Claims (1)
- 【特許請求の範囲】 1.構成性シアリダーゼ遺伝子が機能的に発現されない組み替えセルライン。 2.ヌクレオチドの突然変異、付加および除去のいずれかによる該遺伝子の破 壊のためにシアリダーゼ遺伝子が機能的に発現されない、請求項1に記載の組み 替えセルライン。 3.該遺伝子の除去のためにシアリダーゼ遺伝子が機能的に発現されない、請 求項1に記載の組み替えセルライン。 4.ホモローガスな組み替えによる該遺伝子の破壊のためにシアリダーゼ遺伝 子が機能的に発現されない、請求項1に記載の組み替えセルライン。 5.アンチセンスRNAを用いる転写の調節による該遺伝子機能の破壊のため にシアリダーゼ遺伝子が機能的に発現されない、請求項1に記載の組み替えセル ライン。 6.チャイニーズハムスター卵巣から誘導されるセルラインである、請求項1 に記載の組み替えセルライン。 7.チャイニーズハムスター卵巣セルラインの細胞培養液から取得し得る、実 質上均質なシアリダーゼ。 8.約43kDaの分子量を有する請求項7に記載のシアリダーゼ。 9.そのトリプシン性フラグメントが、アミノ酸配列CRVQAQSPNSG LDFQDN(配列番号13)を有する、請求項7に記載のシアリダーゼ。 10.アミノ酸配列CRVQAQSPNSGLDFQDN(配列番号13)の ペプチドを用いて得られる抗体に結合する、請求項7に記載のシアリダーゼ。 11.(a)アミノ酸配列CRVQAQSPNSGLDFQDN(配列番号1 3)の一部または全てをコードしているオリゴヌクレオチドプローブを製造し; (b)このプローブを哺乳動物DNAライブラリー中の核酸とハイブリダイズ してハイブリッドを形成させ; (c)ハイブリッドを分離し、それによりシアリダーゼをコードしている当該 核酸配列を得る、 ことからなる工程によって得られる、シアリダーゼをコードしている核酸配列 。 12.シアリダーゼがチャイニーズハムスターセルラインの細胞培養液から取 得し得る、請求項11に記載の核酸配列。 13.シアリダーゼコード化遺伝子の取得に有用であり、且つアミノ酸配列C RVQAQSPNSGLDFQDN(配列番号13)の一部または全てをコード している配列を有する、オリゴヌクレオチドプローブ。 14.糖蛋白をコードしている核酸を宿主細胞中で発現させることからなり、 構成性シアリダーゼ遺伝子が機能的に発現されない、糖蛋白の産生のための方法 。 15.ヌクレオチドの突然変異、付加および除去のいずれかによる該遺伝子の 破壊のためにシアリダーゼ遺伝子が機能的に発現されない、請求項14に記載の 方法。 16.該遺伝子の除去のためにシアリダーゼ遺伝子が機能的に発現されない、 請求項14に記載の方法。 17.ホモローガスな組み替えによる該遺伝子の破壊のためにシアリダーゼ遺 伝子が機能的に発現されない、請求項14に記載の方法。 18.アンチセンスRNAを用いる転写の調節による該遺伝子機能の破壊のた めにシアリダーゼ遺伝子が機能的に発現されない、請求項14に記載の方法。 19.チャイニーズハムスター卵巣から誘導されるセルラインである、請求項 14に記載の方法。 20.CHO細胞シアリダーゼについて図10に示されるアミノ酸配列をコー ドしているヌクレオチド配列を含む、分離されたDNA。 21.CHO細胞シアリダーゼについて図10に示されるアミノ酸配列をコー ドしているヌクレオチド配列を含む、発現ベクター。 22.CHO細胞シアリダーゼについて図10に示されるアミノ酸配列をコー ドしている分離されたDNAで形質転換された宿主細胞。 23.CHO細胞シアリダーゼについて図10に示されるアミノ酸配列を有す るポリペプチドを、該ベクターにより形質転換された宿主細胞中で発現すること のできる発現ベクターで宿主細胞を形質転換することからなる方法。 24.形質転換された宿主細胞を培養し、宿主細胞培養からポリペプチドを回 収する工程をさらに含む、請求項23に記載の方法。
