Description
る受容体を発現する。その受容体はヒト阻害KIR受容体に類似している。抗体断片でLy49受容体をブロックすることによって、NK細胞は細胞障害性がより強くなり、インビトロ及び生体内でマウス白血病細胞を死滅させることができることが示されている(Koh et al. 2001)。
患者への細胞の移入直後の免疫学的根絶若しくは場合によっては、対宿主性移植片疾患(GVHD)の発症を回避するためにMHCがマッチしたT細胞が用いられなければならないということである。加えて、うまく移入されたT細胞は循環中で長期にわたって生存することが多く、治療から生じる持続的副作用を制御することを困難にする。

CRISPR/Cas9
細胞を以下の通り調製し、阻害受容体機能を除去した。gRNAコンストラクトを、NK細胞のヒトゲノムにおける「古典的」阻害受容体LIR2及び「チェックポイント」阻害受容体CTLA4をコードする遺伝子を標的とするように設計及び調製した。次いで、CRISPR/Cas9ゲノム編集を利用して、LIR2及びCTLA4標的遺伝子をノックアウトした。
NK細胞のチェックポイント阻害受容体のノックアウトのために使用した別のプロトコールは、Cas9 RNPトランスフェクションのプロトコールであった。このプロトコールを使用することの利点は、CRISPR/Cas9プロトコールのDNAプラスミドの使用と比較して著しく低い毒性で、同程度のトランスフェクション効率が達成できるということであった。
(1)必要な合成crRNA及びtRNA(ダーマコン社から購入)の20μM溶液を調製した。
(2)4μlのcrRNA(20μM)及び4μlのtRNA(20μM)を混合した。
(3)次いで、混合物を2μlのCas9タンパク質(5μg/μl)に加えた。
(4)構成成分のすべてを混合し、室温で10分間インキュベーションした。

NK細胞のチェックポイント阻害受容体のノックアウトのために使用した別のプロトコールは、XTN TALENトランスフェクションのプロトコールであった。このプロトコールを使用することの利点は、野生型CRISPRと比較して、特に高いレベルの特異性が達成できるということであった。
KHYG-1細胞を、トランスフェクション効率、シングルセルクローニング効率、及び核型/コピー数を含む特定の属性について分析した。次いで、細胞を供給社の推奨事項に従って培養した。
細胞にステップ1のヌクレアーゼをトランスフェクションした。高いレベルの切断を得るためにこのステップを最高3回繰り返し、各トランスフェクションの前に培養物を分け、中間培養物を維持した。
一旦細胞が96ウェルプレートでコンフルエントになると、培養物をまとめ、全く同じ3枚の96ウェルプレートに分けた。1枚のプレートはバックアップとして凍らせ、1枚のプレートは再播種してクローンの増殖を続け、最後のプレートは遺伝子型確認のために使用した。
細胞は無菌条件下で貯蔵した。
KHYG-1細胞のCD96のsiRNAノックダウンをエレクトロポレーションによって行った。細胞系の用途に適し、分裂細胞及び分裂していない細胞をうまくトランスフェクションすることができ、最高90%のトランスフェクション効率が達成されることから、ロンザ社製のヌクレオフェクションキットTをAmaxa Nucleofector IIとともに使用した。
CD96)については165μlのRNAseを含まない水に再懸濁した。チューブを
90℃に1分間加熱した後、37℃で60分間インキュベーションした。その後、siRNAストックを-20℃で必要になるまで保存した。
CD96ノックダウンを有する/有しないKHYG-1細胞を、異なるエフェクター:標的(E:T)比でK562細胞と共培養した。
CD96ノックダウンKHYG-1細胞と対照群(n=3)との違いを比較するために対応のあるt検定を用いた。
NK-92細胞におけるCD328のsiRNAによって媒介されるノックダウン
材料、試薬、及び機器
対照siRNA(カタログ番号:sc-37007)及びCD328 siRNA(カタログ番号:sc-106757)は、サンタクルーズバイオテクノロジー社から購入した。NK-92細胞において高い細胞生存率(>75%)とともに最高90%のトランスフェクション効率を得るために、Nucleofector(商標)Device(Nucleofector II、ロンザ社)を用いてロンザ社のNucleofector(商標)キットTを使用した。10%FBS、2mM L-グルタミンを含み、抗生物質を含まないRPMI-1640をヌクレオフェクション後の培養のために用いた。マウス抗ヒトCD328-APC(カタログ番号:339206)はバイオレジェンド社から購入した。
10μMのsiRNAストック溶液を作製する。
