製造例
下記製造例の方法で表1の化合物を製造した。

下記に記載された10段階の反応を経て2-((2-ethylbenzothiophene-3-yl)methyl)hexahydro-2H-pyrazino[1,2-a]pyrazine-6,9-dioneを製造した。各段階での合成方法を以下で具体的に説明する。
1)段階1
反応式1のようにベンゾチオフェン(1.0当量)をテトラヒドロフラン溶媒下で-78℃に冷却し、2.5Mのn-ブチルリチウム(1.2当量)溶液を添加した。-78℃を維持しつつ1時間撹拌し、エチルヨージド(2.0当量)を滴加した後、0℃で1時間撹拌した。反応完結を確認し、アンモニウムクロライド水溶液とエチルアセテートを使用して反応を終了した。有機層を蒸留水で洗浄し、無水マグネシウムサルフェートで乾燥後、減圧濃縮した。残渣をシリカクロマトグラフィーで精製して、2-エチルベンゾチオフェンを収得した。
2)段階2
2-エチルベンゾチオフェン(1.0当量)をメチレンクロライド(MC)に投入した後、0℃に冷却し、0℃を維持しつつ、スズ(IV)クロライド(1.5当量)、ジクロロメチルメチルエーテル(1.5当量)を順に投入した後、1時間撹拌した。反応完結を確認し、アンモニウムクロライド水溶液とメチレンクロライドを使用して反応を終了した。有機層を蒸留水で洗浄し、無水マグネシウムサルフェートで乾燥し、減圧濃縮した。残渣をシリカクロマトグラフィーで精製して、2-ethyl-1-benzothiophene-3-carbaldehydeを収得した。
3)段階3
piperazine-2-carboxylic acid(1.0当量)、ジオキサンおよび蒸留水を入れ、5℃以下に冷却した。その後、di-tert-butyl dicarbonate(1.1当量)を10℃以下を維持しつつ投入した。室温で5時間撹拌後、炭酸ナトリウム(1.1当量)を投入し、5分間撹拌した。9-Fluorenylmethyl chloroformate(1.2当量)を投入し、一晩中撹拌後、減圧濃縮した。残渣にエチルアテートと1M塩酸を入れ、pHを2~3にし、残渣がすべて溶けるまで激しく撹拌した。有機層をbrineで洗浄し、無水マグネシウムサルフェートで乾燥後、減圧濃縮した。EAで再結晶化して、1-(((9H-fluoren-9-yl)methoxy)carbonyl)-4-(tert-butoxycarbonyl)piperazine-2-carboxylic acidを収得した。
4)段階4
1-(((9H-fluoren-9-yl)methoxy)carbonyl)-4-(tert-butoxycarbonyl)piperazine-2-carboxylic acid、ベンジルアルコール(1.02当量)とトリフェニルホスフィン(1.02当量)の無水メチレンクロライド溶液を窒素気流下で0℃に冷却した。0℃以下を維持しつつ、Diethyl Azodicarboxylate(DEAD)(1.02当量)を投入し、1時間撹拌した。水を投入して、20分間激しく撹拌後、有機層を無水マグネシウムサルフェートで乾燥後、減圧濃縮した。残渣をシリカクロマトグラフィーで精製して、1-((9H-fluoren-9-yl)methyl)2-benzyl 4-(tert-butyl)piperazine-1,2,4-tricarboxylateを収得した。
5)段階5
反応式5のように、室温で1-((9H-fluoren-9-yl)methyl)2-benzyl 4-(tert-butyl)piperazine-1,2,4-tricarboxylateのメチレンクロライド(5.0ボリューム)溶液にトリフルオロ酢酸(TFA)(2.0ボリューム)を添加し、室温で30分間撹拌した。重炭酸ナトリウムで中和させた後、メチレンクロライドで抽出し、有機層をbrineで洗浄した。無水マグネシウムサルフェートで乾燥し、濃縮させて、1-((9H-fluoren-9-yl)methyl)2-benzyl piperazine-1,2-dicarboxylateを収得した。
6)段階6
