Description
本出願は、米国特許法第119条(e)に従い、2013年5月10日に提出された米国仮出願第61/822,092号明細書、及び2014年2月21日に提出された米国仮出願第61/943,094号明細書に対する優先権を主張する。尚、これら文献の各々の内容は、その全体を参照により本明細書に組み込むものとする。
本発明は、国立衛生研究所により付与された助成金第R01A1087879号を得て実現された。政府は、本発明に関して一定の権利を有する。
具体的な官能基及び化学用語の定義を以下に、より詳細に記載する。本発明の目的のために、化学元素は、元素の周期表(Periodic Table of the Elements)、CASバージョン、物理化学ハンドブック(Handbook of Chemistry and Physics)、第75版、内表紙(inside cover)に従って同定し、具体的な官能基は、一般に、そこに記載されている通りに定義される。さらに、有機化学の一般原理、並びに機能性部分及び反応性は、以下:Organic Chemistry,Thomas Sorrell,University Science Books,Sausalito,1999;Smith and March March’s Advanced Organic Chemistry,5th Edition,John Wiley&Sons,Inc.,New York,2001;Larock,Comprehensive Organic Transformations,VCH Publishers,Inc.,New York,1989;Carruthers,Some Modern Methods of Organic Synthesis,3rd Edition,Cambridge University Press,Cambridge,1987に記載されている。
I.非遺伝子操作真核ペプチド及び細胞のソルタギング
本開示では、遺伝子操作されていない哺乳動物細胞は、ソルターゼを用いて、すなわち、ソルターゼ触媒アシル基転移により、有効に標識できるという意外な発見を説明する。一部の態様では、本発明は、ソルターゼを用いて、ソルターゼ認識モチーフ又は求核アクセプター配列を含むタンパク質を発現するように遺伝子操作されていない生存哺乳動物細胞に、薬剤を結合させる方法を提供する。一部の実施形態では、哺乳動物細胞は、遺伝子操作されていない。本発明の一部の態様は、ソルターゼ触媒アシル基転移反応が、生存哺乳動物細胞に内在性の1つ又は複数のポリペプチドと薬剤の結合を可能にする、すなわち、ソルターゼを用いて、ソルタギングのために遺伝子操作されていない生存哺乳動物細胞に薬剤を結合させることができるという認識に関する。本明細書で用いられる場合、ポリペプチドは、それが、ソルターゼ触媒反応においてソルターゼ基質又は求核基として作用するのを可能にするように遺伝子操作されていない、すなわち、ポリペプチドが、ソルターゼに接触可能な領域(例えば、C末端若しくはその付近)にソルターゼ認識モチーフを含むように遺伝子操作されておらず、かつ、ソルターゼ触媒反応において求核基の役割を果たし得る求核アクセプター配列、例えば、ポリペプチドのN末端に位置するか、又はポリペプチドの切断によって配列がN末端に配置されるように位置する、1つ又は複数のグリシンを含む配列などを含むように遺伝子操作されていなければ、「ソルタギングのために遺伝子操作されていない」。一部の実施形態では、ポリペプチドは、遺伝子操作されていない。1細胞は、細胞が、ソルターゼ触媒反応においてソルターゼ基質又は求核基の役割を果たすのに好適なポリペプチド(天然か、又は遺伝子操作で得られたかの何れか)を発現するように遺伝子操作されていなければ、「ソルタギングのために遺伝子操作されていない」とみなされる。一部の実施形態では、細胞は、ソルターゼ認識モチーフ又は求核アクセプター配列を含むポリペプチドを発現するように遺伝子操作されていない。一部の実施形態では、細胞は、遺伝子操作されていない。一部の実施形態では、細胞は、人の手により導入された、そのゲノムに対する修飾を含まない。一部の態様では、本発明は、非常に多様な薬剤の何れかを、非遺伝子操作哺乳動物細胞の表面に結合させるためのソルターゼの使用に関する。別途記載されていないか、又は文脈から明らかでない限り、本開示が哺乳動物細胞のソルタギングについて述べる場合、一般に、ソルタギングのために遺伝子操作されていない哺乳動物細胞を意味することが意図される。いくつかの実施形態では、動物細胞は、遺伝子操作されていない。

