Description
以下の詳細な説明、実施例および添付の図面で、本発明をより詳細に説明する:
図1は、IL−2の改変型を発現するプラスミドの概略図を示す。L:リーダー;IL−2:インターロイキン2;ERRS:小胞体保留シグナル。
図2は、短期間(6日)の培養での、IL−2が形質導入されたNK−92細胞、および、親のNK−92細胞の増殖を示す棒グラフである。NK−92細胞を、初期濃度25000細胞/ml、90%CellGro(R)、10%FBS、および、500IU/mlのIL−2で培養した。0日目に、細胞をPBSで徹底的に洗浄し、それまでの培養段階からのわずかな量のIL−2も全て除去した。改変されていないNK−92細胞は、IL−2の非存在下で死滅したが、遺伝学的に改変された細胞は、添加されたIL−2の存在下での細胞と同等に増殖した。
図3は、NK−92GFPと、IL−2を発現するNK−92改変細胞とを1:1の比率で共培養した結果を示す棒グラフである。25000個のIL−2を発現するNK−92細胞を、同数のGFPを発現するNK−92細胞と混合した。それまでの培養物からの外的なわずかな量のIL−2も全て排除するために、どちらの細胞群もPBSで2回徹底的に洗浄した。48時間後に、FACS解析でGFP陽性細胞と陰性細胞との比率を定量した。NK−92IL2ERとの培養で、NK−92GFP細胞は死滅したが、これは、これらの細胞からのIL−2の分泌がないことを証明している。
図4は、インビトロでの遺伝学的に改変されたNK−92のIL−2生産を説明する棒グラフである。細胞を、25000細胞/mlの開始濃度で6日間培養した。培地を交換してから48時間後に上清を回収した。材料および方法の章で説明されているように、酵素結合免疫吸着検査法(ELISA)を用いて上清中のIL−2のレベルを測定した。その結果によれば、NK92IL2WTのIL−2生産および分泌は、T細胞のそれと同等だが、それに対して、NK92IL2ER細胞の上清中のIL−2レベルは、培地のみ(コントロール)と同等であったことが示される。
図5は、4時間の51Cr放出分析におけるK−562細胞に対する親の細胞と遺伝学的に改変された細胞の細胞毒性を示す。遺伝学的に改変された細胞は、改変されていない細胞と同等の細胞毒性能を保持する。
IL−2遺伝子を選択または構築し、
前記IL−2遺伝子を有するレトロウイルスベクターを製造し、
ドナーから細胞を回収し、
前記細胞を遺伝学的に改変し、そして、
場合により、トランスフェクションされた細胞を選択すること、
からなる。
材料および方法
細胞系
レトロウイルス生産には、細胞系フェニックスGP(Phoenix GP)を用いた(Department of Molecular Pharmacology,Stanford University School of Medicine,スタンフォード,カリフォルニア州のGarry P.Nolan博士(Ph.D.)の許可済)。免疫染色には、アフリカミドリザルから誘導された細胞系のCos−7(DSMZ,ブラウンシュバイク,ドイツ)を用いた。両方の細胞系を、グルタマックス(Glutamax)、ピルビン酸ナトリウム、4500mg/lグルコース、および、ピリドキシンを含み、10%ウシ胎仔血清(FBS−インビトロジェン(Invitrogen))が追加されたダルベッコ改変イーグル培地(DMEM−インビトロジェン社,ペーズリー,スコットランド)で培養した。
IL−2cDNAテンプレートを含むpORF−hIL2プラスミドを、インビボジェン(InvivoGen,サンディエゴ,カリフォルニア州,米国)から購入した。必要なIL−2プライマーを、オリゴ(Oligo)6.6ソフトウェア(モレキュラー・バイオロジー・インサイト社(Molecular Biology Insights Inc),コロラド州,米国)を用いて設計し、それらをDNAテクノロジーApS(DNA Technology ApS,オーフス,デンマーク)に注文した。以下のプライマーを用いて、IL−2変異体をPCRでクローニングした:
配列番号1:野生型IL−2[TTA CAA TTG ATC ACC GGC GAA GGA GG](フォワード)、および、
