Description
1)幹細胞を増殖する段階;
2)前記増殖された幹細胞を軟骨細胞に分化させる段階;および
3)軟骨細胞に分化されている幹細胞からエキソソームを分離および精製する段階;を含む方法を用いることができる。
軟骨細胞に分化されている幹細胞からエキソソームを分離するために、ヒト脂肪由来幹細胞を一般培養培地[10%ウシ胎児血清、1%ペニシリン/ストレプトマイシンを含有するDMEM(Dulbecco Modified Eagle Medium high glucose)]で80〜90%程度増殖させた後、分化培地[5%ウシ胎児血清、1%ペニシリン/ストレプトマイシン、100nM デキサメタゾン(dexamethasone)、0.15mM アスコルビン酸(ascorbic acid)、1X ITS(Insulin−Transferrin−Sodium selenite)、10ng/mL TGF−β1(Transforming Growth Factor β1)を含有するDMEM(Dulbecco Modified Eagle Medium high glucose)]に交換し、合計5週間培養することにより、軟骨細胞への分化を誘導した。
軟骨細胞に分化されている幹細胞由来のエキソソームの効能を比較するために増殖するヒト脂肪由来幹細胞からエキソソーム(Adipose tissue−derived stem cell、ASC−EXO)を分離して比較群として使用した。具体的には、分化培地を使用しないことを除いては、実施例1と同じ方法を実行して増殖するヒト脂肪由来幹細胞からエキソソームを分離した。
実施例1の軟骨細胞に分化されている幹細胞から分離されたエキソソームと増殖する幹細胞から分離されたエキソソームを透過電子顕微鏡(transmission electron microscope)とナノ粒子分析器(dynamic light scattering)を使用して大きさおよび形状を確認し、特定のタンパク質の発現有無を示すエキソソームアンチボディアレイ(Exo−CheckTM exosome antibody arrays)を用いて、エキソソーム膜表面タンパク質を確認した。
エキソソームを用いて、ヒト脂肪由来幹細胞の軟骨細胞への分化を誘導するために、増殖している幹細胞由来のエキソソーム(ACS−EXO)を含む培地組成物と、軟骨細胞に分化されている幹細胞由来のエキソソーム(Chondro−EXO)を含む培地組成物を用いた。前記培地組成物は、10μg/mLの濃度のエキソソームを幹細胞培養培地[5%ウシ胎児血清、1%ペニシリン/ストレプトマイシンを含有するDMEM(Dulbecco Modified Eagle Medium high glucose)]に追加して使用した。
エキソソームを用いた幹細胞の軟骨細胞への分化誘導を確認するために、軟骨細胞に分化されている幹細胞由来のエキソソーム(Chondro−EXO)を含む培地組成物を用いた。前記培地組成物は、軟骨細胞に分化される幹細胞由来のエキソソームを5、10、30、50μg/mLの濃度で幹細胞培養培地[5%ウシ胎児血清、1%ペニシリン/ストレプトマイシンを含有するDMEM(Dulbecco Modified Eagle Medium high glucose)]に追加して使用した。
骨関節炎(osteoarthritis)モデルに通常使用されるDMM(Destabilization of the medial meniscus)関節炎誘発モデルマウス(mouse)を用いて、軟骨細胞に分化されている幹細胞由来のエキソソームの実際の生体内軟骨組織再生効果を確認した。

- 軟骨細胞へ分化中の脂肪由来幹細胞由来のエキソソームを有効成分として含む軟骨細胞への分化誘導または軟骨組織再生用組成物であって、
前記軟骨細胞へ分化中の脂肪由来幹細胞は、前軟骨凝縮(precartilage condensation)が現れる時期から軟骨細胞に分化した状態になる直前までの時期の細胞を意味する、軟骨細胞への分化誘導または軟骨組織再生組成物。 - 前記脂肪由来幹細胞は、ヒトまたは動物由来の幹細胞である、請求項1に記載の軟骨細胞への分化誘導または軟骨組織再生用組成物。
- 請求項1または請求項2に記載の組成物を含む軟骨細胞への分化誘導用培地組成物。
- 前記培地組成物は、エキソソームが1〜100μg/mLの濃度で含まれるものである、請求項3に記載の軟骨細胞への分化誘導用培地組成物。
- 請求項1または請求項2に記載の組成物を含む軟骨組織再生用注射剤。
- 前記注射剤は、エキソソームが1〜1000μg/mLの濃度で含まれるものである、請求項5に記載の軟骨組織再生用注射剤。
- 請求項1または請求項2に記載の組成物を含む軟骨疾患治療用薬剤学的組成物。