室温で段階2を通じて収得した2-ethyl-1-benzothiophene-3-carbaldehyde(1.0当量)と段階5を通じて収得した1-((9H-fluoren-9-yl)methyl)2-benzyl piperazine-1,2-dicarboxylate(1.2当量)を1,2-ジクロロエタン(DCE)に投入後、30分間撹拌した。ナトリウムトリアセトキシボロヒドリド(3.0当量)を入れ、室温で一晩中撹拌した。反応液にメチレンクロライドと水を入れ、20分間激しく撹拌した。分離した有機層をbrineで洗浄し、無水マグネシウムサルフェートで乾燥し、濃縮して、1-((9H-fluoren-9-yl)methyl)2-benzyl 4-((2-ethylbenzothiophen-3-yl)methyl)piperazine- 1,2-dicarboxylateを収得した。
7)段階7
室温で1-((9H-fluoren-9-yl)methyl)2-benzyl 4-((2-ethylbenzothiophene-3-yl)methyl)piperazine-1,2-dicarboxylate(1.0当量)をジメチルホルムアミドに溶解させた後、ピペリジン(10当量、溶媒中、25%ピペリジン)を投入し、1時間撹拌した。反応液にエチルアセテートを入れた後、アンモニウムクロライド水溶液で溶液内のピペリジンを洗浄した。無水マグネシウムサルフェートで乾燥後、減圧濃縮して得られた残渣をシリカクロマトグラフィーで精製して、benzyl 4-((2-ethylbenzothiophene-3-yl)methyl)piperazine-2-carboxylateを収得した。
8)段階8
室温でbenzyl 4-((2-ethylbenzofuran-3-yl)methyl)piperazine-2-carboxylate(1.0当量)とN-(tert-Butoxycarbonyl)glycine(1.1当量)、および(Benzotriazol-1-yloxy)tripyrrolidinophosphonium hexafluorophosphate(1.2当量)をジメチルホルムアミドで溶解させた後、N,N-ジイソメチルプロピルエチルアミン(1.5当量)を添加し、室温で一晩中撹拌した。アンモニウムクロライド水溶液とエチルアセテートを使用して反応を終了させた。得られた有機層をbrineで洗浄後、無水マグネシウムサルフェートで乾燥し、減圧濃縮した。残渣をシリカクロマトグラフィーで精製して、benzyl 1-((tert-butoxycarbonyl)glycyl)-4-((2-ethylbenzothiophene-3-yl)methyl)piperazine-2-carboxylateを収得した。
9)段階9
反応式9のように、室温で段階8を通じて収得したbenzyl 1-((tert-butoxycarbonyl)glycyl)-4-((2-ethylbenzothiophene-3-yl)methyl)piperazine-2-carboxylateをメチレンクロライド(5.0ボリューム)に溶解させた後、トリフルオロ酢酸(2.0ボリューム)を添加して室温で30分間撹拌した。反応が完了した後、反応液を重炭酸ナトリウム水溶液で中和させ、メチレンクロライドで抽出した。無水マグネシウムサルフェートで乾燥し、減圧濃縮して、benzyl 4-((2-ethylbenzothiophene-3-yl)methyl)-1-glycylpiperazine-2-carboxylateを収得した。
10)段階10
段階9で収得したbenzyl 4-((2-ethylbenzothiophene-3-yl)methyl)-1-glycylpiperazine-2-carboxylateをイソプロパノール(5.0ボリューム)に溶解させた後、酢酸(1.5ボリューム)を入れて加温して、1時間撹拌した。反応が完結した後、重炭酸ナトリウム水溶液で中和した。反応物をメチレンクロライドで抽出し、無水マグネシウムサルフェートで乾燥した。減圧濃縮後、残渣をシリカクロマトグラフィーで精製して、2-((2-ethylbenzothiophene-3-yl)methyl)hexahydro-2H-pyrazino[1,2-a]pyrazine-6,9-dione(S101)を収得した。

実験例
1.SKOV3とSKOV3-TRの準備
