ヒト樹状細胞のインビトロ発生およびそれらの用途本発明は、概して、ヒト樹状
細胞のインビトロ発生法、更に詳しくは、発生した細胞の治療および診断上の用
途に関する。
樹状細胞は、いくつかの免疫応答、例えば、MHCに制限されたT細胞の感作、
器官移植片の拒絶およびT細胞依存性抗体の形成を開始する働きをする抗原提示
細胞の系である。樹状細胞は、多数の非リンパ系組織において見出されるが、輸
入リンパまたは血流によってリンパ系器官のT細胞依存性区域へ移動することが
できる。それらは皮膚において見出さね、その場合それらをランゲルハンス細胞
と称し、そして粘膜中にも存在する。それらは、それらが抗原を獲得することが
できる末梢組織中の免疫系の見張り役である。これらの細胞はCD4を発現する

のヒト樹状細胞の単離は最近になって達成されたばかりであり、しがち少数の細
胞しか発生させることができない。例えば、フロイデンタール(Freuden

頁(1990)。多数のヒト樹状細胞のインビトロ発生は、インビトロのヒトナ
イーブCD4およびCD8T細胞の感作、HIV感染を妨げることができる物質
のスクリーニング、ヒトTおよびB細胞で再構成された5CID−huマウスに
おけるヒトB細胞の一次および二次インビボ応答の発生、並びに一層感受性の混
合リンパ球反応検定の組み立てに重要な利益を与えるであろう。
同種異系組織および器官の移植に対する主な障害は、移植片受容者による移植片
拒絶である。ドナー組織に対する受容者すなわち宿主の細胞性免疫反応は、拒絶
過程においである重要な役割を果たす。細胞性免疫反応には二つの重要な段階、
すなわち、(i)宿主細胞が、主要組織適合性遺伝子複合体(MMC)の関係に
おいてドナー細胞を異物として認める場合の認識:および(i i)宿主細胞が
異種細胞を攻撃することによって応答する場合の破壊がある。攻撃過程の一部努
として、多数の応答細胞が増殖され、そして細胞障害性、すなわち、適当な抗原
を示すドナー細胞を死滅させる能力を獲得する。したがって、細胞性免疫は、二
つの測定可能な機能、すなわち、増殖および細胞障害活性によって記載すること

細胞培養技術の開発は、インビボ免疫感作を模倣するインビトロの方法の確立を
もたらし、したがってインビトロでの細胞性免疫を評価するための尺度が与えら
れた。移植に関して特に有用であるのは、混合リンパ球反応(MLR)または混
合リンパ球培養である。MLRは比較的簡単な検定法であるが、それには多数の
変法がある。典型的に、検定は、適当な培養系中の応答リンパ球と、照射などに
よって増殖および/または転写機構が損傷された刺激リンパ球とを混合すること
から成る。細胞を数日間培養した後、多数の種々の測定を行なって、刺激細胞に
対する応答細胞の反応性の程度、例えば、トリチウム化チミジンの取込み、芽細
胞の数、分裂細胞の数、サイトカイン生産等を定量することができる。検定にお
いて他に変化するものとしては、応答および刺激細胞源、例えば、末梢血、牌臓
、リンパ節等:応答細胞が刺激細胞に関して同系か、同種異系かまたは異種であ
るかどうか;刺激細胞を損傷させる方法、例えば、照射若しくはDNA合成阻害
斉j(例えば、マイトマイシンC)による処理または類似のものがある。
細胞性免疫反応性についての通常の検定としてのMLRの欠点は感度である。
たとえ特定の読みを用いたとしても、しばしば、MLRにおいて強い効果を得る
ことは難しい。刺激細胞集団中の抗原提示細胞は、応答細胞を刺激する原因とな
っていると考えられるが;しかしながら、大部分の組織源のこのような細胞は数
が少なくおよび/または非能率的に刺激する種類から成る。MLR検定の感度お
よびそれゆえの有用性は、更に強力な刺激細胞集団の入手可能性によって大いに
向上させることができると考えられる。樹状細胞は、それらが強力な抗原提示細
胞として周知であることから、この機能に役立ちうると考えられる。例えば、ス

