Description
脂肪組織由来間質血管分画(SVF)細胞は、下記の段階を通じて収得した。
無菌手術室で医師の主導の下に脂肪吸引術を実施して脂肪組織を抽出した後、これを生理食塩水に混合した。そして、12,000rpm~20,000rpm条件のホモジナイザー(homogenizer)を用いて機械的に脂肪組織からECMを分離した後、これを遠心分離した。遠心分離後の沈殿物をさらに遠心分離する過程を3回~5回繰り返した後、最終的にECMを収得した。
図1は、本発明によるオーダーメード型真皮再生シートを製造するために、第1液および第2液をそれぞれ準備する過程を示すものである。具体的に、前記収得されたSVFとECMを混合注射器(mix syringe)を用いて混合した後、これに4.5mg/mlのフィブリノーゲンが混合されたアプロチニン液1mlと混合注射器(mix syringe)を用いて混合して、第1液を製造した。この際、第1液内のECMの含量を図2のように10wt%または20wt%に調節した。
本発明による真皮再生シートの効能を検証するために、4ヶ月齢の豚の皮膚を2.5cm×2.5cmのサイズに除去した後、前述した方法のように、豚の脂肪組織からSVFおよびECMを収得した。図3は、実験例2のための動物実験での皮膚を除去した写真を示すものである。

- 脂肪組織の間質血管分画、細胞外基質、およびフィブリノーゲンを含む第1液と、
トロンビンを含む第2液と、を含み、
前記第1液での前記細胞外基質の含量は、20wt%~60wt%であり、
前記第2液での前記トロンビンの濃度は、40IU/ml~250IU/mlである、真皮再生シート用バイオインク組成物。 - 前記第1液での脂肪組織の間質血管分画の濃度は、105個/ml~107個/mlであることを特徴とする、請求項1に記載の真皮再生シート用バイオインク組成物。
- 前記細胞外基質は、5μm~100μmの直径の粒子であることを特徴とする、請求項1に記載の真皮再生シート用バイオインク組成物。
- 前記第1液でのフィブリノーゲンの濃度は、4mg/ml~50mg/mlであることを特徴とする、請求項1に記載の真皮再生シート用バイオインク組成物。
- 前記第1液は、アプロチニンをさらに含むことを特徴とする、請求項1に記載の真皮再生シート用バイオインク組成物。
- (a)3Dスキャナを用いて、欠損された真皮部位の3次元データを収得する段階と、
