Description
炒り工程によって1次加工された緑茶の葉と、70%エタノールとを抽出槽に入れて、60℃で3時間抽出した。抽出液から液状部分を分離して濾過した後、酸性白土を添加して撹拌し、次いで、ろ紙を利用して濾過した後に濃縮し、噴霧乾燥して粉末状の緑茶葉抽出物を製造した。製造された緑茶葉抽出物内に含まれたカテキンの含量は、抽出物の全体の重量を基準にして35重量%である。
発酵アルコールを利用して、乾燥した黄漆木の葉を1次抽出した後に、濾過することで1次抽出物を得た。前記1次抽出物を、精製水を利用して2次抽出した後、濾過、濃縮して2次抽出物を得た。前記2次抽出物を殺菌及び乾燥して、粉末状の黄漆木の葉の抽出物を製造した。
発酵アルコールを利用して、粉砕されたたまねぎを抽出した後、濾過及び濃縮を行い、カラム精製及び乾燥を行なうことで粉末状のたまねぎ抽出物を製造した。
下記表1に記載したような組成にしたがって、製造例1ないし3でそれぞれ製造された、緑茶の葉の抽出物と、黄漆木の葉の抽出物と、たまねぎの抽出物とを混合し、カテキン吸収率増進剤を製造した。
製造例1で製造された、カテキンを有効成分として含む緑茶の葉の抽出物を対照群として準備した。
下記表2に記載した組成にしたがって、製造例1及び2でそれぞれ製造された、緑茶の葉の抽出物と、黄漆木の葉の抽出物とを混合して、カテキン吸収率増進剤を製造した。
下記表3に記載した組成にしたがって、製造例1及び3でそれぞれ製造された、緑茶の葉の抽出物と、たまねぎの抽出物とを混合して、カテキン吸収率増進剤を製造した。
実施例及び比較例でそれぞれ製造された増進剤を、In vitroでの生体消化評価モデルである人工消化液で3時間連続培養したのであり、小腸段階の後の消化産物を取って、UPLC-PDA-ESI-MS/MSn装備を利用して、カテキン4種の含量を分析し、消化器官内安定性評価を実施した。前記UPLC-PDA-ESI-MS/MSn装備の分析条件は、下記表3に記載したところである。この際、前記カテキン4種は、(-)-エピカテキン(Epicatechin、EC)、(-)-エピガロカテキン(Epigallocatechin、EGC)、(-)-エピカテキンガレート(Epicatechin gallate、ECG)及び(-)-エピガロカテキンガレート(Epigallocatechin gallate、EGCG)である。
実施例及び比較例でそれぞれ製造された増進剤に対し、カテキンの小腸上皮細胞での吸収率を調べるため、小腸細胞(Caco-2)を選定し、大きさ6-ウェルのモノプレート(mono plate、Cat.140675、sterile、Nunclon)を利用して実験を実施した。
緑茶抽出物60重量%、カテキン吸収率増進用添加剤2重量%、アスコルビン酸2重量%、及び結晶セルロース34.1重量%、二酸化ケイ素1.0重量%、及びステアリン酸マグネシウム0.9重量%を常法で打錠して錠剤を製造する。
緑茶抽出物28重量%、カテキン吸収率増進用添加剤2重量%、クエン酸10重量%、キシリトール10重量%、とうもろこし澱粉10重量%、グリセリン20重量%及びソルビトール20重量%を混合し、製丸器を使って丸剤を製造する。
緑茶抽出物18重量%、カテキン吸収率増進用添加剤2重量%、アスコルビン酸10重量%、キシリトール10重量%、酵素処理ステビア5重量%、及びイソマルト55重量%を混合し、流動層顆粒器を使って顆粒に成形した後、包材に充填する。
緑茶抽出物10重量%、カテキン吸収率増進用添加剤1重量%、酵素処理ステビア10重量%、グレープフルーツ濃縮液10重量%及び精製水残量を混合した後、通常の健康飲料の製造方法にしたがい、1時間85℃で撹拌加熱し、得た溶液を濾過して、滅菌された容器に取り、密封・滅菌した後、冷蔵保管して健康飲料組成物の製造に用いる。
Claims (7)
- カテキンを有効成分として含む緑茶の葉の抽出物;黄漆木(Dendropanax morbiferus)の葉の抽出物;及びたまねぎの皮の抽出物;を含む小腸上皮細胞でのカテキン吸収率が増進されたカテキン剤。
- 前記カテキンを有効成分として含む緑茶の葉の抽出物100重量部に対して、黄漆木の葉の抽出物と、たまねぎの皮の抽出物との混合物1ないし4重量部を含む請求項1に記載の小腸上皮細胞でのカテキン吸収率が増進されたカテキン剤。
- 前記カテキンを有効成分として含む緑茶の葉の抽出物100重量部に対して、黄漆木の葉の抽出物と、たまねぎの皮の抽出物との混合物2ないし4重量部を含む請求項1に記載の小腸上皮細胞でのカテキン吸収率が増進されたカテキン剤。
- 前記黄漆木の葉の抽出物とたまねぎの皮の抽出物との重量比は、1:1ないし1:4であることを特徴にする請求項2に記載の小腸上皮細胞でのカテキン吸収率が増進されたカテキン剤。
- 請求項1ないし請求項4のいずれか一項に記載のカテキン吸収率が増進されたカテキン剤を含む抗酸化健康食品組成物。
- 前記抗酸化健康食品組成物は、錠剤、液状、カプセル、丸剤、顆粒剤、ドリンク剤、キャラメル、ダイエットバー、ゼリー、口腔フィルム及びティーバッグからなる群から選択された1種以上として剤形化されることを特徴にする請求項5に記載の抗酸化健康食品組成物。
- 請求項1ないし請求項4のいずれか一項に記載のカテキン吸収率が増進されたカテキン剤を含む抗酸化薬学組成物。