配列番号2:[TTA ATC GAT GTA TCT TAT CAT GTC G](リバース);
配列番号3:細胞質内の(リーダーなし)IL−2[ACC GCC ATG GCA CCT ACT TCA AGT TCT ACA AA](フォワード)、および、
配列番号4:[TTA ATC GAT GTA TCT TAT CAT GTC G](リバース);および、
配列番号5:小胞体 IL−2[TTA CAA TTG ATC ACC GGC GAA GGA GG](フォワード)、および
配列番号6:[TCA CAG TTC GTC CTT CTC GCT GCC AGT CAG TGT TGA GAT GAT GCT TT](リバース、小胞体保留シグナルを含む)。
野生型IL−2を発現する(分泌する)第一の配列(配列番号8)、
比較として、細胞質に標的化した第二の配列(配列番号9)を構築した。
最後に、小胞体に標的化した第三の配列(配列番号10)を構築した。

フェニックスGP細胞およびCos−7細胞を、35mmの細胞培養ウェルあたり、ベクターコンストラクトプラスミドそれぞれ3μgおよび2μg、ならびに、pMD−G(1μg)(水疱性口内炎ウイルスエンベロープ糖タンパク質をコードする、ジュネーブ大学(University of Geneva,ジュネーブ,スイス)の遺伝学および微生物学教室(Dept.of Genetics and Microbiology)のD.Trono博士によって親切にも提供された)で一時的にトランスフェクションした。このフェニックスGPをトランスフェクションした後、形質導入実験のためにウイルス上清を回収し、免疫染色のためにトランスフェクションされたCos−7細胞を用いた。トランスフェクションのために、フージーン6(Fugene 6)試薬(ロシュ・ベーリンガー・マンハイム(Roche Boehringer Mannheim),ドイツ)を、製造元の説明書に従って用いた。簡単に言えば、DNAプラスミドベクターとフージーン試薬を、細胞培地(体積100μl)中で1/2の質量/体積比で混合し、15分間後に細胞に添加した。ポジティブコントロールとして、レトロウイルス主鎖中にGFPを含むプラスミド(pSF91−GN3g)を用いた。トランスフェクションの24時間および48時間後に上清を回収し、0.45μmのミルレックス−GP(Millex−GP)シリンジトップフィルター(ミリポア社(millipore Corporation),ベッドフォード,マサチューセッツ州)を通過させてろ過し、すぐに形質導入に用いた。ポジティブコントロールにおけるトランスフェクション効率は、常に50%より高かった。
発現されたインターロイキン−2の生物活性を、IL−2依存性細胞系NK−92を用いた細胞増殖分析によって測定した。細胞増殖を、製造元の推奨に従って、トリパンブルー染色による生存率測定と細胞増殖試薬WST−1(ロシュ・ベーリンガー・マンハイム)を用いて、ブッヒャー(Buercher)製チャンバーで細胞を計数することによって定量した。
ヒトインターロイキン−2の定量的な測定のために、OptEIATMヒトIL−2ELISAキットII(BDバイオサイエンシズ・ファーミンジェン(BD Biosciences Pharmingen),サンディエゴ,カリフォルニア州,米国)を、製造元の説明書に従って用いた。
IL−2で改変されたCos−7細胞をPBSで洗浄し、4%パラホルムアルデヒドで室温(RT)で15分間固定した。固定した後、細胞をPBSでリンスし、NP40(Vysis Inc,イリノイ州,米国)の1%PBS溶液と室温で10分インキュベートした。次に、細胞をPBSで3回洗浄し、PBS中に0.1%トゥイーン20(Tween 20)(シグマ−アルドリッチ)、0.1%BSA−c(アウリオン(Aurion),オランダ)、および、5%ヤギ血清(ダコ社(DAKO A/S),グロストルップ,デンマーク)を含むブロッキング緩衝液で30分間ブロックした。細胞を、PBS/0.1%トゥイーン20で3回、4分間洗浄し、ブロッキング緩衝液で希釈した5μg/mlの一次精製ラット抗ヒトIL−2抗体(BDバイオサイエンシズ・ファーミンジェン)と共に45分インキュベートした。