「ソルタギングプロセス」とは、少なくとも一部の実体(例えば、タンパク質、細胞)がソルタギングされるプロセスを指す。一般に、ソルタギングプロセスは、ソルタギングしようとする実体、例えば、真核細胞、例えば、哺乳動物細胞を、トランスアミダーゼ及びソルターゼ基質と、ソルターゼ触媒反応が起こり得る条件下で、接触させるステップを含む。いくつかの実施形態では、生理学的条件下でソルターゼ反応を実施してもよい。一般に、ソルターゼ触媒結合は、アシルドナーとアシルアクセプターの結合が起こる条件下で、トランスアミダーゼ、アシルドナー(ソルターゼ基質)、及び求核アシルアクセプターを互いに接触させることにより実施してもよい。本開示の実施形態において、求核アシルアクセプターは、哺乳動物細胞により発現されるタンパク質であってもよい。前記成分を互いに接触させるのは、単一体の流体にこれらを添加する、及び/又は1つの反応容器中で、例えば、あるいは、これらの成分を互いに近接させて、これらを衝突させることにより、達成することができる。この系中の成分は、必要に応じて、容器の振盪などにより、様々な方法で混合してよい。成分は、任意の順序で前記系に添加してよい。結合は、任意の容器(例えば、マイクロチューブなどのチューブ、フラスコ、皿)、マイクロプレート(例えば、96ウェル若しくは384ウェルプレート)などで実施してよい。反応混合物は、反応を実施することができる任意の好適な温度で維持してよい。一部の実施形態では、約3又は4℃~約15℃の温度で結合を実施する。一部の実施形態では、約15℃~約50℃の温度で結合を実施する。一部の実施形態では、約23℃~約37℃の温度で連結を実施する。いくつかの実施形態では、温度は、室温である(例えば、約25℃)。任意の好適な量及び成分比を使用して、ソルターゼ基質を動物細胞に結合させることができる。一部の実施形態では、アシルドナーは、約5μM~約10mM、約10μM~約5mM、約100μM~約500μM、約200μM~約1mMの濃度で存在する。一部の実施形態では、アシルドナーは、少なくとも約0.25mM、少なくとも約0.5mM、又は少なくとも1mMである。いくつかの実施形態では、トランスアミダーゼは、約1μM~約500μM、約15μM~約150μM、約150μM~約250μM、約250μM~約500μMの濃度で存在する。いくつかの実施形態では、トランスアミダーゼは、10μM超、例えば、11μM~20μM、20μM~30μM、30μM~50μM、50μM~100μMの濃度で存在する。一部の実施形態では、トランスアミダーゼは、10μM超の濃度で存在し、野生型ソルターゼ、例えば、野生型ソルターゼA、例えば、野生型黄色ブドウ球菌(S.aureus)SrtA、又は野生型黄色ブドウ球菌(S.aureus)SrtAの0.5倍~5倍の活性を有するその変異体である。いくつかの実施形態では、トランスアミダーゼは、約10μM~約1mM、約15μM~約1mM、約20μM~約1mM、約25μM~約1mM、30μM~約1mM、約50μM~約1mM、約100μM~約1mM、又は約250μM~約1mMの濃度で存在する。いくつかの実施形態では、これらの濃度は、野生型ソルターゼA、とりわけ、例えば、野生型黄色ブドウ球菌(S.aureus)SrtAに適用する。
一般に、本明細書に記載するように、任意のタイプの細胞をソルタギングしてよく、又はソルタギングしようとする細胞の供給源として用いてよい。一部の実施形態では、細胞は、哺乳動物細胞を含むか、それから構成される。一部の実施形態では、哺乳動物細胞は、霊長類細胞(ヒト細胞若しくはヒト以外の霊長類細胞)、げっ歯類細胞(例えば、マウス、ラット、ウサギ、ハムスター細胞)、イヌ、ネコ、ウシ、ブタ、又はその他の哺乳動物細胞である。一部の実施形態では、細胞は鳥類細胞である。一部の実施形態では、細胞は、無脊椎動物細胞である。一部の実施形態では、細胞は、真菌(例えば、酵母若しくはカビ)又は原生動物細胞である。
一部の実施形態では、真核細胞、例えば、哺乳動物細胞に結合した、又は結合させようとする薬剤は、クリックケミストリーハンドルを含んでもよい。例えば、前述したソルターゼ基質のA1は、クリックケミストリーハンドルを含み得る。クリックケミストリーは、2001年にSharplessにより導入された化学哲学であり、これは、小さな単位を互いに結合することにより、迅速に、しかも高い信頼性で、物質を生成するように設計された化学を表す(例えば、Kolb,Finn及びSharpless Angewandte Chemie International Edition(2001)40:2004-2021;Evans,Australian Journal of Chemistry(2007)60:384-395を参照)。本発明の態様に従い有用な別のクリックケミストリーハンドル、反応条件、及び関連方法の例は、Joerg Lahann,Click Chemistry for Biotechnology and Materials Science,2009,John Wiley&Sons Ltd,ISBN 978-0-470-69970-6に記載されている。一部の実施形態では、クリックケミストリーハンドルは、本明細書の参照文献の何れか及び/又は表1若しくは表2に記載されている。例えば、クリックケミストリーハンドルは、末端アルキン、アジド化物、歪みアルキン、ジエン、ジエノフィル、アルコキシアミン、カルボニル、ホスフィン、ヒドラジド、チオール、又はアルケン部分を含むか、これらから構成されるものでもよい。