発明の概要
本発明は、ヒト樹状細胞のインビトロ発生法に関する。本発明は、更に、本発明
の方法によって生産されたヒト樹状細胞の単離された集団および、樹状細胞の純
粋な集団を刺激細胞として用いる改良されたMLR検定を含む該単離細胞の用途
を包含する。本発明の方法は、CD34+造血系前駆細胞を、腫瘍壊死因子α(
TNF−α)およびインターロイキン−3(IL−3)の存在下または顆粒球−
マクロファージコロニー刺激因子(GM−C8F)の存在下で培養して本発明の
CDlLa+樹状細胞を生成する工程を含む。
本発明は、したがって、ヒト樹状細胞を含む細胞組成物の製造法であって、+
CD34 造血系細胞を、TNF−αおよびIL−3でまたはGM−CSFで処
理し;そして
CD1.a抗原を発現する被処理CD34 造血系細胞を単離する工程を含む上
記方法を提供する。

本発明は、更に、インビトロ培養後にCD1a抗原を発現するCD34+造血系
細胞;および上記に定義の方法によって製造されたヒト樹状細胞を含む細胞組成
物を提供する。
樹状細胞は免疫学的応答を開始する。本発明による樹状細胞に関して本明細書中
に報告されたインビトロのデータは、これらの細胞が実験手段として有用であり
、更には、癌およびウィルス感染を含む養子免疫療法による種々の疾患のインビ
ボ処置において用いることができるということを示している。
更に、本発明は、樹状細胞系を製造するためのまたは樹状細胞を誘導するための
TNF−αとIL−3と一緒のまたはGM−C8Fの使用、CD4 ヘルパーT
細胞を発生させるための樹状細胞の使用;養子免疫療法におけるCD4 ヘルパ
ーT細胞の使用;ヒト抗体産生用の5CID−huマウスの使用;並びに癌特異
的およびウィルス特異的CD8 細胞障害性T細胞を発生させるための謝状細胞
の使用、特に、エイズ(AIDS)ウィルスに対して特異的なCD8 細胞障害
性T細胞を発生させるための樹状細胞の使用に関する。
本発明のもう一つの特徴は、
応答細胞試料を提供し;
刺激細胞が応答細胞に関して同種異系であるような、しかも刺激細胞が、(a)
CD34 造血系細胞をTNF−at6よびIL−3でまたはGM−C3Fで処
理し、そして(b)CD1a抗原を発現する被処理CD34 造血系細胞を単離
する工程を含む方法によって製造された樹状細胞から成るような不活イ」す激細
胞試料を提供し;
該応答細胞および該不活化刺激細胞を同時培養し;そして該応答細胞の応答を測
定する工程を含む混合リンパ球反応を含む。

発明の詳細な説明
本発明の重要な態様は、CD34 造血系細胞からの樹状細胞の発生である。

CD34 造血系前駆細胞は、骨髄などの種々の組織源から得ることができるが
、好ましくは、請帯血試料から以下のように得られる。すなわち、試料からの軽
密度単核細胞をフィコール・ハイパクー(Ficol 1−Hypaque)勾
配分離によって単離しくd=1..077g/mL)、そして例えば、1%w/
v組織培養等級ウシ血清アルブミンを補足したRPMI 1640培地中におい
て37℃で一晩中インキユベーションすることによって付着細胞から除去する。
好ましくは、CD34抗原を有する細胞を、抗CI)34単クロ一ン性抗体、例
えば、イムノチク(Immunotech)(マルセイユ、フランス)から入手
可能なImu−133,3、ベクトン・ディキンソン(BectonDieki
nson)(マウンテン・ビュー、カリフォルニア)から入手可能な抗MylO
または類似のものを用いる間接免疫パニングによる正の選択によって非付着単核
部分から単離する。パニングフラスコは以下のように調製される。すなわち、ト
リス緩衝液(0,05モル/L、pH9,4)中で25μg/mLの濃度のヒツ
ジFab抗マウスIgGを、750m2組織培養フラスコ中に分配しく10mL
) 、4℃で一晩中被覆させる。別個に、(上記で論及の付着細胞から除去され
た)軽密度単核細胞を、2%熱失活プールヒトAB血清(HABS)を補足した
RPMI 1640中において細胞107個/mLで抗CD34抗体5μg/m
Lと一緒に4℃で1時間インキュベートした。その後、細胞を2%HABS含有
冷培地中で洗浄し、そして細胞約5X107個を含む10mLを、上記のように
ヒツジ抗マウスIgGを予め被覆したフラスコに分配する。4℃で2時間インキ
ュベーション後、懸濁液中の非付着細胞(すなわち、CD34除去部分)を静か
にピペッティングし且つ培地で数回洗浄することによって採取する。次に、付着
「パニング済み」細胞(すなわち、CD34豊富部分)を激しくピペッティング
することによって回収する。

