Description
「薬物」という用語は、その最も広い意味で、小分子、ウイルス粒子、ポリペプチド、ポリヌクレオチド、ポリペプチド、ポリペプチドと脂質を含む組成物、及びポリヌクレオチドと脂質を含む組成物を含む。薬物は、鎮痛剤、麻酔剤、抗炎症剤、駆虫剤、抗不整脈剤、抗喘息剤、抗生物質、抗がん剤、抗凝固剤、抗うつ剤、抗糖尿病薬、抗てんかん薬、抗ヒスタミン薬、鎮咳薬、降圧薬、抗ムスカリン薬、抗マイコバクテリア薬、抗腫瘍薬、抗酸化剤、解熱剤、免疫抑制剤、免疫刺激剤、抗甲状腺薬、抗ウイルス薬、抗不安鎮静薬、催眠薬、神経遮断薬、収斂剤、静菌剤、ベータアドレナリン受容体遮断薬、血液製剤、代用血液、気管支拡張薬、緩衝剤、心臓変力剤、化学療法剤、造影剤、コルチコステロイド、咳抑制薬、去痰薬、粘液溶解薬、利尿薬、ドーパミン作動薬、抗パーキンソン病薬、フリーラジカル捕捉剤、成長因子、止血剤、免疫剤、脂質調節剤、筋弛緩剤、タンパク質、ペプチド、ポリペプチド、副交感神経興奮薬、副甲状腺カルシトニン、ビスホスホネート、プロスタグランジン、放射性医薬品、ホルモン、性ホルモン、抗アレルギー剤、食欲刺激剤、食欲抑制剤、ステロイド、交感神経刺激薬、甲状腺剤、ワクチン、血管拡張剤、及びキサンチンからなる群より選択することができる。
「金属酸化物材料」という用語は、その最も広い意味で、金属と見なされる元素と酸素をベースとした酸化剤との反応から形成されるすべての材料を含む。例示的な金属酸化物材料には、酸化アルミニウム、酸化チタン、酸化鉄、酸化ガリウム、酸化マグネシウム、酸化亜鉛、酸化ニオブ、酸化ハフニウム、酸化タンタル、酸化ランタン、及び二酸化ジルコニウムが含まれるが、これらに限定されない。例示的な酸化剤には、水、オゾン、及び無機過酸化物が含まれるが、これらに限定されない。
原子層堆積は、元素又は化合物の自己制御的単層を順次追加することによって原子又は分子単層のレベルに制御された厚さ及び均一性を有する膜の堆積を可能にする、薄膜堆積技術である。自己制御とは、一度に単一の原子層のみが形成されることを意味し、表面を再生してさらなる堆積を可能にするには、後続のプロセスステップが必要である。
化学気相堆積は、元素又は化合物が気相中又は表面上での化学反応によって表面に堆積される薄膜堆積技術である。それは、堆積が自己制御されない、すなわち、化学物質が供給されている限り、膜は成長し続けるという点において、原子層堆積とは異なっている。それは、化学反応の結果、前駆体核種とは化学的に異なる堆積膜が生成されるという点で、物理気相堆積とは異なっている。
その最も広い意味での「リアクタシステム」という用語は、ALD又はALD/CVDの混合又はCVDを実行するために使用することができるすべてのシステムを含む。例示的なリアクタシステムが図1に示されており、以下においてさらに説明される。
最初に、粒子はリアクタシステム10のドラム40にロードされる。粒子は、薬物、例えば上述の薬物の1つを含むソリッドコアを有することができる。アクセスポートが密封されると、コントローラ60は、レシピに従ってリアクタシステム10を動作させ、粒子上に薄膜金属酸化物層を形成する。
i)ガス分配システム30は、第1の指定された圧力が達成されるまで、第1の反応ガス、例えばTMAを、ガス源32aからチャンバ20に流入させるように動作させる。特定の圧力は、反応ガスの飽和圧力の半分に対し0.1Torrとすることができる。
ii)第1の反応物質の流れが停止され、例えば、コントローラのタイマーで測定して、指定された遅延時間の経過が許される。これにより、第1の反応物質がドラム40内の粒子床を通って流れ、ドラム40内の粒子50の表面と反応することが可能になる。
iii)真空ポンプ50は、例えば1Torr未満、例えば1~100mTorr、例えば50mTorrまで圧力が下がるように、チャンバ20を排気する。