本発明を限定することなく、このセクションでは、本開示、ソルタギング済み哺乳動物細胞の調製若しくは使用方法、並びに関連組成物に関して、対象とするいくつかのタンパク質、核酸、脂質、小分子、及び他の実体について述べる。本明細書に記載する実体は、様々な目的に用いることができる。例えば、様々な実施形態において、タンパク質、核酸、脂質、小分子、及び糖を、ソルターゼを用いて哺乳動物細胞に結合させてもよく、ソルターゼを用いて互いに結合させて、細胞培養に使用してもよく(例えば、細胞を増殖、刺激、若しくは分化させるために)、及び/又は被験者に投与してもよい。
一部の態様では、本発明は、ソルタギング済み哺乳動物細胞を作製するのに有用なキットを提供し、前記細胞は、ソルタギングのために遺伝子操作されていない。一部の実施形態では、キットは、以下:(i)ソルターゼポリペプチド、ソルターゼポリペプチドをコード化する核酸若しくはベクター、又はソルターゼポリペプチドを発現する細胞系;及び(ii)ソルタギング済み哺乳動物細胞の調製、特性決定、及び/又は精製に有用な1つ又は複数の品目を含む。sでは、1つ又は複数の品目は、ソルタギングのために遺伝子操作されていないソルタギング済み生存哺乳動物細胞を調製、特性決定、及び/又は精製する方法に関して本明細書に記載する品目の何れであってもよい。一部の実施形態では、1つ又は複数の別の品目は、以下から選択される:(a)トランスアミダーゼ認識配列;(b)ソルタギングのために遺伝子操作されていない哺乳動物細胞;(c)生存哺乳動物細胞をソルタギングするための反応バッファーとしての使用に好適な液体組成物、又はその成分;(d)ソルターゼからソルタギング済み細胞を分離するのに有用な1つ又は複数の試薬;及び(e)対照物質から選択される。一部の実施形態では、キットは、ソルタギングのために遺伝子操作されていない哺乳動物細胞を調製するための説明書を含む。一部の実施形態では、説明書は、キットとは別に入手できるようにしてもよい。例えば、説明書は、インターネットを介して、例えば、ワールドワイドウェブ(「ウェブ」)から提供されるか、又はそこで閲覧することができる。一部の実施形態では、トランスアミダーゼ認識配列を含む薬剤は、腫瘍抗原に結合する結合部分を含む。前記薬剤を用いて、例えば、TAを発現する腫瘍の治療が必要な被験者に投与しようとする細胞をソルタギングすることができる。一部の実施形態では、前述したキットの何れかは、ソルターゼを含まなくてもよい。例えば、トランスアミダーゼ認識配列を含む薬剤は、単独で、又は前述した別の品目の1つ又は複数と一緒に提供してもよい。
ソルターゼ修飾細胞、例えば、ソルターゼ修飾哺乳動物細胞を用いて、非常に多様な疾患及び障害を治療することができる。一部の実施形態では、治療薬と結合したソルターゼ修飾細胞を用いて、非結合治療薬が有用なあらゆる疾患又は病状を治療することができる。このような治療方法は、本発明の一態様である。
標準的方法を用いて、マウス赤血球欠失脾細胞を単離した。約2百万個の細胞を、血清を含まない100マイクロリットルのDME中で、黄色ブドウ球菌(Staphylococcus aureus)由来のソルターゼA(150μM)含有又は非含有何れかのビオチン-LPETGプローブ(1mM)と一緒に、4℃で1時間インキュベートした。次に、細胞をPBSで5回洗浄した後、レムリ(Laemmli)バッファー中で溶解させた。サンプルをSDS-PAGEゲル上に泳動させた。各インキュベーションからの溶解物の約1/4をレーン毎にロードした。標準的方法を用いたストレプトアビジン-HRPとのブロッティングにより、ビオチン化タンパク質を視覚化した。得られた免疫ブロットを図2に示す。ブロットは、ビオチン-LPETGプローブによるいくつかのタンパク質の標識を示している。他の実験に基づき、約30kDの顕著なバンドが、ビオチン-LPETGで標識されたソルターゼ自体であると考えられ、約60kDのバンドは、ビオチン標識ソルターゼ二量体であると思われる。
注:化膿レンサ球菌(S.pyrogenes)ソルターゼAのプローブを作製するために、Fmoc-Gly-OHの代わりにFmoc-Ala-OHを使用する。
i 100μモルのリンク(Rink)アミド樹脂(167mg、0.6mモル/g)を、焼結ガラスフィルター底を備える、キャップしたガラスカラムに添加し、手首作用振盪機で15分間振盪することによって、ジクロロメタン(DCM)(7mL)中で樹脂を溶媒和させた後、真空ろ過によりDCMを除去する。
ii N-メチル-2-ピロリドン(NMP)(7mL)に20%ピペリジン溶液を添加し、15分間振盪することにより、樹脂のFmoc保護基を除去する。
注:全ステップで、NMPの代わりにDMFを用いてもよい。
iii 真空ろ過によりピペリジン溶液を除去した後、樹脂を、NMP(7mL、1分)で3回、DCM(7mL、1分)で3回洗浄した後、NMP(1分)でさらに1回洗浄する。
