Description

【発明の詳細な説明】 本発明は、ＩＱ−ヒドロキシビタミンＤ類の軟カプセル
の製造方法に関する。
近年ＩＱ位に水酸基を有するビタミンＤ類、例えばＩＱ
−ヒドロキシビタミンＤ３，ＩＱ，２５一ジヒドロキシ
ビタミンＤ３等がその強いビタミン○様生物活性から注
目されている。
しかしながら、これらのビタミンＤ類はその医薬として
の施用がきわめて少量であるため錠剤、丸剤等の剤形で
は一銭中のビタミンＤ類の量が不均等になり易く、また
その含有量を正確に定量できない等の不利がある。また
、ビタミンＤ類は光、特に紫外線にきわめて不安定な物
質であり、その取扱いは遮光状態で行われねばならず当
然ながらその製剤は完全遮光状態に置かれることが要求
される。これらのＩＱーヒドロキシビタミンＤ類の性質
からその製剤化方法としては、ビタミンＤ類を油性基剤
に溶解し欧カプセル化する方法が最適でありまた、カプ
セル化することにより紫外線を遮断した透明プラスチッ
クを用いるＰＴＰ包装の形態が可能となる。しかしなが
ら、本発明者らの実験によるとＩＱ−ヒドロキシビタミ
ンＤ類を油性基剤に溶解してカプセル剤とし、紫外線を
遮断した状態で太陽光線を照射するとかなりの量のＩＱ
−ヒドロキシビタミンＤ類の活性がそこなわれる。この
原因について種々検討した結果、軟カプセルのゼラチン
の防腐剤として通常使用されているパラオキシ安息香酸
ェステルの影響によることが判明した。また、剤皮の基
剤として用いるゼラチンは微生物により腐敗を受けやす
く特に軟カプセル剤は剤皮の硬化を防ぐ目的でグリセリ
ン、マクロゴール４００，プロピレングリコール等の軟
化剤が添加されているため微生物による腐敗が促進され
防腐剤の添加が必須である。本発明者は剤皮中に含まれ
るゼラチンの防腐剤について種々検討した結果、ソルピ
ン酸およびその塩類がＩＱーヒドロキシピタミン○類を
失活させることなく、むしろその安定性を増大させる効
果を有することを見出し本発明を完成した。
すなわち本発明は、ＩＱーヒドロキシビタミンＤ類を油
性基剤に溶解し、ついでソルビン酸類を含有する剤皮で
軟カプセル化することを特徴とする経口用ＩＱーヒドロ
キシビタミンＤ類軟カプセルの製造方法である。本発明
で、ＩＱーヒドロキシビタミンＤ類とは、ＩＱ位に水酸
基を有するビタミンＤ類であり、具体的にはＩＱ−ヒド
ロキシビタミンＤ３．ＩＱ，２５一ジヒドロキシビタミ
ンＤ３，ＩＱ，２４−ジヒドロキシビタミンＤ３等であ
る。
油性基剤としては脂肪酸のトリグリセライド等の油脂、
リノール酸等の中頭の不飽和脂肪酸、綿実油、オリーブ
油、トウモロコシ油等が使用されるが、室温付近で油状
のもので、使用するＩＱーヒドロキシビタミンＤ類を分
解しないものであれば特に制限なく使用し得る。ソルビ
ン酸類としてはソルビン酸または、ソルビン酸の塩例え
ばソルビン酸カリウム、ソルビン酸ナトリウム等である
。
また、ソルビン酸またはその塩類を適当な比率で混合し
て使用することも可能である。ソルピン酸類の使用量は
、剤皮に対して０．２〜１重量％程度用いると好適な結
果が得られる。こうして得られたＩＱ−ヒドロキシピタ
ミンＤ類の軟カプセルは、ＩＱーヒドロキシビタミン○
類の安定性がきわめてよい。
その具体的な効果を表に示す。表は実施例により調製し
たＩＱーヒドロキシビタミンＤ３の表面を紫外線を遮断
する透明フィルムで覆い、太陽光線のもとで２週間各試
料とも同一条件で暴露した後ｌｑ−ヒドロキシビタミン
Ｄ３を定量したものである。表中の定量値は防腐剤無添
加の剤皮のＩＱーヒドロキシビタミンＤ３の塁を１００
とした場合の比率である。
表 実施例 ＩＱ−ヒドロキシビタミンＤ３を０．Ｄ．０（日清製油
社製品：中鎖脂肪酸のトリグリセラィド）に１０ｒ夕／
叫の濃度に溶解する。
下記処方による剤皮組成分を加温溶解し、先に調製した
ものを１カプセル中にＩＱーヒドロキシｌビタミンＤ３
として１ムタ含有するように軟カプ・セル製造機を用い
て常法により軟カプセルを製造心た。
剤皮処方例 ゼラチン １の重量部グリセリ
ン ５重量部防 腐 剤
０．０母重量部精 製 水
１４重量部。

Claims (1)

  1. 【特許請求の範囲】 １ １α−ヒドロキシビタミンＤ類を油性基剤に溶解し
    、ついでソルビン酸類を含有する剤皮で軟カプセル化す
    ることを特徴とする経口用１α−ヒドロキシビタミンＤ
    類軟カプセルの製造方法。 ２ ソルビン酸類としてソルビン酸、またはその塩を用
    いる特許請求の範囲第１項記載の製造方法。 ３ ソルビン酸類としてソルビン酸ナトリウム、または
    ソルビン酸カリウムを用いる特許請求の範囲第１項記載
    の製造方法。 ４ １α−ヒドロキシビタミンＤ類が１α−ヒドロキシ
    ビタミンＤ＿３である特許請求の範囲第１項記載の製造
    方法。
