Claims (1)
- 【特許請求の範囲】 1.ルシフェラーゼ活性を有し、かつPhotinuspyralis、Luc iola mingrelica、Luciola cruciataまたはL uciola lateralis由来のルシフェラーゼに対して60%を越え るアミノ酸配列相同性を有するタンパク質であって、Photinus pyr alisルシフェラーゼの残基354またはLuciola mingreli ca、Luciola cruciata及びLuciola lateral isルシフェラーゼの残基356に対応するアミノ酸残基がグルタミン酸以外の アミノ酸であることを特徴とするタンパク質。 2.アミノ酸配列XGDDKPGAを含み、この配列中のXはグルタミン酸以外 のアミノ酸残基であることを特徴とする請求項1に記載のタンパク質。 3.アミノ酸配列TPXGDDKPGAを含み、この配列中のXはグルタミン酸 以外のアミノ酸残基であることを特徴とする請求項2に記載のタンパク質。 4.アミノ酸Xがグリシン、プロリンまたはアスパラギン酸でないことを特徴と する請求項1から3のいずれか1項 に記載のタンパク質。 5.アミノ酸Xがトリプトファン、バリン、ロイシン、イソロイシン及びアスパ ラギンのうちのいずれかであるか、またはこれらのアミノ酸のうちのいずれかの 類似体または修飾体であることを特徴とする請求項1から3のいずれか1項に記 載のタンパク質。 6.アミノ酸Xがリシン及びアルギニンのいずれか一方であるか、またはこれら のアミノ酸の類似体または修飾体であることを特徴とする請求項1から3のいず れか1項に記載のタンパク質。 7.配列番号2に示したアミノ酸配列を含み、前記配列中のXaaは請求項5ま たは6に記載のアミノ酸またはその類似体もしくは修飾体であるタンパク質。 8.請求項1から7のいずれか1項に記載のタンパク質をコードするDNA。 9.配列番号1に示したヌクレオチド配列を含み、前記配列の1063〜106 5位の3個の塩基Nはグルタミン酸以外のアミノ酸をコードするコドンを構成す ることを特徴とする請求項8に記載のDNA。 10.前記コドンが請求項5または6に記載のアミノ酸、 類似体または修飾体をコードすることを特徴とする請求項9に記載のDNA。 11.請求項1から7のいずれか1項に記載のタンパク質をコードするluc遺 伝子を含むベクター。 12.野生型または組み換えluc遺伝子を含むベクターを、Photalis pyralisルシフェラーゼの354位のグルタミン酸またはLuciol a mingrelica、Luciola cruciataもしくはLuc iola lateralisルシフェラーゼの356位のグルタミン酸をコー ドするコドンを代替アミノ酸、該アミノ酸の類似体、または該アミノ酸の修飾体 をコードするコドンに変更する部位特異的突然変異誘発によって処理することに より取得可能であることを特徴とする請求項11に記載のベクター。 13.代替アミノ酸が請求項5または6に記載のアミノ酸、類似体または修飾体 であることを特徴とする請求項12に記載のベクター。 14.内部にluc遺伝子が連結された、pKK223−3、pDR540及び pT7−7の中から選択されることを特徴とする請求項9から13のいずれか1 項に記載のベ クター。 15.請求項8から14のいずれか1項に記載のDNAまたはベクターを保有す る、請求項1から7のいずれか1項に記載のタンパク質を発現させ得る細胞。 16.大腸菌、S.cerevisiaeまたは昆虫細胞であることを特徴とす る請求項15に記載の細胞。 17.ATP測定によるアッセイを実施するための試験キットであって、発光試 薬中に存在する請求項1から7のいずれか1項に記載のタンパク質を含むことを 特徴とするキット。 18.光を発生するルシフェリン及びルシフェラーゼを用いてATPを測定する アッセイ方法であって、前記光の量はATPの量に関連し、ルシフェラーゼが請 求項1から7のいずれか1項に記載のタンパク質であることを特徴とする方法。 19.アッセイを30〜70℃の温度で行なうことを特徴とする請求項18に記 載の方法。 20.アッセイを37〜60℃の温度で行なうことを特徴とする請求項18に記 載の方法。 21.アッセイを40〜50℃の温度で行なうことを特徴 とする請求項18に記載の方法。 22.熱安定化剤の不在下に室温で10日間貯蔵後にそのルシフェラーゼ活性を 85%以上維持しているルシフェラーゼ調製物。 23.請求項1から7のいずれか1項または請求項22に記載のルシフェラーゼ の、特異的結合試薬のためのラベルとしての使用。 24.請求項1から7のいずれか1項に記載のルシフェラーゼで標識した特異的 結合試薬を含むことを特徴とする試験キット。 25.細胞またはDNAの同定のための、請求項8から14のいずれか1項に記 載のルシフェラーゼコーディングDNAまたはベクターの使用。