iv NMP(7mL)中に、Fmoc-Gly-OH(89mg、300μモル)、HBTU(114mg、300μモル)、及びDIPEA(104μL、600μモル)を溶解させた後、樹脂に添加する。懸濁液を2時間室温で振盪する。
v 真空ろ過により反応溶液を除去し、NMP(7mL、1分)で3回、続いてDCM(7mL、1分)で3回、樹脂を洗浄する。カップリング反応は、カイザー(Kaiser)テストを実施することにより確認することができる。
注:反応が不完全であれば、標準的カップリングに使用する試薬の量を半分にして、ステップiv~vを繰り返し、1時間振盪する。
ポーズポイント:樹脂を真空下で乾燥させた後、4℃で保存することができる。この段階では、保存が要望される場合、Fmoc保護形態でペプチドを保存する。
vi Fmoc-Gly-OH(89mg、300μモル)、Fmoc-Thr(OtBu)-OH(119mg、300μモル)、Fmoc-Glu(OtBu)-OH(127mg、300μモル)、Fmoc-Pro-OH(101mg、300mol)、Fmoc-Leu-OH(106mg、300μモル)、Fmoc-ε-(85mg、300μモル)を用いて、ステップi~vを繰り返す。
注:ProのN末端が2級アミンであるため、Leuカップリングの範囲を確認するのにカイザーテストは効果がない。このカップリング反応を試験するために、クロロアニル試験又は微小切断を用いることができる。直交性保護基は、この短縮切断の間に完全に除去されなくてもよいことに留意されたい。
vii εアミノカプリン酸残基でのFmocの除去後、NMP(7mL)中、5(6)-TAMRA(52mg、120μモル)、PyBOP(63mg、120μモル)、及びDIPEA(42μL、240μモル)の溶液を添加した後、室温で一晩振盪する。発蛍光団の光退色を防止するために、カラムをアルミホイルで包む。
viii ステップvを繰り返した後、カイザーテストを実施して、TAMRAカップリングを確認する。
ix 95%TFA、2.5%H2O、及び2.5%TIS(5mL)から構成される切断溶液に樹脂を室温で2時間懸濁させる。
x 90mLの氷冷(-0℃)ジエチルエーテルに切断溶液を溶出させた後、さらに3mLの切断溶液でエーテル中に樹脂をすすぎ流す。
xi このエーテル溶液を-20℃で20分保存して、ペプチドを沈殿させる。懸濁液を1,900gで15分4℃にて遠心分離し、上清を静かに移し、残ったエーテルを減圧下で穏やかに蒸発させる。
ジメチルエーテルは、可燃性かつ揮発性であるため、スパークフリー冷凍庫及び遠心分離機を使用するのが望ましい。

重要なステップ:同一性及び純度は、LC/MS分析(10分にわたる線形勾配5→45%LC/MSバッファーB)により確認することができる。LC/MSによって、粗ペプチドが十分な純度であることが判明した場合には、次のステップ(xii~xiv)は省いてもよく、ペプチドをソルターゼ反応に直接用いてもよい。
xii 乾燥ペプチドをH2O(2mL)に溶解させ、卓上遠心分離機を用いて、14,000rpmで10分遠心分離することにより、粒子状物質を除去する。
注:純粋H2O中に溶解しないペプチドに、最大50%のtert-ブタノールを添加してもよい。また、粒子状物質を除去するために、スピンフィルター又はシリンジフィルターを用いてもよい。
xiii 遠心分離した上澄みを、10~70%バッファーB勾配を用いたC18カラムでの逆相HPLCにより15分間精製した後、90%バッファーBで5分フラッシュする。予め少量の100μLを注入してから、ペプチド純度及び分離しやすさの目的に応じて、勾配を調節することが推奨される。良好な勾配が確立されたら、残った粗材料を400~600μLの注入により精製することができる。
xiv LC/MSにより産物の画分を分析した後、所望の画分を乾燥まで凍結乾燥させる。

標準的方法を用いて単離した約2百万個のマウス赤血球欠失脾細胞を、無血清のDME中、黄色ブドウ球菌(Staphylococcus aureus)由来のソルターゼA(200μM)と共に又はそれなしの何れかで、C末端LPETG(100μM)(C末端LPETGを含まないオリジナルGFP特異的VHHの説明については、Kirchhofer,A.,et al.,Nat Struct Mol Biol.2010 Jan;17(1):133-8.doi:10.1038/nsmb.1727.Epub 2009 Dec 13を参照)を含むGFP特異的VHHと一緒に、4℃で1時間インキュベートした。次に、細胞をPBSで5回洗浄した後、GFPと30分インキュベートし、フローサイトメトリーに付した。結果を図4に示す。青色のヒストグラムは、ソルターゼAと共に(右)又はそれなしで(左)インキュベートした生存細胞にゲーティングしたPEシグナルを示す。黒色のヒストグラムは、GFPと一緒にインキュベートした対照脾細胞に対するバックグラウンド染色を示す。左側パネルの青色及び黒色ヒストグラムは、ほぼ重なり合う。右側パネルの青色ヒストグラムは、矢印で示す。これらの結果は、ソルターゼの存在下で、GFP特異的VHHとインキュベートした細胞のみに対するGFPの特異的結合を示しており、細胞へのVHHのソルターゼ触媒結合による前記細胞の標識を立証するものである。