iv)ガス分配システム30は、第2の指定された圧力が達成されるまで、不活性ガス、例えばN2を、ガス源32cからチャンバ20に流入させるように動作させる。第2の指定された圧力は1~100Torrでありうる。
v)不活性ガスの流れが停止され、例えば、コントローラ内のタイマーで測定して、指定された遅延時間の経過が許される。これにより、不活性ガスがドラム40の細孔を通って流れ、粒子50を通じて拡散して、反応ガスと任意の蒸気副生成物を排除することができる。
vi)真空ポンプ50は、例えば1Torr未満、例えば1~500mTorr、例えば50mTorrまで圧力が下がるように、チャンバ20を排気する。
vii)ガス分配システム30は、第3の指定された圧力が達成されるまで、第2の反応ガス、例えばH2Oを、ガス源32aからチャンバ20に流入させるように動作させる。第3の圧力は、反応ガスの飽和圧力の半分に対し0.1Torrとすることができる。
viii)第2の反応物質の流れが停止され、例えば、コントローラのタイマーで測定して、指定された遅延時間の経過が許される。これにより、第2の反応物質がドラム40内の細孔を通って流れ、ドラム40内の粒子50の表面と反応することが可能になる。
ix)真空ポンプ50は、例えば1Torr未満、例えば1~500mTorr、例えば50mTorrまで圧力が下がるように、チャンバ20を排気する。
1つ以上の金属酸化物材料によって囲まれた薬物含有コアを含む医薬組成物についての2つの例示的な方法が提供される。第1の例示的な方法は、次の一連のステップを含む:(a)薬物を含む粒子をリアクタにロードすること、(b)蒸気又はガス状の金属前駆体をリアクタ内の基質に塗布すること、(c)不活性ガスを使用してリアクタの1つ以上のポンプパージサイクルを実行すること、(d)蒸気又はガス状の酸化剤をリアクタ内の基質に塗布すること、及び(e)不活性ガスを使用してリアクタの1つ以上のポンプパージサイクルを実行すること。本方法を実行している間、粒子の温度は35℃を超えない。
薬学的に許容される添加物には、次のものが含まれるが、これらに限定されない:
(1)次を含む、界面活性剤及びポリマー:ポリエチレングリコール(PEG)、ポリビニルピロリドン(PVP)、ラウリル硫酸ナトリウム、ポリビニルアルコール、クロスポビドン、ポリビニルピロリドン-ポリビニルアクリレート共重合体、セルロース誘導体、ヒドロキシプロピルメチルセルロース、ヒドロキシプロピルセルロース、カルボキシメチルエチルセルロース、ヒドロキシプロピルメチルセルロースフタレート、ポリアクリレート及びポリメタクリレート、尿素、糖類、ポリオール類、カルボマー及びそれらのポリマー、乳化剤、シュガーガム、デンプン、有機酸及びそれらの塩、ビニルピロリドン及び酢酸ビニル;
(2)結合剤、例えば、セルロース、架橋ポリビニルピロリドン、微結晶セルロースなど;
(3)充填剤、例えば、ラクトース一水和物、無水ラクトース、微結晶セルロース、及びさまざまなデンプンなど;
(4)潤滑剤、例えば、コロイド状二酸化ケイ素、タルク、ステアリン酸、ステアリン酸マグネシウム、ステアリン酸カルシウム、シリカゲルを含む、圧縮される粉末の流動性に作用する薬剤など;
(5)甘味料、例えば、スクロース、キシリトール、サッカリンナトリウム、シクラメート、アスパルテーム、及びアセスルファムKなどの任意の天然又は人工甘味料;
(6)香味剤;
(7)防腐剤、例えば、ソルビン酸カリウム、メチルパラベン、プロピルパラベン、安息香酸及びその塩、ブチルパラベンなどのパラヒドロキシ安息香酸の他のエステル類、エチル又はベンジルアルコールなどのアルコール類、フェノールなどのフェノール化合物、若しくは塩化ベンザルコニウムなどの4級化合物など;
(8)バッファー;