樹状細胞はCD34 細胞から、GM−C3FまたはTNF−αおよびIL−3
を含む培地中においてこれらを培養することによって得られる。好ましくは、樹
状細胞はCD34+細胞から、TNF−α並びにGM−C8Fを含む培地中にお
いてこれらを培養することによって得られる。本発明において用いるのに適当な
TNF−α、GM−C8FおよびIL−3は、例えば、ジエンザイム・コー承し
−ション(Genzyme Corp、)(ケンブリッジ、MA)から商業的に
入手可能であるし、または組換え体発現システムによって、例えば、クラーク(
C1a r k)ら、米国特許第4.959.455号明細書(IL−3);ク
ラークら、PCT出願第EP85100326号明細書(公開東WO36100
639号明細書)(GM−C3F);およびマーク(Mark)ら、米国特許第
4.677.063号明細書(TNF−α)に示されたように製造することがで
きる。好ましくは、IL−3およびGM−C8Fは飽和濃度で用いられ;すなわ
ち、それらは、CD34 細胞上のIL−3およびGM−C8F受容体全部が生
物学的に活性なIL−3およびGM−C8F分子で占有される濃度で用いられる
。
当然ながら、実際の濃度は、用いられるIL−3およびGM−C8Fの性質に依
存しうる。好ましくは、特異的活性を有するヒトIL−3を少なくとも5x10
6U/mgで用い、そこにおいて活性の単位は、液体培養中のヒト骨髄細胞によ
って取込まれた3H−チミジンによって決定される半最大増殖活性に対応する。
以下に記載の培養系において、飽和濃度はLong/mL(または50U/mL
)であった。好ましくは、特異的活性を有するヒトGM−C8Fを少なくとも2
x106U/mgで用い、そこにおいて活性の単位は、IL−3に関して上記に
定義の通りである。以下に記載の培養システムにおいて、飽和濃度は1100n
/mL(または200 U/m L)であった。好ましくは、TNF−aを2〜
3ng/mLまたは40〜60U/mLの範囲の濃度で、最も好ましくは、約2
.5ng/mLまたは50U/mLの濃度で用いる。TNF−αの単位は、カー
スウ細胞は、標準的な添加剤を含む標準的な組織培地、例えば、10%(v/v
)熱失活ウシ胎児血清、10mMヘペス、2mM L−グルタミン、5x1o−
5M2−メルカプトエタノール、ペニシリン(100U/mL)およびストレプ
トマイシン(100mg/mL)を補足したRPMI 1640中で同時培養す
ることができる。好ましくは、CD34+細胞はサイトカイン存在下において8
〜12日間培養される。
培養中に生成する樹状細胞は、抗CD1aおよび/または抗CD14抗体を用い
る以外は(抗体は両方共、例えば、ベクトン・ディキンソンから商業的に入手可