・ 凍結乾燥二本鎖siRNAを66μlのRNAseを含まない水(siRNA希釈バッファー:sc-29527)に再懸濁してFITC対照/対照siRNAとし、標的遺伝子siRNA(siRNA CD328)はRNAseを含まない水330μlに再懸濁する。
・ チューブを90℃で1分間熱する。
・ 37℃で60分間インキュベーションする。
・ すぐに使用しない場合は、siRNAストックを-20℃で保存する。
・ 1つのヌクレオフェクション試料(100μl標準キュベット用)は以下を含む。
・ 細胞数:2×106細胞
・ siRNA:4μlの10μMストック
・ Nucleofector溶液:100μl
・ 必要な数の細胞を培養する。(継代数1又はヌクレオフェクションの2日前に細胞は対数増殖期になければならない)。
・ 各試料に対してsiRNAを調製する。
・ Nucleofector溶液(1試料あたり100μl)を予め室温に温める。
・ 血清及び補助剤を含んでいる培養培地のアリコートを50ml容チューブ中で予め37℃に温める。血清及び補助剤を含む、1.5mlの培養培地を入れることよって6ウェルプレートを調製し、加湿した37℃/5%CO2インキュベーター中でプレートを予めインキュベーションする。
・ 細胞培養物のアリコートをとり、細胞をカウントして細胞密度を決定する。
・ 必要な数の細胞を1500RPMで5分間遠心分離する。残留培地が細胞ペレットを覆わないように、完全に上清を捨てる。
・ 細胞ペレットを、室温のNucleofector溶液中に、2×106細胞/100μlの最終濃度に再懸濁する。細胞生存率及び遺伝子導入効率を低下させるので、細胞懸濁液を15~20分より長くNucleofector溶液中に保存することは避ける。
・ 100μlの細胞懸濁液をsiRNAと混ぜる。
・ 試料をamaxa認定キュベットに移す。試料がキュベットの底を覆うことを確認し、ピペットで取りながら気泡の発生を防ぐ。キュベットを青いキャップで閉じる。
・ 適切なNucleofectorプログラムを選択する(NK-92細胞についてはA-024)。キュベットをキュベットホルダーに挿入し(Nucleofector II:回転台を最終位置へ時計回りに回転し)、「x」ボタンを押してプログラムを開始させる。
・ 細胞への損傷を避けるために、(ディスプレイが「OK」を示して)プログラムが完了した直後に試料をキュベットから取り出す。500μlの予め温めた培養培地をキュベットに加えて、試料を準備した6ウェルプレートに移す。
・ 加湿した37℃/5%CO2インキュベーター中で細胞をインキュベーションする。16~24時間後にフローサイトメトリー分析及び細胞傷害性アッセイを行う。
材料、試薬、及び機器
蛍光増強リガンド(カタログ番号:AD0116)に基づくDELFIA EuTDA細胞傷害性キットはパーキンエルマー社から購入した。標的細胞K562を10%FBS、2mM L-グルタミン、及び抗生物質を含むRPMI-1640培地で培養した。96ウェルV底プレート(カタログ番号:83.3926)はザルスタット社から購入した。プレートを遠沈するために(プレートローターを備えた)エッペンドルフ遠心分離機5810Rを使用した。溶解したK562細胞によって生じる蛍光信号を測定するためにVARIOSKAN FLASH(ScanItソフトウェア2.4.3付属)を使用した
。
・ 標的K562細胞に蛍光増強リガンドDELFIA BATDA試薬をロードする。・ K562細胞を培地で洗浄し、培養培地で細胞数を1×106細胞/mLに調節する。2~4mLの細胞を5μlのBATDA試薬に加えて、37℃で10分間インキュベーションをする。
・ 1mM プロベネシド(シグマP8761)を含む培地を用いて3~5回、1500RPMで5分間遠心分離してロードしたK562細胞を洗浄した。
・ 最後の洗浄の後、細胞ペレットを培養培地に再懸濁し、約5×104細胞/mLに調節した。
・ バックグランド、自発的放出、及び最大放出を検出するためにウェルを準備した。
・ 100μLのロードした標的細胞(5,000個の細胞)をV底プレートにピペットで移す。
・ 100μLのさまざまな細胞濃度のエフェクター細胞(NK-92)を加える。エフェクター対標的の比は1:1~20:1の範囲である。
・ プレートを100×gのRCFで1分間遠沈する。
・ 加湿した5%CO2雰囲気で37℃にて2時間インキュベーションする。最大放出ウェルについては、培地を集める15分前に10μLの溶解バッファーを各ウェルに加える。
・ プレートを500×gで5分間遠沈する。