この実施例では、金属酸化物でカプセル化した薬物を調製するために開示された方法の1つが実行され、データが提示される。この実施例では、蒸気又はガス状の金属前駆体はトリメチルアルミニウム(TMA)であり、TMAが粒子上又はコーティングされた粒子の表面上の露出したヒドロキシル基と反応した後に副生成物のガス状メタンが形成され、酸化剤は水蒸気である。
簡単に言えば、この方法は次の一連のステップで構成されている:
(a)薬物を含む粒子をリアクタにロードすること;
(b)リアクタ圧力を1Torr未満に下げること;
(c)リアクタ内の水蒸気のレベルを監視するために残留ガス分析(RGA)を実行することにより、リアクタの内容物が所望の含水量になるまでリアクタの内容物を攪拌すること;
(d)蒸気又はガス状のTMAを加えることによってリアクタを少なくとも1Torrに加圧すること;
(e)リアクタ圧力を安定させること;
(f)リアクタの内容物を攪拌すること;
(g)ガス状メタン及び未反応のTMAを含めた蒸気又はガス状の内容物のサブセットをポンプで排出すること、及びリアクタ内のガス状メタン及び未反応のTMAのレベルを監視するためにRGAを実行することによってポンプによる排出をいつ停止するかを決定すること;
(h)窒素ガスを使用してリアクタに一連のポンプパージサイクルを実行すること;
(i)水蒸気を加えることによってリアクタを少なくとも1Torrに加圧すること;
(j)リアクタ圧力を安定させること;
(k)リアクタの内容物を攪拌すること;

図3は、この方法の1サイクルについて、ステップ(d)、(h)、(i)、及び(m)の間に測定する代表的な残留ガス分析トレースを示している。この方法は、1サイクルあたり2から4オングストロームの金属酸化物コーティングの成長率を再現可能に示す。対照的に、成長をALDに制限する別の方法では、1サイクルあたり平均1オングストロームの平均成長しか示さなかった。特定の理論に縛られはしないが、この方法で観察された成長率を所与とすれば、成長はALDとCVDとの組合せによって媒介されうる。
金属酸化物コーティングによる小分子のカプセル化が構造又は溶解プロファイルを変更したかどうかを評価するために、小分子クラリスロマイシン、クラリスロマイシン・カルボマー複合体、パゾパニブHCl.パルボシクリブ、及びアモキシシリン・クラブラン酸カリウムを金属酸化物コーティングでカプセル化し、得られた粒子を化学分析に供して、金属酸化物コーティングが構造又は溶解プロファイルを変更したかどうかを決定した。粉末形態の小分子を、以下の表に示される以下の変更を加えた本開示で提供される方法によってカプセル化した。
高速液体クロマトグラフィー
アセトニトリルと水の50:50体積:体積の混合液に被分析物を溶解することによって1mg/mLの濃度の試料を調製し、0.45μmフィルターで濾過した。移動相、カラム、及びオーブン温度を含めた分析の正確な条件は、検討する被分析物によって異なる。分析の典型的な例は、アセトニトリル(90:10v/v)と混合した0.05M pH4のリン酸バッファー、Agilent Pursuit XRs 3C-18 3μmカラム、37℃のオーブン温度、流量0.9mL/分、注入量35μL、実行時間5分、214nmのUV検出器で構成される移動相を使用する。
試料は、18:82v/vの比の水-アセトニトリル混合液に溶解することによって調製した。次に、試料をシアノ-4-ヒドロキシケイ皮酸マトリックスと混合し、MALDIチップにロードした。MSデータ取得はリフレクトロンポジティブモードで実行した。
