7日目に500μg/動物のNOD201Mを静脈内投与され、1、7および13日目に200μg/動物の抗PD−L1の静脈内投与を併用された。
〇 5群は1、7および13日目に500μg/動物のNOD201Mを静脈内投与され、1、7、および13日目に200μg/動物の抗CTLA−4の静脈内投与を併用された。
〇 6群は1、7、13、および19日目に500μg/動物のNOD201Mを静脈内投与され、1、7、13、および19日目に200μg/動物の抗LAG−3の静脈内投与を併用された。
〇 7群は1、7、13、および19日目に500μg/動物のNOD201Mを静脈内投与され、1、7、13、および19日目に200μg/動物の抗TIM−3の静脈内投与を併用された。
〇 8群は1、7、13、および19日目に500μg/動物のNOD201Mを静脈内投与され、1、7、13、および19日目に500μg/動物の抗TIGITの静脈内投与を併用された。
〇 9群は1、7、および13日目に500μg/動物のNOD201Mを静脈内投与され、3および9日目に50μg/動物のIFN−αの静脈内投与を併用された。
〇 10群は1、7、および13日目に500μg/動物のNOD201Mを静脈内投与され、3および9日目に250μg/動物の抗CD137の静脈内投与を併用された。
腫瘍は週に2回ノギスを使用して測定された。各動物は、その腫瘍が1000m3のエンドポイント体積に達したとき、または試験終了時(54日目)のいずれか早いときに安楽死処理された。腫瘍体積エンドポイントで本試験を離脱した動物は、安楽死の日付とともに腫瘍進行(TP)のために安楽死されたと記録された。
腫瘍サイズを理由として動物が本試験から離脱した場合、その動物に対して記録された最終腫瘍体積は、その後の時点でのメジアン体積を算出するために使用されるデータとともに含められた。カプランマイヤープロットは、時間に対する、試験に残留した各群中の動物の百分率を示すために作製された(図24および40を参照のこと)。群のメジアン腫瘍体積は、時間関数としてプロットされ、群中の2回目のTR死の後に切り捨てられた。平均腫瘍体積プロットも本試験に含められた(提示される図)。
RNA配列およびTILデータの作製のための実験デザイン
メスのC57BL/6マウス(C57BL/6/NCrl、チャールズリバー社)は試験1日目で9週齢であり、16.4〜23.8gの範囲の体重(BW)であった。動物は水(逆浸透、1ppm Cl)ならびに、18.0%の粗タンパク質、5.0%の粗脂肪、および5.0%の粗繊維からなるNIH 31 Modified and Irradiated Lab Diet(登録商標)を不断給餌された。マウスは照射済みのEnrich−o’cobs(商標)で飼育され、20〜22℃(68〜72°F)および40〜60%の湿度、12時間の明暗サイクルで、据え付けのマイクロアイソレーター中で眠った。
腫瘍体積(mm3)=(w2xl)/2
式中、wは腫瘍の幅、lは長さであり、単位はmmである。腫瘍の重量は、腫瘍体積1mm3は1mgに等しいという仮定で推定され得る。
1)RNAライブラリ調製およびHiSeq配列解析
総RNAは、Qiagen RNeasy Mini Kit(Qiagen社)を使用して抽出された。RNAサンプルは、Qubit 2.0 Fluorometer(Life Technologies社、米国カリフォルニア州カールスバッド)を使用して定量され、2100 Bioanalyzer(Agilent Technologies社、米国カリフォルニア州パロアルト)でRNAの完全性をチェックした。RNAライブラリの調製、配列解析反応、および初期バイオインフォマティクス解析は、GENEWIZ,LLC.(米国ニュージャージー州サウスプレインフィールド)で実施された。
すべての解析の前に、配列リードはトリミングされ、Qiagen CLC Genomics Server 9.0を使用して3’末端のアダプターの可能性のある配列、および低品質(エラー率<0.05)のヌクレオチドを除去した。30塩基よりも短いリードは、その後の解析から取り除かれた。トリミングされたリードはマウス参照ゲノムGRCm38(ftp.ensembl.org/pub/current_fasta/mus_musculus/dna)へと割り当てられた。総リード数とRPKM値が算出された。