・ 20μLの上清を平底96ウェルプレートに移し、200μLの予め温めたユウロピウム溶液を加え、プレートシェーカーを用いて室温で15分間インキュベーションする。・ 蛍光をVARIOSKAN FLASHを用いて時間分解蛍光光度計で測定する。次式を用いて特異的放出を算出する。
上記の実施例において示されたように、チェックポイント阻害受容体機能は、さまざまな方法でノックダウン又はノックアウトすることができる。以下のプロトコールは、血液がん患者を治療するために開発された。
生体外で培養して増殖させる。よって、相異なるチェックポイント阻害受容体の改変を有するさまざまなNK細胞を、特異的ながんに対する細胞傷害性について検査することができる。このステップは、治療に最も適したNK細胞又はその誘導体を選択するために用いることができる。
トランスフェクション後にその生存率及びがん細胞を死滅させる能力を評価するために、KHYG-1細胞にTRAILとTRAILバリアントの両方をトランスフェクションした。
KHYG-1細胞のベースラインTRAIL(CD253)発現をフローサイトメトリー用いてアッセイした。
野生型TRAIL mRNA及びTRAILバリアント(D269H/195R)mRNAはTriLink BioTechnologies社によって合成された。分注して-80℃として保存した。マウス抗ヒトCD253-APC(バイオレジェンド社カタログ番号:308210)及びアイソタイプ対照(バイオレジェンド社カタログ番号:400122)並びにマウス抗ヒトCD107a-PE(eBioscience社カタログ番号:12-1079-42)及びアイソタイプ対照(eBioscience社カタ
ログ番号:12-4714)抗体を使用して細胞試料を染色し、BD FACS Canto IIフローサイトメーターで分析した。DNA染料SYTOX-グリーン(ライフテクノロジーズ社カタログ番号:S7020;DMSO中の5mM溶液)を用いた。KHYG-1細胞において高い細胞生存率とともに最高90%のトランスフェクション効率を得るために、Nucleofector(商標)Device(Nucleofector II、ロンザ社)を用いてロンザ社のNucleofector(商標)キットTを使用した。10%FBS、L-グルタミン(2mM)、及びIL-2(10ng/ml)を含み、抗生物質を含まないRPMI1640をヌクレオフェクション後の培養のために用いた。
マウス抗ヒトCD2-APC抗体(BDファーミンジェン社カタログ番号:560642)を使用した。アネキシンV-FITC抗体(ImmunoTools社カタログ番号:31490013)を使用した。DNA染料SYTOX-グリーン(ライフテクノロジーズ社カタログ番号:S7020)を使用した。24ウェル細胞培養プレート(ザルスタットAG社カタログ番号:83.3922)を使用した。骨髄性白血病細胞株K562、多発性骨髄腫細胞株RPMI8226及びMM1.Sを標的細胞として使用した。K562、RPMI8226、MM1.Sは、10%FBS、2mM L-グルタミン、及びペ
ニシリン(100U/mL)/ストレプトマイシン(100mg/mL)を含むRPMI1640培地で培養した。
実施例6で上に記載した通りに、KHYG-1細胞にTRAILバリアントをトランスフェクションした。以下のプロトコールは血液がん患者の治療用に開発された。
ボルテゾミブ(Bt)は、多発性骨髄腫(MM)の治療に有用なプロテアソーム阻害剤(化学療法様薬)である。ボルテゾミブは、MM細胞を含むいくつかの異なるタイプのがん細胞上のDR5発現を上方制御することが知られている。
ボルテゾミブはミレニアム・ファーマシューティカルズ社から購入した。マウス抗ヒトDR5-AF647(カタログ番号:565498)は、BDファーミンジェン社から購入した。染色した細胞試料はBD FACS Canto IIで分析した。
(2)MM細胞を1×106/mL、2mL/ウェルで6ウェルプレートに播種した。
(3)次いで、MM細胞を相異なる投与量のボルテゾミブで24時間処理した。
(4)次いで、ボルテゾミブ処理/未処理のMM細胞のDR5発現をフローサイトメトリーで分析した(図18)。
実施例6で上に記載した通りに、KHYG-1細胞にTRAILバリアント(TRAIL D269H/E195R)をトランスフェクションした。
(2)TRAILバリアントmRNAのエレクトロポレーションの6時間後に、KHY
G-1細胞をMM細胞と12ウェルプレートで培養した。洗浄後、細胞濃度を1×106/mLに調節した後、KHYG-1とMM1.S細胞を1:1の比で混合し、12時間培養した。
(3)KHYG-1細胞の細胞傷害性のフローサイトメトリー分析を実施した。共培養した細胞を集めて洗浄した後、Annexin V結合バッファーを用いて、CD2-APC抗体(5μL/試験)、Annexin V-FITC(5μL/試験)、及びSYTOX-グリーン(5μL/試験)で染色した。
