(1) アルデヒド、マレイミドまたはスクシンイミド反応基を有する非ペプチド性重合体をオキシントモジュリン誘導体ペプチドのアミン基またはチオール基に共有結合で連結するステップ、
(2)ステップ(1)の反応混合物からアミノ末端以外の位置に非ペプチド性重合体が共有結合されたオキシントモジュリン誘導体ペプチドを分離するステップ、および
(3) 連結された非ペプチド性重合体のもう一方の末端に免疫グロブリンFc領域を共有結合で連結して、非ペプチド性重合体の両末端がそれぞれ免疫グロブリンFc領域およびオキシントモジュリン誘導体ペプチドと結合したペプチド結合体を生成するステップを含むことができる。
(1)各末端にアルデヒド反応基とマレイミド反応基を有する非ペプチド性重合体をそれぞれオキシントモジュリン誘導体のシステイン残基に共有結合で連結するステップ、
(2)ステップ(1)の反応混合物からシステイン残基に非ペプチド性重合体が共有結合されたオキシントモジュリン誘導体を含む結合体を分離するステップ、および
(3) 非ペプチド性重合体のもう一方の末端に免疫グロブリンFc領域を共有結合で連結して、非ペプチド性重合体の両末端がそれぞれ免疫グロブリンFc領域およびオキシントモジュリン誘導体ペプチドと連結されたペプチド結合体を生成するステップを含むことができる。
実施例1-1:GLP-1に対してcAMP反応を示す細胞株の生産
ヒトGLP-1受容体遺伝子のcDNA(OriGene Technologies、Inc. USA)のORF(open reading frame)に該当する部分を鋳型として、HindIII切断部位とEcoRI切断部位をそれぞれ含む正方向プライマーおよび逆方向プライマーを用いてPCRを行って、PCR産物を得た。
逆方向プライマー:5’-GAACGGTCCGGAGGACGTCGACTCTTAAGATAG-3 ‘(配列番号51)
ヒトグルカゴン受容体遺伝子のcDNA(OriGene Technologies、Inc. USA)からORFに(open reading frame)対応する部分を鋳型として、EcoRI切断部位とXhoI切断部位をそれぞれ含む正方向プライマーおよび逆方向プライマーを用いてPCRを行い、PCR産物を得た。
逆方向プライマー:5’-CTAACCGACTCTCGGGGAAGACTGAGCTCGCC-3 ‘(配列番号53)
実施例2-1:オキシントモジュリン誘導体の合成
オキシントモジュリン誘導体のin vitro活性を測定するために、下記アミノ酸配列を有するオキシントモジュリン誘導体を合成した(表1)。
抗肥満ペプチドの効果を測定するために、前記実施例1-1と1-2で製造した形質転換体を用いてin vitroにおける細胞活性を測定した。
MAL-10K-ALD PEG(NOF., Japan)をオキシントモジュリン誘導体(配列番号23)のアミノ酸配列24番のシステイン残基にペグ化するために、オキシントモジュリン誘導体(配列番号23)とMAL-10K-ALD PEGのモル比を1:3、タンパク質の濃度を3mg/mlとし、室温で3時間反応させた。この際、反応はグアニジン1Mを含んだ50mMTris緩衝液(pH8.0)において行われた。反応終了後、前記反応液はSOURCE Sを使用してシステインにモノペグ化されたオキシントモジュリン誘導体を精製した(column:SOURCE S、流速:2.0m/分、勾配:A0→100% 50分 B(A:20mM クエン酸ナトリウム、pH3.0+45%エタノール、B:A +1M KCl))。
MAL-10K-ALD PEGをオキシントモジュリン誘導体(配列番号25)のアミノ酸配列30番のシステイン残基にペグ化するために、オキシントモジュリン誘導体(配列番号25)とMAL-10K-ALD PEGのモル比を1:3、タンパク質の濃度を3mg/mlとし、室温で3時間反応させた。この際、反応は1Mグアニジンを含む50mのMTris緩衝液(pH8.0)において行われた。反応終了後、前記反応液をSOURCE Sを使用して、システインにモノペギル化オキシントモジュリン誘導体を精製した(column:SOURCE S、流速:2.0ml/分、勾配:A0→100% 50分 B(A:20mM クエン酸ナトリウム、pH3.0+45%エタノール、B:A +1M KCl))。
実施例5-1:実験動物の分類
8週齢の雄ハムスター(Golden Syrian hamster、120-130g)は、中国のVital River Chinaから購入した。ハムスターは血中脂質プロファイルが、他のげっ歯類とは異なりヒトと類似しており、高脂肪の食餌に敏感な動物として知られている。