結果が図4~14に示されている。コーティングされていない対照と比較して、酸化チタン又は酸化アルミニウムのいずれかでコーティングされた小分子クラリスロマイシン及びクラリスロマイシン・カルボマー複合体は、HPLC分析で検出して、構造にほとんど又はまったく変化を示さなかった。コーティングされていない対照と比較して、酸化チタン又は酸化アルミニウムのいずれかでコーティングされた小分子パゾパニブHCl.及びパルボシクリブは、MALDI-MS分析で検出して、構造にほとんど又はまったく変化を示さなかった。コーティングされていない対照と比較して、酸化チタン又は酸化アルミニウムのいずれかでコーティングされた小分子クラリスロマイシン、クラリスロマイシン・カルボマー複合体、及びパルボシクリブ、並びに酸化アルミニウムでコーティングされたパゾパニブHClは、溶解プロファイルにほとんど又はまったく変化を示さなかった。対照的に、酸化チタンでコーティングされた小分子パゾパニブHCl及び酸化チタン又は酸化アルミニウムのいずれかでコーティングされたアモキシシリン・クラブラン酸カリウムは、コーティングされていない対照と比較して、変化した溶解プロファイルを示した。酸化チタンでコーティングされたパゾパニブHClは、コーティングされていない対照と比較して、変化した溶解プロファイル(同様の初期放出及びその後の遅延放出)を示す。酸化チタンでコーティングされたアモキシシリン・クラブラン酸カリウムは、コーティングされていない対照と比較して、変化した溶解プロファイル(初期放出が遅く、その後、30分、飽和せずに速やかに放出)を示す。最後に、酸化アルミニウムでコーティングされたアモキシシリン・クラブラン酸カリウムは、コーティングされていない対照と比較して、変化した溶解プロファイル(初期放出が遅く、その後、速やかに放出)を示す。
この実施例は、酸化チタン又は酸化アルミニウムのいずれかによる5つの小分子のカプセル化が、小分子構造に大幅な減少を与えないが、小分子及び/又は金属酸化物コーティングに応じて、溶解プロファイルにほとんど又はまったく変化をもたらさないか、あるいは有意な変化を与えることを実証している。特定の理論に拘束されるわけではないが、出願人は、全体として、1)溶解プロファイルへの影響が(プロセス条件、API、及び溶解条件に基づいて)同じコーティング材料で大きく変動する可能性があること、及び2)同じ基本材料で作られたコーティングは、溶解プロファイルに本質的に影響を与えない可能性があることは、驚くべきことであるということに注目している。これは、同じ基本材料を使用して、特定の用途に合わせた溶解プロファイルを生成することができる多目的なプロセスを意味する。出願人は、本明細書に記載されるように、熟練した当業者がさまざまな方法又はパラメータを試験して、構造が大幅に縮小せず、かつ、コーティングされていない小分子と比較して溶解プロファイルの変化を示す又は示さない、選択した金属コーティングでコーティングされた小分子を生成することができると結論付けている。
金属酸化物コーティングによる小分子のカプセル化が構造又は溶解プロファイルを変更したかどうかを評価するために、小分子インドメタシンを金属酸化物コーティングでカプセル化し、得られた粒子を化学分析に供して、金属酸化物コーティングが非晶質インドメタシンの結晶化度を変化させたかどうかを決定した。粉末形態の小分子を、以下の表に示される以下の変更を加えた本開示で提供される方法によってカプセル化した。
非晶質インドメタシンの結晶化度の評価
標準的なインドメタシン試料を凍結乾燥して非晶質インドメタシンを調製し、水分に曝す前後の結晶含有量を示差走査熱量測定(DSC)で測定した。結晶化ピーク下の面積を使用して、非晶質材料の結晶化の比熱を決定した。部分的に結晶性の材料の結晶化度パーセントは、結晶化熱を完全に非晶質の材料の結晶化熱で割り、1からその値を差し引いて100を掛けることによって決定した。
コーティングされていない対照と比較して、酸化チタン又は酸化アルミニウムのいずれかでコーティングされた小分子インドメタシンは、処理されたままの状態で及び90%相対湿度(RH)への曝露後に、非晶質状態から結晶状態への変換の低下を示した(図15)。
