Abstract
Description
Claims (20)
- 1.誘発性プロモーターの制御下で少なくとも1種の発現可能ヒトクラスIMH C遺伝子を含む真核細胞系。
- 2.発現可能ヒトβ2ミクログロブリン遺伝子をさらに含む請求項1記載の細胞 系。
- 3.少なくとも1種の発現可能ヒトクラスIMHC遺伝子を含む昆虫細胞系。
- 4.前記遺伝子が誘発可能プロモーターの制御下に存在する請求項3記載の細胞 系。
- 5.発現可能ヒトβ2ミクログロブリン遺伝子をさらに含む請求項3記載の細胞 系。
- 6.ヒトクラスIMHC分子モノタイプを発現する請求項1記載の細胞系。
- 7.前記HHC分子が空である請求項6記載の細胞系。
- 8.前記昆虫細胞が、表面発現されたヒトクラスMHC分子を有するドロソフィ ラ細胞を含んで成る請求項3記載の細胞系。
- 9.前記表面一発現されたMHC分子の数が、CD8細胞を活性化するのに十分 な量である請求項8記載の細胞。
- 10.少なくとも1種の発現可能ヒトクラスIMHC遺伝子を含む真核細胞系を 生成するための方法であって;真核細胞の培養物を確立し;そして 誘発性プロモーター及び発現可能ヒトクラスIMHC遺伝子を含んで成る誘発性 発現ベクターにより前記培養物をトランスフェクトすることを含んで成る方法。
- 11.前記ベクターが、発現可能ヒトβ2ミクログロブリン遺伝子をさらに含ん で成る請求項10記載の方法。
- 12.前記ベクターが、転写可能ポリアデニル化部位をコードするヌクレオチド 配列をさらに含んで成る請求項10記載の方法。
- 13.前記細胞がドロソフィラ細胞である請求項10記載の方法。
- 14.前記ベクターがpRmHa−3を含んで成る請求項13記載の方法。
- 15.活性化されたCD8細胞をインビトロで生成するための方法であって、C D8細胞とドロソフィラ細胞上に表面発現された抗原一負荷ヒトクラスIMHC 分子とを、前記CD8細胞を抗原特異的態様で活性化するのに十分な時間、イン ビトロで接触せしめることを含んで成る方法。
- 16.前記抗原一負荷クラス1分子から前記活性化されたCD8細胞を分離し; 前記活性化されたCD8細胞を許容できるキャリヤー又は賦形剤に懸濁し;そし て 前記懸濁液を処置の必要な個人に投与することをさらに含んで成る請求項15記 載の方法。
- 17.ヒト患者における標的細胞を特異的に殺害するための方法であって: 前記患者から前駆体CD8細胞を含む流体サンプルを得;前記CD8細胞とドロ ソフィラ細胞上で表面一発現された抗原一負荷ヒトクラスIMHC分子とを、抗 原特異的態様で前記CD8細胞を活性化するのに十分な時間、インビトロで接触 し;そして前記活性化されたCD8細胞を前記患者に投与することを含んで成る 方法。
- 18.移植された組織の拒絶を阻止するための方法であって:ドロソフィラ細胞 上で表面一発現された空のヒトクラスI分子を毒素にインビトロで接合し; 前記接合されたクラスI分子と前記移植された粗織に由来する抗原性ペプチドと を、前記接合体に前記ペプチドを負荷するのに十分な時間、インビトロで接触し ; 前記ペプチドー負荷された接合体を分離し;そして前記ペプチドー負荷された接 合体を、移植処置を受けた個人に投与することを含んで成る方法。
- 19.誘発性プロモーターに操作的に結合される発現性ヒトクラスIHHC遺伝 子を含んで成る組換えDNA配列。
- 20.ベクターにおける請求項19記載の配列。