VHHの代わりに、ソルターゼ認識配列を有する鎖を含む一般的ヒト抗体を用いる以外は、実施例3~5を繰り返した。
VHHの代わりに、ソルターゼ認識配列を含むscFvを用いる以外は、実施例3~5を繰り返した。
実施例3~7を繰り返すが、以下:(i)ソルタギングの前にMACSビーズ(例えば、CD8+T細胞単離キット(Cell Isolation Kit)、ヒト(♯130-096-495)、Miltenyi Biotecを使用)を用いて、CD8+細胞を富化するステップ;(ii)ソルタギング済みリンパ球の投与前に、CD28に対する抗体を用いて、共刺激を加えるステップ;並びに/又は(iii)グランザイム及び/若しくはパーホリンの分泌を測定することにより、及び/又は抗CD107mAbを用いて、CD107細胞表面を評価することにより、ソルタギング済みリンパ球のサンプルの細胞傷害活性をin vitroで評価するステップをさらに追加する。
実施例9、10、11、及び/又は28で用いる抗体:抗PGK(クローン22C5D8、Invitrogen)、抗マウス/ヒトアクチン(クローンAb-5、BD biosciences)、抗トキソプラズマ(Toxoplasma gondii)アクチン、セイヨウワサビペルオキシダーゼ結合ヤギ抗ウサギIg(Southern Biotech、品番4041-05)、セイヨウワサビペルオキシダーゼ結合抗マウスIg(GE Healthcare、品番NXA931)、抗TCRbeta(クローンH57、BD Pharmingen)、抗CD4(クローンGK1.5、ebiosciences)、抗CD19(クローン1D4、BD Pharmingen)、抗TER119(クローンTER-119、BD Pharmingen)、アロフィコシアニン結合ストレプトアビジン(ebiosciences、品番17-4317)、フィコエリスリン結合ストレプトアビジン(Southern Biotech、品番7100-09S)、ヨウ化プロピジウム(Sigma-Aldrich、品番P4864)。
この実施例では、OTI Rag-/-マウス由来の赤血球欠失脾細胞が、刺激時に適切な標的細胞に対して細胞媒介傷害性を及ぼすことができる細胞集団を含むことを立証すると共に、細胞傷害性を定量する方法を示す。脾細胞は、リンパ球、NK細胞、マクロファージなどの多様な細胞集団を含む免疫系細胞の混合集団である。OTI Rag-/-マウスは、Ragを欠失しており、H2bに関連して、SIINFEKLペプチドを認識するT細胞受容体のトランスジェニックマウスである。これらのマウスは、SIINFEKLに特異的なCD8+細胞を産生する。上記マウスは、Rag欠失状態であるために、B細胞及びCD4+T細胞を欠失している。
この実施例は、ソルターゼを用いて、ターゲティング部分を非遺伝子操作免疫系細胞に結合させると、ターゲティング部分が結合する標的を担持する細胞に特異的に向かう細胞傷害性が増大することを立証する。実施例10と同様に、本実施例に記載する実験でも、OTI Rag-/-マウス由来の赤血球欠失脾細胞を使用する。この脾細胞をマウスMHCクラスIに結合するVHH(VHH7)とソルタギングした後、これらの細胞が、MHCクラスIを発現するマウスB細胞に対して細胞傷害性を及ぼす能力を、実施例9に記載したのと同様の手法を用いて評価した。
2つの異なるプローブをリンパ球に設置することができるかどうかを調べるために、ソルターゼAの存在下で、エンハンサー-LPETGと共に又はそれなしで赤血球欠失脾細胞を60分インキュベートした後、ビオチン-LPETGを反応物に15分かけて添加した(図11)。ビオチン-LPETGの前にエンハンサー-LPETGとインキュベートした細胞は、ビオチン-LPETGのみとインキュベートした細胞と比較して、同様の量の表面結合ビオチンを有した(図11)。これらのデータは、細胞に対するVHHのソルタギングは、後のビオチン-LPETGの結合にわずかにしか影響しないことを示唆している。より小さいLPETGタグ付けプローブの方が、より大きなLPETGタグ付けタンパク質によって占められずに残った表面提示求核基に容易に接触し得る可能性がある。
B細胞慢性リンパ芽球性白血病を有する患者から取得したヒトT細胞を、キメラ抗原受容体(CD3-ζ及びCD28のシグナル伝達ドメインを含む、CD3-ζの膜貫通及び細胞内ドメインに融合した抗CD20一本鎖Fv領域)を発現するように、遺伝子改変する。抗CD3/抗CD28ビーズとの培養で細胞を10日間拡大してから、ソルターゼ、並びに(i)PD-1の細胞外ドメインと、(ii)ソルターゼ認識配列とを含む薬剤と一緒にインキュベートした後、前記被患者に戻して移入する。白血病に対する投与細胞の効果をモニターする。