能)、上記のようにパニングによって単離される。
本発明のMLR法は、以下、すなわち、(1)応答細胞試料を提供し; (2)
刺激細胞が該応答細胞に関して同種異系であるような、しかも刺激細胞が、+
(a)CD34 造血系細胞をTNF−aおよびIL−3でまたはGM−CS
Fで処理し、そして(b)CD1a抗原を発現する被処理CD34+造血系細胞
を単離する工程を含む方法によって製造された樹状細胞から成るような不活化刺
激細胞試料を提供し; (3)該応答細胞および該不活化刺激細胞を同時培養し
:そして(4)該応答細胞の応答を測定する工程を含む。
好ましくは、応答細胞試料は、移植片の受容者であるべき患者の末梢血からの+
CD4 T細胞から成る。T細胞集団の入手は、ディサバト(DiSabat。
)ら監修、Meth、in Enzymol、、108巻(1984)で十分に
記載されている当該技術分野において周知の技法を用いる。例えば、CD4 T
細胞は以下のように単離することができる。すなわち、最初に単核細胞を末梢血
から単離し且つ付着細胞から除去し;次に、例えば、イムノマグネチック除去(
例えば、ダイナビーズ(Dynabeads)、ダイナル(Dynal)、オス
口、ノルウェイを用いる)または類似のものにより、商業的に入手可能な単クロ
ーン性抗体、例えば、抗CD14、抗CD16、抗CD20、抗CD8、抗CD
40(ベクトン・ディキンソンおよび/またはオルト・ダイアグノスティック・
システムズ(Ortho Diagnostic Systems)、=ニーシ
ャーシーから入手可能)の混合物を用いて他の細胞種を除去することによってC
+
D4 T細胞を精製する。95%より高純度のCD4+集団は、典型的に、2回
のイムノマグネチック除去後に達成される。
本発明の刺激細胞は、応答細胞を採取された人とは別の人から得られたCD34
+造血系前駆細胞由来の樹状細胞であり;すなわち、刺激細胞は、応答細胞に関
して同種異系である。これらの細胞は前記のように得られる。
本発明の刺激細胞を不活化して、それらがそれらの刺激機能を依然として遂行し
つるが、応答細胞から測定される応答を不明瞭にすることがあった何等かの他の
機能から阻害されるようにする。したがって、不活化の性状は、検定の「読み」
にある程度依存する。好ましくは、読み、すなわち応答細胞で測定される応答は
細胞増殖である。他の読みとしては、更に、サイトカイン生産、細胞溶解能等の
ような現象を挙げることができる。好ましくは、刺激細胞を処理して、それらは
複製不能であるが、それらの抗原処理機構は依然として機能するようにする。
これは、便宜上、細胞を応答細胞と混合する前に、例えば、約1500〜500
0R(ガンマ線またはX線)、好ましくは3000〜4000Rの照射によって
達成される。

