Description

本開示は、4−アルコキシ−3−ヒドロキシピコリン酸の製造方法に関する。より詳細には、本開示は、2−置換フランからの4−アルコキシ−3−ヒドロキシピコリン酸の製造方法に関する。

概要
本開示は、式A
a)臭素化剤、塩基および水を式B
b)混合物から、式Aの化合物を単離するステップ、を含む方法で製造される。
a)式I
b)第1の混合物から、式D
c)式Dの化合物をアルコールおよび酸化合物または酸形成化合物と混合し、次いで加熱して、第2の混合物を形成するステップ、ならびに
d)第2の混合物から式E
e)還元剤を式Eの化合物に添加して、第3の混合物を形成するステップ、
f)第3の混合物から、式F
g)鉱酸を式Fの化合物に添加して、第4の混合物を形成するステップ、ならびに
h)第4の混合物から、式Bの化合物を単離するステップ、を含む方法で製造される。

b)第1の混合物から、式H
c)鉱酸を式Hの化合物に添加して、第2の混合物を形成するステップ、ならびに
d)第2の混合物から、式Bの化合物を単離するステップ、を含む方法で製造してもよい。一部の実施形態において、本方法で使用するルイス酸は三フッ化ホウ素エーテラートである。一部の実施形態において、酸開裂性基はベンジル基である。また、一部の実施形態において、本方法で使用する強酸は、塩酸および臭化水素酸からなる群から選択される少なくとも1種の酸である。
本明細書で使用される用語「単離する」、「単離すること」または「単離」は、限定するものではないが、ろ過、抽出、蒸留、結晶化、遠心分離、粉砕、液−液相分離または当業者に公知の他の方法等の標準的な方法を使用して、終了した化学的プロセスの混合物の他の成分から、所望の生成物を、部分的または完全に取り出すことを意味する。単離した生成物は、≦50%〜≧50%の範囲の純度を有していてよく、標準的な精製方法を使用して、より高純度に精製してよい。単離した生成物は、精製して、または精製することなく、次のプロセスステップで使用してもよい。
本明細書に記載の方法において、式Aの4,6−ジブロモ−3−ヒドロキシピコリネートエステルは、臭素化−転位反応の使用による1つの化学的ステップで、式Bのアルキル2−アミノ−2−(フラン−2−イル)アセテートのハロゲン化水素酸塩から製造される。出発のHC1またはHBr塩のいずれかとしての式Bのフラン化合物(R2はC1〜C4アルキルを示す)を、臭素、1,3−ジブロモ−5,5−ジメチルヒダントイン、またはN−ブロモスクシンイミド等の適切な臭素化剤で処理する。反応は、好ましくは、約4モル当量の臭素を使用して行う。反応が終了まで進行することを確実にするために、5%、10%または15%モル過剰等の過剰の臭素化剤を使用することが好都合であり得る。
式EのO−アルキルオキシムエステルは、最初に、O−アルキル−ヒドロキシルアミンのハロゲン化水素酸塩、2−(フラン−2−イル)−2−オキソ酢酸(式C)、塩基および溶媒を一緒に混合し、得られた混合物を加熱して、式Dのオキシム酸を得ることによって製造される(化学的ステップaおよびb)。当該反応において、1〜約3モル当量のO−アルキル−ヒドロキシルアミンのハロゲン化水素酸塩(R3はC1〜C4アルキルである)および約2〜約6モル当量の塩基を使用してよい。適切な塩基としては、トリアルキルアミン、炭酸ナトリウム、または炭酸カリウム等のアルカリ金属炭酸塩等が挙げられる。適切な溶媒としては、メタノール、エタノール、または2−プロパノール等のアルコールが挙げられる。本反応は、典型的には、実質的に均一な反応物の混合物を維持するために、十分な撹拌を伴って行い、終了まで進行させるために、一般に約1〜約10時間、好ましくは約1〜約5時間が必要である。
経路B(スキーム1)を用いて、式Gの2−メトキシアミノ酸誘導体を、ルイス酸の存在下でフランとカップリングして、式Hの2−置換フランを得る。式Gの化合物(式中、R2はC1〜C4アルキルであり、R4は、アリル基、ベンジル基、または置換アリル基もしくは置換ベンジル基から選択される酸開裂性基である)を溶媒中に入れ、次いで、室温で、ルイス酸で処理し、続いてすぐにフランを添加する。当該反応に使用するための適切な溶媒としては、ジエチルエーテル、メチルt−ブチルエーテル、2−メチルフラン、ジオキサン等が挙げられる。適切なルイス酸としては、三フッ化ホウ素エーテラート、三塩化アルミニウム、四塩化スズ等が挙げられる。当該カップリング反応において、化合物Gに対して、約1.0〜2.0、好ましくは約1.2〜1.7モル当量のルイス酸、および約2.0〜6.0、好ましくは約3.0〜5.0モル当量のフランを典型的には使用する。反応は、典型的には、約10時間〜約48時間、好ましくは約18〜約32時間、室温で行う。反応が終了した後、標準的な単離および精製手法を使用することによって式Hの2−置換フランを回収する。
Claims (3)
- 式A
[式中、R2はC1〜C4アルキルである]の化合物の製造方法であって、以下のステップ:
a)臭素化剤および水を式B
[式中、XはClまたはBrであり、R2はC1〜C4アルキルである]の化合物に添加することによって混合物を作製するステップ、ならびに
b)前記混合物から、前記式Aの化合物を単離するステップ、
を含む、方法。 - 前記臭素化剤が臭素である、請求項1に記載の方法。
- さらに塩基を含有する、請求項1に記載の方法。