B細胞慢性リンパ芽球性白血病を有する患者から取得したヒトT細胞を、キメラ抗原細胞受容体(CD3-ζ及びCD28のシグナル伝達ドメインを含む、CD3-ζの膜貫通及び細胞内ドメインに融合した抗CD20一本鎖Fv領域)を発現するように、遺伝子改変する。抗CD3/抗CD28ビーズとの培養で細胞を10日間拡大しから、ソルターゼ、並びにソルターゼ認識配列を含むように修飾されたPD-1に結合するscFvを含む薬剤と一緒にインキュベートした後、前記患者に移入する。白血病に対する投与細胞の効果をモニターする。
骨髄腫患者からNK細胞を単離して、培養により拡大する。これらの細胞を、ソルターゼ、並びにソルターゼ認識配列を含むように修飾された抗CS-1又は抗BCMA抗体とインキュベートした後、患者に移入する。この治療を12週間にわたって毎週繰り返す。多発性骨髄腫に対する投与細胞の効果をモニターする。
III期結腸癌と最近診断された患者からPMBCを単離する。これらの細胞を、ソルターゼ、並びにソルターゼ認識配列を含むように修飾された抗CS-1又は抗BCMA抗体とインキュベートした後、患者に移入する。この治療を12週間にわたって毎週繰り返す。局所再発又は転移の存在について患者をモニターする。
SS1Fvの修飾形態である、メソテリンに特異的なFvを作製する。修飾Fvは、C末端にソルターゼ認識配列を有する。ヒトT細胞(遺伝子改変していない)をソルターゼ及び修飾SS1Fvと一緒にインキュベートした。
ソルターゼ、並びにソルターゼ認識配列を含むように修飾した抗CA-125抗体と一緒に、ヒトNKをインキュベートする。結合後、NK細胞をOVCAR細胞(CA125及びメソテリンを発現する上皮卵巣癌細胞(EOC)株)と一緒に培養する。細胞をヨウ化プロピジウムで染色した後、FACSにより分析する。CA125ターゲティングNK細胞が、OVCAR細胞をin vitroで死滅させる能力を確認する。
患者から得たヒトT細胞を用いて、前記実施例を繰り返し、培養により拡大する。
III期原発性卵巣癌を有する患者からNK細胞を単離し、培養して拡大する。細胞のアリコートを、ソルターゼ、並びにソルターゼ認識配列を含むように修飾した抗CA-125抗体と一緒にインキュベートする。第2アリコートを、ソルターゼ、並びにソルターゼ認識配列を含むように修飾した抗メソテリン抗体と一緒にインキュベートする。
患者から得たPBMCを用いる以外は、前記実施例を繰り返す。
NK細胞、この場合、NK-92細胞を用いる以外は、前記実施例を繰り返す。
III期原発性卵巣癌を有する患者から得たヒトT細胞を、キメラT細胞受容体(CD3-ε及びCD28のシグナル伝達ドメインを含む、TCRの膜貫通及び細胞内ドメインに融合した抗CA125一本鎖Fv領域)を発現するように、遺伝子改変する。抗CD3-抗CD28ビーズとの培養で細胞を10日間拡大してから、ソルターゼ、並びにC末端にソルターゼ認識配列を有する単一ポリペプチドとしてヒトインターロイキン-12の2つのサブユニットを含む融合タンパク質と一緒にインキュベートした後、手術後の患者に戻して移入する。12週間にわたって毎週治療を繰り返す。残留する癌及び転移に対する投与細胞の効果をモニターする。CA-125の血中レベルについて患者をモニターし、高いCA-125レベルが検出されたら、再治療する。
CA-125及びCD3εに特異的な単一ドメイン抗体を組み換え細胞発現系において作製する。C末端にソルターゼ認識配列を組み込んだCD3εに特異的なsdAbを作製する。ソルターゼを用いて、クリックケミストリーハンドルを各sdAbのN末端に設置する。次に、2つのsdAbを互いに結合させて、二重特異性抗体を作製する。III期原発性卵巣癌を有する患者から取得したヒトT細胞を、CD3-CD28ビーズと一緒に培養して拡大した後、ソルターゼ及び二重特異性抗体と一緒にインキュベートした後、手術後の患者に戻して移入する。12週間にわたって毎週治療を繰り返す。残留する癌及び転移に対する投与細胞の効果をモニターする。CA-125の血中レベルについて患者をモニターし、高いCA-125レベルが検出されたら再治療する。
B細胞急性リンパ芽球性白血病を有する患者から取得したヒトT細胞を、抗CD3/抗CD28ビーズとの培養で10日間拡大してから、CD19に結合するscFvとソルタギングした後、患者に戻して移入する。白血病に対する投与細胞の効果をモニターする。
B細胞急性リンパ芽球性白血病を有する患者から取得したヒトT細胞を、キメラ抗原細胞受容体(CD3-ζ及びCD28のシグナル伝達ドメインを含む、TCRの膜貫通及び細胞内ドメインに融合した抗CD19一本鎖Fv領域)を発現するように、遺伝子改変する。抗CD3/抗CD28ビーズとの培養で細胞を10日間拡大しから、CD22に結合するscFvとソルタギングした後、患者に移入する。白血病に対する投与細胞の効果をモニターする。
B細胞急性リンパ芽球性白血病を有する患者から取得したヒトT細胞を、キメラ抗原細胞受容体(CD3-ζ及びCD28のシグナル伝達ドメインを含む、TCRの膜貫通及び細胞内ドメインに融合した抗CD22一本鎖Fv領域)を発現するように、遺伝子改変する。抗CD3/抗CD28ビーズとの培養で細胞を10日間拡大しから、CD19に結合するscFvとソルタギングした後、患者に移入する。白血病に対する投与細胞の効果をモニターする。