個に対して加え且つ前記の培地中で4日間インキュベートする。インキュベーシ
ョン後、細胞をトリチウム化チミジン1μCiで6時間パルスした後、それらを
採取し且つトリチウム化チミジン取込みを、例えば、シンチージョン計数によっ
て測定する。
実験
以下の実施例は、本発明を例証するのに役立つ。細胞系、試薬およびそれらの濃
度、温度並びに他の変数値は、本発明の応用を単に例示するものであり、本発明
を制限するものと考えられるべきではない。
造血系前駆細胞から発生した細胞を二色蛍光測定用に処理した。簡単にいうと、
細胞を、非結合単クローン性抗体、フィコエリトリン結合(PE結合)抗マウス
免疫グロブリン、正常マウス血清および、フルオレセインイソチオシアネート(
FITC)で直接的に標識された単クローン性抗体0KT6 (オルトからの抗
CD1a)またはLeu−M3(ベクトン・デイキンソンからの抗CD14)と
−緒に逐次的にインキュベートした。図1で示したように、CD34 細胞をG
M−C3FおよびTNF−αの存在下で12日間培養することは、CD1a抗原
をは一貫して観察された。単球系統内でのCD1a抗原の発現は、ランゲルノ1
ンス細胞に限定される。図1aは、IgG −FITC7ソタイブ対照対1 g
G 28−PEイソタイプ対照からの二色蛍光強度を示す。図IBは、0KT
6−FITC(CDla発現に比例する)対しeu−M3−PE (CD14発
現に比例する)からの二色蛍光強度を示す。図2で示したように、CD1a 細
胞は培養の開始時に検出されなかったし、CD1a抗原はTNF−α存在下の4
日間の培養後に始めて観察することができ、そして発現は20日目止で増加した
。5%未満のCD1a 細胞がIL−3存在下で観察されたが、5〜15%のC
D1a 細胞はGM−C3Fにおいて検出された。大部分のCD1a 細胞はI
L−3およびTNF−αにおいて(10〜25%)、主としてGM−C8Fおよ
びTNF−αにおいて(20〜60%)検出された(12日間の培養後の5回の
実験の範囲)。増殖効率に関して、LL−3およびTNF−αはIL−3のみよ
りも2〜3倍TNF−αにおいては細胞2〜3x106個の培養物が回収された
。GM−C8FおよびTNF−αは、CD1a 細胞を発生させるのに最も強力
な因子組合せであると考えられたので、これらの細胞の特性決定用の培養はいず
れも、GM−C8FおよびTNF−αを含んだ。図2は、(i)IL−3(中実
三角)、(ii)IL−3およびTNF−α(中空三角)、(i i i)GM
−C8F (中実四角)および(iv)GM−C3FおよびTNF−a(中空四
角)の存在下のCD34 細胞105個の増加を示す。実線は3回の実験におけ
る全細胞数を示し、+
破線はCD1a抗原の発現を示した。図2Aおよび2Bは、2組の別個の実験に
よるデータを示す。
本発明の方法によって発生したCD1a 細胞の形態学を、光学顕微鏡および電
子顕微鏡両方を用いて研究した。GM−C8Fのみにおいて観察された付着細胞
は、規則的な形状のマクロファージの古典的様相を示すが、GM−C3Fおよび
TNF−αの存在下で得られたものは、高度に分岐した樹状突起、小葉に分かれ
た核および樹状突起のある絨毛表面を有する樹状細胞の典型的な様相を有する。
若干のCD1a+細胞(5個中約1個)は二重膜結合のある小器官を有し、バー
ベック顆粒の構造を思い起こさせる。
GM−C3FおよびTNF−αの存在下での12日間培養後に発生したCD1a
+細胞の表現型は二色蛍光分析によって決定された。実験に応じて、CD14を
同時発現するCD1a 細胞の百分率は10〜70%に変化した。下記の表1細
胞はCD1cを発現しなかったし、CD4およびCD40を僅かしか発現しなか
った。CD1a およびCD14 細胞は両方ともFC7RI I (CD32
)、FCγRI I I (CD16)およびCR3(CD11b)を有するこ
とが分かったが、CD1a CD14 細胞だけがFC7RI (CD64)お
よびCR1(CD 35)を発現した。CD1.a およびCD1a CD14
細胞は、LFAl a (CDI 1 a)およびLFΔ1β(CD18)の
両方を発現したが、CD1a+細胞はCD1a CD14 細胞よりも高濃度の
ICAMI (CD54)を示した。CD1a 細胞は極めて高濃度のHLA−
DRを発現した(CD14“細胞よりも5〜10倍大きい)。対照的に、CD1