本実施例は、提供された方法が、具体的には、コーティングされた粒子中の薬物が、処理されたままの状態で及び例えば90%RHなどのストレスへの曝露後に、非晶質状態から結晶状態への変換の低下を示すように、より安定な金属酸化物材料でコーティングされた小分子を生成することができるという指針を提供する。
提供された方法が、小分子に均一でコンフォーマルな薄い金属酸化物コーティングを可能にするかどうかを評価するため、本開示の方法によってアセトアミノフェンを金属酸化物材料でコーティングし、原子層顕微鏡法及びXPS分析によって分析した。粉末形態の小分子を、本開示に提供される方法でカプセル化した。
透過電子顕微鏡法
TEM用の試料を、FEI Strata 400 Dual Beam FIB/SEM上でインシトゥFIBリフトアウト技術を使用して調製した。粉砕する前に、試料を保護カーボン及びe-Pt/I-Ptでキャップした。TEMラメラの厚さは約100nmであった。試料を、明視野(BF)TEMモード及び高解像度((HR)TEMモードで200kVで動作する、FEI Tecnai TF-20 FEG / TEMで画像化した。エネルギー分散分光法(EDS)を使用して、画像の定性元素マップを取得した。
X線光電子分光法(XPS)を試料に行い、コーティングの前後の表面化学の詳細を取得した。粉末の試料を接着性の基板に取り付け、機器にロードした。軟X線(1486eV)を使用して試料を励起し、X線の浸透深さは5nmであり、スポットサイズは200μmであった。
コーティングされたアセトアミノフェン粒子の集束イオンビーム(FIB)ミリングによって調製された断面の直接TEM画像化は、粒子上の位置にかかわらず、ナノメートルスケールで酸化アルミニウムによる薬物粒子の均一かつコンフォーマルなコーティングを示している(図16)。エネルギー分散分光法(EDS)は、コーティングが実質的にアルミニウム及び酸素からなることを定性的に証明した(図17)。
本実施例は、提供された方法がナノメートルレベルの精度で小分子上の均一かつコンフォーマルな薄い金属酸化物コーティングを可能にするというガイダンスを提供する。
金属酸化物コーティングによる凍結乾燥mAbのカプセル化が、mAbの構造又は安定性を変化させるかどうかを評価するため、mAbトラスツズマブ(ハーセプチン(登録商標))及びベバシズマブ(アバスチン(登録商標))を金属酸化物コーティングでカプセル化し、得られた粒子を生化学的及び化学的分析に供して、金属酸化物コーティングがmAbの構造、安定性、又は標的ポリペプチドに結合する能力を変化させたかどうかを決定した。2つのmAbsを、以下の変更を伴う本開示で提供される方法によってカプセル化した:(1)ハーセプチン(登録商標)では、コーティングされた粒子を薬学的に許容される希釈剤又は担体と混合する前の連続サイクルを99回行い、蒸気又はガス状の金属前駆体は酸化アルミニウム(Al2O3)であった;(2)アバスチン(登録商標)では、コーティングされた粒子を薬学的に許容される希釈剤又は担体と混合する前の連続サイクルを49回行い、蒸気又はガス状の金属前駆体は酸化チタン(TiO2)であった。
質量分析と組み合わせた液体クロマトグラフィー