上皮性卵巣がん細胞株であるSKOV3とこれに由来してパクリタキセル(Paclitaxel)抗がん剤に抵抗性を有する耐性がん細胞株で製作されたSKOV3-TRを対象に細胞実験を進め、図1(a)は、SKOV3のパクリタキセル抗がん剤IC50値を示す図であり、(b)は、SKOV3-TRのパクリタキセル抗がん剤IC50値を示す図である。図2(a)は、SKOV3のドセタキセル抗がん剤IC50値を示す図であり、(b)は、SKOV3-TRのドセタキセル抗がん剤IC50値を示す図である。
12-Wellで成長している二つのがん細胞株に102種のS-formsを低濃度(0.5μM)および高濃度(2μM)で4時間前処理(pretreatment)した後、パクリタキセルをSKOV3-TR細胞株には4μM、SKOV3細胞株には0.05μM処理した。細胞の形態学的分析のために、1day、2day、3day後にNone(何も処理しない群)、DMSO(パクリタキセル溶媒)、エタノール(ethanol,EtOH)(S-forms溶媒)、DMSO+EtOH、パクリタキセル(Paclitaxel)、パクリタキセル+エタノール(Pacli+EtOH)と102種のS-formsを処理した細胞を分析し、イメージをキャプチャーした。細胞の物質代謝分析のために、パクリタキセルとS-formsを併用処理してから3日後にWST-1分析を実施して、図3~図10に示し、同一実験群を活用してCrystal violet染色法を通じて細胞生存率を分析して、図13~図20に示した。
ドセタキセルは、12-wellで成長している二つの細胞株にS-formを(0.5μM)および高濃度(2μM)で4時間前処理(pretreatment)した後、ドセタキセルをSKOV3-TR細胞株には0.9μM、SKOV3細胞株には0.5nM処理した。細胞の形態学的分析のために、1day、2day、3day後にNone(何も処理しない群)、DMSO(ドセタキセル溶媒)、エタノール(ethanol,EtOH)(S-forms溶媒)、DMSO+EtOH、ドセタキセル(Doxetaxel)、ドセタキセル+エタノール(Doxetaxel+EtOH)とS-formsを処理した細胞を分析し、イメージをキャプチャーした。細胞の物質代謝分析のために、ドセタキセルとS-formsを併用処理してから3日後にWST-1分析を実施して、図21~図24に示し、同一実験群を活用してCrystal violet染色法を通じて細胞生存率を分析して、図27~図30に示した。

2.WST-1分析
細胞の物質代謝を分析するWST-1分析結果によれば、パクリタキセル処理によってSKOV3-TRおよびSKOV3で物質代謝の減少を誘発した。パクリタキセルのSKOV3-TRの102種に対するWST-1分析結果とSKOV3の102種に対するWST-1分析結果を図3~図10に示した。
すなわち、パクリタキセル処理群は、SKOV3-TRおよびSKOV3で細胞死滅の誘発および細胞成長の抑制などの現象によってNoneやDMSO処理群に比べて450nmの吸光度が減少した。S-formsをパクリタキセルと併用処理時に、耐性がん細胞株であるSKOV3-TRでは、パクリタキセルの抗がん効能を増強してパクリタキセル+エタノール(Pacli+EtOH)処理群に比べてさらに低い吸光度を示したが、SKOV3では、パクリタキセルの抗がん効能を増強しなかった。また、SKOV3-TRでは、0.5μMのS-formsより2μMのS-formsにおいてパクリタキセルによる物質代謝減少をさらに促進することから見て、用量-反応関係(dose-response relationship)が成立することが分かった。比較例であるパクリタキセル+エタノール(Pacli+EtOH)処理群の吸光度に基準線を入れた。パクリタキセル抗がん増強効能は、パクリタキセル+エタノール処理群(Pacli+EtOH)に比べてパクリタキセル+S-forms処理時にWST-1分析で吸光度(OD Value at 450nm)が有意に減少した。
細胞の物質代謝を分析するWST-1分析結果によれば、ドセタキセル処理によってSKOV3-TRおよびSKOV3で物質代謝の減少を誘発した。ドセタキセルのSKOV3-TRのS-Formに対するWST-1分析結果とSKOV3のS-Formに対するWST-1分析結果を図21~図24に示した。