本発明による持続型オキシントモジュリン誘導体の結合体がハムスターの脂質減少に及ぼす影響を調べるために、次のような実験を行った。前記実施例5-1で分類した持続型オキシントモジュリン誘導体の投与または非投与ハムスターの血液を採取して血液内の脂質含有量を分析した(HITACHI7020を使用)。分析結果は図1〜5に示した。
VICTOZA(登録商標)は、持続型グルカゴン様ペプチド-1(GLP-1)であり、現在糖尿病治療薬として市販されており、肥満治療効果とHDLコレステロール値の増加効果が知られている。
本発明のまた別の態様は、以下のとおりであってもよい。
〔1〕オキシントモジュリン誘導体を有効成分として含む高脂血症、脂肪肝疾患または動脈硬化症の予防または治療のための組成物。
〔2〕前記オキシントモジュリン誘導体が、配列番号2〜34からなる群から選択されるアミノ酸配列を有する、前記〔1〕に記載の組成物。
〔3〕前記オキシントモジュリン誘導体が、免疫グロブリン断片、抗体、エラスチン、アルブミン、フィブロネクチンからなる群から選択されたものに連結した結合体の形態である、前記〔1〕に記載の組成物。
〔4〕前記結合体は、配列番号2〜34からなる群から選択されるアミノ酸配列を有するオキシントモジュリン誘導体が非ペプチド性重合体を介して免疫グロブリンFc領域に連結されたものである、前記〔3〕に記載の組成物。
〔5〕前記非ペプチド性重合体が、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコールとプロピレングリコールの共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、ポリサッカライド、デキストラン、ポリビニルエチルエーテル、 ポリ乳酸(PLA)、 ポリ乳酸−グリコール酸共重合体(PLGA)、脂質重合体、キチン類、ヒアルロン酸、およびこれらの組み合わせからなる群から選択される、前記〔4〕に記載の組成物。
〔6〕前記非ペプチド性重合体の両末端が、それぞれ免疫グロブリンFc領域およびオキシントモジュリンのアミン基またはチオール基に連結する、前記〔4〕に記載の組成物。
〔7〕前記組成物が、高脂血症、脂肪肝疾患、もしくは動脈硬化症の予防または治療効果を示す薬学的製剤をさらに含む、前記〔1〕に記載の組成物。
〔8〕前記脂肪肝疾患が、非アルコール性脂肪肝疾患、アルコール性脂肪肝疾患、栄養性脂肪肝疾患、飢餓性脂肪肝疾患、肥満性脂肪肝疾患、糖尿病性脂肪肝疾患または脂肪性肝炎である、前記〔1〕に記載の組成物。
〔9〕前記非アルコール性脂肪肝疾患が、高脂血症、糖尿病または肥満から発生する、前記〔8〕に記載の組成物。
〔10〕前記非アルコール性脂肪肝疾患が、単純性脂肪肝、非アルコール性脂肪肝炎、肝繊維症および肝硬変症からなる群から選択される、前記〔8〕に記載の組成物。
〔11〕オキシントモジュリン誘導体を個体に投与するステップを含む、高脂血症、脂肪肝疾患または動脈硬化症を予防または治療する方法。
〔12〕高脂血症、脂肪肝疾患または動脈硬化症の予防または治療のための医薬の製造におけるオキシントモジュリン誘導体の用途。
Claims (28)
- オキシントモジュリン誘導体結合体を有効成分として含む高脂血症、脂肪肝疾患または動脈硬化症の予防または治療のための組成物であって、
オキシントモジュリン誘導体結合体が、
配列番号24、25又は26のいずれかのアミノ酸配列を含むオキシントモジュリン誘導体;
免疫グロブリンFc領域;及び
非ペプチド性重合体
を含み、
非ペプチド性重合体が、オキシントモジュリン誘導体と免疫グロブリンFc領域とを共有結合によって連結した、組成物。 - 前記非ペプチド性重合体が、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコールとプロピレングリコールの共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、ポリサッカライド、ポリビニルエチルエーテル、ポリ乳酸(PLA)、ポリ乳酸−グリコール酸共重合体(PLGA)、脂質重合体、ヒアルロン酸、又は、これらの組み合わせから選択される、請求項1に記載の組成物。
- 前記非ペプチド性重合体が、ポリエチレングリコールである、請求項2に記載の組成物。
- 前記非ペプチド性重合体が、デキストラン、キチン、又は、これらの組み合わせである、請求項2に記載の組成物。
- 前記非ペプチド性重合体の一末端が、免疫グロブリンFc領域のアミン基またはチオール基に連結し、前記非ペプチド性重合体のもう一方の末端が、オキシントモジュリン誘導体のアミン基またはチオール基と連結する、請求項1〜4のいずれか1項に記載の組成物。
- 高脂血症、脂肪肝疾患、もしくは動脈硬化症の予防または治療効果を示す薬学的製剤をさらに含む、請求項1〜5のいずれか1項に記載の組成物。