進行性卵巣癌を有する患者に対し、手術後、以下:(1)プラチナ-タキサンを単独で用いた標準的アジュバント化学療法;(2)プラチナ-タキサンによる標準的アジュバント化学療法に加えて、遺伝子改変はせずに、ソルターゼを用いて抗CA-125抗体と結合させたオートロガスT細胞の静脈内投与による治療;又は(3)プラチナ-タキサンによる標準的アジュバント化学療法に加えて、遺伝子改変はせずに、ソルターゼを用いて抗CA-125抗体と結合させたオートロガスT細胞の静脈内及び腹腔内投与の両方による治療の何れかをランダムに受けさせる。3つの治療群における無増悪生存期間及び5年生存率をモニターし、比較する。
B細胞慢性リンパ芽球性白血病を有する患者及び免疫適合性ドナーからヒト赤血球を取得し、ソルターゼ、並びに(i)PD-1の細胞外ドメイン及び(ii)ソルターゼ認識配列を含む薬剤と一緒にインキュベートする。ソルタギングの後、赤血球を患者に移入する。白血病に対する投与細胞の効果をモニターする。
B細胞慢性リンパ芽球性白血病を有する患者及び免疫適合性ドナーから取得したヒト赤血球を、ソルターゼ、並びにPD-1に結合し、かつ、ソルターゼ認識配列を含むように修飾されたscFvを含む薬剤と一緒にインキュベートする。ソルタギングの後、赤血球を患者に移入する。白血病に対する投与細胞の効果をモニターする。
骨髄腫患者又は免疫適合性ドナーから赤血球を取得する。これらの細胞を、ソルターゼ、並びにソルターゼ認識配列を含むように修飾された抗CS-1又は抗BCMA抗体と一緒にインキュベートした後、患者に移入する。この治療を12週間にわたって毎週繰り返す。多発性骨髄腫に対する投与細胞の効果をモニターする。
赤血球をIII期結腸癌と最近診断された患者から単離するか、又は免疫適合性ドナーから取得する。これらの細胞を、ソルターゼ、並びにソルターゼ認識配列を含むように修飾された抗CS-1又は抗BCMA抗体とインキュベートした後、患者に移入する。この治療を12週間にわたって毎週繰り返す。局所再発又は転移の存在について患者をモニターする。
ソルターゼを用いて、ソルターゼ認識配列を含むように修飾されたミエリン塩基性タンパク質(MBP)又はソルターゼ認識配列を含むように修飾されたオボアルブミンの何れかを、SJLマウスから得られた非遺伝子操作RBCに結合させる。
黒色腫と診断された患者から単離するか、又は免疫適合性ドナーから取得したT細胞を拡大し、黒色腫に対する抗体、例えば、MART-1、並びに2つのチェックポイント阻害剤、例えば、イピリムマブ(抗CTLA4)及び抗PD1ニボルマブとソルタギングする。ソルタギング済みT細胞を、黒色腫患者に投与する。黒色腫に対する投与細胞の効果をモニターする。
HER2陽性乳癌と診断された患者から単離するか、又は免疫適合性ドナーから取得したRBC細胞をハーセプチン(Herceptin)、抗PD1抗体、及び抗VEGFR2抗体とソルタギングして、HER2陽性乳癌の患者に投与する。癌に対する投与細胞の効果をモニターする。
HER2陽性乳癌と診断された患者から単離するか、又は免疫適合性ドナーから取得したRBC細胞をTRAIL及びハーセプチン(Herceptin)とソルタギングして、HER2陽性乳癌の標準的化学療法/ハーセプチン療法の補助薬として投与する。癌に対する投与細胞の効果をモニターする。
成人患者の再発性/難治性前駆B細胞性ALLは、予後不良で、しかもアンメット・メディカル・ニーズ(unmet medical need)の侵襲性悪性疾患である。ブリナツモマブ(B細胞とT細胞を連結させて、T細胞活性化及びCD19発現細胞に対する傷害性T細胞応答をもたらすように設計された二重特異性一本鎖抗体構築物)を担持するようにソルタギングした赤血球を、再発性/難治性前駆B細胞性ALLの成人患者に投与する。患者は、最大5カ月サイクルの静脈内RBC-ブリナツモマブ治療を受ける。
前述したように、ソルタギングによるCD8T細胞の修飾は、細胞傷害機能を顕著には妨害しない。寄生体による宿主細胞への侵入は、別のタイプの細胞-細胞相互作用を表している。ソルターゼを用いて修飾したトキソプラズマ(Toxoplasma gondii)タキゾイト(tachyzoite)が、細胞に侵入することができるかどうかを調べるために、寄生体をTAMRA-修飾LPETGペプチドでソルタギングしてから、これらをヒト包皮線維芽細胞と一緒にインキュベートした。タキゾイト(ミリリットル当たり2千万~4千万個)をHHE(ハンクス(Hanks)バッファー+Hepes+EDTA)中の500μMのTAMRA-LPETG及び20μMのCa2+非依存性ソルターゼA(実施例11で用いるものと同じ)と15分インキュベートした。次に、寄生体を洗浄した後、ヒト包皮線維芽細胞(HFF)と一緒にインキュベートした。ソルタギングした寄生体を蛍光顕微鏡検査により視覚化して、線維芽細胞に侵入するそれらの能力をモニターし、記録した。ソルタギングした寄生体は、線維芽細胞に完全に侵入することができることが認められた(図8(A);要請に応じてビデオ閲覧可能)。