すなわち、ドセタキセル処理群は、SKOV3-TRおよびSKOV3で細胞死滅の誘発および細胞成長の抑制などの現象によってNoneやDMSO処理群に比べて450nmの吸光度が減少した。S-formsをドセタキセルと併用処理時に耐性がん細胞株であるSKOV3-TRでは、ドセタキセルの抗がん効能を増強して、ドセタキセル+エタノール(Doxetaxel+EtOH)処理群に比べてさらに低い吸光度を示したが、SKOV3では、ドセタキセルの抗がん効能を増強しなかった。また、SKOV3-TRでは、0.5μMのS-formsより2μMのS-formsにおいてドセタキセルによる物質代謝減少をさらに促進することから見て、用量-反応関係(dose-response relationship)が成立することが分かった。比較例であるドセタキセル+エタノール(Doxetaxel +EtOH)処理群の吸光度に基準線を入れた。ドセタキセル抗がん増強効能は、ドセタキセル+エタノール処理群(Doxetaxel +EtOH)に比べてドセタキセル+S-forms処理時にWST-1分析で吸光度(OD Value at 450nm)が有意に減少した。
3.細胞生存率および細胞株の分析
細胞生存率および細胞数を分析するCrystal violet染色によれば、パクリタキセル処理によってSKOV3-TRおよびSKOV3で細胞生存率減少を通した細胞数減少を誘発した。代表的にパクリタキセルとドセタキセル抗がん増強効能を示したS101とS105をSKOV3-TRおよびSKOV3細胞株でパクリタキセルとドセタキセルを3日間単独処理したり、パクリタキセルまたはドセタキセルと3日間併用処理した後、イメージをキャプチャーした。パクリタキセルに対するSKOV3-TRのCrystal violet染色結果を図11に示し、SKOV3のCrystal violet染色結果を図12に示した。ドセタキセルに対するSKOV3-TRのCrystal violet染色結果を図25に示し、SKOV3のCrystal violet染色結果を図26に示した。

4.抗がん効能の測定
1)細胞数変化の分析(In vitro)
幹細胞性がん細胞であるselected-MDA-MB231 cellとselected-MCF7に対してER stressがないG(+):glucoseが存在するときとER stressがあるG(-):glucoseが存在しないときのS101化合物100、50、10nMを処理後に細胞数の変化を図31に示し、Crystal violetで染色したイメージをキャプチャーした結果を図32に示した。また、glucose deprivation後、S101のがん細胞死滅効果をTUNEL assayで測定して図33に示した。
2)Mouse in vivoモデルの検証
Cancer stem cellで証明されたselected-MDA-MB231 cellをin vitroで培養してharvest後、mouse in vivoモデルを構築して抗がん効果を検証した。Control groupとsevere ER stressを誘発し、抗がん剤の代用として2-deoxy-d-glucoseを使用したものを比較例1(2DG)とした。S101 2mgを単独使用したものを実施例2(S101単独)とし、2-deoxy-d-glucose 10mgとS101 2mgを使用したものを実施例3(S101+2DG、2mg:10mg)として検証した。tumor体積の変化結果を図34に示した。Dissected tumorの重さとサイズを各group間に比較した結果をそれぞれ図35の(a)および(b)に示した。selected-MDA-MB231 cell xenograft modelでS101と2DG併用抗がん効果の測定時に各group間のbody wight変化量を測定して図36に示した。
S101、2DGの単独処理および併用処理時にcytotoxicity程度を読み取るために、各groupからliverを摘出してparaffin embedded tissue blocksを製作した。その後、切片してslideを製作した後、H&E stainingを実行して、liver tissue degradationの有無を病理的に読み取って、cytotoxicity程度を類推した。図37は、Magnification x 400で示す図である。