移動相A(0.1%(v/v)FA)及び10%移動相B(アセトニトリル中、0.1%(v/v)FA)を用いた、Agilent 6230エレクトロスプレーイオン化飛行時間型質量分析計(ESITOF-MS)機器に接続されたAgilent 1260 Infinity Bio-inert Quaternary LCシステムを使用して、AdvanceBio RP mab C4(Agilent Technologies)カラムで逆相クロマトグラフィー(RPC)を行った。試料を、10kDaのMWCOセントリコン(Pall Corporation)を介してバッファー交換し、カラムにロードし、0.5ml/分の流量で10%-65%Bの直線勾配を使用して分離した。MSスペクトルは正イオンモードで較正し、TICは1,000~7000m/zで記録した。キャピラリーガスの温度/電圧(Vcap)をそれぞれ350℃/5,500Vに設定し、フラグメンター電圧(Vfrag)は400Vであった。Agilent MassHunter定性分析及びBioConfirmソフトウェアの一部として最大エントロピー(MaxEnt)アルゴリズムを使用して、MSスペクトルをデコンボリューションした。
移動相A(0.1%(v/v)TFA)及び移動相B(アセトニトリル中、0.1%(v/v)TFA)を用いて、Agilent 6230 ESI-TOF-MS機器に接続されたAgilent 1260 Infinity Bio-inert Quaternary LCシステムを使用して、55℃で動作するAdvanceBioペプチドマッピングC18(Agilent Technologies)カラムで、逆相クロマトグラフィー(RPC)を行った。消化された試料をカラムに注入し、0.3ml/分の流速で5%~65%Bの直線勾配を使用して分離した。MSスペクトルは正イオンモードで較正し、TICは100~3200m/zで記録した。キャピラリーガスの温度/Vcapをそれぞれ300℃/4500Vに設定し、Vfragは300Vであった。Agilent MassHunter定性分析及びBioConfirmソフトウェアのタンパク質分子特徴抽出(MFE)アルゴリズムを使用してMSスペクトルを分析し、可能性のあるペプチドのリストを取得し、in-silicoで消化させたmAbペプチドと照合して、配列包括度を取得した。
FTIRスペクトルは、減衰全反射(ATR)法を使用して室温で記録した。0.5mg/mlのmAbのFTIR吸光度スペクトルを、500~4000cm-1の範囲で収集した。元のスペクトルの11ポイントのサビツキー・ゴーレイ平滑化を適用することによって、二次微分スペクトルを得た。1600サビツキー・ゴーレイ平滑で700cm-1の範囲の二次微分スペクトルを、レーベンバーグ・マーカートのアルゴリズムを使用した曲線近似法によってデコンボリューションし、α-ヘリックス(1660~1654cm-1)、β-シート(1637~1614cm-1)、ターン(1678~1670cm-1)、ランダムコイル(1648~1638cm-1)及びβ-逆平行(1691~1680cm-1)に対応するピークを調整し、ガウス関数を用いて面積を測定した。次に、所与の配座に割り当てられたすべての構成成分のバンドの面積を合計し、総面積で割った。
50nm/分の走査速度で0.1cmの経路長の石英セルを使用して、5nmのスペクトルバンド幅で、25℃で200~250nmの範囲で、遠紫外線CDスペクトルを記録した。試料濃度を0.2mg/mlに維持し、3つのスペクトルを走査し、平均化し、最後にバッファーのベースラインを差し引いた後にプロットした。平均残基楕円率(MRE、deg cm2/dmole)を計算した。
タンパク質溶液(0.5mg/ml)を295nmで励起することによって、固有の蛍光を測定した(トリプトファンの励起のみ)。発光スペクトルを300~450nmの範囲で記録した。ANS(8-アニリノナフタレン-1-スルホン酸)染料を使用した蛍光分光光度計で、380nmの励起と400~600nmの発光を用いて、試料の外部蛍光強度(0.5mg/ml)を記録した。すべての測定は3回繰り返して行った。各スペクトルは3回の走査の平均を表している。
