- 【特許請求の範囲】 1.配列番号46のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 2.配列番号5のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 3.配列番号7のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 4.配列番号9のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 5.配列番号11のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 6.配列番号13のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 7.配列番号15のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 8.配列番号17のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 9.配列番号19のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 10.配列番号21のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 11.配列番号55のヌクレオチド配列を有する組換えDNA分子、またはそのハイ ブリッド形成可能なフラグメント。 12.配列番号47のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 13.配列番号6のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 14.配列番号8のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 15.配列番号10のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 16.配列番号12のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 17.配列番号14のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 18.配列番号16のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 19.配列番号18のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 20.配列番号20のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 21.配列番号22のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 22.配列番号56のアミノ酸配列を有する単離されたポリペプチド、またはその抗 原性フラグメント。 23.動物被験者中の遺伝子の発現を誘導できるプロモーターと操作により関連し たグラム陽性細菌の輸出タンパク質をコードする遺伝子を含むベクターを含み、 その輸出タンパク質が付着関連タンパク質、毒力決定基、毒素及び主要抗原タン パク質からなる群から選ばれることを特徴とするグラム陽性細菌による感染から の動物被験者の保護用のワクチン。 24.動物被験者中の遺伝子の発現を誘導できるプロモーターと操作により関連し たグラム陽性細菌の種の多くの株に共通の抗原である輸出タンパク質をコードす る遺伝子を含むベクターを含むことを特徴とするグラム陽性細菌による感染から の動物被験者の保護用のワクチン。 25.グラム陽性細菌がS.ニューモニアエである請求の範囲第23項または第24項に 記載のワクチン。 26.遺伝子によりコードされたタンパク質がアドヘジンである請求の範囲第23項 または第24項に記載のワクチン。 27.遺伝子によりコードされたタンパク質がアドヘジンである請求の範囲第25項 に記載のワクチン。 28.動物被験者中の遺伝子の発現を誘導できるプロモーターと操作により関連し たS.ニューモニアエの輸出タンパク質をコードする遺伝子を含むベクターを含み 、その遺伝子が配列番号1、3、5、7、9、11、13、15、17、19、22、46、55、ami A及びponAからなる群から選はれたヌクレオチド配列を含むことを特徴とするS. ニューモニアエによる感染からの動物被験者の保護用のワクチン。 29.動物被験者がヒトである請求の範囲第23項、第24項または第28項に記載のワ クチン。 30.免疫原量のグラム陽性細菌の輸出タンパク質及びアジュバントを含み、その 輸出タンパク質が付着関連タンパク質、毒力決定基、毒素及び主要抗原タンパク 質からなる群から選ばれることを特徴とするグラム陽性細菌による感染からの動 物被験者の保護用のワクチン。 31.免疫原量のグラム陽性細菌の種の多くの株に共通の抗原である輸出タンパク 質及びアジュバントを含むことを特徴とするグラム陽性細菌による感染からの動 物被験者の保護用のワクチン。 32.グラム陽性細菌がS.ニューモニアエである請求の範囲第30項または第31項に 記載のワクチン。 33.遺伝子によりコードされたタンパク質がアドヘジンである請求の範囲第30項 または第31項に記載のワクチン。 34.