Abstract

세포 배양 기술의 개발로 인해, 생체내 면역 과정을 모방한 시험관내 방법을 확립하게 됨으로써, 시험관내에서 세포-중재된 면역성을 평가하는 측정법이 제공되었다. 특히, 이식과 관련한 유용성은 혼합 임파구 반응(MLR), 또는 혼합 임파구 배양이다. [발명의 요약]

[발명의 상세한 설명]
본 발명의 중요한 양상은 CD34+조혈 세포로부터 수지상 세포의 생성이다.
CD34+조혈원 세포는 다양한 조직원, 예를 들어 골수로부터 수득되나, 바람직하게는 다음과 같이 제대 혈액 샘플로부터 수득된다: 샘플로부터 정밀도 단핵구를 피콜-하이파큐(Ficoll-Hypaque) 구배 분리(d=1.077g/ml)로 분리하고, 예를 들어 1% w/v 조직-배양 등급의 소 혈청 알부민으로 보충된 RPMI 1640 배지에서 37℃로 밤새 배양하여 유착 세포를 제거시킨다. 바람직하게는, CD34 항원을 포함하는 세포를 항-CD34 모노클로날 항체, 예를 들어 Immunotech(Marseille, France)로부터 입수가능한 Imu-133.3, Becton Dickinson(Mountain View, California)으로부터 입수가능한 항-My 10등을 사용하는 간접적 면역 패닝(panning)에 의한 포지티브 선별을 통해 비-유착 단핵구 분획으로부터 분리시킨다. 패닝 플라스크는 다음과 같이 제조한다: 트리스 완충액(0.05mol/l, pH 9.4)중의 25μg/ml 농도의 양(sheep) Fab 항마우스 IgG를 75-㎠ 조직 배양 플라스크에 분배시켜(10ml) 4℃에서 밤새 코팅시킨다. 별도로, 경-밀도 단핵구(상술한 바와 같이 유착 세포를 제거시킴)를 2% 열-불활성화시킨 푸울링된 사람 AB 혈청(HABS)로 보충된 RPMI 1640중에서 107개 세포/ml로 5μg/ml의 항-CD34 항체와 함께 4℃에서 1시간 동안 배양한다. 이어서, 세포를 2% HABS를 포함하는 찬 배지내에서 세척하고, 약 5×107개 세포를 포함하는 10ml을, 상술한 바와 같이 양의 항마우스 IgG로 미리 피복시킨 플라스크에 분배시킨다. 4℃에서 2시간 배양후, 현탁액중의 비-유착 세포(즉, CD34-제거된 분획)를 가만히 피펫팅하고 배지로 수회 세정함으로써 수거한다. 이어서, 유착 분리된(panned) 세포(즉, CD34-풍부 분획)을 격렬한 피펫팅으로 회수한다.

배양물중에 형성되는 수지상 세포는 상술한 바와 같이 패닝시켜 분리시키며, 단 항-CD1a 및/또는 항-CD14 항체가 사용된다는 것을 예외로 한다(두 항체는 예를 들어 Becton-Dickinson에서 상업상 입수가능하다).

바람직하게는, 반응 세포의 샘플은 이식조직을 수령할 환자의 말초혈로부터의 CD4+T 세포로 이루어진다. T 세포 모집단을 수득하는 데 사용되는 기술은 당 분야에 널리 공지되어 있으며, 문헌[참조: DiSabato et al.. eds.. in Meth. in Enzymol.. Vol. 108(1984)]에 충분히 기술되어 있다. 예를 들어, CD4+T 세포는 하기와 같이 분리될 수 있다: 먼저 단핵구를 말초혈로부터 분리시키고 유착 세포를 제거시킨다. 이어서 CD4+T 세포를, 예를 들어 시판되는 모노클로날 항체, 예를 들어 항-CD14, 항-CD16, 항-CD20, 항-CD8, 항-CD40(Becton-Dickinson 및/또는 Ortho Diagnostic Systems, New Jersey 시판)의 칵테일을 사용하여 면역자기 제거[예를 들어 Dynabeads(Dynal. 본 발명의 자극 세포는 반응 세포를 취한 사람이 아닌 다른 사람으로부터 취한 CD34+조혈원 세포로부터 유도되는 수지상 세포이다. 즉, 자극 세포는 반응 세포에 대해 동종이다. 이들 세포는 상술한 바와 같이 수득한다.
본 발명의 자극 세포는, 그들의 자극 작용은 여전히 포함하되 반응 세포로부터 측정되는 반응은 감출 수 있는 다른 작용은 억제되도록 불활성화된다. 따라서, 불활성화의 성질은 다소 검정의 판독(read-out)에 따라 달라진다. 바람직하게는, 반응 세포에서 측정되는 반응, 또는 판독은 세포 증식이다. 기타의 판독은 또한 사이토킨 생성, 세포분해능 등과 같은 현상을 포함한다. 바람직하게는, 자극 세포는, 복제될 수 없으나 이들의 항원-프로세싱 기구가 여전히 작용성이도록 처리된다. 이는, 통상적으로, 반응 세포와 혼합하기 전에, 예를 들어 약 1,500내지 5,000R(감마 또는 X-방사선), 바람직하게는 3,000 내지 4,000R로 세포를 조사하여 수행한다.

