Abstract

PURPOSE: To obtain the titled substance, by tritylating (S)-(+)-5-hydroxymethyltetrahydrofuran-2-one, hydrolyzing the tritylation product, and reacting the hydrolyzed substance with triphenylphosphine and a dialkyl azodicarboxylate.
CONSTITUTION: (S)-(+)-5-Hydroxymethyltetrahydrofuran-2-one of formulaIderived from natural (S)(+) glutamic acid is tritylated with triphenylmethyl chloride, and the resultant compound is then hydrolyzed in the presence of an alkali, e.g. KOH, to give an oxy acid derivative of formula 3, which is then reacted with and cyclized triphenylphosphine and a dialkyl azodicarboxylate to afford the titled substance. The resultant substance is then hydrolyzed to give (R)-(-)-5- hydroxymethyltetrahydrofuran-2-one of formula 5. The purifying operation is preferably carried out in the respective steps to give a high yield.