サイズ排除クロマトグラフィーは、300mMのNaCl及びpH6.8で0.05%のNaN3のバッファーを用いたDionex Ultimate 3000 UHPLCシステム(Thermo Scientific)を使用して、Superdex200カラムで行った。検出は、280nmでのUV吸光度を監視することによって行った。Chromeleonソフトウェアを使用して、ピーク積分及びピーク面積を決定した。
イオン交換クロマトグラフィーは、Agilent Bio MAb NP5カラムと、300mMのNaCl及びpH6.8で0.05%のNaN3のバッファーとを用いた、Dionex Ultimate 3000 RSLCシステム(Thermo Scientific)を使用して行った。検出は、280nmでのUV吸光度を監視することによって行った。
さまざまなmAb試料のヒトFcRn受容体への結合速度論的相互作用は、Biacore X100(商標)(GE Healthcare)にHBS-EPバッファー(GE Healthcare Life Sciences)を用いて表面プラズモン共鳴を使用して測定した。組換えヒトFcRn抗体を固定化し、試料を一連の濃度で注入した。速度定数は、BIA Evaluation2.0.1ソフトウェアを使用した1:1フィットモデルを使用してセンサーグラムから計算した。

金属コーティングでカプセル化されたmAbの質量及び配列同一性を確認するために、質量分析と組み合わせた液体クロマトグラフィーを行った。アバスチン(登録商標)の結果が図18~20に示されている。データは、酸化チタンコーティングが、アバスチン(登録商標)のインタクトなタンパク質質量にほとんど又はまったく影響を及ぼさないことを示唆している。1つの変更を除き、コーティングされたアバスチン(登録商標)の予測される変更は、コーティングされていないアバスチン(登録商標)のものと一致した。ハーセプチン(登録商標)の結果が図21~23に示されている。データは、酸化アルミニウムコーティングが、コーティングされていないアバスチン(登録商標)と比較して、ハーセプチン(登録商標)のインタクトなタンパク質質量にほとんど又はまったく影響を及ぼさないことを示唆している。
この実施例は、金属コーティングによる2つの異なる凍結乾燥mAbのカプセル化が、mAbの構造、安定性、又は標的ポリペプチド(例えば、ヒトFcRn)に結合する能力に有意な低下をもたらさないことを実証している。注目すべきことに、酸化チタンでコーティングされたハーセプチン(登録商標)は、コーティングされていないアバスチン(登録商標)と比較して、標的のヒトFcRn受容体に対して4倍強力な結合(KDの低減によって反映される)を示した。また、注目すべきは、酸化チタンでコーティングされたアバスチン(登録商標)が、コーティングされていないアバスチン(登録商標)と比較して、SECで決定して、80℃で10日間、凝集体の蓄積率の低下を示すことである。出願人は、当業者が本明細書に記載のさまざまな方法又はパラメータを試験して、mAb構造、安定性、又は標的ポリペプチドに結合する能力が大幅に低下しない、選択した金属コーティングでコーティングされた凍結乾燥mAbを生成することができると結論付けている。
Claims (17)
- 1つ以上の金属酸化物材料によって囲まれた薬物含有コアを含む医薬組成物を調製する方法であって、
(a)薬物を含む、体積平均に基づいて0.1μmから1000μmの間のメジアン粒径を有する粒子をリアクタにロードすること;
(b)蒸気又はガス状の金属前駆体をリアクタ内の粒子に塗布すること;
(c)不活性ガスを使用してリアクタの1つ以上のポンプパージサイクルを実行すること;
(d)蒸気又はガス状の水を酸化剤としてリアクタ内の粒子に塗布すること;
(e)不活性ガスを使用してリアクタの1つ以上のポンプパージサイクルを実行すること;及び
(f)ステップ(b)~(e)を1回又は複数回繰り返し、コアを囲む1つ以上の金属酸化物材料の全体の厚さを増加させること
の一連のステップを含み、