次に、細胞を再度、PBS/0.1%トゥイーン20で5分間、4回洗浄し、ブロッキング緩衝液中の5μg/mlの二次オレゴングリーン488nmヤギ抗ラットIgG抗体と共に1時間インキュベートした。最後に、細胞をPBS/トゥイーン20でリンスし、PBSで1:4000に希釈したヘキスト染色(Hoechst stain)(モレキュラープローブスBV(Molecular Probes BV),ライデン,オランダ)を用いて室温で5分間、対比染色し、続いてPBSで1回洗浄した。CCDカメラ(モデルS/N370KL0565,クック社(Cooke Corporation),ニューヨーク州,米国)を備えたライカ製のDMRXA顕微鏡(ライカ・マイクロシステムズ社(Leica Microsystems,GmbH),ヴェッツラー,ドイツ)を用いて、蛍光顕微鏡検査法によって細胞を可視化した。DAPI/ヘキストに設定されたフィルター、FITC、Cy3およびCy5を、クロマ(Chroma Technology,ブラットルボロ,バーモント州,米国)から得た。スライドブック(Slidebook)2.1.5ソフトウェア(インテリジェント・イメージング・イノベーションズ社(Intelligent Imaging Innovations Inc,デンバー,コロラド州,米国)、および、アドビ・フォトショップ(Adobe Photoshop)5.0(アドビ・システムズ(Adobe systems,シアトル,ワシントン州,米国)を用いて画像を得た。
緑色蛍光タンパク質(GFP)を発現するように改変されたNK−92細胞を、IL−2で改変されたNK−92細胞と1:1の比率で混合した。共培養実験を、6ウェルプレートで以下のように行った:25000個のIL−2を発現するNK−92細胞を、同数のGFPを発現するNK−92細胞と混合した。それまでの培養物から外的なわずかな量のIL−2も全て排除するために両方の細胞群をPBSで2回徹底的に洗浄した。48時間後に、FACS解析でGFP陽性細胞と陰性細胞との比率を定量した。
細胞毒性の機能を、標準的な4時間の51Cr放出分析で3連で測定した。簡単に言えば、1×106個のK562細胞を51Cr(100μl)で標識し、37℃で1時間インキュベートした。トリパンブルー排除法の色素を用いてエフェクター細胞を計数し、標的細胞と混合して、エフェクターを得た:標的の比率は10:1、3:1、1:1、および、0.3:1とした。ネガティブコントロールとして、セルグロ培地を用い、ポジティブコントロール細胞を、1%トリトンXとインキュベートした。V底型の96ウェルプレートで37℃で4時間インキュベートした後、上清70μlを各ウェルから吸引して、パッカード(Packard)のコブラ・オート−ガンマ(Cobra Auto−Gamma)5000シリーズカウントシステム(メリデン,コネチカット州,米国)を用いて計数した。自然発生による放出の比率を以下の式から計算した:%51Cr放出量=(サンプル−自然発生)/(最大放出量−自然発生)×100。
優先権主張年中に、健康なドナー由来の一次細胞を用いてさらなる実験を行った。
軟膜の細胞を、カロリンスカ大学病院(Karolinska University Hospital,フッディンゲ,ストックホルム,スウェーデン)の健康な血液バンクのドナーから得て、同じ日に培養を開始した(0日目)。PBMCを、リンホプレップ(Lymphoprep,ニエガード(Nyegaard),オスロ,ノルウェイ)を用いた濃度勾配遠心分離によって単離した。リン酸緩衝食塩水(PBS)(ギブコ(Gibco))で2回洗浄した後に、トリパンブルー色素排除法で細胞生存率を評価し、細胞を6ウェルの培養皿(ファルコン(Falcon),ベクトン・ディッキンソン(Le Pont de Claix,フランス)製)上で0.25×106細胞/mlで平板培養した。全ての培養で、5%ヒト血清(バイオウィッタカー(Biowhittaker),キャンブレックス・バイオサイエンス(Cambrex Bio Science),ウォーカーズビル,メリーランド州,米国)、500IU/mlインターロイキン−2(IL−2)(ペプロテック(Peprotech),ニュージャージー州,米国)、および、10ng/ml抗CD3抗体、OKT−3(オルソ・バイオテック社(Ortho Biotech Inc.),ラリタン,ニュージャージー州,米国)を添加したセルグロSCGM培地(セルジェニックス,フライブルグ,ドイツ)を用いた。5日目に、OKT−3を洗浄して除き、その後、細胞を500IU/mlのIL−2、および、5%ヒト血清が追加されたセルグロ培地(OKT−3非含有)中で培養した。次に、1〜2日ごとに、21日目まで培養に新しい培地を添加した。細胞の部分集合の絶対細胞数(ACC)を、細胞の部分集合のパーセンテージに、同じ時点での培養の細胞の総数を掛けることにより得た。