Claims (26)
- ヒト細胞にソルターゼ認識配列を介して結合された薬剤を有するヒト細胞であって、前記薬剤が、前記ヒト細胞の内在性の非操作ポリペプチドの細胞外部分と結合しており、前記ヒト細胞が、ソルターゼ認識配列を含むポリペプチドを発現するように遺伝子操作されておらず、前記薬剤が、サイトカイン、抗原、共刺激分子、又はアジュバントを含むことを特徴とするヒト細胞。

- 請求項1に記載のヒト細胞において、前記ヒト細胞が、骨髄系前駆細胞又はリンパ球前駆細胞であることを特徴とするヒト細胞。
- 請求項1に記載のヒト細胞において、前記ヒト細胞が、赤血球であることを特徴とするヒト細胞。
- 請求項1に記載のヒト細胞において、前記ヒト細胞が、リンパ球、単球、樹状細胞、マクロファージ、好中球、マスト細胞、好酸球、好塩基球、及びナチュラルキラー(NK)細胞からなる群から選択されることを特徴とするヒト細胞。
- 請求項1に記載のヒト細胞において、前記薬剤が、共刺激分子を含むことを特徴とするヒト細胞。
- 請求項9に記載のヒト細胞において、前記共刺激分子が、CD28ファミリー受容体に結合する分子、CD2ファミリー受容体に結合する分子、ICOSに結合する分子、CD27に結合する分子、又は4-1BB(CD137)に結合する分子であることを特徴とするヒト細胞。
- 請求項9に記載のヒト細胞において、前記共刺激分子が、B7分子、ICOSリガンド、TNFαファミリーメンバー、4-1BBL、又はOX40Lであることを特徴とするヒト細胞。
- 請求項11に記載のヒト細胞において、前記共刺激分子が、4-1BBLであることを特徴とするヒト細胞。
- 請求項1に記載のヒト細胞において、前記薬剤が、アジュバントを含むことを特徴とするヒト細胞。
- 請求項13に記載のヒト細胞において、前記アジュバントが、CD40リガンド、抗CD40抗体、TLRのリガンド、病原体由来分子パターン(PAMP)、PAMP模倣物、免疫刺激核酸、又はカチオンポリマーであることを特徴とするヒト細胞。
- 請求項13に記載のヒト細胞において、前記アジュバントが、TLRのリガンドを含み、前記TLRが、TLR3、TLR4、及び/又はTLR9であることを特徴とするヒト細胞。
- 請求項14に記載のヒト細胞において、前記アジュバントが、カチオンポリマーを含み、前記カチオンポリマーが、ポリ(アミノ酸)であることを特徴とするヒト細胞。
- 請求項1に記載のヒト細胞において、前記薬剤が、サイトカインを含むことを特徴とするヒト細胞。
- 請求項17に記載のヒト細胞において、前記サイトカインが、インターロイキン、TNFα、コロニー刺激因子、インターフェロンα2a、インターフェロンα2b、インターフェロンβ-1a、インターフェロンβ-1b、白血病阻害因子(LIF)、又はオンコスタチンMであることを特徴とするヒト細胞。
- 請求項18に記載のヒト細胞において、前記サイトカインが、インターロイキンであり、前記インターロイキンが、IL-1、IL-2、IL-3、IL-4、IL-5、IL-6、IL-7、IL-9、IL-10、IL-11、IL-12、IL-13、IL-14、膜結合IL-15、IL-15、IL-17、IL-21、IL-23、IL-27、IL-35、又はIL-38であることを特徴とするヒト細胞。
- 請求項18に記載のヒト細胞において、前記サイトカインが、インターロイキンであり、前記インターロイキンが、IL-15又は膜結合IL-15であることを特徴とするヒト細胞。
- 請求項18に記載のヒト細胞において、前記サイトカインが、コロニー刺激因子であり、前記コロニー刺激因子が、顆粒細胞コロニー刺激因子(G-CSF)、顆粒細胞マクロファージコロニー刺激因子(GM-CSF)、又はマクロファージコロニー刺激因子(M-CSF)であることを特徴とするヒト細胞。
- 請求項1に記載のヒト細胞において、前記ソルターゼ認識配列が、配列LPXTGを含むことを特徴とするヒト細胞。
- 対象の免疫反応を調節するための、請求項1乃至22の何れか1項に記載のヒト細胞。
- 疾患を有する対象を治療するための、請求項1乃至22の何れか1項に記載のヒト細胞。
- 請求項24に記載のヒト細胞において、前記疾患が、感染性疾患、癌、自己免疫疾患、アレルギー、炎症状態、又は免疫不全を含むことを特徴とするヒト細胞。
- ヒト細胞に薬剤を結合させる方法であって、当該方法が、前記ヒト細胞を、ソルターゼ認識配列及び薬剤の両方を含むソルターゼ基質と接触させるステップを備え、
前記接触させるステップが、ソルターゼの存在下で、前記ソルターゼが前記ソルターゼ基質を前記ヒト細胞の内在性の非操作ポリペプチドの細胞外部分と結合させるのに好適な条件下で行われ、前記ヒト細胞が、ソルターゼ認識配列を含むポリペプチドを発現するように遺伝子操作されておらず、前記薬剤が、サイトカイン、抗原、共刺激分子、又はアジュバントを含むことを特徴とする方法。