3.末端修復、5’リン酸化、およびdAテーリング(Tailing)
4.アダプターのライゲーション、PCR富化、および配列解析。
1.配列のQC、2.低品質塩基のトリミング、アダプター配列の切り出し、3.ゲノムおよびスプライスジャンクションへリードをマッピング、4.遺伝子/エクソン上のリード密度、およびアノテーション、5.スプライスアイソフォームID、6.転写物/遺伝子に対する総ヒット数およびRPKM値の算出、7.転写物発現の比較、7.GOアノテーション、Uniprotアノテーション。
3.1 参照ゲノムへの配列リードのマッピング、および遺伝子ヒット数の抽出:配列リードはトリミングされ、末端のアダプターの可能性のある配列および低品質ヌクレオチド(エラー率<0.05)が除去された。トリミング後、30ヌクレオチドよりも短い配列リードは廃棄された。残りの配列リードをマウスの参照ゲノム(GRCm38、ftp.ensembl.org/pub/current_fasta/mus_musculus/dna)に対して割り当てた。総遺伝子ヒット数が測定され、RPKM値が算出された。
CLC Genomicsを使用したマッピングおよび総遺伝子ヒット数算出の後、総遺伝子ヒット数を使用して、遺伝子差次的発現を比較した。
教師無し階層的クラスター化は、全てのサンプル、および標準化後のすべての遺伝子を用いて行われた。
以下のサンプル群間の遺伝子発現値の以下の比較が実施された(注記:1群=PBS、2群=抗PD1、3群=NOD201、4群=抗PD1/NOD201の組み合わせ

Wald検定を使用して、P値および倍数変化が生成された。偽発見率(FDR:False Discovery Rate)<0.05、および絶対倍数変化>1.5の遺伝子は、各比較に対し、差次的発現遺伝子と呼称された。
遺伝子オントロジー解析(GO:Gene Ontology analysis)は、各比較に関し、有意に発現された遺伝子に対して行われた。FDR<0.05のGO生物学的プロセスのリストが取得された。
スプライスバリアントに関し、その発現レベルが測定され、遺伝子発現比較と同じように発現比較が行われた。差次的発現転写物のリストが各比較に対して取得された(FDR<0.05、倍数変化>1.5)。
抗体融合体:
Ab融合体の様々なアプリケーション/用途;1)Ab融合体は、サイズの増加またはFcRnリサイクルの増加を介したノッチン(knottins)の半減期延長に使用することができる。2)ノッチンを、癌標的に特異的な抗体に結合させて、インテグリンおよび別の標的に結合する多重特異的タンパク質を生成し、両標的を調節する、または相乗効果をもたらすことができる(例としては、抗EGFR、抗VEGF、または抗CTLA−4および他のチェックポイントなどがありえる)。3)これに基づき、ノッチンを抗体に結合させ、より効率的に抗体を腫瘍に送達させ、さらなる有効性を得ることができる。4)より効率的に送達することで、Abの副作用を軽減し得る。
Claims (40)
- 対象のがんを治療する方法であって、
インテグリン結合性ポリペプチド−Fc融合体の有効量を前記対象に投与するステップを含み、
前記インテグリン結合性ポリペプチド−Fc融合体は、治療有効量が投与され、配列番号130(GCPRPRGDNPPLTCSQDSDCLAGCVCGPNGFCG)と配列番号131(GCPRPRGDNPPLTCKQDSDCLAGCVCGPNGFCG)からなる群から選択される配列を含み、
前記インテグリン結合性ポリペプチドはFcドメインと共役させている、方法。 - 前記Fcドメインは、IgG1、IgG2、IgG3、IgG4からなる群から選択される、請求項1に記載の方法。
- 前記Fcドメインは、ヒトFcドメインである、請求項2に記載の方法。

- (i)IL−2、もしくは、(ii)免疫チェックポイント阻害物質または免疫チェックポイント刺激物質の追加投与は、骨髄由来免疫抑制細胞(MDSC)の減少を引き起こす、請求項17に記載の方法。
- (i)IL−2、および(ii)免疫チェックポイント阻害物質または免疫チェックポイント刺激物質の両方の追加投与は、(i)IL−2、および/または(ii)免疫チェックポイント阻害物質または免疫チェックポイント刺激物質の個別投与に比べて、CD8+T細胞の腫瘍浸潤をより増加させる、請求項18に記載の方法。
- (i)IL−2、および(ii)免疫チェックポイント阻害物質または免疫チェックポイント刺激物質の両方の追加投与は、(i)IL−2、および/または(ii)免疫チェックポイント阻害物質または免疫チェックポイント刺激物質の個別投与に比べて、骨髄由来免疫抑制細胞(MDSC)をより減少させる、請求項18に記載の方法。