본 발명의 방법에 의해 생성된 CD1a+세포의 형태를 광학현미경 및 전자 현미경으로 조사한다. GM-CSF 단독에서 관측된 유착 세포는 일정 모양의 대식구에 대한 일반적 양상을 나타내는 반면, GM-CSF+TNF-α의 존재하에서 수득된 세포들은 고분지 수지상돌기, 소엽 핵, 및 수지상 돌출을 갖는 융모상 표면이 있는 수지상 세포의 전형적 양상을 갖는다. CD1a+세포중 일부(5개중 약 1개)는 버백(Birbeck) 과립의 구조를 생각나게 하는, 이중막을 연결시키는 소기관을 갖는다.
GM-CSF와 TNF-α의 존재하에서 배양 12일후 생성된 CD1a+세포의 표현형은 2-색 형광 분석으로 결정한다. 실험에 따라서, CD14를 공동-발현하는 CD1a+세포의 백분율(%)은 10 내지 70%로 다양하다. 하기 표1에 나타낸 결과는, CD1a+세포 15%미만이 CD14를 공동-발현하는 실험으로부터 수득된 것이다: 이로써, CD1a+세포와 CD1a-CD14+세포 모두가 특정분석되었다. 수지상 세포에 의한 자극후 CD4+ T 세포의 증식
CD34+세포를 본 발명에 따라 12시간 동안 배양한 후, 4,000Rad로 조사시켜 실험용 자극 세포를 만든다. 10 내지 (2.4×104)개 자극 세포를, 후술하는 바와 같이 10% 사람 AB+혈청으로 보충된 배지중의 환저 미세시험 조직 배양 플레이트의 2.5×104개 휴지 CD4+T 세포 또는 기타 반응 세포에 대해 시이딩시킨다.
본 발명의 수지상 세포를, (반응 세포로서의) 휴지 동종 CD4+T 세포의 증식을 유도하는 능력에 대해 검정한다. 제3a도에 나타낸 바와 같이, IL-3 단독의 존재하에서 배양된 세포(▲)는 최저의 동종 CD4+T 세포 증식을 유도하였다. 이와는 대조적으로, GM-CSF 단독(■), IL-3+TNF-α(△), 및 GM-CSF+TNF-α(□)의 존재하에서 배양된 세포는 동종 CD4+T 세포의 증식을 강력히 유도하였다. 제3b도에 나타난 실험에서, CD34+세포는 모든 실험에 대해 GM-CSF 및 TNF-α의 존재하에서 배양하며, 반응 세포는 성인 말초혈(□), 코드혈(△ 및 ▲), 및 동계 코드혈 CD4+T 세포(■)이다. 제3b도는, 코드혈 또는 성인 말초혈로부터 유도된 동종 CD4+T 세포가 동일하게 자극되었으며, 동계 CD4+T 세포가 매우 낮은 정도(동종 세포보다 20배 약한 반응)로 자극되었다는 것을 나타낸다.
GM-CSF 및 TNF-α의 존재하에서 12일 배양한 후, CD1a+세포를 면역자기 제거에 의해 제거시키는 경우(제4도). 유도능의 강력한 상실이 관측되었다. CD4+T 세포 증식의 50배 향상은 각각 CD1a+세포의 제거 전후에 1:200(1:200 내지 1:400의 3회 실험 범위) 및 1:8(1:8 내지 1:40의 3회 실험 범위)의 비[(자극세포)/(동종 CD4+T 세포)]에 대해 관측되었다. 조사후, 자극 세포로서 다음이 사용되었다: 총모집단(■), CD1a+세포를 제거시켜 포함시키지 않은 모집단(□), 및 항-IgG1이소타입 항체로 제거시킨 대조군 모집단(△).
[실시예 3]
수동 면역 치료용의 암-특이적 CD8 세포독성 T 세포의 생성
CD34+세포를 상술한 바와 같이 암 환자의 말초혈로부터 분리시키고, GM-CSF 와 TNF-α의 존재하에 성장시킨다. 말초혈 CD8 세포를 냉동보관한다. 수지상 세포를 생성시킨후, CD8 T 세포를 해동시킨 다음, 이를 환자의 암 세포와 혼합한다. 감작화시킨후, CD8 T 세포를, 예를 들어 문헌[참조: Rosenberg의 미합중국 특허 제4,690,915호]에 기술되어 있는 바와 같이 IL-2의 존재하에 증식시킨다. 총 세포를, 이들이 종양-특이적 세포독성 활성을 나타내는 경우 환자에게 재주입시킨다. 종양 세포를 특이 종양 항원으로 대체시킨다[참조: van der Bruggen et al.. GM-CSF 및 TNF-α의 존재하에서 생성된 수지상 세포는 휴지 동종 CD8+T 세포 증식의 강력한 자극제이다.
CD34+코드혈원으로부터 생성된 수지상 세포를 12일 동안 배양하여 조사(4,000Rad)시키고, 이어서 휴지 CD4+및 CD8+T 세포에 대한 자극 세포로서 사용한다. 10 내지 2.5×105개 자극 세포를 10% 사람 AB+혈청으로 보충된 배지(20U/ml IL-2의 존재 또는 부재)중의 환저 미세시험 조직 배양 플레이트내의 2×104개 휴지 T 세포에 대해 시이딩시킨다. 5일간 배양후, 세포를 8시간 동안3H-티미딘 1μCi로 펄스를 발생시키고, 수거하여 계수한다. 시험을 삼중으로 수행하고, 그 결과를 평균 수/분으로 나타낸다.