(4)データをFlowJo 7.6.2ソフトウェアを使用してさらに解析した。CD2陰性集団はMM1.S細胞を表わす。KHYG-1細胞は強いCD2陽性である。最後に、CD2陰性集団のAnnexinV-FITC及びSYTOX-グリーン陽性細胞を分析した。
TRAILバリアント発現からNK細胞の細胞傷害性の増加が生じるという前述の実施例における決定的証拠にもかかわらず、我々は、細胞傷害性の増加が、がん細胞アポトーシスを誘導することから生じたのか(可能性が最も高い)、又はNK細胞が意図せず活性化されて、より強い細胞障害性表現型を呈し、よってパーフォリン分泌を介してがん細胞を死滅させることによるのかを確認したいと望んだ。
マウス抗ヒトパーフォリン-AF647(カタログ番号:563576)は、BD pharmingenから購入した。コンカナマイシンA(カタログ番号:SC-202111)は、サンタクルーズバイオテクノロジー社から購入した。染色した細胞試料は、BD FACS Canto IIを使用して分析した。
(2)KHYG-1細胞(エレクトロポレーション後の6時間、ペニシリン/ストレプトマイシンを含まないRPMI1640培地で培養)をさらに、100nM CMA又は等量の溶媒(DMSO)で2時間処理した。
(3)細胞を遠心分離(1500RPM×5分)によって集め(1×106細胞/試験)、上清を吸引した。
(4)細胞をPBS溶液中の4%パラホルムアルデヒドで室温にて15分間固定した。
(5)細胞を4mLのFACSバッファー(PBS、0.5~1%BSA、0.1%ア
ジ化ナトリウム)で2回洗浄した。
(6)細胞を1mLのPBS/0.1%サポニンバッファーで室温にて30分間透過処理した。
(7)細胞を4mLのPBS/0.1%サポニンバッファーで洗浄した。
(8)細胞を100μLのPBS/0.1%サポニンバッファーに再懸濁した後、5uLの抗体を各チューブに加えて、氷上で30分間インキュベーションした。
(9)細胞を1500RPMでの5分間遠心分離によって、PBS/0.1%サポニンバッファーで3回洗浄した。
(10)細胞を500uLの氷冷FACSバッファーに再懸濁して、分析まで一時的に暗所で氷上又は冷蔵庫内に4℃で置いた。
(11)細胞をフローサイトメーター(BD FACS Canto II)で分析した。データをFlowJo 7.6.2ソフトウェアを使用して処理した。
実施例6で上に記載した通り、KHYG-1細胞にTRAILバリアント(TRAIL
D269H/E195R)をトランスフェクションした。
(2)TRAIL mRNAのエレクトロポレーションの6時間後に、KHYG-1細胞を100mM CMA又は等量の溶媒で2時間処理した。
(3)KHYG-1細胞を遠心分離によってRPMI1640培地で洗浄し、IL-2を含むRPMI1640培地に再懸濁し、細胞濃度を1×106/mLに調節した。
(4)IL-2を含むRPMI1640培地にMM1.S細胞を再懸濁し、細胞濃度を1×106/mLに調節した。
(5)KHYG-1とMM1.S細胞を1:1の比で混合し、12時間共培養した。
(6)KHYG-1細胞の細胞傷害性のフローサイトメトリー分析を実施した。共培養した細胞を洗浄し、CD2-APC抗体(5uL/試験)で染色した。
(7)洗浄後、Annexin V結合バッファーを用いてAnnexinV-FITC(5uL/試験)及びSYTOX-グリーン(5uL/試験)でさらなるなる染色を行った。
(8)さらに、データをFlowJo 7.6.2ソフトウェアを使用して解析した。CD2陰性集団はMM1.S細胞を表す。KHYG-1細胞は強いCD2陽性である。次いで、AnnexinV-FITC及びCD2陰性集団中のSYTOX-グリーン陽性細胞を解析した。
チェックポイント阻害受容体CD96発現をノックダウンした場合及びTRAILバリアントを発現した場合にNK細胞の細胞傷害性の増加が観察された。我々は2つの遺伝子改変を組み合わせてNK細胞の細胞傷害性の相乗効果を起こすことをさらに試した。
(2)共培養の12時間後に、細胞を集め、洗浄し、CD2-APCで染色し、再度洗浄し、Annexin V結合バッファーを用いてAnnexinV-FITC(5uL/試験)及びSYTOX-グリーン(5uL/試験)でさらに染色した。
(3)細胞試料を、BD FACS Canto IIフローサイトメーター)を使用して分析した。データをFlowJo 7.6.2ソフトウェアを使用してさらに解析した。CD2陰性集団はMM1.S細胞を表す。KHYG-1細胞は強いCD2陽性である。次いで、AnnexinV-FITC及びCD2陰性集団中のSYTOX-グリーン陽性細胞を解析した。