SF91g−IL2wt、SF91g−IL2−L、SF91g−IL2ER、および、SF91g−GN3(GFPコントロール)レトロウイルスベクターを生産する安定して形質導入されたレトロウイルス産生細胞系を、10%FCSが追加されたDMEM中で成長させた。細胞がやや密集状態になったら、新しい新鮮な培地を24時間添加した。最後に、上清を回収し、0.45μmフィルター(ミリポア,ビレリカ,マサチューセッツ州,米国)でろ過し、−70℃で凍結した。回収および採取した上清は、0.5×106ウイルス粒子/mlのタイター(HeLa細胞で測定)を有していた。この産生細胞系から生産されたウイルス粒子は、GALV(テナガザル(Gibbon Ape)白血病ウイルス)エンベロープを含む。培地を、8μg/mlポリブレン(シグマ)と500U/mlのIL−2の存在下で、1.000gで室温で2時間遠心分離して、レトロウイルスを含む上清(3の重複感染度(MOI))で交換することによって、全ての形質導入を行った。遠心分離の後、上清を、NK細胞培地(セルグロ培地,500U/mlのIL−2、および、5%ヒト血清を含む)で交換し、細胞を21日目まで増殖させた。コントロールとして、同じドナーのPBMCを成長させ、形質導入した細胞と同様の条件下で擬似感染させた。
NK−92細胞におけるIL−2発現による細胞増殖に対する作用
IL−2発現のための3種のコンストラクトを評価した;1種は、野生型IL−2を発現し、1種は、IL−2を細胞質に標的化し、1種は、IL−2を小胞体に標的化する(図1)。これらの細胞質内コンストラクトは、生物学的に不活性であった(データ示さず)。
形質導入後に、遺伝子発現させるために細胞をIL−2の存在下で4日間培養し、次に、PBSで十分に洗浄し、培養条件下でIL−2添加または無添加のいずれかで維持した。この実験には、発明者等により開発された2種のIL−2が形質導入された細胞系;NK92IL2WT、および、NK92IL2ERが選択された。図2で示されるように、形質導入されていないNK−92コントロール細胞は、IL−2を除去してから培養6日目に死滅した。それに対して、NK92IL2WTとNK92IL2ERは、外的なIL−2が追加されたNK−92細胞と同様の増殖曲線を示した(図2)。
NK92IL2WTとNK92IL2ERの増殖の特徴は、ほぼ1年間の連続培養の後でも安定であった。
まず、小胞体に局所化された発現のためのベクターの機能性を、Cos−7細胞への一過性トランスフェクションによって解析した。Cos−7細胞は、顕微鏡イメージングにおいて明確な形態を示すため、形質導入されたタンパク質の発現をモニターするために、該細胞を選択した。モノクローナルラット抗ヒトIL−2抗体を用いたIL−2ERで形質導入されたCos−7細胞の間接的な免疫蛍光法によれば、小胞体での局在化に伴い明るい緑色の染色が示された。(データ示さず)。
続いて本発明者等は、少量のIL−2が形質導入された細胞から周囲の培地に漏出するかどうか、加えて、それが、親のNK−92細胞の増殖の促進に十分であり得るのかどうかを調査しようとした。そのために、同数のNK−92のIL−2で改変されたNK−92細胞系と、GFPで改変されたNK−92細胞系とを共培養し、増殖に関して評価した。培養してから48時間後、NK−92GFP細胞の比率は、NK92IL2ERと混合した場合に著しく減少した(45%)。それに対して、NK92IL2WT細胞と混合した場合、増殖の促進が観察された(図3)。培養してから2日後に、両方の群の比率はそれでもなお最初のほぼ50%を保持しており、これは、NK92IL2WTは、上清にIL−2を分泌して、NK92GFPの生存を促進していることを示す。