CD34 細胞を本発明にしたがって12時間培養した後、それらに4000ラ
ドを照射して、実験用の刺激細胞を生成した3、刺激細胞10〜2.4x104
個を、以下に記載したように、丸底マイクロテスト組織培養プレート中において

10%ヒトAB 血清を補足した培地中の休止CD4 T細胞または他の応答細
胞2.5xlO4個に対して播種した。
本発明の樹状細胞の、休止同種異系CD4+T細胞を(応答細胞として)増殖さ
せる能力を検定した。図3Aで示したように、IL−3のみの存在下(中実玉子
角)で培養された細胞は、最低の同種異系CD4 T細胞増殖を誘導した。対照
的に、GM−C8Fのみの存在下(中実四角)、IL−3およびTNF−αの存
在下(中空三角)並びにGM−C8FおよびTNF−αの存在下(中空四角)で
培養された細胞は、同種異系CD4+T細胞の激しい増殖を誘導した。CD34
+
“前駆細胞の培養条件に応じて、CD34 T細胞の最適増殖は、異なる値の比
率(刺激細胞)/(同種異系CD34+T細胞)に対して観察された。対照値(

刺激細胞不含CD34 T細胞)との比較において、CD4 T細胞によるトリ
チウム化チミジン取込みの50倍の増加は、GM−C3Fのみ、IL−3および
TNF−α、並びにGM−C3FおよびTNF−αのそれぞれの存在下で培養さ
れた細胞に関して1:3.8(1:3〜1:25の範囲)、1:12.5 ci
:10〜1:35の範囲)および1 : 360 (1: 100〜1 : 4

図3Bで図示した実験において、CD34 細胞を全部の実験に関してGM−C
8FおよびTNF−αの存在下で培養し、そして応答細胞は成人末梢血(中空十
四角)、謄帯血(中空および中実三角)および同系謄帯血CD4 T細胞(中実
四角)であった。図3Bは、謄帯血由来かまたは成人末梢血由来の同種異系CD
4+T細胞が同様に刺激されたこと、および同系CD4+T細胞がはるかに低い
程度に刺激されたこと(同種異系細胞より20倍低い応答)を示す。
GM−C3FおよびTNF−αの存在下の12日間の培養後に、CD1a+細胞
がイムノマグネチック除去によって除かれた場合(図4)、誘導能力の激しい+

減損が観察された。CD4 T細胞増殖の50倍の増加は、CD1a 細胞が除
去された前後に、それぞれ1:200(3回の実験の1+200〜1:400の
範囲)および1:8(3回の実験の1:8〜1:40の範囲)の比率(刺激細胞

)/(同種異系CD4 T細胞)に対して観察された。照射後、以下、すなわち

、全集団(中実四角)、CD1a 細胞が除去によって除かれた集団(中空四角
)および、抗IgG1イソタイプ抗体を用いて除去が行なわれた対照集団(中空
三角)を刺激細胞として用いた。
CD34+細胞を癌患者の末梢血から単離し且つ前記のようにGM−C3Fおよ
びTNF−αの存在下で増殖させる。末梢血CD8細胞を冷凍保存する。樹状細
胞がいったん発生したら、CD8T細胞を解W+フ且つ患者の癌細胞と混合する
。感作後、例えば、ローゼンバーグ(Rosenberg)、米国特許第4,6
90.915号明細書に記載されたように、CD8T細胞をIL−2存在下で増
加させる。全細胞を、それらが腫瘍特異的細胞障害活性を示すという条件で患者
に再注入する。腫瘍細胞は、特異的腫瘍抗原で置換されることができる。例えば
1フアン0デア0プルジエン(van der Bruggen)ら、CD34
+謄帯血前駆細胞から発生した樹状細胞は、12日間培養され且つ照射(400
0ラド)された後、休止CD4+およびCD8+T細胞に対する刺激細胞として
用いられた。刺激細胞10〜2.5xlO3個を、丸底マイ知テスト組織培養プ
レート中において、10%ヒトAB+血清を補足した、200/m1のIL−2
含有または不含培地中の休止T細胞2xlO4個に対して播種した。5日間のイ
ンキュベーション後、細胞を3H−チミジン1μCiで8時間パルスし、採取し
そして計数した。試験は三重反復で行ない、そして結果を1分間当たりの平均数
として表わした。
CD4 およびCD8+T細胞を培養した培地に対してIL−2を加えること十
は、実際に、それらの3H−チミジン取込みによって示されるように、CD4+
およびCD8 T細胞の増殖を刺激した。図5で示したように、培地のみの存在
下(中空四角)で培養されたCD8 細胞は極めて少ない増殖を示したが、TL

