前述ベクターは、例えばPvuI(NEB)などの酵素消化部位がベクターバックボーンの原核領域のみにある制限エンドヌクレアーゼを使用して線形化され、37℃で一晩保存した。翌日にフェノール‐クロロホルム法によりDNAを回収してトランスフェクトに用いた。
Claims (16)
- 外来ヌクレオチド配列と、SEQ ID NO:4、5、6、7、9、17、20、21、22及び23のいずれかのヌクレオチド配列とを含むベクター。
- 前記ベクターが組換え発現ベクターである請求項1に記載のベクター。
- (iii)~(iv)からなる群から選択されるヌクレオチド配列をさらに含む、
(iii)SEQ ID NO:13に対して少なくとも90%の配列同一性を有する配列;
(iv)SEQ ID NO:13の逆相補配列に対して少なくとも90%の配列同一性を有する配列;
である、請求項1または2に記載のベクター。 - CMVプロモーターをさらに含む、請求項1~3のいずれか1項に記載のベクター。
- 前記CMVプロモーターは、SEQ ID NO:16に対して少なくとも90%の配列同一性を有するヌクレオチド配列を含む、請求項4に記載のベクター。
- 前記CMVプロモーターは、SEQ ID NO:16と同一であるヌクレオチド配列を含む、請求項4に記載のベクター。
- 前記(iii)~(iv)からなる群から選択されるヌクレオチド配列は、SEQ ID NO:13と同一である、請求項3~6のいずれか1項に記載のベクター。
- 前記ヌクレオチド配列は、SEQ ID NO:4である、請求項1~7のいずれか1項に記載のベクター。
- 前記ヌクレオチド配列は、SEQ ID NO:5である、請求項1~7のいずれか1項に記載のベクター。
- 前記ヌクレオチド配列は、SEQ ID NO:6である、請求項1~7のいずれか1項に記載のベクター。
- 前記ヌクレオチド配列は、SEQ ID NO:7である、請求項1~7のいずれか1項に記載のベクター。
- 前記ヌクレオチド配列は、SEQ ID NO:9である、請求項1~7のいずれか1項に記載のベクター。
- SEQ ID NO:13に対して少なくとも90%の配列同一性を有するヌクレオチド配列と、SEQ ID NO:4、5、6、7、9、17、20、21、22及び23のいずれかのヌクレオチド配列とを含むベクター。
- CMVプロモーターをさらに含む、請求項13に記載のベクター。
- CMVプロモーターと、SEQ ID NO:4、5、6、7、9、17、20、21、22及び23のいずれかのヌクレオチド配列とを含むベクター。
- 前記CMVプロモーターは、SEQ ID NO:16に対して少なくとも90%の配列同一性を有するヌクレオチド配列を含む、請求項15に記載のベクター。