SCID-hu 마우스에서 제1 및 제2 항체 반응의 발생
SCID-hu 마우스는 제2 반응이 항원을 상기시키되 제1 반응은 확립시키지 않도록 한다(참조: Moller, The SCID-hu Mouse, Vol. 124(Munksgaard. Copenhagen. 1991); Duchosal et al.. Nature. Vol. 355. pp. 258(1992); and Mosier et al.. Curr. Top. Microbiol. Immunol.. Vol. 152. pp. 195(1989)]. 이는 수지상 세포 푸울의 재구성이 부족하기 때문인 것으로 믿어진다. 항원-펄스 발생시킨 수지상 세포는 생체내에서 항체 반응을 효과적으로 유도할 수 있기 때문에[참조: Sornasse et al.. J. Exp. Med.. Science. Vol. 251. pp. 70(1991)]에 교시되어 있는 바와 같이 엡스타인-바르 바이러스의 존재하에 CD40 시스템에서 배양함으로써 죽지 않도록하였다.

시험관내에서 생성된 수지상 세포를 사용한 수동 면역 치료에 의한 AIDS 치료
수지상 세포를 시험관내에서 HIV에 의해 감염시키면, HIV를 포함하는 배양물에서 3 내지 5일후 세포 표면으로부터 HIV가 발아하는 것으로 밝혀졌다[참조: Patterson. J. Gen. Virol.. Vol. 68. pgs. 1177-1181(1987)]; Macatonia et al.. Immunology. Vol. 71. pgs. 38-45(1989); and Knight et al.. Immunol. Lett.. Vol. 19. pgs. 177-182(1988)]. HIV에 의한 수지상 세포의 생체내 감염에 대한 증거가 피부의 랑게르한스 세포에 대한 감염의 기술 및 피부 세포내 MHC 제11형 분자의 양 감소로부터 나타난다[참조: Tschachler et al.. J. Invest. Dermatol.. Vol. 88. pgs. 233-237(1987); and Belsito et al.. N. Engl. J. Claims (6)
- 사람 CD34+조혈세포를 GM-CSF , TNF-α와 IL-3 또는 GM-CSF와 TNF-α로 처리함으로써 상기 CD34+조혈세포로부터 사람 수지상(dendritic) 세포의 생성으로 유도하는 단계; 및 이러한 사람 수지상 세포를 회수하는 단계를 포함하여, 사람 수지상 세포를 포함하는 세포성 조성물을 제조하는 방법.
- 암 또는 바이러스 항원으로 펄스 발생시킨 수지상 세포를 시험관내에서 생성시키고, CD8+세포독성 T 세포를 감작화시켜 이의 증식을 유도하기에 충분한 양의 상기 시험관내에서 생성된 수지상 세포로 사람 CD8+T 세포를 처리하는 단계; 및 암-특이적 또는 바이러스-특이적 사람 CD8+세포독성 T 세포를 회수하는 단계를 포함하여, 암-특이적 또는 바이러스-특이적 사람 CD8+세포독성 T 세포를 생성하는 방법.
- 제2항에 있어서, CD8+T 세포가 암 항원으로 감작화되어 암 특이적 CD8+세포독성 T 세포가 생성되는 방법.
- 제2항에 있어서, CD8+T 세포가 바이러스 항원으로 감작화되어 바이러스 특이적 CD8+세포독성 T 세포가 생성되는 방법.
- 제4항에 있어서, CD8+T 세포가 AIDS 바이러스의 바이러스 항원으로 감작화되고, 생성된 CD8+세포독성 T 세포가 HIV 바이러스에 대해 특이적인 방법.
- 항원으로 펄스 발생시킨 수지상 세포를 시험관내에서 생성시키고, 항원-특이적 CD4+헬퍼 T 세포를 감작화시켜 이의 증식을 유도하기에 충분한 양의 상기 시험관내에서 생성된 수지상 세포로 사람 CD4+T 세포를 처리하는 단계; 및 항원 특이적 사람 CD4+헬퍼 T 세포를 회수하는 단계를 포함하여, 항원 특이적 사람 CD4+헬퍼 T 세포를 생성하는 방법.