- 前記脂肪肝疾患が、非アルコール性脂肪肝疾患、アルコール性脂肪肝疾患、栄養性脂肪肝疾患、飢餓性脂肪肝疾患または脂肪性肝炎である、請求項1〜6のいずれか1項に記載の組成物。
- 前記非アルコール性脂肪肝疾患が、高脂血症から発生する、請求項7に記載の組成物。
- 前記非アルコール性脂肪肝疾患が、単純性脂肪肝、非アルコール性脂肪肝炎、肝繊維症または肝硬変症である、請求項7に記載の組成物。
- 高脂血症、脂肪肝疾患または動脈硬化症の予防または治療のための医薬の製造におけるオキシントモジュリン誘導体結合体の使用であって、
オキシントモジュリン誘導体結合体が、
配列番号24、25又は26のいずれかのアミノ酸配列を有するオキシントモジュリン誘導体;
免疫グロブリンFc領域;及び
非ペプチド性重合体
を含み、
非ペプチド性重合体が、オキシントモジュリン誘導体と免疫グロブリンFc領域とを共有結合によって連結した、使用。 - 前記非ペプチド性重合体が、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコールとプロピレングリコールの共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、ポリサッカライド、ポリビニルエチルエーテル、ポリ乳酸(PLA)、 ポリ乳酸−グリコール酸共重合体(PLGA)、脂質重合体、ヒアルロン酸、又は、これらの組み合わせから選択される、請求項10に記載の使用。
- 前記非ペプチド性重合体が、ポリエチレングリコールである、請求項11に記載の使用。
- 前記非ペプチド性重合体が、デキストラン、キチン、又は、これらの組み合わせである、請求項11に記載の使用。
- 前記非ペプチド性重合体の一末端が、免疫グロブリンFc領域のアミン基またはチオール基に連結し、前記非ペプチド性重合体のもう一方の末端が、オキシントモジュリン誘導体のアミン基またはチオール基と連結する、請求項10〜13のいずれか1項に記載の使用。
- 高脂血症、脂肪肝疾患、もしくは動脈硬化症の予防または治療効果を示す薬学的製剤をさらに含む、請求項10〜14のいずれか1項に記載の使用。
- 前記脂肪肝疾患が、非アルコール性脂肪肝疾患、アルコール性脂肪肝疾患、栄養性脂肪肝疾患、飢餓性脂肪肝疾患または脂肪性肝炎である、請求項10〜15のいずれか1項に記載の使用。
- 前記非アルコール性脂肪肝疾患が、高脂血症から発生する、請求項16に記載の使用。
- 前記非アルコール性脂肪肝疾患が、単純性脂肪肝、非アルコール性脂肪肝炎、肝繊維症または肝硬変症である、請求項16に記載の使用。
- 中性脂肪濃度を減少するため、血中総コレステロール濃度を減少するため、血中LDLコレステロール濃度を減少するため、血中HDLコレステロール濃度を増加するため、または、血中HDL/LDLコレステロールの割合を増加するための組成物であって、
活性成分としてオキシントモジュリン誘導体結合体を含み、
オキシントモジュリン誘導体結合体が、
配列番号24、25又は26のいずれかのアミノ酸配列を含むオキシントモジュリン誘導体;
免疫グロブリンFc領域;及び
非ペプチド性重合体
を含み、
非ペプチド性重合体が、オキシントモジュリン誘導体と免疫グロブリンFc領域とを共有結合によって連結した、組成物。 - 前記非ペプチド性重合体が、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコールとプロピレングリコールの共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、ポリサッカライド、ポリビニルエチルエーテル、ポリ乳酸(PLA)、 ポリ乳酸−グリコール酸共重合体(PLGA)、脂質重合体、ヒアルロン酸、又は、これらの組み合わせから選択される、請求項19に記載の組成物。
- 前記非ペプチド性重合体が、ポリエチレングリコールである、請求項20に記載の組成物。
- 前記非ペプチド性重合体が、デキストラン、キチン、又は、これらの組み合わせである、請求項20に記載の組成物。
- 前記非ペプチド性重合体の一末端が、免疫グロブリンFc領域のアミン基またはチオール基に連結し、前記非ペプチド性重合体の別の末端が、オキシントモジュリン誘導体のアミン基またはチオール基と連結する、請求項19〜22のいずれか1項に記載の組成物。
- 中性脂肪濃度を減少するため、血中総コレステロール濃度を減少するため、血中LDLコレステロール濃度を減少するため、血中HDLコレステロール濃度を増加するため、または、血中HDL/LDLコレステロールの割合を増加するための医薬の製造におけるオキシントモジュリン誘導体結合体を含む組成物の使用であって、