遺伝子によりコードされたタンパク質がアドヘジンである請求の範囲第32項 に記載のワクチン。 35.免疫原量のS.ニューモニアエの輸出タンパク質及びアジュバントを含み、輸 出タンパク質が配列番号2、4、6、8、10、12、14、16、18、20、22、47、56、Po nA及びAmiAからなる群から選ばれたアミノ酸配列を含むことを特徴とするS.ニュ ーモニアエによる感染からの動物被験者の保護用のワクチン。 36.動物被験者がヒトである請求の範囲第30項、第31項または第35項に記載のワ クチン。 37.a.グラム陽性DNA分子の重複突然変異誘発が起こり得るベクター中のシグ ナル配列を欠いているインジケータータンパク質遺伝子及びプロモーターを含む 読み取り枠の上流及び読み取り枠中にグラム陽性細菌から得られたDNA分子を 翻訳挿入する工程(そのインジケータータンパク質は細菌により輸出されなけれ ば非機能性である)、 b.ベクターをグラム陽性細菌に導入する工程、 c.グラム陽性細菌を増殖し、それによりグラム陽性細菌の輸出タンパク質とイ ンジケータータンパク質の融合タンパク質を発現できる工程、 d.インジケータータンパク質が機能性である細菌を選択し、インジケータータ ンパク質の輸出を指示する工程、 e.グラム陽性細菌が通常付着する真核生物細胞に対しグラム陽性細菌の付着性 の損失につきスクリーニングする工程、そして f.付着性の損失を示すグラム陽性細菌を選択し、それにより輸出付着関連タン パク質をコードする突然変異遺伝子を含むグラム陽性細菌を選択する工程 を含むことを特徴とするグラム陽性細菌の付着関連輸出タンパク質をコードする 遺伝子の部分の同定方法。 38.a.グラム陽性DNA分子の重複突然変異誘発が起こり得るベクター中のシグ ナル配列を欠いているインジケータータンパク質遺伝子及びプロモーターを含む 読み取り枠の上流及び読み取り枠中にグラム陽性細菌から得られたDNA分子を 翻訳挿入する工程(そのインジケータータンパク質は細菌により輸出されなけれ ば非機能性である)、 b.ベクターをグラム陽性細菌に導入する工程、 c.グラム陽性細菌を増殖し、それによりグラム陽性細菌の輸出タンパク質とイ ンジケータータンパク質の融合タンパク質を発現できる工程、 d.インジケータータンパク質が機能性である細菌を選択し、インジケータータ ンパク質の輸出を指示する工程、 e.動物LD50モデル中でグラム陽性細菌の毒力の損失につきスクリーニングする 工程、そして f.毒力の損失を示すグラム陽性細菌を選択し、それにより輸出タンパク質毒力 決定基をコードする突然変異遺伝子を含むグラム陽性細菌を選択する工程 を含むことを特徴とするグラム陽性細菌の毒力決定基である輸出タンパク質をコ ードする遺伝子の部分の同定方法。 39.インジケータータンパク質がエシェリキア コリPhoAである請求の範囲第37 項または第38項に記載の方法。 40.グラム陽性細菌がS.ニューモニアエである請求の範囲第37項または第38項に 記載の方法。 41.輸出タンパク質がアドヘジンである請求の範囲第37項または第38項に記載の 方法。 42.動物被験者中の遺伝子の発現を誘導できるプロモーターと操作により関連し た請求の範囲第37項または第38項に記載の方法により同定された遺伝子を含むベ クターを含むことを特徴とするグラム陽性細菌による感染からの動物被験者の保 護用ワクチン。 43.免疫原量の請求の範囲第37項または第38項に記載の方法により同定された遺 伝子によりコードされたタンパク質及びアジュバントを含むことを特徴とするグ ラム陽性細菌による感染からの動物被験者の保護用ワクチン。 44.配列番号2、6、8、10、12、14、16、18、20、22、47及び56からなる群から 選ばれたアミノ酸配列を有するタンパク質と反応性の抗体またはそのフラグメン ト。 45.免疫原投薬量の請求の範囲第23項、第24項、第28項、第30項または第31項に 記載のワクチンを投与することを特徴とするグラム陽性細菌による感染から被験 者を保護する方法。 46.免疫原投薬量の請求の範囲第42項に記載のワクチンを投与することを特徴と するグラム陽性細菌による感染から被験者を保護する方法。 47.免疫原投薬量の請求の範囲第43項に記載のワクチンを投与することを特徴と するグラム陽性細菌による感染から被験者を保護する方法。 48.グラム陽性細菌の存在を請求の範囲第44項に記載の抗体またはそのフラグメ ントで検出することを特徴とするグラム陽性細菌による感染の診断方法。 49.配列番号1、5、7、9、11、13、15、17、19、21、46及び55からなる群か ら選ばれたヌクレオチド配列を有する組換えDNA分子のハイブリッド形成可能 なフラグメントである核酸プローブで被験者からの試料中のグラム陽性細菌の存 在を検出することを特徴とするグラム陽性細菌による感染の診断方法。 50.配列番号1、5、7、9、11、13、15、17、19、21、46及び55からなる群から選 ばれたヌクレオチド配列を有する組換えDNA分子のハイブリッド形成可能なフ ラグメントであるプライマーを使用するポリメラーゼ連鎖反応によりグラム陽性 細菌の存在を検出することを特徴とするグラム陽性細菌による感染の診断方法。 51.治療有効投薬量の請求の範囲第43項に記載の抗体を投与することを特徴とす るグラム陽性細菌による感染から被験者を保護する方法。 52.治療有効投薬量の請求の範囲第37項に従って単離された遺伝子によりコード されたグラム陽性アドヘジンを投与することを特徴とするグラム陽性細菌による 感染から被験者を保護する方法。