殖を示さなかったが、IL−2の存在下(中実丸)で培養されたCD4 細胞は
かなり多い増殖を示した。
5CID−huマウスは、二次応答で抗原を呼び戻すことを可能にしたが、−次
応答の確立はできなかった。例えば、モラー(Mo l l e r) 、Th
、e、5CID−hu Mouse、124巻仏ンクスガード(Munk、sg
gard)、コペンハーゲン、1991);デュコサル(Duchosal)ら
、195頁(1989)。これは樹状細胞プールが再構成されなかったことに依
ると考えられる。抗原でパルスされた樹状細胞はインビボで抗体応答を効率よく
誘導することができるので(例えば、ソーナス(Sornasse)ら、エエE
xp、Med、、1.75巻、15頁(1992))、ヒト細胞のドナーのCD
34細胞からまたは適合性MHCを共有する別のドナーからインビトロで発生し
た樹状細胞を用いて5CID−huマウスを再構成する。樹状細胞は、注入前に
適当な抗原でパルスされる。マウスがいったん抗原に対して抗体を示したら、B
細胞を血液および他の再構成された器官から単離し、そして例えば、バンチエル
−(Banchereau)ら、5cience、251巻、70頁(1991
)で示されたようにエプスタイン・バールウィルス存在下のCD40系において
培養することによって不死化させる。
実施例5
インビトロで発生した樹状細胞を用いる養子免疫療法によるエイズの治療樹状細
胞をインビトロにおいてHIVで感染させ、そしてHIVは、HIVを用いる培
養において3〜5日後に、細胞表面から出芽するのが見出される。例えば、バダ

Claims (20)
- 1.ヒト樹状細胞を含む細胞組成物の製造法であって、CD34+造血系細胞を 、TNF−αおよびIL−3でまたはGM−CSFで処理し;そして CD1a抗原を発現する被処理CD34+造血系細胞を単離する工程を含む上記 方法。
- 2.前記CD34+細胞をGM−CSFの他にTNF−αで処理する請求項1に 記載の方法。
- 3.インビトロ培養後にCD1a抗原を発現するCD34+造血系細胞。
- 4.細胞組成物であって、 CD34+造血系細胞を、TNF−αおよびIL−3でまたはGM−CSFで処 理し;そして CD1a抗原を発現する被処理CD34+造血系細胞を単離する工程を含む方法 によって製造されたヒト樹状細胞を含む上記細胞組成物。
- 5.前記CD34+細胞がGM−CSFの他にTNF−αで処理されている請求 項4に記載の細胞組成物。
- 6.樹状細胞系を製造するためのTNF−αとIL−3と一緒のまたはGM−C SFの使用。
- 7.樹状細胞を誘導するためのTNF−αとIL−3と一緒のまたはGM−CS Fの使用。
- 8.CD4+ヘルパーT細胞を発生させるための樹状細胞の使用。
- 9.養子免疫療法におけるCD4+ヘルパーT細胞の使用。
- 10.癌特異的およびウイルス特異的CD8+細胞障害性T細胞を発生させるた めの樹状細胞の使用。
- 11.エイズウイルスに対して特異的なCD8+細胞障害性T細胞を発生させる ための樹状細胞の使用。
- 12.混合リンパ球反応であって、 応答細胞試料を提供し; 刺激細胞が該応答細胞に関して同種異系であるような、しかも刺激細胞が、(a )CD34+造血系細胞をTNF−αおよびIL−3でまたはGM−CSFで処 理し、そして(b)CD1a抗原を発現する被処理CD34+造血系細胞を単離 する工程を含む方法によって製造された樹状細胞から成るような不活化刺激細胞 試料を提供し; 該応答細胞および該不活化刺激細胞を同時培養し;そして該応答細胞の応答を測 定する工程を含む上記混合リンパ球反応。
- 13.前記CD34+細胞をTNF−αおよびGM−CSFで処理する請求項1 2に記載の方法。
- 14.前記応答細胞の前記応答が増殖である請求項12に記載の方法。
- 15.IL−3およびGM−CSFを飽和濃度で用いる請求項12に記載の方法 。
- 16.TNF−αを2〜3ng/mLまたは40〜60U/mLの範囲の濃度で 用いる請求項12に記載の方法。
- 17.TNF−αを約2.5ng/mLまたは50U/mLの濃度で用いる請求 項12に記載の方法。
- 18.癌に罹患している患者の治療法であって、該患者に対して請求項3に記載 の細胞を投与することを含む上記方法。
- 19.ウイルス感染に罹患している患者の治療法であって、該患者に対して請求 項3に記載の細胞を投与することを含む上記方法。
- 20.細胞が患者由来である請求項18または請求項19に記載の方法。