NK92IL2WT、および、NK92IL2ER系は、51Crで標識されたK562細胞を、親のNK−92細胞に匹敵するレベルで溶解させることができた。(図5)。1:1のエフェクター:標的の比率により、NK−92細胞が59%であったのに対して、平均59.2%(NK92IL2WT)、および、平均46.4%(NK92IL2ER)の細胞毒性活性が得られた。

Claims (20)
- 実質的に生理学的なレベルのIL−2を発現する遺伝学的に改変された細胞を用いた免疫治療の方法であって、ここでIL−2を発現する哺乳動物細胞は、
IL−2遺伝子を選択または構築し、
該IL−2遺伝子を有するレトロウイルスベクターを製造し、
ドナーまたは患者から細胞を回収し、
細胞を遺伝学的に改変し、
場合により、遺伝学的に改変された細胞を選択する
ことからなる方法によって製造され、さらに、該細胞は、それを必要とする患者に投与される、上記方法。 - IL−2遺伝子は、配列番号11で示されるタンパク質、または、その機能的等価体をコードする遺伝子である、請求項1に記載の方法。
- IL−2遺伝子は、配列番号13で示されるタンパク質、または、その機能的等価体をコードする遺伝子である、請求項1に記載の方法。
- IL−2遺伝子は、配列番号8、配列番号10、および、それらの機能的等価体から選択される遺伝子である、請求項1に記載の方法。
- 哺乳動物細胞は、ナチュラルキラー(NK)細胞、および、T細胞から選択される、請求項1〜4のいずれか一項に記載の方法。
- 哺乳動物細胞は、ナチュラルキラー(NK)細胞である、請求項5に記載の方法。
- IL−2遺伝子は、IL−2が細胞の小胞体に向けて発現されるように改変される、請求項1に記載の方法。
- 実質的に生理学的なレベルのIL−2を発現する遺伝学的に改変された細胞の製造方法であって、
IL−2遺伝子を選択または構築し、
該IL−2遺伝子を有するレトロウイルスベクターを製造し、
ドナーまたは患者から細胞を回収し、
細胞を遺伝学的に改変し、
場合により、遺伝学的に改変された細胞を選択する、
ことからなる、上記方法。 - IL−2遺伝子は、配列番号11で示されるタンパク質、または、その機能的等価体をコードする遺伝子である、請求項8に記載の方法。
- IL−2遺伝子は、配列番号13で示されるタンパク質、または、その機能的等価体をコードする遺伝子である、請求項8に記載の方法。
- IL−2遺伝子は、配列番号8、配列番号10、および、それらの機能的等価体から選択される遺伝子である、請求項8に記載の方法。
- 請求項8〜11のいずれか一項に記載の方法によって得られる、IL−2の生産が可能なトランスジェニック哺乳動物細胞。
- 請求項8〜11のいずれか一項に記載の方法によって得られるトランスジェニック哺乳動物細胞であって、改変されていない状態の細胞は、増殖に関してIL−2依存性であり、それらを有意な量で生産することができないが、該トランスジェニック細胞は、外的なIL−2を必要とすることなく増殖を維持するのに十分な量でIL−2を生産する、上記細胞。
- IL−2が発現され、そして細胞の小胞体中に保留される、請求項13に記載のトランスジェニック哺乳動物細胞。
- 患者に、治療上有効で生理学的に許容できる量の、請求項12〜14のいずれか一項に記載のトランスジェニック細胞を投与することを含む癌の治療方法。
- 細胞は、患者から採取され、トランスフェクションされ、そして同じ患者に戻される、請求項1〜7または15のいずれか一項に記載の方法。
- 細胞は、ドナーから採取され、トランスフェクションされ、そして患者に投与される、請求項1〜7または15のいずれか一項に記載の方法。
- トランスジェニック細胞の使用は、付加的、または、補足的な治療の一つであり、その他の治療の前に、その後に、または、実質的にそれと同時に行われる、請求項1〜7または15のいずれか一項に記載の方法。
- トランスジェニック細胞の使用は、癌の治療における一次療法の一つである、請求項1〜7または15のいずれか一項に記載の方法。
- 免疫促進は、感染の治療または予防における処理の一つである、請求項1〜7のいずれか一項に記載の方法。
